Genetic Relationship of Genus Rosa Germplasm and Genetic Diversity of Rosa rugosa in Korea (장미속 유전자원의 유전적 관계와 국내 해당화의 유전적 다양성)

  • Fifty-nine accessions of 23 species in genus Rosa were collected, and 15 accessions of Rosa rugosa were collected throughout 10 regions of Korea. Their genetic relationship was investigated by using morphological analysis and RAPD marker. The morphological analysis was measured for 7 quantitative and quantified for 4 qualitative traits. RAPD analysis obtained a total of 959 polymorphic bands by using twenty primers. Morphological analysis classified most according to the rose section system except for several species. The cluster analysis of genus Rosa based on RAPD data could identify the subgenus $Platyrhodon$ and $Eurosa$. The subgenus $Eurosa$ separated five sections; $Gallicanae$, $Cinnamomeae$, $Pimpinellifoliae$, $Synstylae$ and $Caninae$. Correlation analysis between morphological and RAPD analysis showed low significance ($r$ = 0.35). The accessions of R. rugosa belonged to the section $Cinnamomeae$ clustered into three groups at genetic distance ranging from 0.28 on the base of RAPD analysis. In conclusion, the genetic relationship of the genus Rosa was consistent to the previously reported rose section system, and domestic collections of $R.$ $rugosa$ were separated from 3 groups on the base of RAPD marker.

A taxonomic study of Abeliophyllum Nakai (O1eaceae) based on RAPD analysis (RAPD분석에 의한 미선나무속의 분류학적 연구)

  • Abeliophyllum distichum Nakai (O1eaceae) is a monotype of Korea and is distinguished from related genus Forsythia Vahl by the morphological characters such as fruit shape, flower color and etc. Even though several intraspecific taxa were reported according to the color of flowers and shape of fruits, there have been many controversals on the taxonomic indentity and status of rank on those taxa. In the present study, we performed the RAPD analysis to delimit the infraspecific taxa of Abeliophyllum distichum and to investigate the genetic polymorphism and relationships among 12 populations. 212 scorable RAPD markers with 70 common markers were found from the PCR reactions with 16 random oligoprimers and were analyzed by Nei's genetic distance. From 0.108 to 0.321 of genetic variations were showed among the taxa. Some regional groups instead of same taxa were clustered from the phonogram of UPGMA analysis. Also, we could not find distinct lineage among intraspecific taxa. The result from RAPD analysis supported that the infraspecific taxa of Abeliophyllum distichum might be the individual variations and treated as the same taxa. RAPD analysis was very useful to confirm the high gene pool with diverse genetic polymorphism among Abeliophyllum distichum populations.

A Phylogenetic Relationships of Araliaceae Based on PCR-RAPD and ITS Sequences (PCR-RAPD와 ITS 서열 분석에 의한 두릅나무과 (Araliaceae) 의 유연관계 분석)

  • Phylogenetic relationships among species in Araliaceae were analyzed using PCR-RAPD and sequence of ITS region of nuclear ribosomal DNA based on samples collected in Korea. RAPD analysis showed various polymorphic bands which were able to differentiate species and genus, and specific bands showing variations among individuals within species. Cluster analysis using gel images revealed high molecular variability within species of Aralia eleta. No significant variation was found among cultivated species of Panax ginseng, but they showed high genetic differences with wild type of the species. In ITS analysis, specific sequences for each genus and species were observed and these were allowed to differentiate species and genus. Phylogenetic analysis using ITS sequences showed that Acanthopanax and Kalopanax had a close relationship, and Aralia and Panax are monophyletic, but genus Hedera is different species from other species in family Araliaceae in this study. The results showing close relationship between genera Aralia and Panax were also observed in RAPD analysis. Contrary to the results of RAPD analysis of Panax ginseng, sequence analysis of ITS showed no significant difference between wild mountain ginseng and cultivated species of P. ginseng. Also, both RAPD and ITS analysis of P. ginseng showed no significant genetic variability among cultivation sites. Results indicate that P. ginseng cultivating in Korea is monophyletic. The molecular analysis used in this study agreed on classification using morphological feature. These results suggest that molecular techniques used in this study could be useful for phylogenetic analysis of Araliaceae.

A Systematic Relationship of the Korean Symplocaceae Based on RAPD analysis (RAPD에 의한 한국산 노린재나무과 식물의 유연관계 분석)

  • In order to estimate the genetic relationships among four taxa of Korean Symplocaceae and their regional populations, RAPDs analysis was performed. The length of the amplified DNA fragments ranged from 150 to 1,900 bp. Ninety-two RAPD bands were scored for 11 primers and the genetic distance was calculated with by Nei-Li's genetic dissimilarity. Deciduous and evergreen groups were clearly separated in the UPGMA analysis. Symplocos coreana was clustered as a distinct group, and S. sawafutagi and S. tanakana were also clustered at specific level, respectively. The RAPD data was useful to cognize the genetic variation and to discuss the relationships among Korean Symplocaceae

A Parametric Study of Random Amplified Polymorphic DNA (RAPD) Analysis: A Lactobacillus Model (유산균 Lactobacillus 종간의 분류를 위한 RAPD 분석법의 매개변수에 관한 연구)

  • A study was carried out to understand some parameters affecting on RAPD analysis with Lactobacillus species. From the results, we found that appearance of specific DNA bands were very influenced by the concentration of $MgCl_2$ but it was overcome by applying enough amount of Taq DNA polymerase. Other parameters such as concentrations of template DNA, random primers and Taq DNA polymerase have enhanced the production of specific DNA bands by increasing their concentration applied. However, we noticed that G/C contents of random primers did not show any correlations with number of specific RAPD bands generated but the RAPD results were heavily influenced by the characteristics of the random primers, that is, the sequences of the oli.

Genetic Analysis of Somaclones Derived from Different Cytotype Plants of Scilla scilloides Complex using RAPD (PCR-RAPD 기법을 이용한 세포 유형이 다른 무릇 (Scilla scilloides Complex) 체세포클론의 유전적 분석)

  • The usefulness of randomly amplified polymorphic DNA (RAPD) was evaluated to access the genetic variation in somaclones derived from different cytotype plants of Scilla scilloides Complex, AA (2n=16), BB (2n=18) and AABB (2n=34). Three arbitrary decamer primers were successfully used to amplify genomic DNA from the somaclones. DNA polymorphism was observed between cytotypes. The total number of bands in AA, BB and AABB somaclones were 110, 116 and 103, and marker bands examined were 15, 19 and 26, respectively. The diversity of types using PCR in AA, AABB and BB somaclones were 39.2%, 72.3% and 45.7%, respectively. RAPD band patterns suggest that type AA is more stable than type BB and AABB. The frequencies of specific band in AA, BB or AABB somaclones were 0.9%, 4.3% and 4.9%, respectively. The applicability and reliability of RAPD markers for evaluating the somaclones are discussed.

  • RAPD analysis was examined to estimate the availability as a genetic marker. The availability was evaluated in terms of genetic divergence and relationships among Haliotis discus hannai, H. rufescens, H. rubra and H. midae in both hemispheres of the world. In results, RAPD analysis showed a clear genetic divergence between every pair of species. However, genetic relationships among the four species estimated by RAPD analysis unreflected to geographical distribution and morphological characteristics. In conclusion, RAPD is suitable genetic markers for estimates of genetic divergence and differences among abalone species.

RAPD-PCR Analysis in Fusarium species (Fusarium 종에서의 RAPD-PCR분석)

  • To assess genetic diversity amoug 21 strains from sixleen Frrsn~i~nn species , we used RAPD(rando1n amplified pol.ymorphic DNA) analysis based on PCR(po1ymerase chain reaction). Eleven primers showing Ule polymorphism were chosen from the 40 random pnmers-tcstcd. A total of 263 polymorphic bands were generated by the primers and the size of amplified DNA fragments ranged from 0.1 lo 3.0 kb. Sirnilku-it), coefficients between strains were calcnlatcd, and UPGMA cluster analysis was used to generate a dendrogram showing relationships among them. The results from RAPD-PCR analysis were grouped into four main groups at the si~nilarity level of 0.627.

Genetic Variations and Relationships of Persicaria thunbergii(Sieb. & Zucc.) H. Gross ex Nakai(Polygonaceae) by the RAPD Analysis (RAPD 분석에 의한 고마리(마디풀과)의 유전적 변이 및 유연관계)

  • RAPD analyses were performed from twenty-four populations of Persicaria thunbergii(Sieb. & Zucc.) H. Gross ex Nakai. The length of amplified DNA fragments ranged from 200 to 1,900bp. 184 scorable RAPD markers were found from PCR reactions with sixteen random oligoprimers. Based on the results, populations of Persicaria thunbergii were classified into disturbance streams of urban and rural streams as well as natural streams. And the populations from natural streams showed having higher genetic similarites than those from highly disturbed streams, Also, the heterogenetic differences between the populations from natural and disturbed areas could be represented the results of the stream environmental changes.

Random Amplified Polymorphic DNA Analysis for Typing Extended-Spectrum-β-Lactamase of Klebsiella pneumoniae

  • Fifty-one extended-spectrum-${\beta}$-lactamase(ESBL) producing Klebsiella pneumoniae strains were isolated from national university hospitals. All K. pneumoniae strains showed resistance to broad-spectrum antibiotic and most of them presented resistance to amikacin, gentamicin and ciprofloxacin. The results of amplified polymorphic DNA (RAPD) pattern for randomly isolated fifty-one strains were as follows; both twenty-one strains from Chungnam National University hospital and ten strains from Chungbuk National University hospital showed RAPD type Ia and Ib. However, twenty strains isolated from Gyeongsang National University hospital belonged to RAPD type IIa and IIb. All isolates were divided into four molecular types and showed high level of genetic diversity. These results suggested that RAPD analysis provided a rapid and simple method for analysing genotypes of ESBL.
