In July 2021, we surveyed diseases affecting wild vegetables grown in Hongcheon, Gangwon Province, Korea. During this survey, we observed severe seedling rot symptoms in Kamchatka goatsbeard (Aruncus dioicus) grown in a vinyl greenhouse. The incidence of the disease in the plant seedlings was 1-10%. Diseased seedlings from this population were collected, and fungi were isolated from leaf and petiole lesions. Rhizoctonia sp. was consistently isolated from the lesions. We examined the morphological and cultural characteristics and anastomosis groups of nine Rhizoctonia sp. isolates obtained from the lesions. The results revealed that all isolates corresponded to Rhizoctonia solani AG-1(IB). Three isolates of R. solani AG-1( IB) were evaluated to determine their pathogenicity towards Kamchatka goatsbeard seedlings through artificial inoculation. The tested isolates caused rot symptoms on the inoculated plant seedlings. The symptoms were similar to those observed in plant seedlings from the vinyl greenhouse. We found that R. solani AG-1(IB) caused seedling rot of Kamchatka goatsbeard.
Rhizoctonia solani Kuhn[Thanatephorus cucumeris (Frank) Donk]is a destrutive soil-borne plant pathogen affecting many agricultural crops worldwide. R. solani is divided into anastomosis groups based on the ability of the hyphae to fuse, and into subgroups based on morphological, physiological characteristics. AG classifications are convenient and useful in identifying primary causal agents of Rhizoctonia diseases, although the mechanism of anastomosis is not fully understood. Beacause of the difficulties, we sought to develop a more direct method for genetic identification and charaterization of R. solani. Twenty nine isolates of R. solani were used for the analysis of genetic relationships among themselves and for rapid anastomosis grouping with AFLP method. All isolates studied were divided into five groups. Isolate 6 was included in AG-3 with 67% genetic similarity. When isolates 3 was compared with 13 and 10 each, they showed more than 84% and 83% similarity, respectively. Isolates 3, 4, 5, 13, and 16 were included in AG-1 with 83% genetic similarity. Isolates 1, 7, and 8 were included in AG-1(IB).
Twenty-three strains of Serratia sp., isolated from surface-sterilized rice roots collected in Chonbuk and Chungnam province, were identified and characterized. They were Gram-negative, rod shaped and red pigmented typically and their endophytism was confirmed by inoculation and reisolation of the strains in planta. Their antifungal activity against 4 rice pathogenic fungi was compared and ranged from 62.4 to 85.2% against Rhizoctonia solani and 68.0 to 88.5% against Pyricularia grisea. Among the 23 strains tested, strain Rsm220 showed the strongest inhibition activity against 4 pathogenic fungi. The strain was, therefore, selected as a biocontrol candidate for both the pathogens and its bacteriological characteristics and 165 rDNA sequences were analyzed. Phenotypic and biochemical characteristics of the selected Rsm220 were highly related to the type strain of S. marcescens and 165 rDNA sequencing of Rsm220 showed a homology of 98.2% to the type strain of S. marcescens. The strain Rsm220 was identified as S. marcescens and the inhibition result of this endophytic strain indicates that it is a potential biocontrol agent for R. solani and R grisea.
The objective was to examine the antifungal activity of Pinus densiflora extract for the control of turfgrass fungal diseases. Antifungal activities of the various fractions of n-hexane, methylene chloride (Ch), ethyl acetate (EtOAc), and n-butanol from P. densiflora were evaluated against Rhizoctonia solani AG1-1B, R. solani AG2-2IV, Sclerotinia homoeocarpa, R. cerealis, Pythium spp., and Colletotrichum graminicola. The Ch and EtOAc fractions showed antifungal activity against Pythium sp. and C. graminicola in paper disc assay. The effective concentration to produce 50% mycelial inhibition ($EC_{50}$) using five discriminatory concentrations of pinosylvin (1) from the Ch fraction of P. densiflora was evaluated on R. solani AG1-1B, R. solani AG2-2IV, R. cerealis, and S. homoeocarpa. S. homoeocarpa showed the highest sensitivity with the lowest mean $EC_{50}$ value ($8.426{\mu}g/mL$) among the four pathogens. Among the three Rhizoctonia pathogens, R. cerealis had the highest mean $EC_{50}$ value ($99.832{\mu}g/mL$) and R. solani AG2-2IV, with the lowest sensitivity, had the lowest $EC_{50}$ value ($39.696{\mu}g/mL$). These results suggested that pinosylvin (1) from P. densiflora could be a valuable lead compound in the improvement of a novel antifungal agent.
A fungal strain of Trichoderma having strong chitinolytic activity was isolated from field soil enriched with crabshell for several years. Based on 5.8S rRNA, partial 18S, 28S rRNA genes, ITS1, ITS2 sequence analysis and morphological characteristics, the fungus was identified as Trichoderma asperellum and named as Trichoderma asperellum T-5 (TaT-5). The fungus released lytic enzymes such as chitinase and ${\beta}$-1, 3-glucanse, and produced six antifungal substances in chitin broth medium. To demonstrate the protective effect of TaT-5 against damping-off in cucumber plant caused by Rhizoctonia solani, TaT-5 culture broth (TA), chitin medium (CM) and distilled water (DW) were applied to each pot at 10 days after sowing, respectively. Then, the homogenized hyphae of R. solani were infected to each pot at 1 week after TaT-5 inoculation. During experimental period, fresh weight of shoot and root in cucumber plant more increased at TA treatment compared to other treatments. PR-proteins (${\beta}$-1, 3-glucanase and chitinase) activities in cucumber leaves markedly increased at CM and DW treatments, but the activity slightly increased and then decreased at TA treatment at 3 days after infection of R. solani. The activity of PR-proteins activities in cucumber roots at all treatments decreased with time where the degree of decrement was more alleviated at TA treatment than CM and DW. These results suggest that the lytic enzymes (chitinase and ${\beta}$-1, 3-glucanse) and antifungal substances produced by TaT-5 can reduce the pathogenic attack by R. solani in cucumber plants.
Fifty three diseased root samples of Chinese cabbage and 22 soil samples were taken from six alpine areas in Kangwon province to examine the organisms causing root rot of Chinese cabbage. Among thirteen microorganisms detected from diseased roots samples, Pythium ultimum, Pythium echinocarpum and Rhizoctonia solani were detected frequently and were pathogenic to Chinese cabbage. Five to 100% of young Chinese cabbage plants grown in the sample soils taken from alpine cabbage fields were infected either alone or together with P. ultimum, P. echinocarpum and R. solani, depending upon the or gin of sample soils. When relative density of the three organsims in the sample soils was estimated based on the infection frequency on the plants grown in those sample soils, P. ultimum was prevalent only in Chahang-i-li area. P. echinocarpum was prevalent in Yongsan-il-li, Hoengwe-li, and Munmaek areas. R. solani was solely found in Maebongsan area where both P. ultimum and P. echinocarpum were not detected. In the three areas where both P. ultimum and P. echinocarpum were present, density of P. echinocarpum was generally greater than that of P. ultimum. Seedlings of thirty major Chinese cabbage varieties were inoculated independently with the three organisms in the greenhouse. Six varieties were resistant to P. ultimum. Three and two varieties were moderately resistant to P. ultimum and P. echinocarpum, respectively. All varieties were highly susceptible to R. solani.
Herein, we report the first occurrence of web blight of rosemary caused by Rhizoctonia solani AG-1-IB in Gangneung, Gangwon Province, Korea, in August 2014. The leaf tissues of infected rosemary plants were blighted and white mycelial growth was seen on the stems. The fungus was isolated from diseased leaf tissue and cultured on potato dextrose agar for identification. The young hyphae had acute angular branching near the distal septum of the multinucleate cells and mature hyphal branches formed at an approximately $90^{\circ}$ angle. This is morphologically identical to R. solani AG-1-IB, as per previous reports. rDNA-ITS sequences of the fungus were homologous to those of R. solani AG-1-IB isolates in the GenBank database with a similarity percentage of 99%, thereby confirming the identity of the causative agent of the disease. Pathogenicity of the fungus in rosemary plants was also confirmed by Koch's postulates.
This study was undertaken to find a new formulation of soil amendment, and selection of antogonists and to effectively control brown and large patch of turfgrasses caused by Rhizoctoniz solani AG1-1 and AG 2-2. Fourteen inorgainc chemicals (1%, w/w) were added individually in vitro, and some chemicals showed suppressiveness to R. solani. Alum suppressed effectively mycelial growth of R. solani in the range of 17 to 77% as compared with control. The four chemicals such as Al2(SO4)3, alum, CaO, and NH4NO3 were finally selected. Out of three organic compounds, composted pine bark (CPB) showed prominent suppressive effect as compared with milled alfalfa and pine leaves. After inoculation of R. solani isolates AG-1 and AG2-2 on the turf seedlings, water soaked lesions and blight symptoms were developed on the whole seedlings. According to inhibition zone method, mycelial growth of the fungus were greatly suppressed by culture filterates of the antagonists, Gliocladium virens (Gl1-) and Pseudomonas sp. (P713). CPB soil amendment mixed with antagonists (1% w/w) controlled not only brown and large patch of turfgrasses, but also promote the good growth of the seedlings. In addition, the controlling effect was maintained more than 30 days. Especially, the controlling effect of two antagonists was similar to Cㅖㅠ soil amendment with the antagonists and also stimulated a favorable growth of the seedlings. Therefore, its is expected that continuous control of Rhizoctonia blight of turfgrasses can be obtained in field by subsequent applications of the antagonists.
Incidence of radish disease caused by Rhizoctonia solani ranged from 1 to 30% in fields located at Hwaseong, Naju and Yeoncheon in Korea during the growing seasons in 1989, 1990 ad 1993. A total of 133 isolates of R. solani was obtained from the diseased seedlings, leaves and roots of radish collected. The fungus was most commonly isolated from the roots. Among 133 isolates of R. solani, 56 isolates were classified as anastomosis group AG-1 by anastomosis test, 37 isolates as AG-2-1, and 40 isolates as AG-4. Among the isolates of AG-1, 26 isolates were grouped as cultural type IA, and the others as cultural type IB. Cultural types IA and IB of AG-1, were isolated from the leaves, AG-2-1 from the roots, and AG-4 from the seedlings, leaves and roots. Pathogenicity tests revealed that the AG-1(IA) isolates were highly virulent on leaves of radish, but avirulent on the seedlings, petioles and roots. The AG-1(IB) isolates were highly virulent on the leaves, but mildly virulent on the seedlings and avirulent or mildly virulent on the petioles and roots. The AG-2-1, isolates were mildly virulent on the leaves and seedlings and mildly or highly virulent on the petioles and roots. The AG-4 isolates were highly virulent on the seedlings and mildly or highly virulent on the leaves, petioles and roots.
A large patch disease caused by Rhizoctonia solani AG2-2 (IV) is a serious problem in Korean lawngrass (Zoysia japonica) sites including golf courses and sports fields in Korea. Antagonistic microorganisms against R. solani AG2-2 (IV) were isolated from various forest and crop soil sources in Southern Korea. Among the 61 isolates, I-009, FRIN-001-1, and YPIN-022 strains showing dramatic inhibition of the mycelial growth of R. solani AG2-2 (IV) in the pairing culture were selected as the most potential antagonistic microorganisms for this study. Based on the 16s RNA sequence comparison, I-009 and FRIN-001-1 isolates were identified as Bacillus spp., while YPIN-022 isolate belongs to the genus Pseudomonas. The greater inhibition (clear) zone between two edges of the selected and pathogenic microbes ranged from 11 to 15 mm in three selections, but the others averaged to 7 mm out of 30 mm distance. In another antifungal test using culture filtrate, those three isolates represented a range of 51.7 to 63.5% suppression potential. The selected isolates also inhibited significantly the stem-segment colonization by R. solani AG2-2 (IV) in vivo test by 28.1%, 43.0%, and 23.7% when inoculated with I-009, FRIN-001-1, and YPIN-022, respectively. The highest antagonistic activity for the large patch disease was demonstrated by the isolate FRIN-001-1, which will be useful for developing a bio-pesticide against Rhizoctonia.
