밤나무 잎차(茶)의 항알레르기 효과 (Anti-allergic Effects of Castanea crenata Leaf Tea)

  • 밤나무 잎을 이용하여 차(茶)로 제조한 다음, 제조된 차에서 항알레르기 효과를 실험하였다. 밤나무 잎 차는 대표적인 차의 형태인 불발효차와 반발효차의 제조공정을 이용하여 증제차와 유념차로 제조하였으며, 항알레르기 효과는 제조된 증제차와 유념차의 각각의 물추출물을 RBL-2H3 세포로부터 hexosaminidase의 방출 억제 정도를 측정하는 enzyme assay를 이용하여 실시한 결과, 물추출물의 $300\;{\mu}/mL$의 수준에서 증제차는 50.4%의 저해효과를 나타냈으며, 유념차는 35.4%의 저해효과를 나타내어 항알레르기 효과가 있음을 확인하였다.

Compound 48/80로 유발된 Allergy에 대한 생혈윤부음(生血潤膚飮)의 효과 (Inhibitory Effect of Saingheylyunbooem on Compound 48/80 Stimulated Allergic Reaction)

  • The present study was conducted to investigate the anti-allergic activity of Saingheylyunbooem(SHU)). We investigated the anti-allergic effects of SHU in RBL-2H3 basophilic leukemia cells by compound 48/80, a mast cell degranulator in mice. SHU inhibited ${\beta}$-hexosaminidase and histamine release from compound 48/80 stimulated RBL-2H3 cells. The in vitro anti-inflammatory activities of SHU in LPS-stimulated RAW 264.7 cells were investigated. SHU inhibited NO production effectively dowregulated the expression of iNOS mRNA and iNOS protein expression in LPS-stimulated RAW 264.7 cells. These result provide evidences that SHU may be beneficial in the treatment of allergic inflammtory disease.

  • RBL-2H3 cell에서 HRE에 의하여 histamine이 분비되는 과정에서 ROS가 생성되는지 실험해 본 결과, ROS가 HRF를 처리한지 5분대에 최대치를 보이며 생성되었다가 소멸되는 것을 관찰할 수 있었다. 따라서 HRF가 세포내 second messenger로써의 ROS를 생성하였다고 확인할 수 있었다. 또한 ROS는 단백질 정제 과정에서의 endotoxin오염에 의해 영향을 받지만,본 실험에서 규명한 HRF에 의한 ROS 생성은 endotoxin에 의한 것이 아닌, 순수하게 HRF에 의한 signaling의 결과라는 것도 확인할 수 있었다.

호염구세포주와 복강 비만세포에서 유색미 겨 추출물의 알레르기 염증 억제활성 (Inhibitory Activity of Pigmented Rice Bran Extract to the Allergic Inflammation in Basophilic Cell Line and Peritoneal Mast Cells)

  • 유색미 겨 추출물이 염증반응에 미치는 효과를 호중구 세포주 및 비만세포가 분비하는 염증매개물질인 histamine과 ${\beta}-hexosaminidase$의 분비 및 염증 cytokine 생성에 대한 억제활성을 측정하여 평가하였다. 호중구 세포주인 RBL-2H3 세포와 rat에서 분리 추출한 복강 비만세포(RPMC, peritoneal mast cell)를 이용하여 histamine과 ${\beta}-hexosaminidase$ 분비에 대한 억제활성을 조사한 결과, 일반미 겨 추출물은 RBL-2H3 세포에서 염증매개물질들의 분비량을 증가시킨 반면, 유색미 겨 추출물은 이들의 분비를 억제하였다(histamine 10.19% 억제, ${\beta}-hexosaminidase$ 110.03% 억제). RPMC에서는 유색미가 histamine과 ${\beta}-hexosaminidase$의 분비를 일반미보다 각각 8배와 3배나 높게 억제하는 것으로 나타났다. 염증관련 cytokine으로서 $IL-1{\beta},\;TNF-{\alpha}$ 및 IL-6의 발현을 RT-PCR로 측정한 결과, 유색미가 일반미 보다 $TNF-{\alpha},\;IL-1{\beta}$ 및 IL-6의 발현을 억제하는 효과가 우수하였다. ELISA를 이용하여 cytokine의 분비량을 측정한 실험에서도 유색미가 일반미보다 $TNF-{\alpha},\;IL-1{\beta}$ 및 IL-6의 분비를 효과적으로 억제하였다. 이상의 결과는 유색미가 염증반응의 원인 물질인 histamine과 ${\beta}-hexosaminidase$의 분비에 대한 억제효과 뿐 아니라, 염증 cytokine의 발현을 저해하는 효과도 일반미보다 우수하다는 사실을 보여주었다.

  • The rat mast cell line RBL-2H3 contains both phospholipase D (PLD)1 and PLD2. Previous studies with this cell line indicated that expressed PLD1 and PLD2 are both strongly activated by stimulants of secretion. We now show by use of PLDs tagged with enhanced green fluorescent protein that PLD1, which is largely associated with secretory granules, redistributes to the plasma membrane in stimulated cells by processes reminiscent of exocytosis and fusion of granules with the plasma membrane. 자주쓴풀 메탄올추출물의 IgE-매개 알레르기 반응 억제 및 기전 (Swertia pseudochinensis Methanol Extract Inhibits IgE-mediated Allergic Response In vitro and In vivo)

  • Mast cells play a key role in IgE-mediated allergic response. We investigated whether Swertia pseudochinensis Hara extract (SPE) inhibits IgE-mediated allergic response in mast cells and an allergic animal model. Additionally, we explored SPE's mechanism of action in mast cells. Our results showed that SPE inhibited both antigen-stimulated degranulation and the production of TNF-α and IL-4 in bone marrow-derived mast cells (BMMCs) and rat basophilic leukemia (RBL)-2H3 cells. SPE also suppressed allergic response in IgE-mediated passive cutaneous anaphylaxis (PCA) in mice. As for the mechanism of action of SPE in mast cells, it inhibited the activation of Syk kinase, a critical signaling protein in the FcεRI-mediated signaling pathway, and also the activation of LAT, a downstream adaptor protein of Syk. We further observed the reduced activation of mitogen-activated protein (MAP) kinases (P38, ERK1/2, and JNK) and Akt in mast cells. Our results described for the first time that SPE has an anti-allergic effect by suppressing mast cells through the inhibition of Syk kinase. Therefore, SPE may be useful for the treatment of type I allergic diseases.

량혈제습탕(凉血除濕湯)의 비만세포 활성화 억제작용 (Anti-inflammatroy Activity of YHJST in Activated Mast Cells)

  • Activated mast cells play pivotal roles in allergic and non-allergic inflammatory responses through the release of inflammatory mediators such as histamin, cysteinyl leukotriens, pro-inflammatory cytokines as well as chemokine. We tested whether YHJST, which is clinically prescribed for the treatment of various inflammatory disease including allergic disease, modulate inflammatory reactions in activated mast cells. YHJST decreased the release of histamine and b-hexosamidase in pholbol-12-myristate 13-acetate and/or calcium ionophore A23187 stimulated HMC-1 and RBL-2H3 cells, respectively. Further, the gene expressions of tumor necrosis factor-a, IL-6 and IL-8 were significantly reduced by YHJST. YHJST suppressed powerful induction of NF-kB promoter-mediated luciferase activity. Taken together, these data suggested that YHJST showed it's anti-inflammatory effects through the down-regulation of mast cell activation.

Anti-allergic Effect of Seungmagalgeun-tang through Suppression of NF-${\kappa}B$ and p38 Mitogen-Activated Protein Kinase Activation in the RBL-2H3 Cells

  • In previous report, Seungmagalgeun-tang (SGT) could exert its anti-inflammatory actions in the BV-2 microglial cells. However, study on the anti-inflammatory effect of SGT in mast cells has not been identified. Therefore, we examined on the anti-inflammatory effect of SGT on the phorbol 12-myristate 13-acetate (PMA) and calcium ionophore A23187-induced rat basophilic leukemia (RBL-2H3) cells. SGT inhibited the release of ${\beta}$-hexosaminidase and secretion and expression of pro-inflammatory cytokines such as tumor necrosis factor (TNF)-${\alpha}$ and interleukin (IL)-4 on RBL-2H3 cells, without affecting cell viability. The protein expression level of nuclear factor (NF)-${\kappa}B$ (p65) was decreased in the nucleus by SGT. In addition, SGT suppressed the degradation of inhibitory protein $I{\kappa}B-{\alpha}$ protein, the activation of p38 mitogen-activated protein kinase (MAPK), and the expressions of cyclooxygenase (COX)-2 mRNA and protein level in RBL-2H3 cells. These results suggest that SGT could be involved anti-allergic effect by control of NF-${\kappa}B$ (p65) translocation into the nucleus through inhibition of $I{\kappa}B-{\alpha}$ degradation and suppression of COX-2 expression.

Processed Xanthii fructus increases cell viability of mast cell line, RBL-2H3

  • The effect of aqueous extract of processed Xanthii fructus (PXF) on cell viability and histamine release from mast cell has been investigated. PXF showed higher cell viability than unprocessed Xanthii fructus (XF) at the concentrations of 5 and 10 mg/ml. Aqueous extract of PXF and unprocessed XF inhibited compound 48/80-induced systemic anaphylaxis in mouse. Both PXF and unprocessed XF dose-dependently inhibited histamine release from rat peritoneal mast cells by compound 48/80 to the similar extent at 0.01, 0.1 and 1 mg/ml. Our studies provide evidence that processing of XF may be beneficial to reduce cytotoxicity in high concentration (at 5 and 10 mg/ml) but does not affect on anti-allergic activity.

Na,K-ATPase와 IgE-Dependent Histamine Releasing Factor의 결합에 영향을 미치는 Protein Kinase C Isotype에 관한 연구 (PKC Isotype that Affects the Interaction of HRF with Na, K-ATPase)

  • 본 실험에서는 MRF가 다량 존재하는 RBL-2H3 세포주에 다양한 PKC isotype별 억제제를 처리하여 in vitro상에서 Na, K-ATPase $\alpha$1L3를 이용한 pull-down assay와 RBL-2H3 세포를 이용한 membrane fractionation을 실시하였다. 그 결과 HRF는 in vitro에서 $\alpha$1L3와 결합한다는 사실을 재확인 할 수 있었고 실제 세포주 내에서 Na,K-ATPase와 결합한다는 것을 알 수 있었다. 사용한 약물로부터 PKC $\alpha,\;\beta,\;\delta$뿐 아니라 protein phosphatase 2B(PP2B)도 HRF와 $\alpha$1L3의 결합에 관여한다는 사실을 알 수 있었다. 또한 이들 PKC, PP2B에 의해 인산화된 HRF 분자는 cytosolic fraction으로 이행할 수 있으며 이러한 결과는 탈인산화된 HRF가 Na,K-ATPase와 결합하여 Na, K-ATPase의 기능을 조절한다고 추정할 수 있다. 그러나 약물자체가 histamine 분비에 영향을 미칠 수 있으며 cytosolic HRF보다 exocytosis된 HRF가 histamine를 더 분비하도록 할 수 있으므로, 약물을 전처리한 세포에 외부에서 HRF를 첨가하여 histamine이 유리되는 정도가 어떻게 변화하는지를 HRF를 가하지 않은 결과와 비교해야 할 것이다.
