Isolation and Characterization of Antioxidative Compounds from the Aerial Parts of Angelica keiskei

  • Ethyl acetate-soluble neutral fraction of hot water extracts from the aerial parts of Angelica keiskei showed a 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical-scavenging activity. Six antioxidative compounds were purified and isolated by various chromatographic procedures. Based on the analyses of FAB-MS and NMR, the isolated compounds were structurally elucidated as luteolin 7-O-${\beta}$-D-glucopyranoside (1), quercetin 3-O-${\beta}$-D-galactopyranoside (2), quercetin 3-O-${\beta}$-D-glucopyranoside (3), quercetin 3-O-${\alpha}$-D-arabinopyranoside (4), kaempferol 3-O-${\alpha}$-D-arabinopyranoside (5), and luteolin 7-O-rutinoside (6). The glycosides of flavonols and luteolin showed DPPH radical-scavenging activity. One molecule of 2, 3, 4, 6, 1, and 5 scavenged 4.2, 4.2, 4.1, 2.5, 2.2, and 1.4 molecules of DPPH radical, respectively.

Effect of Environmental Factors on Flavonol Glycoside Production and Phenylalanine Ammonia-lyase Activity in Cell Suspension Cultures of Ginkgo biloba

  • A study was carried out to elucidate the relation between the production of flavonol glycosides and the change of phenylalanine ammonia-lyase activity in cell suspension cultures of Ginkgo biloba by the unassisted and synergistic effects of various factors. The quercetin production showed a mixed-growth-associated pattern in cell suspension cultures. Fluorescent light and UV radiation increased phenylalanine ammonia-lyase (PAL) activity, and resulted in the increase of the production of quercetin and kaempferol ten- and four-fold, respectively, as compared to that obtained in the normal culture condition. The cell growth of Ginkgo biloba was enhanced .at higher temperatures whereas the quercetin production was at its maximum at low temperatures. Moreover, the quercetin production was increased by temperature change during the culture period. In particular, the quercetin production was at the highest level when the culture temperature was elevated from $10^{\circ}C\;to\;30^{\circ}C$. The addition of phenylalanine as a precursor in the culture medium stimulated an 8-fold increase in the production of quercetin; the addition of naringenin caused a l0-fold increase. The quercetin production was also greatly increased by feeding enzyme cofactors such as 2-ketoglutarate and ascorbic acid in the culture medium, but specific PAL activity was not increased except with phenylalanine feeding. The synergistic effect of UV radiation and naringenin feeding was observed, resulting in the increase of flavonol glycoside production at a rate higher than in any other case investigated.

  • The antioxidative constituents isolated from Hedyotis diffusa were identified as quercetin 3-O-${\beta}$-rutinoside (1) and quercetin 3-O-${\beta}$-glucoside (2). We also isolated asperuloside (3) from this plant. Identification was done based on spectroscopic analysis. Quercetin 3-O-${\beta}$-rutinoside was the stronger antioxidant than quercetin 3-O-${\beta}$-glycoside while asperuloside was inactive.

  • For the investigation of medicinal resources the studies were carried out to evaluated the pharmaco-constituents in the Leaves of Parthenocissus tricuspidata(Vitaceae), of which leaves have been used in Korea as folk remedies for the treatments of arthritis, jaundice, toothache, neuralgia, and etc. From 1-butanol fraction of the MeOH extract, Compound I ($C_{21}H_{18}O_{13}$, Quercetin-3-O-$\beta$-D-glucuronopyranoside), Compound II ($C_{21}H_{20}O_{12}$, Quercetin-3-O-$\beta$-D-glucopyranoside) and Compound III ($C_{25}H_{28}O_{12}$, Quercetin-3-O-(6"-n-butyl)-$\beta$-D-glucuronopyranoside) were isolated by column chromatographic separation using Sephadex LH-20 and ODS gel. Their structures were elucidated through instrumental analyses, such as $^{1}H$-NMR, $^{13}C$-NMR, IR, UV, El-Mass, FAB-Mass and GC. Especially compound III was Flavonol glycoside and named parthenosin.

Flavonoid chemistry of Fallopia sect. Reynoutria (Polygonaceae) in Korea (한국산 닭의덩굴속 호장근절(마디풀과)의 화학분류학적 연구)

  • Fifteen populations comprising three taxa and a putative hybrid of Fallopia sect. Reynoutria in Korea were examined for their leaf flavonoid constituents. Nineteen flavonoid compounds were isolated and identified; they were glycosylated derivatives of the flavonols quercetin and kaempferol, and of the flavones apigenin and luteolin. Among them, quercetin 3-O-galactoside and quercetin 3-O-glucoside were major flavonoid constituents, and present in all taxa. The flavonoid data appear to be very useful for taxon delimitation, and all taxa examined are readily distinguished by their flavonoid profiles. In addition, the flavonoid data suggest that the Nonsan population may be of hybrid origin involving F. japonica var. japonica, F. forbesii, and F. sachalinensis. In F. japonica var. japonica, there is no apparent correlation between their levels of polyploidy and flavonoid chemistry, but geographical variation of the flavonoid profiles among some populations was detected.

  • Chemical investigation of the plant Tristemma hirtum P. Beauv (Melastomataceae) resulted to the isolation of a new flavonol glycoside named quercetin-7-O-${\alpha}$-D-arabinofuranoside (1), together with nine known compounds including 3'-hexadecanoyl-2'-(9aZ)-tetradecanoyl-glycerol 1'-O-[${\beta}$-D-galactopyranosyl-(1'' ${\rightarrow}$ 6'')-${\alpha}$-D-galactopyranoside] (2), arjunolic acid (3), ${\beta}$-sitosterol-3-O-${\beta}$-D-glucopyranoside (4), terminolic acid (5), quercetin (6), asiatic acid (7), maslinic acid (8), $1{\beta}$-O-galloylpedunculagin (9) and 6-hydroxyapigenin 7-O-${\beta}$-D-glucopyranoside (10) from the methanol extract using normal and reversed phase column chromatography. The structures of these compounds were determined by comprehensive interpretation of their spectral data mainly including 1D- 2D-NMR ($^1H-^1H$ COSY, HSQC, and HMBC) spectroscopic and ESI-TOF-MS mass spectrometric analysis.

Acid treatment effects on the contents of quercetin glycosides and aglycone in red onion powder (산처리에 의한 적양파 분말의 quercetin 배당체와 aglycone의 농도변화)

  • Flavonoids are bioactive plant metabolites that have a range of beneficial effects on human health. Quercetin 4'-glycoside (Q4'G), quercetin 3,4'-diglycoside (Q3,4'G), and quercetin aglycone (QA) are the main flavonoids found in onions. QA, in particular, is likely to have a greater biological effect than glycosides. To develop an onion extract with high quercetin content, the optimal extraction conditions for red onion powder containing the outer layer of the onion were determined. The effects of acid treatment on the concentration of quercetin glycosides and QA were evaluated. The flavonoids of red onion powder were optimally extracted under 60-70% ethanol at 70℃ for 2 h. The deglycosylation of Q3,4'G and an increase in Q4'G content occurred within 6 h of 0.2% acetic acid treatment. The QA content and deglycosylation of Q4'G eventually peaked at 24 h. In addition, QA content and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity were highly correlated, with a correlation coefficient of 0.90.

  • To use onion(Allium cepa L.) residue as raw materials of food product, yields of onion-juice according to various extracting methods and optimal extracting condition of quercetin and its related glycosides were carried out. Onion juices gained by the methods of pressing, rotary crushing, freeze pressing and enzyme treatment. The yield by the method of enzyme treatment was higher than others. The yields of juice from fresh onion and heat-treated onion(8$0^{\circ}C$/10min) by pressing were 66% and 83%, respectively. Ethanol extraction of onion was efficient at 75$^{\circ}C$ and acetic acid extraction was proper at 3% concentration for 3 hours at 11$0^{\circ}C$. The onion extract was fractionated in the order of hexane, chloroform, ethylacetate and butanol to test flavonoids. The highest abundant flavonoids were found in ethylacetate and butanol fraction.

  • Cedrela sinensis is a broadleaf tree that is widely cultivated in Korea and China. It was used for treating enteritis, dysentery, and skin itch in oriental medicine. In this study, three major flavonoids, kaempferol-3-O-rhamnoside (1), quercetin-3-O-rhamnoside (2), and quercetin-3-O-glucoside (3), were isolated from the leaf of Cedrela sinensis. The biological activities of these compounds were tested by inhibitory activity of matrix metalloproteinases-2 (Type IV collagenase) method together with a cytotoxicity and a apoptosis test against human cancer cell lines.
