Mercury-Specific Effects on Photosynthetic apparatus of Barley Chloroplasts Compared with Copper and Zinc Ions (구리${\cdot}$아연과 비교한 보리 엽록체의 광합성 기구에 미치는 수은 이온의 특이한 효과)

  • To find heavy metal-specific effects on the photosynthetic apparatus of higher plants, we investigated effects of $CuCl_2$, HgCl_2$ and $ZnCl_2$ on electron transport activity and chlorophyll fluorescence induction kinetics of chloroplasts isolated from barley seedlings. Effects on some related processes such as germination, growth and photosynthetic pigments of the test plants were also studied. Germination and growth rate were inhibited in a concentration-dependent manner by these metals. Mercury was shown to be the most potent inhibitor of germination, growth and biosynthesis of photosynthetic pigments of barley plants. In the inhibition of electron transport activity, quantum yield of PS II, and chlorophyll fluorescence induction kinetics of chloroplasts isolated from barley seedlings, mercury chloride showed more pronounced effects than other two metals. Contrary to the effects of other two metals, mercury chloride increased variable fluorescence significantly and abolished qE in the fluorescence induction kinetics from broken chloroplasts of barley seedlings. This increase in variable fluorescence is due to the inhibition of the electron transport chain after PS ll and the following dark reactions. The inhibition of qE could be attributed to the interruption of pH formation and do-epoxidation of violaxathin to zeaxanthin in thylakoids by mercury. This unique effect of mercury on chlorophyll fluorescence induction pattern could be used as a good indicator for testing the presence and/or the concentration of mercury in the samples contaminated with heavy metals.

Efficiency Factors of Singlet Oxygen Generation from Core-Modified Expanded Porphyric : Tetrathiarubyrin in Ethanol

  • The photophysical properties and the singlet oxygen generation efficiency of tetrathiarubyrin have been investigated to elucidate the possibility of its use as a photodynamic therapy (PDT) photosensitizer by steady-state and time-resolved spectroscopic methods. The observed photophysical properties were affected by various molecular aspects, such as extended ${\pi}conjugation$, structural distortion, and internal heavy atom. The steady-state electronic absorption spectrum was red-shifted due to the extended $\pi-conjugation$, and the spin orbital coupling was enhanced by the structural distortion and the internal heavy atom effect. As a result of the enhanced spin orbital coupling, the triplet quantum yield increased to 0.90 $\pm$ 0.10 and the triplet state lifetime was shortened to 7.0 $\pm$ 1.2 ${\mu}s$. Since the triplet state decays at a relatively faster rate, the efficiency of the oxygen quenching of the triplet state decreases. The singlet oxygen quantum yield was estimated to be 0.52 $\pm$ 0.02, which is somewhat lower than expected. On the other hand, the efficiency of singlet oxygen generation during the oxygen quenching of triplet state, $f{\Delta}^T$, is near unity. Such high efficiency of singlet oxygen generation can be explained by the following two possible factors: The hydrogen bonding of ethanol which impedes the deactivation pathway of the charge transfer complex with oxygen to the ground state, the less probability of the aggregation formation.

  • The cmc's of sodium dodecyl sulfate (SDS)/Triton X-100 surfactant mixtures were determined by surface tension measurement at various surfactant compositions. The cmc values were lower than those predicted from ideal mixture. The regular solution theory was applied to calculate the interaction parameter, micellar composition, and the activity coefficients of surfactants in the mixed micelle. The interaction parameter (${\beta}$) was - 2.1. The nonideality arised largely from decreased activity of SDS in the mixed micelle. The mean aggregation numbers (${\bar{n}}$) and micropolarity of hydrocarbon region of the mixed micelles were determined by luminescence probe techniques. The total aggregation number (${\bar{n}}_{SDS}+{\bar{n}}_{TX}$) in mixed micelles showed little dependency on the composition of the micelle. The apparent dielectric constant of the hydrocarbon region of the micelle vs micellar composition plot showed positive deviation from linearity. Emission and emission quenching of excited tris(2,2'-bipyridine)ruthenium(Ⅱ) cation, $(Ru(bpy)_3^{2+})$, by methylviologen ($MV^{2+}$) were also investigated in the mixed micellar solutions. The quenching rate was lowest when the mole fraction of SDS in the surfactant mixtures (${\alpha}_{SDS}$) is about 0.25 and highest at ${\alpha}_{SDS}$ = 0.85. This was explained in terms of combined effects of binding of the cations with the micelle and mobility of the bound cations on the surface of the micelles.

  • The effect of carbon doping contents on the microstructure, hardness, and corrosion properties of heat-treated AISI steel grades of plain carbon steel was investigated in this study. Various microstructures including coarse ferrite-pearlite, fine ferrite-pearlite, martensite, and bainite were developed by different heat treatments i.e. annealing, normalizing, quenching, and austempering, respectively. The developed microstructures, micro-hardness, and corrosion properties were investigated by a light optical microscope, scanning electron microscope, electromechanical (Vickers Hardness tester), and electrochemical (Gamry Potentiostat) equipment, respectively. The highest corrosion rates were observed in bainitic microstructures (2.68-12.12 mpy), whereas the lowest were found in the fine ferritic-pearlitic microstructures (1.57-6.36 mpy). A direct correlation has been observed between carbon concentration and corrosion rate, i.e. carbon content resulted in an increase in corrosion rate (2.37 mpy for AISI 1020 to 9.67 mpy for AISI 1050 in annealed condition).

  • The decomposition of chlorinated methanes including $CCl_4$, $CCl_3H$, and $CCl_2H_2$ was carried out using a thermal plasma process and the characteristics of the process were investigated. The thermal equilibrium composition was analyzed with temperature by Fcatsage program. The decomposition rates at various process parameters including the concentration of reactants, flow rate of carrier gas, and quenching rate, were evaluated, where sufficiently high conversion over 92% was achieved. The generation of main products was strongly influenced by the reaction atmosphere; carbon, chlorine, and hydrogen chloride at neutral condition; carbon dioxide, chlorine, and hydrogen chloride at oxidative condition. The decomposition mechanism was speculated considering the results from Factsage and the identification of generated radicals and ionic species. The main decomposition pathways were found to be dissociative electron attachment and oxidative by radicals formed in a plasma state.

The Inactivation Effects of UV Light on Bacteriophage f2 (박테리오파지 f2에 대한 자외광선의 살균효과)

  • The effects of ultraviolet light on bacteriophage f2 were investigated to determine the inactivation kinetics and its mechanism. The 260nm light showed a little higher inactivation rate than the one of 300 nm. In this work, our main concern was whether structural and/or conformational changes in the protein capsid could occur by UV irradiation. The inactivation for the first 20 minutes irradiation was rapid with a loss of about 4 logs and followed by a slower rate during the next 40 minutes with no survival noted in the samples irradiated for 90 minutes or longer. The structural change of the protein capsid was examined by optical spectroscopic techniques and electron microscopy. The absorption spectra of the UV irradiated phages showed no detectable differences in terms of the spectral shape and intensity from the control phage. However, the fluorescence emission spectroscopic data, i.e. 1) fluorescence quenching of tryptophan residues upon irradiation of 300 nm light, 2) enhancement of fluorescence emission of ANS (8-aniline-1-naphthalene sulfonate) bound to the intact phages compared to the one in the UV-treated phages, and 3) decrease of energy transfer efficiency from tryptophan to ANS in the UV-treated samples, presented remarkable differences between the intact and UV-treated phages. Such a structural alteration was also observed by electron microscopy The UV-treated phages appeared to be broken and empty capsids. Therefore, the inactivation of the bacteriophage f2 by UV irradiation is thought to be attributed to the structural change in the protein capsid as well as damage in the viral RNA by UV irradiation.

Development and Characterization of Helium Microwave Plasma Torch (헬륨 마이크로파 플라즈마 토치의 개발과 특성에 관한 연구)

  • MPT, which has been developed recently, is very tolerant to aqueous samples. Several types of MPT have been investigated and is found that the double concentric tube could sustain a stable plasma at a low plasma gas flow rate. However, the tip of torch is easily ruined. Triple concentric torch has shown the best stability and the plasma shape, much like that of ICP, especially when the central channel is quartz. The plasma is exposed and mixed with air as is suggested from the background spectrum, which leads to quenching of He MPT. Sensitivity of helium MPT equipped with a membrane desolvator has shown 10 times lower than that of Argon MPT for most of elements except for the ones with relatively high excitation energy. He MPT requires small plasma flow rate (about 1.6 L/min), stable and simple to use. Excitational temperature and electron number density measured are 4950 K and 3.28 ${\times}$ $10^{14}cm^{-3}$, respectively.

  • The ring gears of automobile parts are manufactured generally process chart of which is as follows : forging ${\rightarrow}$ annealing or normalizing ${\rightarrow}$ rough machining ${\rightarrow}$ hardening(Quenching-Tempering or carburizing process) ${\rightarrow}$ finish machining. Isothermal annealing process after forging is most effective in the side of improvment of machinability. On this study we selected two kinds of steel;SCM415, SCM435 of most universal and investigated microstructures to find out most suitable condition of heat treatment in proportion continuous cooling and isothermal annealing. As the cooling rate is $5^{\circ}C$ per minute in continuous cooling process, martensite and bainite are coexisted with ferrite and pearlite in SCM435 steel. If the cooling rate is slower than $5^{\circ}C$ per minute, microstructure were only ferrite and pearlite but formation of band structure can't be avoid. On the other hand, microstructure is only ferrite and pearlite regardless of cooling rate because carbon content of SCM415 steel is low. Moreover formation of band structure isn't exposed by faster cooling rate. Most optimal temperature of the isothermal annealing is from $650^{\circ}C$ to $680^{\circ}C$ in SCM435 steel. When holding time is 60 minute with $650^{\circ}C$, the identical ferrite and pearlite microstructures can be obtained.

Application of Non-photochemical Quenching on Screening of Osmotic Tolerance in Soybean Plants (콩의 삼투 저항성 검정에 있어서 Non-photochemical quenching의 적용)

  • Non-photochemical quenching (NPQ) values for utilizing them to detect osmotic tolerance in plants were examined with two different soybean cultivars, an osmotic tolerant soybean (Shinpaldalkong 2) and a control soybean (Taekwangkong). Two different stresses were applied to the cultivars as the restricted irrigations of 200 and 50 ml water $pot^{-1}\;d^{-1}$ for 5 days for a control and a drought stress, respectively, and a sodium chloride solution of 200 mmol for 6 days for a salt stress. The intact leaves of the two cultivars after treatment were used to measure chlorophyll fluorescence parameters, maximum efficiencies of photosystem II photochemistry (Fv/Fm), efficiencies of photosystem II photochemistry (${\Phi}_{PSII}$), $CO_2$ assimilation rate ($P_N$), and NPQ. Leaf water potentials of the two cultivars decreased from - 0.2 to - 0.8MPa by a drought treatment and from - 0.7 to - 1.7MPa by a salt treatment. Leaf water content of Shinpaldalkong 2 after a salt treatment was less decreased than that of Taekwangkong. $F_v/F_m$ values of both cultivars were not changed, while ${\Phi}_{PSII}$ and $P_N$ were decreased proportionally to leaf water potential decrease. The response of NPQ was occurred in Shinpaldalkong 2 under the drought and salt stresses. With Taekwangkong cultivar, only drought stress referred NPQ response. The cultivar differences on chlorophyll fluorescence parameters were found in the relationships between ${\Phi}_{PSII}$ and $P_N$, and between NPQ and ${\Phi}_{PSII}$. Although the positive relationships between ${\Phi}_{PSII}$ and $P_N$ were established on all treatments of both cultivars, the decreasing rate of ${\Phi}_{PSII}$ to $P_N$ was smaller in Shinpaldalkong 2 than Taekwangkong. The NPQ was increased according to the decrease of ${\Phi}_{PSII}$ by osmotic treatments in Shinpaldalkong 2. The complementary relationships between NPQ and ${\Phi}_{PSII}$ were well maintained at all treatments in Shinpaldalkong 2, while these relationships were lost at a salt treatment in Taekwangkong. Taken together, the results suggest that analysis of complementary relationships between ${\Phi}_{PSII}$ and NPQ could be more valuable and applicable for determining osmotic tolerance than single analysis of each parameter such as $F_v/F_m$, ${\Phi}_{PSII}$ and NPQ.

Lipid Peroxidation of Ginseng Thylakoid Membrane (인삼 틸라코이드 막의 지질과 산화)

  • In order to elucidate the mechanism of the leaf-burning disease of ginseng (Panax ginseng C.A. Meyer), the relationships between thylakoid membrane peroxidation and chlorophyll bleaching were investigated in comparison with the ones of soybean (Glycine max L). When I measured the rate of lipid peroxidation in the thylakoids of ginseng and soybean by irradiation of light(60 w.m-2), it was identified that, the remarkably lower rate of lipid peroxidation was found in the ginseng thylakoid than the case of soybean. When lipid peroxidation of ginseng thylakoid was induced in the dark, chlorophyll contents of thylakoid was not changed. The results suggest that lipid peroxidation does not affect the chlorophyll bleaching in ginseng thylakoid. Thylakoid membrane peroxidation as well as chlorophyll bleaching was closely related with photosynthetic electron transport. But, according to the quenching experiment active oxygen species induced lipid peroxidation may be different species in the case of chlorophyll bleaching.
