  • This study was conducted to determine the inheritance of resistance in some rice varieties such as WASE AIKOKU 3, ZENITH, TETEP and TKM 6 to isolate 71-23 of Xanthomonas Oryzae in Korea. From the test in $F_1$ and $F_2$ populations of crosses between resistant and susceptible parents, it was observed that resistance of them were controlled by one dominant gene.

  • Several plant and microbial genes that could confer disease resistance in transgenic rice plants are being cloned and characterized. We are currently constructing transgenic rice lines that overexpress the gene products, such as a galactinol synthase, a defensin, and a bacterial ACC deaminase. Subtractive hybridization of a rice cDNA library constructed from the Xanthomonas oryzae-infected ice leaves resulted in isolation of many inducible cDNA clones including a elongation factor EF2, a oryzain alpha, a catalase, a aldehyde dehydrogenase, a S-adenosylmethionine synthetase, a caffeic acid O-methyltransferase, a glyceraldehyde-3-phosphate dehydrogenase, a light-regulated protein, nKY transcription factors, and a nucleotide diphosphate kinase. Some genes among those may be useful genetic sources for construction of disease resistant transgenic rice. Full lengths of the rice OsFIERG and a rice oryzain genomic clones were cloned, and serial deletion fragments of the promoter regions of these genes were fused with GUS reporter gene in pCAMBIA1201, respectively. Promoter activities of these constructs will be examined upon various stresses and Pathogen infections to obtain the pathogen specific inducible-promoter. Studies on Rice Varietal Resistance to Rice Stem Maggot, Chlorops oryzae Matsumura (벼줄기굴파리에 대한 수도(水稻)의 품종저항성(品種抵抗性))

  • To establish a method for measuring varietal resistance against rice stem maggot in field and pot, 20 varieties were used in $1981{\sim}1983$. The degree of injured stems by the 1st generation larvae was significantly correlated with that of injured panicles by the 2nd ($r=0.864^{**}$). There was no ovipositional preference in rice stem maggot. The larval mortality after artificial infestation varied widly from 5.6 to 100% in varieties and that of the resistant varieties was high after boring. There was a negative correlation between number of injured leaves and larval mortality after artificial infestation ($'81:r=-0.833^{**},\;'82:r=-0.918^{**}$). It is considered that larval mortality mainly caused by antibiosis and larval mortality 20 days after infestation on rice seedlings is ideal for evaluating the varietal resistance to rice stem maggot. Resistant varieties are Nagdongbyeo, Jinjubyeo and Chucheongbyeo susceptible varieties, Keumgangbyeo, Milyang 23, Baigyangbyeo and Milyang 30.

Diversity Analysis of Diazotrophic Bacteria Associated with the Roots of Tea (Camellia sinensis (L.) O. Kuntze)

  • The diversity elucidation by amplified ribosomal DNA restriction analysis and 16S rDNA sequencing of 96 associative diazotrophs, isolated from the feeder roots of tea on enriched nitrogen-free semisolid media, revealed the predominance of Gram-positive over Gram-negative bacteria within the Kangra valley in Himachal Pradesh, India. The Gram-positive bacteria observed belong to two taxonomic groupings; Firmicutes, including the genera Bacillus and Paenibacillus; and Actinobacteria, represented by the genus Microbacterium. The Gram-negative bacteria included ${\alpha}$-Proteobacteria genera Brevundimonas, Rhizobium, and Mesorhizobium; ${\gamma}$-Proteobacteria genera Pseudomonas and Stenotrophomonas; and ${\beta}$-Proteobacteria genera Azospira, Burkholderia, Delftia, Herbaspirillum and Ralstonia. The low level of similarity of two isolates, with the type strains Paenibacillus xinjiangensis and Mesorhizobium albiziae, suggests the possibility of raising species novum. The bacterial strains of different phylogenetic groups exhibited distinct carbon-source utilization patterns and fatty acid methyl ester profiles. The strains differed in their nitrogenase activities with relatively high activity seen in the Gramnegative strains exhibiting the highest similarity to Azospira oryzae, Delftia lacustris and Herbaspirillum huttiense.

Characterization of a heat-resistant antimicrobial peptide secreted by Bacillus subtilis A405 (Bacillus subtilis A405 균주가 생성하는 내열성 항균 peptide의 특성 검정)

  • An antimicrobial peptide producing bacterium, Bacillus subtilis A405, was screened and identified among 700 of antagonistic bacteria. The heat-resistant antimicrobial peptide, AMP-405, was purified from the broth culture of B. subtilis A405 through $20{\sim}40%$ ammonium sulfate precipitation and ultrafiltration. The AMP-405 exhibited strong antimicrobial activities against Botrytis cinerea, Cercospora sp., Fusarium oxysporum, Penicillium digitatum, Celletotrichum gloeosporioides, Rhizoctonia solani, Pythium ultimum, Pyricularia oryzae, Escherichia coli, Pseudomonas spp. and Candida albicans. The molecular weight of the peptide was about 3.0 kDa determined by SDS-PAGE, Native-PAGE and Tris-Tricine gradient electrophoresis, and composed of 9 kinds of amino acid such as aspartic acid, glycine, serine, glutamine, valine, leucine, isoleucine, proline, tyrocine. To determine the efficiency of AMP-405 as a potential maintenance of fruits freshness, cherry tomato was srored at $25^{\circ}C$ for 2 weeks after treatment with $50{\mu}g/ml$ of AMP-405 and $10^{5}$ spores/ml of Botrytis cinerea simultaneously. Treatment with AMP-405 resulted in significantly less infection by Botrytis cinerea, than the treatment with tap water as a control.

  • The experiments were carried out to investigate the selection effects on the drug resistance to Agrepto in Xanthomonas oryzae, the causal bacteria of rice bacterial leaf blight. The results obtained were as follows. 1, The Agrepto was stable at the heat treatment of $105^{\circ}C$ for 20 minutes, when the drug was added in the media. 2. The local isolates of the bacteria, 75-6 and 75-9, showed the different resistante reaction, when they were selected by the Agrepto contained media which concentrations of 10r9/m1 and 100ug/ml. 3. The individuals shelving high degree of resistance, which can grow on the media contained 10,000ug/m1 of Agrepto, could selected by the concentrations as low as 10ug/ml and 100ug/m1, in one generation. 4. The highly resistant isolates which selected by 3-stepwise selections such as 100ug/ml, 3,000ug/ml and 10,000/ml plots, showed nearly normal growth at the media contained 100ug/ml of Agrepto 5. When the isolate 75-9 was selected at the 100ug/m1 of concentration, showed various degrees of resistance, indicating that the isolate may be composed of resistance groups that lower than 500ug/ml, between 500-1,000ug/ml and 1,000-3,000ug/ml, to the Agrept.

  • The study has been carried to investigate the nature of resistance in rice varieties against bacterial leaf blight caused by Xanthomonas oryzae (Uyeda et. Ishiyama) Dowson. In this study, differences on several morphological histological and bacterial growth in rice leaf extract from different varieties were examined. Shirogane and Norm 6 as resistant varieties, Paldal as moderately resistant, and Jinheung, Kimmaze and Suwon 213 as susceptible varieties were used throughout the experiment. 1. Susceptible varieties Jinheung, Kimmaze have more hydathods in flag leaf than resistant varieties but there were no difference between resistant varieties and moderatly resistant varieties. 2. Average length of xylem vessel elements were 50r longer in leaf veins of susceptible varieties Jinheung, Kimnaze and Suwon 213 than those of resistant varieties, but there were no difference among Paldal, Norm 6 and Shirogane. 3. Kimmaze and Suwon 213 have larger diameter of xylem vessel elements than those of other varieties examined. However, it did not differ significantly between resistant varieries and susceptible varieties statistically. 4. Jinheung and Kimmaze have more secondary xylem vessels in root of 6-7th leaf stage than those of Shirogane and Norm 6. Suwon 213, however, showed least number of secondary xylem vessels exceptionally. 5. Leaf extract from resistnat variety Shirogane, suppressed bacterial growth significantly when compared with those from other varieties. 6. Bacterial growth in autoclaved leaf extract from resistant varieties and susceptible varieties did not show any noticeable difference. In general, fresh extract maintained more bacteria than in autoclaved extract after 48 hours of incubation period.

Microbial Floral Dynamics of Chinese Traditional Soybean Paste (Doujiang) and Commercial Soybean Paste

  • Traditional soybean paste from Shandong Liangshan and Tianyuan Jiangyuan commercial soybean paste were chosen for analysis and comparison of their bacterial and fungal dynamics using denaturing gel gradient electrophoresis and 16S rRNA gene clone libraries. The bacterial diversity results showed that more than 20 types of bacteria were present in traditional Shandong soybean paste during its fermentation process, whereas only six types of bacteria were present in the commercial soybean paste. The predominant bacteria in the Shandong soybean paste were most closely related to Leuconostoc spp., an uncultured bacterium, Lactococcus lactis, Bacillus licheniformis, Bacillus spp., and Citrobacter freundii. The predominant bacteria in the Tianyuan Jiangyuan soybean paste were most closely related to an uncultured bacterium, Bacillus licheniformis, and an uncultured Leuconostoc spp. The fungal diversity results showed that 10 types of fungi were present in the Shandong soybean paste during the fermentation process, with the predominant fungi being most closely related to Geotrichum spp., an uncultured fungal clone, Aspergillus oryzae, and yeast species. The predominant fungus in the commercial soybean paste was Aspergillus oryzae.

Bacillus oryzicola sp. nov., an Endophytic Bacterium Isolated from the Roots of Rice with Antimicrobial, Plant Growth Promoting, and Systemic Resistance Inducing Activities in Rice

  • Biological control of major rice diseases has been attempted in several rice-growing countries in Asia during the last few decades and its application using antagonistic bacteria has proved to be somewhat successful for controlling various fungal diseases in field trials. Two novel endophytic Bacillus species, designated strains YC7007 and $YC7010^T$, with antimicrobial, plant growth-promoting, and systemic resistance-inducing activities were isolated from the roots of rice in paddy fields at Jinju, Korea, and their multifunctional activities were analyzed. Strain YC7007 inhibited mycelial growth of major rice fungal pathogens strongly in vitro. Bacterial blight and panicle blight caused by Xanthomonas oryzae pv. oryzae (KACC 10208) and Burkholderia glumae (KACC 44022), respectively, were also suppressed effectively by drenching a bacterial suspension ($10^7cfu/ml$) of strain YC7007 on the rhizosphere of rice. Additionally, strain YC7007 promoted the growth of rice seedlings with higher germination rates and more tillers than the untreated control. The taxonomic position of the strains was also investigated. Phylogenetic analyses based on 16S rRNA gene sequences indicated that both strains belong to the genus Bacillus, with high similarity to the closely related strains, Bacillus siamensis KACC $15859^T$ (99.67%), Bacillus methylotrophicus KACC $13105^T$ (99.65%), Bacillus amyloliquefaciens subsp. plantarum KACC $17177^T$ (99.60%), and Bacillus tequilensis KACC $15944^T$ (99.45%). The DNA-DNA relatedness value between strain $YC7010^T$ and the most closely related strain, B. siamensis KACC $15859^T$ was $50.4{\pm}3.5%$, but it was $91.5{\pm}11.0%$ between two strains YC7007 and $YC7010^T$, indicating the same species. The major fatty acids of two strains were anteiso-$C_{15:0}$ and iso $C_{15:0}$. Both strains contained MK-7 as a major respiratory quinone system. The G+C contents of the genomic DNA of two strains were 50.5 mol% and 51.2 mol%, respectively. Based on these polyphasic studies, the two strains YC7007 and $YC7010^T$ represent novel species of the genus Bacillus, for which the name Bacillus oryzicola sp. nov. is proposed. The type strain is $YC7010^T$ (= KACC $18228^T$). Taken together, our findings suggest that novel endophytic Bacillus strains can be used for the biological control of rice diseases.

Chemical Characteristics and Efficacy of Combined Pesticide Granules Formulated by Different Ways (제제방법별(製劑方法別) 혼합입제(混合粒劑) 농약(農藥)의 특성(特性)과 약효(藥效))

  • A comparative study of chemical characteristics, efficacy and relative merits of extruded and coated granules, containing fungicide and insecticide, was conducted under laboratory and field conditions. Probenzole (3-allyloxy-1,2-benzisothiazole 1,1-dioxide) and carbofuran (2,3-dihydro-2,2-dimethyl-7-benzofuranyl methyl carbamate) were chosen as toxicants for rice blast(Pyricularia oryzae) and brown planthopper (Nilaparvata lugens) control, respectively. Stability of active ingredients in coated granule was superior to the extruded one under accelerated temperature. Active ingredient dissolution of coated one into distilled water showed slow release pattern. Pesticide residues in rice (Oryzae sativa, Chucheongbyeo) shoot applied with extruded one at the dosage of 3kg/10a retained higher levels than those with coated one on two days after application, while the residue levels were a reversed tendency on eight days after application. Efficacy on rice blast exhibited minute differences between the granules, on the other hand efficacy on brown planthopper by coated one was of higher rank than that by extruded one. Production cost of the combined pesticide granule by coating method could be cut down by 6% as compared to extrusion method.
