Genetically modified (GM) rice Agb0101, which expresses the insecticidal toxin modified cry1Ac (mcry1Ac1) gene, was developed by the Rural Development Administration in Korea. To monitor the probable release of Agb0101 in the future, it is necessary to develop a reliable detection method. Here, we developed the PCR detection method for monitoring and tracing of GM rice. The primer pair (RBEgh-1/-2) from a starch branching enzyme (RBE4) gene was designed as an endogenous reference, giving rise to an expected PCR amplicon of 101 bp. For the qualitative PCR detection, construct- and event-specific primers were designed on the basis of integration sequence of T-DNA. Event-specific PCRs amplified specifically 5'- or 3'-junction region spanning the native genome DNA and the integrated gene construct, while none of amplified product was shown on crops, rice varieties, and other insect-resistant transgenic rice lines. The event-specific real-time PCR method was performed using TaqMan probe and plasmid pRBECrR containing both rice endogenous gene RBE4 sequence and 5'-junction sequence as the reference molecule. The absolute limit of quantification (LOQ) of real-time PCR was established with around 10 copies for one plasmid molecule pRBECrR. Thereafter, the different amounts of transgenic rice (1, 3, 5, and 10%, respectively) were quantified by using the established real-time PCR method, with a range below 19.55% of the accuracy expressed as bias, 0.06-0.40 of standard deviation (SD) and 3.80-7.01% of relative standard deviations (RSD), respectively. These results indicate that the qualitative and quantitative PCR methods could be used effectively to detect the event Agb0101 in monitoring and traceability.
This research aims to establish the finite-element modeling techniques for computational modal analyses of liquid propellants and flange joints of launch-vehicle structures. MSC.NASTRAN is used for the present computational modal analyses of the liquid-propellant tank and the small-scaled first-stage model. By means of the correlation between the measured and computed natural frequencies, the finite modeling techniques for liquid propellants and flange joints of launch-vehicle structures are established appropriately. This modal analysis using the virtual-mass method predicts well the bell mode of the liquid-propellant tank containing liquid. In addition, the present computation using RBE2 elements for modeling of flange joints predicts the first and second bending-mode frequencies within a relative error of 10%, which is better than the measured frequencies obtained from the modal test, for the small-scaled first-stage model containing liquid.
Portions of chloroplast genes(psbD and rbcL) were amplified from Pinus thunbergii(Japanese black pine : black pine) and Pinus densiflora(Japanese red pine : red pine) by PCR and digested by a restriction enzyme, HaeIII, respectively. Two species specific cpDNA markers were identified. With the observed cpDNA markers, paternal inheritance of cpDNA in pine hybrids was verified in an artificial hybrid family between black pine(Chollanam 37) and red pine(Chungchongbuk 3). On the basis of paternal inheritance of chloroplast genome in a hybrid, 2 portions of cpDNA amplified from 115 individuals of Pinus densiflora for. erecta were screened to detect any traces of black pine specific cpDNA markers in P. densiflora for. erecta which has been postulated as an introgressive hybrid between red pine and black pine(Hyun el al., 1967). All the analyzed individuals of Pinus densiflora for. erects revealed the identical profiles of HaeIII digested psbD and rbcL genes to red pine. This result suggests that there is no introduced chloroplast genome of black pine in Pinus densiflora for. erecta and that there is no concrete evidence of treating P. densiflora for, erecta as an introgressive hybrid between red pine(♀) and black pine(♂).
Proton therapy is known for its superior treatment method due to Bragg peak. To enhance the therapeutic effects of protons, research has been conducted on distributing gold nanoparticles within tumors to increase the absorbed dose. While previous studies focused on handling gold nanoparticles at micrometer and nonometer scale, this study proposes a method to computationally estimate the effect of gold nanoparticles at the millimeter scale. The Geant4 toolkit was applied to computational modeling. Assuming a uniform distribution of water, similar to the human body, and gold nanoparticles, the concentration of gold nanoparticles was adjusted using density ratios. When the density ratio was 5%, the gain in absorbed energy due to gold nanoparticles was nearly twice that of the pure water phantom at the Bragg peak. As the density ratio increased, the gain in absorbed energy linearly increased. When gold nanoparticles were distributed in only one voxel at the Bragg peak, the energy of the protons affected only the neighboring voxels. However, in cases where gold nanoparticles were distributed over a wide area, the volume showing 95% of the maximum absorbed energy (9.46 keV) for the pure water phantom (9.95 keV) exhibited an improvement in absorbed energy over a region 16 times larger, and this region increased as the density ratio increased. Further research is needed to quantify the relationship between the density ratio of gold nanoparticles and the relative biological effect (RBE) in the millimeter scale.

In this paper, we proposed the novel method for the recognition of English calling cards by using the projection method and the enhanced RBF (Radial Basis Function) network. The recognition of calling cards consists of the extraction phase of character areas and the recognition phase of extracted characters. In the extraction phase, first of all, noises are removed from the images of calling cards, and the feature areas including character strings are separated from the calling card images by using the horizontal smearing method and the 8-directional contour tracking method. And using the image projection method, the feature areas are split into the areas of individual characters. We also proposed the enhanced RBF network that organizes the middle layer effectively by using the enhanced ART1 neural network adjusting the vigilance threshold dynamically according to the homogeneity between patterns. In the recognition phase, the proposed neural network is applied to recognize individual characters. Our experiment result showed that the proposed recognition algorithm has higher success rate of recognition and faster learning time than the existing neural network based recognition.
We obtained Surviving Fraction (SF) after irradiation carbon beam to compare the applicability of the Linear-Quadratic model, Incomplete Repair model, Marchese model. Mathematica software(ver 9.0) used to calcurate parameters and compared result. LQ model could not explain the entire response of fractionated carbon beam irradiation. It becomes necessary to construct models that extend the LQ model of conventional radiotherapy for the carbon beam therapy. By combining both Potentially Lethal Damage Repair (PLDR) and Sublethal Damage Repair (SLDR) a new LQ model can develop that aptly modeled the cellular response to fractionated irradiation.
Dose response curves using absorbed dose to the biological effect are usually available in case of conventional X beam. However, absorbed dose is not consider in treatment planning for carbon beam such as heavy ions. Because the biological effects also depend on other quantities such as the local variation, which is often characterized by the linear energy transfer (LET). So LQ model cannot explain the entire response of fractionated carbon beam irradiation. The variation in LET with penetration depth leads to substantial differences in biological effect of carbon beam. And it is therefore essential in treatment planning to calculate not only the absorbed dose but also the LET to estimate the biological outcome of the radiation of interest. LET variation plays an important role in the fractionated irradiations. It is suggested that consideration of LET is necessary in biophysical model.
To evaluate if the apoptotic fragment assay could be used to estimate the dose prediction after radiation exposure, we examined apoptotic mouse crypt cells per 1,000 cells after whole body $^{60}Co$$\gamma$-rays and 50MeV ($p{\rightarrow}Be^+$) cyclotron fast neutron irradiation in the range of 0.25 to 1 Gy, respectively. The incidence of apoptotic cell death rose steeply at very low doses up to 1 Gy, and radiation at all doses tigger rapid changes in crypt cells in stem cell region. These data suggest that apoptosis may play an important role in homeostasis of damaged radiosensitive target organ by removing damaged cells. The curve of dose-effect relationship for the data of apoptotic fragments was obtained by the linear-quadratic model $y=0.18+(9.728{\pm}0.887)D+(-4.727{\pm}1.033)D^2$ ($r^2=0.984$) after $\gamma$-rays irradiation, while $y=0.18+(5.125{\pm}0.601)D+(-2.652{\pm}0.7000)D^2$ ($r^2=0.970$) after neutrons in mice. The dose-response curves were linear-quadratic, and a significant dose-response relationship was found between the frequency of apoptotic cell and dose. These data show a trend towards increase of the numbers of apoptotic crypt cells with increasing dose. Both the time course and the radiation dose-response curve for high and low linear energy transfer (LET) radiation modalities were similar. The relative biological effectiveness (RBE) value for crypt cells was 2.072. In addition, there were significant peaks on apoptosis induction at 4 and 6h after irradiation, and the morpholoigcal findings of the irradiated groups were typical apoptotic fragments in crypt cells that were hardly observed in the control group. Thus, apoptosis in crypt cells could be a useful in vivo model for studying radio-protective drug sensitivity or screening test, microdosimetric indicator and radiation-induced target organ injury. Since the apoptotic fragment assay is simple, rapid and reproducible in the range of 0.25 to 1 Gy, it will also be a good tool for evaluating the dose response of radiation-induced organ damage in vivo and provide a potentially valuable biodosimetry for the early dose prediction after accidental exposure.
This study was carried out to obtain basic information on breeding using PCR-aided RFLP technology which can identify the variation inter- and intra-species of ginseng in the level of DNA. It was intended to investigate banding pattern on psbA and rbeL genes of chloroplast DNA in ginseng after treating with restriction enzymes. To isolate psbA and rbcL genes of chloroplast, both psbA-N, psbA-C primer and rbcL-N, PX-1 primer were used. As a result, 1,008 bp band of psbA gene and 1,336 bp band of rbcL gene were appeared, which was optimal and expected molecular weight. In addition, primers to isolate atpB, rpoB, trnL, and trnF genes were used, resulting in the expected 1366, 900, 1500 and 1008 bp bands. Genes of psbA and rbcL isolated by PCR were cut by restriction enzymes, Sau3A, TaqI, AluI, HaeIII, and RFLP pattern was investigated. KG line and other species of ginseng were cut by TaqI treatment, and bands were located in 800 bp. The treatment treated by AluI also showed the same 800 bp band in KG line and other species. In HaeIII treatment, 500 bp of faint bands were shown in case of KG line, whereas any bands were not observed in other species. All chloroplast genes formed bands by PCR amplification. However, it was not evident to distinguish intra-or inter-species of ginseng after restriction enzyme treatment. Therefore, more restriction enzyme treatment or sequence comparison method should be considered for further experiment.
The purpose of this study is to investigate whether gold nanoparticles had radiosensitization when combined with gamma and x ray beam. Cytotoxicity was mearsured with comparing survival fraction after incubated time 6,12,18 and 24 hours. Clonogenic assay was employed to assess survival fraction of cells with and without gold nanoparticles treatment following gamma ray irradiation. The most of gold nanoparticles were distributed in the cytoplasm. And the toxicity of gold nanoparticles used this study were found to be non-cytotoxic. And we also observed enhancement by about 40% in RBE value for gamma ray irradiation of cells treated with gold nanoparticles. Dose reduction of about half for gamma ray irradiation is demonstrated for gold nanoparticles treated cells as compared to untreated cells. In cells with exposed to gamma ray, DNA damage was increased when compared to only radiation exposed cells. The study revealed a significant reduction in radiation dose for killing the cells with internalized gold nanoparticles as compared to the cells without gold nanoparticles. The gold nanoparticles treatment resulted in enhancement of radiation effect as evident from increase in relative biological effectiveness values for photon irradiated cells.
