Cloning and Sequence Analysis of Two Catechol-degrading Gene Clusters from a Phenol-utilizing Bacterium Pseudomonas putida SM25

  • A 6.1 kb Sph I fragment from the genomic DNA of Pseudomonas putida SM 25 was cloned into the veetor pUC19. The open reading frame of catB was found to consist of 1,122 nucleotides. The sequence alignment of the catB gene products from different kinds of bacteria revealed an overall identity ranging from 40 to 98%. The catC gene contained an open reading frame of 96 codons, from which a protein with a molecular mass of about 10.6 kDa was predicted. The amino acids in the proposed activesite region of CatC were found to be almost conserved, including the charged residues. Since the catBC genes in P. putida SM25 were tightly linked, the could be regulated under coordinate transcription, and transcribed from a single promoter located upstream of the catB gene, as in P. putida RBI.

  • Ginseng root has been considered to prevent neuronal degeneration associated with brain ischemia, but experimental proof in support of this speculation is limited. Moreover, few studies have compared the neuroprotective actions of ginseng ingredients with those of peptide growth factors and cytokines isf vivo. Using a gerbil forebrain ischemia model, we demonstrated that the oral administration of red ginseng powder before an ischemic insult prevents delayed neuronal death in the hippocampal CAI field and that a neuroprotective molecule within red ginseng powder is ginsenoside Rbl. The neurotrophic effect of ginsenoside Rbl, when examined in the gerbil ischemia model and in neuronal cultures was as potent as or more potent than the effects of epidermal growth factor, ciliary neurotrophic factor, erythropoietin, prosaposin, interleukin-6 and interleukin-3. Besides the protection of hippocampal CAI neurons against brain ischemia/repercussion injuries, ginsenoside Rbl was shown to prevent place navigation disability, cortical infarction and secondary thalamic degeneration in stroke-prone spontaneous hypertensive rats with permanent occlusion of the unilateral middle cerebral artery distal to the striate branches. These findings may validate the empirical use of ginseng root for the treatment of cerebrovascular diseases

Fraud Scenario Prevalent in the Banking Sector: Experience of a Developing Country

  • Banks are the engines that drive the operations in financial sector, money markets and growth of economy. With growing banking industry in India, frauds in Banks are increasing and fraudsters are becoming more sophisticated and ingenious. Shockingly, banking industry in India dubs rising fraud as "an inevitable cost of doing business." As part of study, a questionnaire-based survey was conducted in 2012-13 among 345 Bank employees "to know their perception towards bank frauds and evaluate factors that influence the degree of their compliance level." The study reveals, "there are poor employment practices and lack of effective employee training; usually over-burdened staff, weak internal control systems, and low compliance levels on the part of Bank Managers, Offices and Clerks. Although banks cannot be 100% secure against unknown threats, a certain level of preparedness can go a long way in countering fraud risk. Internal audit professionals should play an integral role in organization's fraud-fighting efforts. Some other promising steps are: educate customers about fraud prevention, make application of laws more stringent, leverage the power of data analysis technologies, follow fraud mitigation best practices, and employ multipoint scrutiny.

  • 인진호탕(茵蔯蒿湯)은 한의학에서 여러 가지 질환에 사용되어 왔다. 특히 간경의 습열(濕熱)로 인한 질환에 습열을 제거함으로써 증(證)의 완화에 많은 응용이 되어 왔다. 본 연구는 습열(濕熱)로 인한 질환중의 하나인 아토피 피부염에 그동안 응용이 적은 인진호탕(茵蔯蒿湯)을 실험적으로 응용함으로써 임상적인 가치를 가질 수 있는 지에 대한 기초연구를 진행하였다. 본 실험에 사용된 세포주는 rat leukemia (RBL-2H3) cells로 인진호탕(茵蔯蒿湯)은 알레르기와 관련된 사이토카인인 tumor necrosis factor (TNF)-$\alpha$, interleukin (IL)-4를 용량의존적으로 억제하였지만 세포독성은 일으키지 않았다. 그리고 동물실험에서 인진호탕(茵蔯蒿湯)은 PCA반응에서 충분한 억제효과를 나타내었다. 또한 Compound 48/80으로 유도된 anaphylaxis shock도 용량 의존적으로 억제함이 밝혀졌다.

Consequence Analysis for Accidental Releases of Toxic Gases through Risk Based Inspection using API-581 (API-581에 의한 위험기반검사에서 독성가스의 누출사고 결과분석)

  • Accidental releases of toxic gases heavily affect to the risk of process facilities. In this research, consequence analysis for accidental releases of ammonia and chlorine gases was studied using the risk based inspection, based on API-581 BRD. It was found out that consequence areas (toxic areas) decrease as temperature increases and as the pipe diameter and pressure decrease. For the same release condition, the toxic area by the release of chlorine gas was larger than that by the release of ammonia gas.

  • The consequence analysis (CA) is widely using in the petrochemical plant through adoption of the process safety management (PSM) system, but it has not practical problem that the CA was not reflected effects for employee count, business interruption loss, utility usage, and etc.. In this study, to establish the practical emergency response plan and to achieve risk based management, the consequence based on the damage area and the financial loss were estimated and compared through application on facilities in the petrochemical plant. If the damage area is used, the consequence category must be determined by safety area considering simultaneously damage area, fatality area and toxic area. Also, the consequences based on the financial loss is more practical method for the case of considering process properties and circumstances.

Long term trends in the Korean professional baseball (한국프로야구 기록들의 장기추세)

  • This paper offers some long term perspective on what has been happening to some baseball statistics for Korean professional baseball. The data used are league summaries by year over the period 1982-2013. For the baseball statistics, statistically significant positive correlations (p < 0.01) were found for doubles (2B), runs batted in (RBI), bases on balls (BB), strike outs (SO), grounded into double play (GIDP), hit by pitch (HBP), on base percentage (OBP), OPS, earned run average (ERA), wild pitches (WP) and walks plus hits divided by innings pitched (WHIP) increased with year. There was a statistically significant decreasing trend in the correlations for triples (3B), caught stealing (CS), errors (E), completed games (CG), shutouts (SHO) and balks (BK) with year (trend p < 0.01). The ARIMA model of Box-Jenkins is applied to find a model to forecast future baseball measures. Univariate time series results suggest that simple lag-1 models fit some baseball measures quite well. In conclusion, the single most important change in Korean professional baseball is the overall incidence of completed games (CG) downward. Also the decrease of strike outs (SO) is very remarkable.

  • Major components of refinery plants are exposed to various damage mechanisms depending on the operation condition, material selection and kinds of internal fluid. Inspection techniques and damage monitoring methods should be selected considering the damage mechanisms of the components. Hence, it is quite necessary to have an information system with a standardized database on the various damage mechanisms. In this study a damage information system with contents on the damage mechanisms included in API 571 code was developed. Concept of the screening table employed in USA and Japan was also adopted to identify the probable damage mechanisms from the information on operating temperature, internal fluid, metal used for the component and stress condition. This system can be used before the risk based inspection planning to identify key damage mechanisms involved.

  • In the present paper forward projection is proposed as a new approach to determine the preform shape in rib-web type forging. In the forward projection technique an optimal billet is determined by applying some mathematical relationship between geometrical trials in the initial billet shape and the final products. In forward projection a volume difference between the desired product shape and the final computed shape obtained by the rigid-plastic finite element method is used as a measure of incomplete filling of working material in the die. At first linear inter-/extrapolation is employed to find a proper trial shape for the initial billet and the method is successfully applied to some cases of different aspect ratios of the initial billet. However, when the initial guesses are not sufficiently near the optimal value linear inter-/extrapolation does not render complete die filling. For more general application, a fuzzy system is used in the forward projection technique in order to determine the initial billet shape for rib-web type forging. It has been thus shown that the fuzzy system is more reliable for the preform design in the rib-web type forging process.

Risk Ranking Analysis for the City-Gas Pipelines in the Underground Laying Facilities (지하매설물 중 도시가스 지하배관에 대한 위험성 서열화 분석)

  • In this article, we are to suggest the hazard-assessing method for the underground pipelines, and find out the pipeline-maintenance schemes of high efficiency in cost. Three kinds of methods are applied in order to refer to the approaching methods of listing the hazards for the underground pipelines: the first is RBI(Risk Based Inspection), which firstly assess the effect of the neighboring population, the dimension, thickness of pipe, and working time. It enables us to estimate quantitatively the risk exposure. The second is the scoring system which is based on the environmental factors of the buried pipelines. Last we quantify the frequency of the releases using the present THOMAS' theory. In this work, as a result of assessing the hazard of it using SPC scheme, the hazard score related to how the gas pipelines erodes indicate the numbers from 30 to 70, which means that the assessing criteria define well the relative hazards of actual pipelines. Therefore. even if one pipeline region is relatively low score, it can have the high frequency of leakage due to its longer length. The acceptable limit of the release frequency of pipeline shows 2.50E-2 to 1.00E-l/yr, from which we must take the appropriate actions to have the consequence to be less than the acceptable region. The prediction of total frequency using regression analysis shows the limit operating time of pipeline is the range of 11 to 13 years, which is well consistent with that of the actual pipeline. Concludingly, the hazard-listing scheme suggested in this research will be very effectively applied to maintaining the underground pipelines.
