Effect of Local Softening for Spring-back Reduction of Ultra High Strength Steel on Microstructure and Mechanical Properties (스프링백 저감을 위한 초고강도강의 국부적 연화 열처리에 따른 미세조직과 기계적 특성 변화에 관한 연구)

  • In order to improve excessive spring-back behavior as a result of the roll forming process using ultra high strength steel (UHSS) sheet, local softening in region of a partial area expected to be deformed on an initial blank is considered in this study. With SPFC1470 UHSS sheet with initial blank thickness of 1.20mm, the local softening is performed with the following conditions: temperatures of 500℃, 550℃, 600℃ and 650℃, and holding time of 20s, 40s, 80s and 160s. Mechanical properties, such as yield stress and tensile strength, as well as elongation, are evaluated through uniaxial tensile tests, while the microstructural characteristics as a result of local softening are also investigated using the heat-treated specimens. As a result, it is shown that the spring-back behavior of the roll-formed prototype was reduced about by 78.9%, when the local softening at about 500℃ was performed for 160s considering the practical manufacturing condition.

  • Water-soluble crude polysaccharide (TM-1) prepared from the leaves of Teucrium viscidum var. miquelianum was fractionated into three polysaccharide fractions, TM-2, TM-3 and TM-4 by the addition of cetyltrimethylammonium bromide. The major polysaccharide fraction, TM-2, consisted of glucose, galactose, rhamnose, mannose, glucuronic acid and galacturonic acid, and all fractions contained galactose and glutose as the major neutral sugars. TM-4 showed the highest anti-complementary activity. When TM-4 was futher fractionated by anion exchange chromatography, TM4- II a and TM4-II b showed the most Potent anti-complementary activity. TM4- II a was composed mainly of galactose, arabinose and glucose in the molar ratios of 2.13 : 0.94 : 1.00 respectively, and contained a small amount of galacturonic acid and glucuronic acid.

Antimicrobial Effects of Retort and Gamma Irradiation on Bacterial Populations in Spicy Chicken Sauce (레토르트 및 감마선 조사에 의한 화닭 덮밥 소스의 미생물 제어 효과 비교)

  • This study evaluated the antimicrobial effects of retort process and gamma irradiation on reduction of total bacterial populations in spicy chicken sauce, which is served on top of the steamed rice. Commercial spicy chicken sauce was treated with retort and gamma ray at 0, 1, 3, 5, and 10 kGy. Total aerobic bacterial populations were then enumerated on plate count agar and isolated bacteria from the test samples were identified using PCR analysis. Moreover, gamma ray sensitivity of identified bacteria was evaluated by $D_{10}$ values, and genotoxicity of gamma-irradiated samples was examined. Gamma irradiation at 3 kGy reduced total aerobic bacterial cell counts in spicy chicken sauce below detection limit, but total aerobic bacterial cell counts in test samples treated with retort were 2.1 log CFU/g. Identified bacteria from the samples were Bacillus subtilis, B. amyloiquefaciense, and B. pumils, and the $D_{10}$ values for B. subtilis and B. cereus were 0.39 ($R^2\;=\;0.921$) and 0.28 log CFU/g ($R^2\;=\;0.904$), respectively. The SOS chromotest showed that the gamma-irradiated spicy chicken sauce did not cause mutagenicity. These results indicate that gamma irradiation of spicy chicken sauce could be useful in ensuring microbial safety.

Determination of Color Value (L, a, b) in Green Tea Using Near-Infrared Reflectance Spectroscopy (근적외 분광분석법을 이용한 녹차의 색도 분석)

  • Near infrared spectroscopy (NIRS) is a rapid and accurate analytical method for determining the composition of agricultural products and feeds. The applicability of near infrared reflectance spectroscopic method was tested to determine the color value (L, a, b) of green tea. A total of 162 green tea calibration samples and 82 validation samples were used for NIRS equation development and validation, respectively. In the developed NIRS equation for analysis of the color value (L, a, b), the most accurate equation for L value was obtained at 2, 8, 6, 1 (2nd derivative, 8 nm gap, 6 points smoothing, and 1pointsecond smoothing), and for a, and b value were obtained at 1, 4, 4, 1 (1st derivative, 4 nm gap, 4points smoothing, and 1 point second smoothing) math treatment condition with SNVD (Standard Normal Variate and Detrend) scatter correction method and entire spectrum ($400{\sim}2,500\;nm$) by using MPLS (Modified Partial Least Squares) regression. Validation results of these NIRS equations showed very low bias (L: 0.005%, a: 0.003%, b: -0.013%) and standard error of prediction (SEP, L: 0.361%, a: 0.141%, b: 0.306%) as well as high coefficient of determination ($R^2$, L: 0.905, a: 0.986, b: 0.931). Therefore, these NIRS equations can be applicable and reliable for determination of color value (L, a, b) of green tea, and NIRS method could be used as a mass screening technique for breeding programs and quality control in the green tea industry.

  • Finite element method is very effective method to simulate the forming processes with good prediction of the deformation behaviour. For the finite element modeling of sheet mental forming the accurate tool model is required. Due to the geometrical complexity of real-size part stamping tools it is hard to make FE model for real-size auto-body stamping parts. In this paper, it was focussed on the drawability factors on auto-body panel stamping by AUTOFORM with using tool planing alloy to reduce law price as well as high precision from Design Optimization of die. According to this study, the results of simulation will give engineers good information to access the Design Optimization of die.

Estimation of Standard Length-Weight Relationships of 10 Freshwater Fish in the South Korea for Application of Relative Weight Index (한국 담수어류 10종의 상대 무게지수(Relative weight index) 활용을 위한 표준 길이-무게 상관관계(Standard Length-Weight relationships) 산출)

  • The Relative weight index is an index for evaluating nutritional condition of fish by calculating a ratio of observed weight to standard weight. The purpose of this study is estimating standard Length-Weight relationships (standard LWRs) to calculate standard weight for application of relative weight index and analyze frequency distribution of relative weight index of 10 freshwater fish in the South Korea. The standard LWRs for each species was calculated by the Length-Weight data sets from 798 times of captures at 141 sites. The correlation of determination (r2) for all species was over 0.9 and values of parameter b were within the expected range according to Froese (2006). We calculated in increments of 10 from 10 to 90 percentile of relative weight index for each species, and it could be used as a standard for quantitative assessment of nutritional condition of freshwater fish.

Screening of Tyrosinase Inhibiting Activity from the Marine-Derived Fungus (해양균류의 Tyrosinase 저해활성 검색)

  • In order to screen new tyrosinase inhibiting principle which is expected to be a new biofunctional skin whitening cosmetics, we have isolated 600 strains of the marine- derived fungi and investigated tyrosinase inhibiting activity for their acetone extracts. The significant activities (>70% Inhibition) were observed in the extract of 10 strains of fungi (MFA7, MFA27, MFA58, MFA317, MFA318, MFA345, MFA412, MFA552, MFA562, MFA581). These active strains were cultured in SWS medium with 1 L scale and the resulting broth and mycelium were extracted to afford mycelium extract (000M) and broth extract (000B), respectively. Tyrosinase inhibiting activity for all extracts has been tested. As the results, the broth extracts of 4 strains (27B, 58B, 552B and 581B) exhibited relatively high levels of activity of $IC_{50}$ values of $3.0-19.0\;{\mu}g/mL$. The active component of 581B was purified by assay-guided isolation to yield the known kojic acid (1), and its structure was determined by physicochemical evidence. Kojic acid showed the significant tyrosinase inhibitory activity with $IC_{50}$ values of $12.0\;{\mu}M$.

Characterization of Quorum-Sensing Signaling Molecules Produced by Burkholderia cepacia G4

  • In many Gram-negative bacteria, autoinducers, such as N-acyl-L-homoserine lactone(acyl-HSL) and its derivative molecules, mediate the cell-density-dependnet expression of certain operons. The current study identified the autoinducers produced by Burkholderia cepacia G4, a trichloroethylene-degrading lagoon isolate, using TLC bioassays with Agrobacterium tumefaciens NT1(pDCI141E33) and Chromobacterium violaceum CVO26, and a GC-MS analysis. The ${R_f}\;and\;{R_t}$ values and mass spectra were compared with those of synthetic compounds. Based on the analyses, it was confirmed that G4 produces N-hexanoyl (C6)-, N-octanoyl (C8)-, N-decanoyl (C10)-, N-dodecanoyl (C12)-HSL, and an unknown active species. The integration of the GC peak areas exhibited a ratio of C8-HSL:C10-HSL:C12-HSL at 3:17:1 with C6-HSL and C10-HSL production at trace and micromolar levels, respectively, in the culture supernatants. Nutants partially defective in producing acyl-HSLs were also partially defective in the biosynthesis of an antibiotic substance. These results indicate that the autoinducer-dependent gene regulation in G4 is dissimilar to the clinical B. cepacia strains isolated from patients with cystic fibrosis.

Role of Bevel Angles Influenced on the Fatigue Life of Butt-welded Joints (맞대기 이음 용접의 피로수명에 베벨 각도가 미치는 역할)

  • This study aims to investigate the influence of bevel angles on the fatigue life of V-groove butt-welded joints with back-plates made by SM490A steel material, generally used for excavators, because changes in the geometry, material and surface properties of welded regions affect the fatigue life of welded structures. Butt type test specimens were prepared by the $CO_2$ welding of rolled steel plates (SM50A steel) with a thickness of 13.5 mm at a welding speed of 30 cm/min and these Butt type test specimens had two different groove angles, which are $40^{\circ}$ (A type) and $30^{\circ}$ (B type). In order to investigate differences in fatigue life between two types, 4-point bending fatigue tests were conducted with a stress ratio of R=0.1 under the cyclic loading environment at a frequency of 5 Hz at room temperature. The fatigue life of A type specimens was approximately 7% higher than that of B type specimens. The stress concentration factors calculated by finite element analysis were 2.16 for A type and 2.25 for B type, whose difference was caused by the influence of the back-plates of butt-welded structures. The current results could provide important guidelines to determine the V-groove angle of butt-welded joints with a satisfactory fatigue life, although under severe operating conditions.

Novel Diagnostic Algorithm Using tuf Gene Amplification and Restriction Fragment Length Polymorphism is Promising Tool for Identification of Nontuberculous Mycobacteria

  • Nontuberculous mycobacteria (NTM) are a major cause of opportunistic infections in immunocompromised patients, making the reliable and rapid identification of NTM to the species level very important for the treatment of such patients. Therefore, this study evaluated the usefulness of the novel target genes tuf and tmRNA for the identification of NTM to the species level, using a PCRrestriction fragment length polymorphism analysis (PRA). A total of 44 reference strains and 17 clinical isolates of the genus Mycobacterium were used. The 741 bp or 744 bp tuf genes were amplified, restricted with two restriction enzymes (HaeIII/MboI), and sequenced. The tuf gene-PRA patterns were compared with those for the tmRNA (AvaII), hsp65 (HaeIII/HphI), rpoB (MspI/HaeIII), and 16S rRNA (HaeIII) genes. For the reference strains, the tuf gene-PRA yielded 43 HaeIII patterns, of which 35 (81.4%) showed unique patterns on the species level, whereas the tmRNA, hsp65, rpoB, and 16S rRNA-PRAs only showed 10 (23.3%), 32 (74.4%), 19 (44.2%), and 3 (7%) unique patterns after single digestion, respectively. The tuf gene-PRA produced a clear distinction between closely related NTM species, such as M. abscessus (557-84-58) and M. chelonae (477-84-80-58), and M. kansasii (141-136-80-63-58-54-51) and M. gastri (141-136-117-80-58-51). No difference was observed between the tuf-PRA patterns for the reference strains and clinical isolates. Thus, a diagnostic algorithm using a tuf gene-targeting PRA is a promising tool with more advantages than the previously used hsp65, rpoB, and 16S rRNA genes for the identification of NTM to the species level.
