Autodisplay of a multimeric protein complex on a cell surface is limited by intrinsic factors such as the types and orientations of anchor modules. Moreover, improper folding of proteins to be displayed often hinders functional cell surface display. While overcoming these drawbacks, we ultimately extended the applicability of the autodisplay platform to the display of a protein complex. We designed and constructed a cell surface attachment (CSA) system that uses a non-covalent protein-protein interaction. We employed the high-affinity interaction mediated by an orthogonal cohesin-dockerin (Coh-Doc) pair from Archaeoglobus fulgidus to build the CSA system. Then, we validated the orthogonal Coh-Doc binding by attaching a monomeric red fluorescent protein to the cell surface. In addition, we evaluated the functional anchoring of proteins fused with the Doc module to the autodisplayed Coh module on the surface of Escherichia coli. The designed CSA system was applied to create a functional attachment of dimeric α-neoagarobiose hydrolase to the surface of E. coli cells.
During the monolayer assembly of zeolite microcrystals using sonication with stacking (SS) method, the factors that govern the degree of close packing (DCP) between the microcrystals, the rate of attachment (rA) of microcrystals onto the substrate, the degree of coverage (DOC), and the binding strength (BS) between each crystal and the substrate were investigated for each mode of binding linkage (MBL). The tested MBLs were covalent linkage (CL), ionic linkage (IL), and polyelectrolyte-mediated ionic linkage (p-IL). Unlike the monolayers of zeolite crystals assembled on glass with a covalent linkage, the strong BS, very high DOC, and very high DCP do not decrease during monolayer assembly on glass through ionic linkages. This results indicate that the surface migration of crystals undergo linkage-nondestructively when crystals were attached to the substrates through ionic linkages.
Absorption and accumulation of heavy metals in plants were determined by phytoavailable contents rather than total contents of heavy metals. Therefore, phytoavailability-based management protocol should be prepared for safe food crop production in contaminated agricultural lands. This study was conducted to understand the distribution and phytoavailability of heavy metal in the Korean agricultural soils affected by abandoned mining sites along with investigation of soil properties (soil pH, OM, DOC, clay content, Al/Fe/Mn content) influencing on the metal phytoavailability. For this, 142 agricultural soils located nearby 39 abandoned mining sites distributed in five province in Korea, were analyzed. Among the four different heavy metals, cadmium (Cd) and zinc (Zn) appeared to exist in more phytoavailable form than cupper (Cu) and lead (Pb). Soil pH was the main factor governing phytoavailable Cd, Pb, and Zn showing positive relationship with partitioning coefficients of the corresponding metals; Cd (r = 0.66, P < 0.001), Pb (r = 0.70, P < 0.001), and Zn (r = 0.62, P < 0.001). This implied higher phytoavailability of the corresponding metals with higher soil pH. In contrast, phytoavailability of Cu (r = 0.41, p < 0.01) was only negatively related with soil DOC (dissolved organic carbon).

A microcosmal experiment using a metagenomic technique was designed to assess the effect of BTEX (benzene, toluene, ethylbenzene, and xylenes) on an indigenous bacterial community in a Daejeon forest soil. A compositional shift of bacterial groups in an artificial BTEX-contaminated soil was examined by the 16S rDNA PCR-DGGE method. Phylogenetic analysis of 16S rDNAs in the dominant DGGE bands showed that the number of Actinobacteria and Bacillus populations increased. To confirm these observations, we performed PCR to amplify the 23S rDNA and 16S rDNA against the sample metagenome using Actinobacteria-targeting and Bacilli-specific primer sets, respectively. The result further confirmed that a bacterial community containing Actinobacteria and Bacillus was affected by BTEX.
Characteristics of organic matters (OM) in wastewater and the removal efficiencies were investigated using the influent and the effluent samples collected from 21 wastewater treatment plants. The OM characteristics investigated included biodegradability, humic content, specific UV absorbance (SUVA), the distribution percentage of refractory OM (R-OM), and synchronous fluorescence spectra. The types of wastewater (sewage, livestock waste/night soils, industrial waste) were easily distinguished by comparing the synchronous fluorescence spectra of the influent wastewater. The prominent peak of protein-like fluorescence (PLF) was observed for livestock waste/night soils whereas sewage exhibited a unique fluorescence peak at a wavelength of 370 nm. Irrespective of the wastewater types, the distribution percentage of R-OM increased from the influent to the effluent. Livestock waste/night soils showed the highest removal efficiency among all the three types of wastewater. There was no statistical difference of the removal efficiency between a traditional activated sludge and biological advanced treatment processes. Removal efficiency based on dissolved organic carbon DOC presented good correlations with the distribution percentage of R-OM and fulvic-like fluorescence (FLF) of the influent. The prediction for DOC removal efficiency was improved by using multiple regression analyses based on some selected OM characteristics and mixed liquid suspended solid (MLSS).

In this study, pretreatment of organic matters with $MIEX^{(R)}$ was evaluated using bench-scale experimental procedures on four organic matters to determine its effect on subsequent UF membrane filtration. For comparison, coagulation process was also used as a pretreatment of UF membrane filtration. Moreover, the membrane fouling potential was identified using different fractions and molecular weights of organic matters. From the removal property of MW organic matters by coagulation process for the sample water NOM and AOM, the removal efficiency of high MW organic matters were much higher than those of low MW organic matters. It was shown that the removal efficiency of high MW organic matter more than 10 kDa was lower than that of low MW organic matter for $MIEX^{(R)}$ process. For the change of permeate flux by the pretreatment process, $MIEX^{(R)}$+UF process showed high removal efficiency of organic matter as compared with coagulation-UF processes, but high reduction rate of permeate flux was presented through the reduction of removal efficiency of high MW organic matter. From sequential filtration test results in order to examine the effect of MW of organic matter on membrane fouling, it was found that the membrane foulant was occurred by high MW organic matter, and the DOC of organic matter less than 0.5 mg/L was working as the membrane foulant. In the case of sample water composed of low MW organic matter less than 10 kDa, since the low MW organic matter less than 10 kDa has high removal efficiency by $MIEX^{(R)}$, low reduction rate of permeate flux is obtained as compared with coagulation-UF processes. In summary, it is required to conduct the research on physical/chemical characteristic of original water before pretreatment process of membrane process is selected, and a pertinent pre-treatment process should be employed based on the physical/chemical characteristic of original water.
The samples for organic carbon analysis were collected between $5^{\circ}\;and\;17^{\circ}N$ along $131.5^{\circ}W$ in the northeast Pacific KODOS (Korea Deep Ocean Study) area. The mean concentration of total organic carbon (TOC) in the surface mixed layer $({\sim}50 m)$ was $100.13{\pm}2.05{\mu}M-C$, while the mean concentration of TOC in the lower 500m of the water column was $50.19{\pm}4.23{\mu}M-C$. A strong linear regression between TOC and temperature $(r^2=0.70)$ showed that TOC distribution was controlled by physical process. Results from the linear regression between chlorophyll-a and TOC, and between chlorophyll-a and particulate organic carbon (POC), decreasing of dissolved organic carbon (DOC) in the surface layer caused by non-biological photo-oxidation process. Below the surface layer, biological production and consumption occurred. DOC accumulation dominated in the depth range of $30{\sim}50m$ and DOC consumption occurred in the depth range of $50{\sim}200m$. TOC was inversely correlated with apparent oxygen utilization (AOU) and TOC/AOU molar ratios ranged from -0.077 to -0.21. These ratios indicated that TOC oxidation was responsible fur $10.9{\sim}30.1%$ (mean 20.2%) of oxygen consumption in the NE Pacific KODOS area. In the euphotic zone, distributions of dissolved and particulate organic matter were controlled by photo-chemical, chemical, biological and physical processes.
As the total organic carbon (TOC) becomes a new water quality standard as an organic matter (OM) index for public sewage treatment facilities (PSTFs) in Korea from 2021, a comparison study needs to be conducted by examining the correlation between TOC and the existing OM indices (DOC, BOD5, CODMn, CODCr). 500 PSTFs were categorized by process configuration and capacity, and correlation between OM indices in influent and effluent was analyzed. The CODMn/TOC showed higher correlation than other OM indices. This results can be used to basic data for various research associated with TOC.

In this study, we investigated the characteristics of the conversion efficiency and the effect of ageing Diesel Oxidation Catalyst (DOC). The DOC was composed of Warm-up Catalytic Converter (WCC) and Underbody Catalytic Converter (UCC). As the result, the conversion efficiency of THC was 10$\~$50$\%$ on WCC and 30$\~$40$\%$ on UCC .The conversion efficiency of CO was 80$\~$90$\%$ on WCC and remained 10$\~$20$\%$ of CO was purified on UCC. The WCC shows high conversion efficiency on CO. After 20 hours aging process of engine bench, conversion efficiencies of THC and CO were improved a little, because it was activated catalyst surface by 20 hours aging. In case of 80 hours aging, the conversion efficiencies of THC and CO were decreased on WCC. However, the UCC was not affected by aging process .
