약용식물 추출물의 Helicobacter pylori에 대한 항균활성 (The Antimicrobial Activity of Medicinal Plants Extracts against Helicobacter Pylori)

  • 41종의 약용식물을 열수추출하여 위궤양 원인균으로 보고된 Helicobacter pylori에 대해 항균활성을 탐색하였다. 2-fold dilution 방법에 의해서는 백련초, 어성초, 방기, 황련은 100 ppm이하, 백두옹, 연교, 종대황, 소엽은 200 ppm 이하, 사간, 우방자, 결명자, 귤피, 희침, 소목은 300 ppm 이하와 농도에서 H. pylori에 대해 항균활성을 나타냈다. 이중 3가지 추출물만이 disc 방법을 통해 항균활성이 재확인되었으며, 소목, 황련, 소엽의 순으로 강한 항균활성을 나타냈다. 항균활성이 확인된 3가지 추출물을 chloroform, ethyl acetate, butanol로 순차 분획하여 H. pylori에 대한 항균활성을 확인하였다. 소목의 경우는 ethyl acetate 분획물에서, 소엽의 경우는 butanol 분획물에서, 황련의 경우는 butanol과 chloroform 분획물에서 가장 강한 항균활성을 나타내었다. 추출물의 분획에 따른 항균활성의 상승효과는 모든 추출물에서 확인할 수 있었다. H. pylori의 crude-urease를 분리하여 urea R broth에서 흡광도와 pH를 조사한 결과, 반응 2시간 후 pH가 8.15까지, 560 nm에서의 흡광도는 1.7까지 증가하는 것으로 나타났다. H. pylori의 urease 활성은 소목, 소엽, 황련에 의해 80%이상 억제되었으며, 이중 소목은 urease 활성을 95%이상 억제시켰다.

근적외선분광법을 이용한 수입건초의 이물질 혼입판정 가능성 평가 (Evaluation of the Potential for the Adulteration Screening of Imported Hay by Near Infrared Reflectance Spectroscopy)

  • Near-infrared reflectance spectroscopy (NIRS) was used to study the potential of adulteration of imported forage. Hay samples were prepared two set ; calibration set and validation one. The former were mixed 12 sets from 100% to 50% with Yangcho (Chinese leymus, leymus chinensis Trin.) and the latter were adulterated with 6 set of 8% to 38% in 5% interval. Mixed materials with Yangcho were rice straw, reed and alfalfa. Stand error of prediction (SEP) in calibration equation for alfalfa, reed and rice straw were 0.97, 0.97 and 0.99 also 0.54, 0.86 and 1.26%. Multiple correlation coefficient ($R^2$) for alfalfa, reed and rice straw were 0.99, 0.97 and 0.99. SEP in the same samples were 1.88, 2.15 and 1.49, respectively.

A Novel Glycosyl Hydrolase Family 16 β-Agarase from the Agar-Utilizing Marine Bacterium Gilvimarinus agarilyticus JEA5: the First Molecular and Biochemical Characterization of Agarase in Genus Gilvimarinus

  • The agarase gene gaa16a was identified from a draft genome sequence of Gilvimarinus agarilyticus JEA5, an agar-utilizing marine bacterium. Recently, three agarase-producing bacteria, G. chinensis, G. polysaccharolyticus, and G. agarilyticus, in the genus Gilvimarinus were reported. However, there have been no reports of the molecular characteristics and biochemical properties of these agarases. In this study, we analyzed the molecular characteristics and biochemical properties of agarases in Gilvimarinus. Gaa16A comprised a 1,323-bp open reading frame encoding 441 amino acids. The predicted molecular mass and isoelectric point were 49 kDa and 4.9, respectively. The amino acid sequence of Gaa16A showed features typical of glycosyl hydrolase family 16 (GH16) ${\beta}$-agarases, including a GH16 domain, carbohydrate-binding region (RICIN domain), and signal peptide. Recombinant Gaa16A (excluding the signal peptide and carbohydrate-binding region, rGaa16A) was expressed as a fused protein with maltose-binding protein at its N-terminus in Escherichia coli. rGaa16A had maximum activity at $55^{\circ}C$ and pH 7.0 and 103 U/mg of specific activity in the presence of 2.5 mM $CaCl_2$. The enzyme hydrolyzed agarose to yield neoagarotetraose as the main product. This enzyme may be useful for industrial production of functional neoagaro-oligosaccharides.

Apoptosis-Induced Cell Death due to Oleanolic Acid in HaCaT Keratinocyte Cells -a Proof-of-Principle Approach for Chemopreventive Drug Development

  • Oleanolic acid (OA) is a naturally occurring triterpenoid in food materials and is a component of the leaves and roots of Olea europaea, Viscum album L., Aralia chinensis L. and more than 120 other plant species. There are several reports validating its antitumor activity against different cancer cells apart from its hepatoprotective activity. However, antitumor activity against skin cancer has not beed studied well thus far. Hence the present study of effects of OA against HaCaT (immortalized keratinocyte) cells - a cell-based epithelial model system for toxicity/ethnopharmacology-based studies - was conducted. Radical scavenging activity ($DPPH{\cdot}$) and FRAP were determined spectrophotometrically. Proliferation was assessed by XTT assay at 24, 48 and 72 hrs with exposure to various concentrations (12.5-200 ${\mu}M$) of OA. Apoptotic induction potential of OA was demonstrated using a cellular DNA fragmentation ELISA method. Morphological studies were also carried out to elucidate its antitumor potential. The results revealed that OA induces apoptosis by altering cellular morphology as well as DNA integrity in HaCaT cells in a dose-dependent manner, with comparatively low cytotoxicity. The moderate toxicity observed in HaCaT cells, with induction of apoptosis, possibly suggests greater involvement of programmed-cell death-mediated mechanisms. We conclude that OA has relatively low toxicity and has the potential to induce apoptosis in HaCaT cells and hence provides a substantial and sound scientific basis for further validation studies.

Dual effects of a mixture of grape pomace (Campbell Early) and Omija fruit ethanol extracts on lipid metabolism and the antioxidant defense system in diet-induced obese mice

  • BACKGROUND/OBJECTIVES: We investigated the effects of a combination of grape pomace (Vitis labrusca, Campbell Early) and Omija fruit (Schizandra chinensis, Baillon) ethanol extracts on lipid metabolism and antioxidant defense system in diet-induced obese mice. MATERIALS/METHODS: Forty male C57BL/6J mice were divided into four groups and fed high-fat diet (control group, CON) or high-fat diet added 0.5% grape pomace extract (GPE), 0.05% Omija fruit extract (OFE) or 0.5% GPE plus 0.05% OFE (GPE+OFE) for 12 weeks. RESULTS: In contrast to the GPE- or OFE-supplemented groups, the GPE+OFE group showed significantly lower body weight and white adipose tissue weights than the CON group. Moreover, GPE+OFE supplementation significantly decreased plasma total cholesterol and increased the plasma HDL-cholesterol/total-cholesterol ratio (HTR) compared to the control diet. The hepatic triglyceride level was significantly lower in the GPE+OFE and GPE groups by increasing ${\beta}$-oxidation and decreasing lipogenic enzyme compared to the CON group. Furthermore, GPE+OFE supplementation significantly increased antioxidant enzyme activities with a simultaneous decrease in liver $H_2O_2$ content compared to the control diet. CONCLUSIONS: Together our results suggest that supplementation with the GPE+OFE mixture may be more effective in improving adiposity, lipid metabolism and oxidative stress in high-fat diet-fed mice than those with GPE and OFE alone.

지질 과산화에 대한 전통약용 식물의 항산화 효과 (Antioxidative effects of traditional medicinal plants on lipid peroxidation)

  • To assess the antioxidative activity of 12 medicinal plants on lipid peroxidation, twelves traditional medicinal plants extracted with 95% methanol were investigated the antioxidative activity using DPPH, thiocyanate acid method, and thiobarbituric acid (TBA) methods. Out of 12 medicinal plants extracted with methanol, the extraction yields of Sedum kamtschaticum was the highest values (49.46%) among them and Geranicum sibiricum, Saururus chinensis root (R), Agrimonia pilosa leaf (L), Agrimonia pilosa root was the lowest value (9.97%). Radical scavenging effect of the selected traditional medicinal plants extracted from different extract solution were examined by 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical method. Antioxidative activity of methanolic extracts was higher than those of ethanol and n-hexane extracts. Scavenging effects in Sedum kamtaschaticum (R) determined by DPPH radical showed the highest among the 12 plants. The antioxidative effects of the first four medicinal plants were similar to those of butylated hydroxyanisole (BHA), and butylated hydroxytoluene (BHT), but higher than that of tocopherol, which was used as a handled control. Antioxidative effects of each indicated concentration of the methanolic extracts on linoleic acid by thiocyanate method was the highest in Sedum kamtschaticum and followed by Geum japonicum and Agrimonia pilosa and their antioxidative effect were similar to those of BHA, and BHT, but higher than that of tocopherol. Antioxidative effects of the selected medicinal methanolic extract on linoleic acid by thiocyanate acid method were examined for 15 days. Peroxidation of control and tocopherol group occurred on days 5 and 9, respectively, but BHA, BHT, selected medicinal methanolic extract group did not occur until on day 15. Antioxidative effects of the selected medicinal methanolic extract on linoleic acid by TBA method were examined for 15 days. Antioxidative activity was similar to those obtained by thiocyanate acid method.

시네라리아 꽃으로부터 에탄올 생산성 및 내열성이 우수한 효모 Hanseniaspora opuntiae 균주 분리 (Isolation of Ethanol-producing Thermotolerant Yeast Hanseniaspora opuntiae from Senecio cruentus)

  • 다양한 꽃으로부터 분리된 160점의 효모 균주 중 시네라리아 꽃에서 분리된 MBY/L6793은 0.48 ± 0.00 g ethanol/g glucose 으로 분리된 균주 중 가장 우수한 에탄올 생산 수율을 나타냈으며, 과꽃으로부터 분리된 MBY/L6986 균주의 약 1.5배 수준이었다. MBY/L6793 균주의 18s rRNA 유전자의 염기서열을 분석한 결과 Hanseniaspora opuntiae로 동정되었으며, 분석된 염기서열은 GenBank (MN859968)에 등록하였다. 40℃에서 배양하였을 때, H. opuntiae MBY/L6793 균주는 20 g의 포도당으로부터 3.82 ± 0.98 g의 에탄올을 생산하였으며, 60g의 포도당으로부터 10.05 ± 0.06 g의 에탄올을 생산하여 대조구 균주인 H. opuntiae KCCM50747 균주의 약 2.45배와 5.74배 수준이었다. H. opuntiae MBY/L6793 균주는 한국생물자원센터에 KCTC37025로 기탁하였다.

더덕에서 약용식물을 이용한 당근뿌리혹선충 증식 억제 효과 (Some Medicinal Plants Suppressed Reproduction of Meloidogyne hapla on Codonopsis lanceolata Trautv)

  • 약용식물 25종 중 지모, 창포, 쇠무릎, 짚신나물, 패랭이, 큰뱀무, 약모밀, 원추천인국, 아주까리, 현삼, 흑참깨, 기린초, 오이풀 등 13종의 식물체 추출액 5배와 10배의 희석농도에서 살선충 효과가 있는 것으로 나타났다. 더덕을 Pot에 파종하여 13종의 식물 추출액 5배액의 살선충 효과를 평가한 결과 짚신나물을 제외한 12종의 식물체 추출액에서 살선충 효과가 있었고, 지모, 쇠무릎, 창포, 패랭이, 원추천인국, 아주까리 등 6종의 추출액은 80% 이상의 억제효과를 나타내었다. 선발된 약용식물을 재배했었던 포장에 더덕을 파종하고 당근뿌리혹선충의 감염정도를 조사한 결과, 대조구에 비해 짚신나물을 제외한 지모 등 12종의 식물 포장에서 뿌리혹 및 토양 내 유충밀도가 적게 발생하였으며, 80% 이상의 방제가를 나타내었다. 선발된 약용식물을 퇴비처럼 처리한 포장에 더덕을 파종하였을 때는, 뿌리혹수 및 난낭수, 토양 내 유충밀도가 무처리에 비해서 흑참깨 처리를 제외하고는 적게 발생하였으나, 방제효과는 선발된 약용식물을 재배했던 포장에 비해 크게 떨어졌으며, 쇠무릎과 창포 처리의 방제가만 70% 이상으로 다른 식물에 비해 좋았다.

계방산의 고도와 사면방위별 하층식생구조 및 지표종 분석 (Understory Vegetation Structure by Altitude and Azimuth Slope and Indicator Species Analysis in Mt. Gyebang)

  • 본 연구는 유전자원보호구역 및 국립공원으로 지정된 계방산의 하층식생구조와 고도 및 사면별 지표종을 구명하기 위해 실시하였다. 조사지 출현식물은 80과 203속 303종 38변종 5품종 4아종 총 350분류군으로 확인되었다. 중요치 분석결과, 관목층에서는 미역줄나무(9.143%), 당단풍나무(7.594%), 노린재나무(6.347%) 등이 우점 하였고, 초본층에서는 조릿대(8.653%), 오리방풀(2.936%), 대사초(2.897%) 등이 우점 하는 것으로 나타났다. NMS 분석결과, 주요종의 분포범위는 고도별 영향이 있는 것으로 밝혀졌다(관목층: $R^2$ >0.3, 초본층: $R^2$ >0.6). Plexus diagram 분석결과, 당단풍나무는 함박꽃나무, 청시닥나무, 참회나무 등과 연관성이 있으며, 초본층의 벌깨덩굴은 투구꽃, 박새, 귀룽나무 등과 연관성이 있는 것으로 분석되었다. 고도에 따른 유의성 있는 지표종은 60종, 사면방위별 유의성 있는 지표종은 30종이 분석되었다. MRPP-test 결과, 고도에 따른 집단 간 종조성은 이질적이며, 사면방위에 따른 종조성은 NE와 SW 간에 이질성이 큰 것으로 분석되었다.

꽃차용 꽃 추출물의 구강위생균에 대한 항균특성 (Antibacterial Activities of Flower Tea Extracts against Oral Bacteria)

  • In this study, we analyzed flower tea activity against oral bacteria. Lagerstroemia indica, Paeonia suffruticosa and Hemerocallis fulva showed high extract yields. Bellis perennis, Punica granatum and Cercis chinensis showed the high rates of yield by ethanol extraction. Extract yield seemed to be related to the characteristics of the specimens rather than to the solvent. Streptococcus mutans, Streptococcus obrinus, Porphyromonas gingivalis and Prevotella intermedia were used to investigate extracts activity against bacteria; the former two cause dental caries and the latter two cause halitosis. Cornus officinalis, L. indica, P. granatum and P.s uffruticosa showed high antibacterial activities against S. mutans. In specimens extracted with ethanol, P. suffruticosa, Camellia sinensis, Camellia japonica L. and Rosa hybrida showed high antibacterial activities. L. indica, P. granatum and C. officinalis showed high antibacterial activities against S. sobrinus. C. officinalis, P. granatum, L. indica and P. suffruticosa showed high activities for specimens extracted with ethanol. The results show that the warm extracts of C. officinalis, L. indica and P. granatum may be effective to prevent dental caries. In particular, the ethanol-based extracts of C. officinalis, P. suffruticosa and C. sinensis were effective to prevent dental caries and thus may be highly marketable. Chrysanthemum zawadskii, R. hybrida, P. granatum, C. japonica L. and Zinnia elegans showed high antibacterial activity against P.gingivalis. R. hybrida showed the highest ethanol extract activity, followed by P. suffruticosa, P. granatum, C. japonica L. and L. indica. R. hybrida, P. granatum, C. morifolium and C. japonica showed high activity against Pr.intermedia in the order named. C. zawadskii, P. granatum, L. indica, C. japonica and A. princeps showed high ethanol extract activity. Thus, the warm extracts of R. hybrida, P. granatum and C. japonica may be helpful to reduce halitosis. In addition, the ethanol-based extracts of P. granatum, C. japonica and L. indica are expected to be highly marketable as mouthwashes.
