목재를 분해시키는 담자균류들은 목재 및 목질복합체에 쉽사리 침투하여 복잡한 리그노셀룰로오스 복합체를 분해시킨다. 이러한 분해에는 많은 효소시스템들이 복합적으로 작용하면서 상호 협동하는 것으로 보고되고 있다. 지금까지 일려진 효소들은 통상 3개의 그룹으로 나눌 수 있는데 그 하나는 목재성분을 직접적으로 공격하는 효소균들, 예를 들면 cellulase complex, laccase(LAC), lignin peroxidase(LIP), horse-radish peroxidase(HRP), manganese-independent peroxidase(MIP) 및 protocatechuate 3,4-dioxygenase(PCD) 등이 있고, 두번째 그룹으로서 manganese-dependent peroxidase(MnP), aryl alcohol oxidase(AAO) 및 glyoxal oxidase(GLO) 등인데, 이들 효소들은 목질을 직접적으로 공격하지 않고 제1그룹의 효소들과 협동하여 작용하는 것으로 알려지고 있다. 제3그룹의 효소들은 glucose oxidase(GOD) 및 cellobiose : quinone oxidoreductase(CBQ)로서 feedback type의 효소들로서 목재고분자의 분해시 대사의 고리를 결합시켜 주는 매우 중요한 기능을 하는 효소군들이다. 그러나 이 이외에도 다른 분해기구가 밝혀지고 있으며 기타 효소들에 의한 리그노셀룰로오스의 분해반응기구의 해명에는 상당한 시간이 걸릴 것으로 사료된다.
As a cytosolic transcription factor, the aryl hydrocarbon (Ah) receptor is involved in several pathophysiological events leading to immunosuppression and cancer; hence antagonists of the Ah receptor may possess chemoprevention properties. It is known to modulate carcinogen-metabolising enzymes, for instance the CYP1 family of cytochromes P450 and quinone reductase, both important in the biotransformation of many chemical carcinogens via regulating phase I and phase II enzyme systems. Utilising chemically-activated luciferase expression (CALUX) assay it was revealed that intact glucosinolates, glucoraphanin and glucoerucin, isolated from Brassica oleracea L. var. acephala sabellica and Eruca sativa ripe seeds, respectively, are such antagonists. Both glucosinolates were poor ligands for the Ah receptor; however, they effectively antagonised activation of the receptor by the avid ligand benzo[a]pyrene. Indeed, intact glucosinolate glucoraphanin was a more potent antagonist to the receptor than glucoerucin. It can be concluded that both glucosinolates effectively act as antagonists for the Ah receptor, and this may contribute to their established chemoprevention potency.
We examined the effects of polysaccharides extracted from Asterina pectinifera on the activities of quinone reductase (QR), glutathione S-transferase (GST), ornithine decarboxylase (ODC), cyclooxygenase (COX)-2 and glutathione (GSH) levels in HT-29 human colon adenocarcinoma cells. We found that the polysaccharides extract induced QR activity in a dose-dependent manner over a concentration range of $20-60\;{\mu}g/ml$ and increased GST activity as much as 1.4-fold over controls. GSH levels were increased 1.3- and 1.5-fold with the extract at 40 and $60\;{\mu}g/ml$, respectively. The activity and protein expression of ODC in 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced colon cancer cells was inhibited by the extract. The polysaccharides suppressed TPA-induced prostaglandin (PG) production. These data indicate that polysaccharides from A. pectinifera increase phase II detoxification enzyme activity and inhibit ODC and COX-2 activities in HT-29 human colon adenocarcinoma cells. Consequently, this effect may contribute to the protective effect of polysaccharides from A. pectinifera against colon cancer.
In the oligomerization of p-aminophenol under the catalytic action of the metallic complexes, the effects of the ligands are studied. When the initial velocity of $O_2$ uptake at pH 8 using Fe(Ⅲ) as the central metal and N-hydroxylethylethylenediaminetriacetic acid (HEDTA), ethylenediaminetetraacetic acid (EDTA), diethylenetriaminepentaacetic acid (DTPA), 1,2-cyclohexanediaminetetraacetic acid(CyDTA) as the ligands respectively are compared, the velocities are as the following order: HEDTA > EDTA > DTPA > CyDTA. Further when the effect of the ligands, nitrilotriacetic acid (NTA), HEDTA, EDTA, and DTPA, on the yields of oligomers are compared, the result shows as the following order: NTA > HEDTA > EDTA > DTPA. These are nearly reverse order of the stability constants of the complexes. In order to determine the composition of the mixed complexes at the initial step, the method of continuous variation is used, and it is found that the composition ratio of Fe-EDTA complex to monomer in the mixed complexes is one at pH 5-8 range. It is also found that at pH 9 or in the more alkaline range, side reactions occur to form water soluble dimer of quinone type and the catalytic action of the metallic complex markedly decreases on account of the hydrolysis of the central metal by the $OH^-$ ion.
Experiments were carried out to investigate whether P, deficiency in detached 25 mM mannose-feeding led to a decline of the photosynthetic electron transport rates through acidification of the thylakoid lumen. With increasing mannose-feeding time, the maximal CO2 exchange rates and the maximal quantum yields of photosynthesis decreased rapidly up to 6 h by 73% then with little decrease up to 12 h. The ATP/ADP ratio declined by 54% 6 h after the treatment and then recovered to the control level at 12 h. However, the NADPH/NADP~ ratio was not significantly altered by mannose treatment. Electron transport rates of thylakoid membranes isolated from 6 h treated leaves did not change, but they decreased by 30% in 12 h treated leaves. The quenching analysis of Chl fluorescence in mannose-treated leaves revealed that both the fraction of reduced plastoquinone and the degree of acidification of thylakoid lumen remained higher than those of the control. The reduction of PSI in mannose fed leaves was inhibited due to acidification of thylakoid lumen (high qE). The reduction of primary quinone acceptor of PSII was inhibited by mannose feeding. Mannose treatment decreased the efficiency of excitation energy capture by PSII. Fo quenching was induced when treated with mannose more than 6 h, and had a reverse linear correlation with (Fv)m/Fm ratio. These results suggest that Pi deficiency in Chinese cabbage leaves reduce photosynthetic electron transport rates by diminishing both PSII function and electron transfer from PSII to PSI through acidification ofthylakoid lumen, which in turn induce the modification of photosynthetic apparatus probably through protein (de)phosphorylation.
Parkinson's disease (PD) is the second most common neurodegenerative disorder after Alzhemier's disease. Neuropathologically, PD is characterized by the selective loss of dopaminergic neurons. The neuronal toxicity of cytosolic excess dopamine (DA) has been described in many studies using several cell lines. In dopaminergic neurons, cytosolic excess DA is easily oxidized via monoamine oxidase (MAO)-B, tyrosinase or by auto-oxidation to produce neurotoxic metabolites such as DA quinone. So, in the present study, we induced cell death by treatment of DA ($600{\mu}M$) in human neuroblastoma SH-SY5Y cell which was treated samples before 24 hr, and cell viability was measured by fluorescence activated cell sorter (FACs) analysis. Of those tested, the extracts of Poria cocos (赤茯笭)(whole), Gastrodia elata (rhizomes), Eucommia ulmoides (炒)(barks), Syneilesis palmata (whole), Acorus gramineus (rhizomes), Ligustrum japonicum (leaves) showed neuroprotective effects in dose dependent manner.
Oxidative stress activates several intracellular signaling cascades that may have deleterious effects on neuronal cell survival. Thus, controlling oxidative stress has been suggested as an important strategy for prevention and/or treatment of neurodegenerative diseases. In this study, we found that ginsenoside Rh1 inhibited hydrogen peroxide-induced reactive oxygen species generation and subsequent cell death in rat primary astrocytes. Rh1 increased the expression of phase II antioxidant enzymes, such as heme oxygenase-1 (HO-1), NAD(P)H:quinone oxidoreductase 1, superoxide dismutase-2, and catalase, that are under the control of Nrf2/ARE signaling pathways. Further mechanistic studies showed that Rh1 increased the nuclear translocation and DNA binding of Nrf2 and c-Jun to the antioxidant response element (ARE), and increased the ARE-mediated transcription activities in rat primary astrocytes. Analysis of signaling pathways revealed that MAP kinases are important in HO-1 expression, and act by modulating ARE-mediated transcriptional activity. Therefore, the upregulation of antioxidant enzymes by Rh1 may provide preventive therapeutic potential for various neurodegenerative diseases that are associated with oxidative stress.
Coal tar pitch was chemically modified with 10 wt% benzoquinone (BQ) to raise the softening point of isotropic pitch precursor and the precursor was melt-spun into pitch fibers, stabilized, carbonized and activated with steam at $900^{\circ}C$. The weight loss of carbon fiber-benzoquinone (CF-BQ) increased with the increase of activation time like other fibers, but was lower than those of Kureha fiber at the same activation time in spite of larger geometric surface area. Those adsorption isotherms fitted into 'Type I' according to Brunauer, Deming, Deming and Teller classification. However, there was very thin low-pressure hysteresis that lower closure points of the hysteresis are about 0.42-0.45. From the pore size distribution curves, there might be some micropores having narrow-necked bottle; a series of interconnected pore is more likely than discrete bottles. FT-IR studies showed that the functional groups such as carboxyl, quinone, and phenol were introduced to ACFs-BQ surface after steam activation. Methylene blue decolorization and iodine adsorption capacity of ACF-BQ increased linearly with the increase of specific surface area and was larger than that of ACF-Kureha at the same specific surface area.
Two biopolymers, silk fibroin (SF) and chitosan, were conjugated by tyrosinase (EC 1.14.18.1), a polyphenolic oxidase, to improve their physicochemical properties, such as their thermal properties and morphological stabilities in organic solvents. The crosslinking between SF and chitosan took place mainly through Michael addition reactions. A main reaction between the amino groups in chitosan and o-quinone, the oxidation product of the tyrosyl residue in SF, was confirmed by UV spectroscopy. Measurements of viscosity and light scattering indicated that the crosslinked SF/chitosan conjugate was compact: it had a smaller particle size because of tight bonding forces between the SF and chitosan molecular chains. Thermal decomposition of SF/chitosan conjugates crosslinked by tyrosinase occurred at higher temperatures. The adhesiveness of the SF/chitosan conjugates decreased steadily as the crosslinking reaction progressed. We propose that this new crosslinking method be used for the preparation of silk fibroin/chitosan conjugates using tyrosinase. We expect that SF/chitosan conjugates crosslinked by tyrosinase can be used preferentially in biomedical applications because of its unique properties and non-toxicity.
