긴 세장한 R/C 벽체의 연성능력 (Ductility Capacity of Slender-Wind R/C Walls)

  • This study investigates the ductility capacity of slender-wide reinforced concrete walls under predominant flexural moment loading. The experimental work for this study aims to provide design guidelines for bar detailing in critical regions under compressive stress in particular in case of slender-wide RC walls. According to the experimental observation the Bernoulli hypothesis of linear strain distribution is no longer valid and the ultimate compressive strain of concrete is significantly reduced, It is postulated that the nonlinear strain distribution causes the concentrated compressive stressed region and hence the premature crushing failure at the toe of walls. The reduced ultimate strain and nonlinear strain distribution need transverse reinforcement for confinement and more realistic models for the strength and displacement estimation of slender-wide RC wall.

  • Urbanization and development of industry makes people concerned about quality of circumstances. Problems of vibration are on the rise. Vibration makes inhabitants feel unpleasant and involves structural damage. The purpose of this study is to assess damage of reinforced concrete structures due to ground motions as the parameters of frequency, duration time and aspect ratio of structures are changed. Ground motions were modeled as sine waves. To compare sine waves with real ground motions, two cases are selected; one is blast loading case and the other is earthquake loading. It was intended to provide means to assess R/C structure damage due to ground motions.

대장균에서 xylA 유전자의 발현조절 (Regulation of xylA Gene Expression in Escherichia coli)

  • 대장균에서 xylose isomerase(XI) 생산의 조절양상을 밝히기 위한 연구의 일환으로 유도물질인 xylose에 의한 XI 생산유도 및 glucose에 의한 이화물 억제 양상을 조사하였다. XI 생산 유전자인 xylA 유전자의 발현을 조절하는 xylR 유전자가 염색체에 존재하는 상태에서 xylA 유전자가 고복제수 유래의 플라스미드에 존재하는 경우 (pEX202/DH77)와 저복제수 유래의 플라스미드에 존재하는 경우(pEX102/DH77)에는 염색체에 존재하는 경우 (JM109)보다 0.4% xylose 첨가에 의한 XI의 유도생산이 각각 1.9 및 1.7배 정도 증가하였다. 염색체에 존재하는 xylR 유전자에 의해 생산된 xylR유전자 산물이 xylA 유전자가 플라스미드에 존재할 경우에도 염색체에 존재할때와 마찬가지로 작용하는 것으로 나타났다. 형질전환주 pEX202/DH77과 pEX102/DH77 및 친주 JM109에서 다 같이 0.2% glucose 첨가에 의해 완전히 XI 유도생산이 억제되었으며 이와같은 glucose에 의한 이화물 억제는 1 mM cAMP의 첨가로 해제되었다. DM 최소배지에서 xylose에 의한 XI 유도시 1 mM CAMP를 첨가하면 0.4% xylose만 첨가했을때 보다 XI 생산이 1.7 내지 2배 정도 증가하었다. Xylose isomerase와 cAMP 생산 변이주(xyl, cya ; TP2010)에 xylA 유전자를 형질전환시킨 pEX13/TP2010은 xylose 첨가로 Xl가 유도생산되지 않았고 cAMP를 함께 첨가해야만 XI가 유도되었다. 이와같이 대장균의 xylA 유전자에서 XI의 생산조절에는 xylose이외에 cAMP도 필수적인 효과물질임을 알 수 있었다.

Complete $^1H$-NMR and $^{13}C$-NMR spectral analysis of the pairs of 20(S) and 20(R) ginsenosides

  • Background: Ginsenosides, the major ingredients of Panax ginseng, have been studied for many decades in Asian countries as a result of their wide range of pharmacological properties. The less polar ginsenosides, with one or two sugar residues, are not present in nature and are produced during manufacturing processes by methods such as heating, steaming, acid hydrolysis, and enzyme reactions. $^1H$-NMR and $^{13}C$-NMR spectroscopic data for the identification of the less polar ginsenosides are often unavailable or incomplete. Methods: We isolated 21 compounds, including 10 pairs of 20(S) and 20(R) less polar ginsenosides (1-20), and an oleanane-type triterpene (21) from a processed ginseng preparation and obtained complete $^1H$-NMR and $^{13}C$-NMR spectroscopic data for the following compounds, referred to as compounds 1-21 for rapid identification: 20(S)-ginsenosides Rh2 (1), 20(R)-Rh2 (2), 20(S)-Rg3 (3), 20(R)-Rg3 (4), 6'-O-acetyl-20(S)-Rh2 [20(S)-AcetylRh2] (5), 20(R)-AcetylRh2 (6), 25-hydroxy-20(S)-Rh2 (7), 25-hydroxy-20(S)-Rh2 (8), 20(S)-Rh1 (9), 20(R)-Rh1 (10), 20(S)-Rg2 (11), 20(R)-Rg2 (12), 25-hydroxy-20(S)-Rh1 (13), 25-hydroxy-20(R)-Rh1 (14), 20(S)-AcetylRg2 (15), 20(R)-AcetylRg2 (16), Rh4 (17), Rg5 (18), Rk1 (19), 25-hydroxy-Rh4 (20), and oleanolic acid 28-O-b-D-glucopyranoside (21).

전극질량 변화에 따른 SMD형 변성 PbTiO3세라믹 공진자의 공진특성 (Resonant Characteristics of SMD Type - Modified PbTiO3 Ceramic Resonator with the Variations of Electrode Weight)

  • In this study, modified PbTi $O_3$ ceramics was manufactured to apply for 30MHz SMD type ceramic resonator with the variations of electrode weight. To investigate the effects of electrode weight on resonant characteristics of ceramic resonator using 3$^{rd}$ overtone thickness vibration mode, ceramic wafers for resonator were fabricated by evaporating electrode weights of 0.66, 1.765, 2.32, 3.87$\times$ 10$^{-4}$ g/c $m^2$ with silver, respectively. And then, SMD type ceramic resonators were fabricated with the size of 3.7$\times$3.1mm and electrode radius size of 0.77mm. With increasing electrode weight, resonant resistance was gradually decreased. At the electrode weight of 2.32$\times$10$_{-4}$ g/c $m^2$, mechanical quality factor( $Q_{mt3}$) and dynamic range(D.R) showed the maximum value of 2,152 and 49dB, respectively.

Lactic Acid Bacteria from Gamecock and Goat Originating from Phitsanulok, Thailand: Isolation, Identification, Technological Properties and Probiotic Potential

  • From independent swab samples of the cloaca of indigenous gamecocks (CIG), anus of healthy baby goats (AHG), and vagina of goats (VG) originating from Phitsanulok, Thailand, a total of 263 isolates of lactic acid bacteria (LAB) were collected. Only three isolates, designated C707, G502, and V202, isolated from CIG, AHG, and VG, respectively, exhibited an excellent inhibitory zone diameter against foodborne pathogenic bacteria when evaluated by agar spot test. Isolates C707 and G502 were identified as Enterococcus faecium, whereas V202 was identified as Pediococcus acidilactici, based on 16S rRNA sequence analysis. When foodborne pathogenic bacteria were co-cultured with chosen LAB in mixed BHI-MRS broth at 39℃, their growth was suppressed. These LAB were found to be capable of surviving in simulated stomach conditions. Only the isolate G502 was able to survive in the conditions of simulated intestinal juice. This research suggests that selected LAB could be used as a food/feed supplement to reduce foodborne pathogenic bacteria and improve the safety of animal-based food or feed.

Regulatory B Subunits of Protein Phosphatase 2A Are Involved in Site-specific Regulation of Tau Protein Phosphorylation

  • Overexpression of amyloid precursor protein with the Swedish mutation causes abnormal hyperphosphorylation of the microtubule-associated protein tau. Hyperphosphorylated isoforms of tau are major components of neurofibrillary tangles, which are histopathological hallmarks of Alzheimer's disease. Protein phosphatase 2A (PP2A), a major tau protein phosphatase, consists of a structural A subunit, catalytic C subunit, and a variety of regulatory B subunits. The B subunits have been reported to modulate function of the PP2A holoenzyme by regulating substrate binding, enzyme activity, and subcellular localization. In the current study, we characterized regulatory B subunit-specific regulation of tau protein phosphorylation. We showed that the PP2A B subunit PPP2R2A mediated dephosphorylation of tau protein at Ser-199, Ser-202/Thr-205, Thr-231, Ser-262, and Ser-422. Down-regulation of PPP2R5D expression decreased tau phosphorylation at Ser-202/Thr-205, Thr-231, and Ser-422, which indicates activation of the tau kinase glycogen synthase kinase 3 beta ($GSK3{\beta}$) by PP2A with PPP2R5D subunit. The level of activating phosphorylation of the $GSK3{\beta}$ kinase Akt at Thr-308 and Ser-473 were both increased by PPP2R5D knockdown. We also characterized B subunit-specific phosphorylation sites in tau using mass spectrometric analysis. Liquid chromatography-mass spectrometry revealed that the phosphorylation status of the tau protein may be affected by PP2A, depending on the specific B subunits. These studies further our understanding of the function of various B subunits in mediating site-specific regulation of tau protein phosphorylation.

한국노인의 연구참여를 위한 동의능력평가척도의 타당성 연구 (The Validation of a Measurement for Assessing the Capacity of Korean Older Adults to Consent to Research)

  • 본 연구의 목적은 국내에서 노인복지연구 참여를 위한 고지된 동의과정에 동의능력평가가 활발하게 적용될 수 있도록 한국형 동의능력척도를 실증적으로 검증하는 것이다. 본 연구를 위해 재가복지서비스를 이용하거나 노인복지시설에서 보호를 받고 있는 404명의 노인을 조사하였다. 연구 1(n=202)에서는 동의능력검사척도의 신뢰도, 탐색적 요인분석, 수렴 및 기준 타당도 분석을 하였다. 연구 2(n=202)는 연구 1을 통해 수정된 척도의 신뢰도와 타당도는 내적 신뢰도, 확증적 요인분석, 수렴 및 기준 타당도 분석 등을 통해 검증하였다. 연구결과 본 척도의 내적신뢰도 계수가 .90로 높은 수준이었고, 모델 적합도 지수(RMSEA=.057, NNFI=.94, IFI=.96, CFI=.96)도 양호하게 나타났다. 또한 관련변수와의 높은 상관관계 계수를 통해 수렴 및 기준타당도가 검증되었다. 한국형 동의능력평가척도가 우리나라 노인을 대상으로 임상(clinical) 및 조사(survey)연구 참여를 위해 동의능력을 평가하는 척도로서 적합함을 통계적으로 확인하였다. 본 척도는 노인을 대상으로 수행될 연구와 프로그램에서 고지된 동의과정을 위해 활용될 것으로 기대된다.

저열량 감미료 Tagatose의 이화학적 특성 (Physicochemical Properties of a Low Calorie Sweetener, Tagatose)

  • 새로운 저열량 감미료인 tagatose를 식품에 적용하기 위해 여러 가지 이화학적 특성을 조사하였다. Tagatose 용액은 Bingham 유체의 점성 특성을 보여주었고 용액의 농도가 $25^{\circ}C$에서 10%에서 50%로 증가할수록 점도가 1.65 cp에서 5.14 cp로 증가하여 비교적 저 점도를 보여주었다. 또한, 온도가 $15^{\circ}C$에서 $55^{\circ}C$로 증가함에 따라 점도가 4.59 cp에서 2.33 cp로 감소함을 보여주었다. Differential scanning calorimetry에 의하여 용융 특성을 조사한 결과 tagatose의 용융 개시 온도는 $130.4^{\circ}C$, 흡열 엔탈피는 -202.3 J/g이었다. 수분활성도는 60% 농도에서 tagatose는 0.892, sucrose는 0.957로 tagatose가 더 낮은 수분활성도를 보여주었다. 흡습성은 상대습도 85%에서 100%까지 tagatose가 sucrose보다 높았다. $20^{\circ}C$에서 $70^{\circ}C$까지의 온도 변화에 따른 용해도를 보았을 때는 tagatose가 sucrose보다 낮았다. pH 2에서 pH 12까지 조정한 용액을 3일 후 관찰하였는데 pH에 대하여 안정하였다. 또한 $154^{\circ}C$까지 4시간 열처리하였을 때 거의 변성이 되지 않았다. 그러나 높은 온도에서 열처리를 한 경우 육안으로 보이는 갈변화에 의하여 흡광도가 증가하는 것으로 나타났다.

Crystal Structures of Dehydrated $Ag^{+}\;and\;Ca^{2+}$ Exchanged Zeolite A, $Ag_{3.3}Ca_{4.35}$-A and of Its Ethylene Sorption Complex

  • Two crystal structures of dehydrated $Ag_{3.3}Ca_{4.35}-A ({\alpha} = 12.256(2){\AA})$ and of its ethylene sorption complex (${\alpha} = 12.259(2){\AA}$) have been determined by single-crystal X-ray diffraction techniques in the cubic space group Pm3m at 21(l)$^{\circ}$C. Both crystals were dehydrated at 360$^{\circ}$C and $2{\times}10^{-6}$ Torr for 2 days and one crystal was treated with 200 Torr of ethylene at 24(2)$^{\circ}$C. The structures were refined to final error indices, $R_1$=O.065 and $R_2$ = 0.088 with 202 reflections and $R_1$=0.049 and $R_2$ = 0.044 with 259 reflections, respectively, for which I>3${\sigma}$(I). In these structures, all Ag$^+$ and Ca$^{2+}$ ions are located on two and three different threefold axes associated with 6-ring oxygens, respectively. In $Ag_{3.3}Ca_{4.35}-A{\cdot}6.65\;C_2H_4,\;3.3\;Ag^+\;and\;3.35\;Ca^{2+}$ ions are recessed 1.09 ${\AA}$ and 0.21 ${\AA}$, respectively, into the large cavity from the (111) plane at O(3). Each Ag$^+$ and Ca$^{2+}$ ion in the large cavity forms a complex with one $C_2H_4$$^{2+}$ ions and ethylene molecules are longer than those between Ag$^+$ ions and ethylene molecules.
