  • A sequential design of experiments was used to optimize MOF-801 synthesis process. For the initial screening, a general 2k factorial design was selected followed by the central composition design, one of the response surface methods. A 23 factorial design based on the molar ratio of fumaric acid, dimethylformamide (DMF), and formic acid was performed to select the more suitable response variable for the design of experimental method among the crystallinity and BET specific surface area of MOF-801. After performing 8 synthesis experiments designed by MINITAB 19 software, the characteristic analysis was performed using XRD analysis and nitrogen adsorption method. The crystallinity with R2 = 0.999 was found to be more suitable for the experimental method than that of BET specific surface area. Based on analysis of variance (ANOVA), it was confirmed that the molar ratio of fumaric acid and formic acid was a major factor in determining the crystallinity of MOF-801. Through the response optimization and contour plot of two factors, the optimal molar ratio of ZrOCl2·8H2O : fumaric acid : DMF : formic acid was 1 : 1 : 39 : 35. In order to optimize the synthesis process, the central composition design on synthesis time and temperature was performed under the identical molar ratio of precursors. The results derived through the designed 9 synthesis experiments were calculated using the quadratic model equation. Thus, the maximum crystallinity of MOF-801 predicted under the synthesis time and temperature of 7.8 h and 123 ℃, respectively.

  • This study examines the effect of collector well installed to reduce groundwater level in the regions with the occurrence of landcreep, a soil mass movement triggered by instability on slopes. Slopes are prone to failure as a result of instability caused by its internal, topographic and geological properties as well as due to external factors such as rainfall and earthquake. In Korea during the rain season, rainfall infiltration affects the groundwater level in soil, building up porewater pressure and load, and finally drives slopes to collapse. Slope failure caused by rainfall infiltration has been leading to a drastic forest degradation. The studied slope is located adjacent to a valley, its terrain corresponds to piedmont gentle slope, while the upper part of the failure surface is steep. After reinforcing the terrain where landcreep had occurred and installing collector well on the slope, we measured the changes in the groundwater level. In order to analyze the relationship between the well and the slope, we calculated the ratio of groundwater level to rainfall before and after the installation of the collector well. As a result, it is confirmed that the ratio increases after the installation of the well, which in turn reduces the groundwater level. Analysis of the change in groundwater level after 3, 7, 15 days antecedent rainfall showed that the higher the overall groundwater level, the less the value ($r_p$) of groundwater level-rainfall ratio is, while the value becomes relatively greater when the groundwater level is low. In particular, if a slope has a large catchment basin as is in the case of the studied site, antecedent rainfall affects groundwater level in the order of 3 < 7 < 15 days.

  • The use of 16S rDNA is commonplace in the determination of prokaryotic species. However, it has limitations, and there are few studies at the genus level. We investigated conserved genes and metabolic pathways at the genus level in 28 strains of 13 genera of prokaryotes using the COG database (conserved genes) and MetaCyc database (metabolic pathways). Conserved genes compared to total genes (core genome) at the genus level ranged from 27.62%(Nostoc genus) to 71.76%(Spiribacter genus), with an average of 46.72%. The lower ratio of core genome meant the higher ratio of peculiar genes of a prokaryote, namely specific biological activities or the habitat may be varied. The ratio of common metabolic pathways at the genus level was higher than the ratio of core genomes, from 58.79% (Clostridium genus) to 96.31%(Mycoplasma genus), with an average of 75.86%. When compared among other genera, members of the same genus were positioned in the closest nodes to each other. Interestingly, Bacillus and Clostridium genera were positioned in closer nodes than those of the other genera. Archaebacterial genera were grouped together in the ortholog and metabolic pathway nodes in a phylogenetic tree. The genera Granulicella, Nostoc, and Bradyrhizobium of the Acidobacteria, Cyanobacteria, and Proteobacteria phyla, respectively, were grouped in an ortholog content tree. The results of this study can be used for (i) the identification of common genes and metabolic pathways at each phylogenetic level and (ii) the improvement of strains through horizontal gene transfer or site-directed mutagenesis.

  • Six populations of the diamondback moth, Plutella xylostella, were collected from the different national areas for resistance and reared in laboratory for two sensitive population. These populations of P. xylostella were examined the developed resistance against commercial products of Bacillus thuringiensis. There were 3 products with B. thuringiensis subsp. kurstaki including Tyuneup$^{(R)}$, Thuricide$^{(R)}$ and Geumulmang$^{(R)}$ and 2 products with B. thuringiensis subsp. aizawai including Tobagi$^{(R)}$ and Scorpion$^{(R)}$. The sensitive population of diamondback moths were provided from National Academy of Agricultural Science (NP) and Highland Agriculture Research Center (GR population) and field populations were caught from 6 different national areas. Resistance against Tyuneup$^{(R)}$ was developed 4.8 and 2.5 times in SP and HS compared with GR population of diamondback moth, respectively. In case of Geumulmang$^{(R)}$, it was developed 9.9 and 6.8 times in SP and NM population compared with NP population, respectively. Otherwise, Tobagi$^{(R)}$ was showed higher resistance in HS than any other population compared with GR population, however, Scorpion$^{(R)}$ that is a same strain with Tobagi$^{(R)}$, was showed only double resistance to SP population. It was supposed that the development of resistance to B. thuringiensis might be caused by the continuous application of the specific commercial product at the specific area. So, we need to use the commercial products of B. thuringiensis in rotation with different B. thuringiensis strains. In the other hand, when HS population with highest resistance were reared in laboratory, their resistance ratio was rapidly dropped to 1.1 times at second generation. We have to examined the resistance mechanism of the diamondback moth to B. thuringiensis strains.

  • For screening of autoregulator receptor gene from Saccharopolyspora erythraea, PCR was performed with primers of receptor gene designed on the basis of amino acid sequences of autoregulator receptor proteins with known function. PCR products were subcloned into the BamHI site of pUC19 and transformed into the E. coli DH5$\alpha$. The isolated plasmid from transformant contained the fragment of 120 bp, which was detected on 2% gel after BamHI treatment. The insert, 120 bp PCR product, was confirmed as the expected internal segment of gene encoding autoregulator receptor protein by sequencing. Southern and colony hybridization using Saccha. erythraea chromosomal DNA were performed with the insert as probe. The plasmid (pEsg) having 3.2 kbp SacI DNA fragment from Saccha. erythraea is obtained. The 3.2 kbp SacI DNA fragment was sequenced by the dye terminator sequencing. The nucleotide sequence data was analyzed with GENETYX-WIN (ver 3.2) computer program and DNA database. frame analyses of the nucleotide sequence revealed a gene encoding autoregulator receptor protein which is a region including KpnI and SalI sites on 3.2 kbp SacI DNA fragment. The autoregulator receptor protein consisting of 205 amino acid was named EsgR by author. In comparison with known autoregulator receptor proteins, homology of EsgR showed above 30%.

  • The phycocyanin pigment (PC) is a marker for cyanobacterial presence in eutrophic inland water. Accurate estimation of low PC concentration in turbid inland water is challenging due to the optical complexity and criticalforissuing an early warning of potentialrisks of cyanobacterial bloom to the public. To monitor cyanobacterial bloom in eutrophic inland waters, an approach is proposed to partition non-water absorption coefficient from measured reflectance and to retrieve absorption coefficient of PC with the aim of improving the accuracy in remotely estimated PC, in particular for low concentrations. The proposed inversion model retrieves absorption spectra of PC ($a_{pc}({\lambda})$) with $R^2{\geq}0.8$ for $a_{pc}(620)$. The algorithm achieved more accurate Chl-a and PC estimation with $0.71{\leq}R^2{\leq}0.85$, relative root mean square error (rRMSE) ${\leq}39.4%$ and mean relative error(RE) ${\leq}78.0%$ than the widely used semi-empirical algorithm for the same dataset. In particular, low PC ($PC{\leq}50mg/m^3$) and low PC: Chl-a ratio values of for all datasets used in this study were well predicted by the proposed algorithm.

  • Interorder heterokaryons were obtained by polyethylene glycol induced fusion of protoplasts from auxotrophic mutants of Pleurotus ostreatus in agaricales and Ganoderma applanaturm in aphyllophorales. When transferred to MMM plates, all fusion colonies exhibited an extremely growth rate. During three times subcultivation on MCM the growth rate of fusants showed faster little by little. Seventy-five % fusion products of potoplasts showed mixed morphologies between those of P. ostreatus and G. applannatum in the first subcultivation on MCM and MGM. The phenotype of these fusants changed similar those of P. osteatus type after three times subcultivation on MCM. However, phenotype of 25% stable strains did not change on subcultivation. Hyphae of all fusion products did not form true clamp connection. All these types did not produce primordia. A comparrison of interorder somatic hybrids was made using isozyme analysis of esterase, malate dehydrogenase and peroxidase. In most cases the enzyme patterns of G. applanatum were not distinct, however, fusant showed non-parental bands.

  • Objectives: Pulse-Respiration Ratio has been used for estimating subject's Han-Yeol [寒熱] status since it mentioned in suwen [素問]. In practicing Pulse-Respiration Ratio over 5 means the status of Yeol [熱], Pulse-Respiration Ratio below 3 means the status of Han [寒]. We performed this study to examine the Optimum Standard for Measuring Pulse-Respiration Ratio on the Basis of Repeatability and Reproducibility. Methods: After subject's 5 minutes rest we measured subject's ECG, respiration pattern, EEG, EMG simultaneously. In this research examiner's number is two, subject's number is four, and the number of repeat is two. We calculated Pulse-Respiration Ratio through dividing Respiration cycle average by Pulse cycle average according to each standard including time section, $EEG(relative-{\alpha}$ density, $relative-{\beta}$ density, ${\alpha}/{\beta}$ and EMG. We analyzed these data through Gage R&R study using MINITAB 13.20 program and considered the results of below 30 %R&R and over 4 Number of Distinct Categories to have a significance. Results: 1. In the applying of time standard, Pulse-Respiration Ratio from section 3, 4, 6, 8 had a significant meaning in the aspect of Repeatability and Reproducibility. 2. In the applying of $EEG({\alpha}$ I , ${\beta}$ I , ${\alpha}/{\beta})$, EMG(E I) standard, there was no significant results. 3. In the applying of time standard(section 5, 6, 7), $EEG({\alpha}$ I , ${\beta}$ I , ${\alpha}/{\beta})$ and EMG(E I) standard simultaneously, Pulse-Respiration Ratio from ${\alpha}/{\beta}$ in section 6, ${\beta}$ I in section 8 had a significant meaning in the aspect of Repeatability and Reproducibility. Conclusions: We can suggest the Optimum Standard for Measuring Pulse-Respiration Ratio on the basis of Repeatability and Reproducibility as followings; 1. Pulse-Respiration Ratio Measuring time should be at least 15 minutes. 2. Applying of time(section 6, 8) and $EEG({\beta}$ I, ${\alpha}/{\beta})$ standard simultaneously is recommended considering reliability and validity but more study is needed. 3. EMG(E I) may be helpful to detect the segment of physical rest and exclude artifacts but more study is needed.

  • This study was conducted to investigate the prevalence of childhood obesity and to figure out the correlation between percentage of body fat and various criteria used for evaluating childhood obesity, and to suggest a new convenient criterion applicable to obesity assessment for large population. subject of 309(male: 153, female: 156) from Uisung elementary school in Kyungbuk, in the age range from 10 to 12 were recruited. The prevalence of obese was evaluated by the methods of Slaughter skinfold equations for children, BMI, and RI. fourteen(4.5%) by Slaughter eq, 39(12.6%) by BMI, 31 (10%) by RI were found to be obese. The results indicate that the evaluation of child obesity is influenced by the methods applied; that is, BMI and RI tend to overpredict the obese while percentage of body fat tend to underestimate. Percentage of body fat by Slaughter equations using triceps and calf skinfold is highly correlated with triceps plus subscapular skinfold thickness(male: r=0.91, female: r=0.88) and with Kim equations for Korean children(male: r=0.93, femlae: r=0.88). The results suggest that, if necessary, practitioners can adequately predict body composition without exposing the upper torso of females.

  • Food safety management should be based on scientific evidences. FAO and WHO presented risk analysis as one of four principles in food safety management. WTO also admits the self safety regulation only when it is made on the basis of risk assessment. Without scientific analysis, tracing and eliminating the cause of food poisoning is impossible. Research and development plays a key role to produce scientific evidences. The Korean government ran over 40 programs in 11 agencies from 2008 to 2010. However, there is no statistics on food safety R&D at present. In this research, food safety projects conducted from 2008 to 2010 are listed up by means of analysing National Science and Technology Information Service (NTIS). The analytical criteria are the name of programs, national standard classification of science and technology, and keywords. As result, Korea Food and Drug Administration, Ministry for Food, Agriculture, Forestry and Fisheries, and Rural Development Administration play major role in the food safety R&D. The portion of more than one year projects should rise up in order to achieve the data for risk assessment, which is strongly required to improve. Besides, the research should be deeper so as to publish more SCI papers. The R&D portfolio should be changed in direction to raise up the portion of biological hazards such as norovirus. In order to do so, a large number of food safety programs should be emerged. The categories of food safety management and the hygiene/quality management of the agricultural and livestock products in the national standard classification of science and technology should be emerged because they are set up reflecting agencies' interests in spite of few differences between them.
