Brucellosis in sheep and goat of Bogra and Mymensingh districts of Bangladesh

  • Brucellosis is the most important bacterial disease of livestock in Bangladesh. The present study was conducted to determine the seroprevalence of brucellosis in goat and sheep in Mymensingh and Bogra districts of Bangladesh using slow agglutination test and Rose Bengal test as screening test and indirect enzyme linked immunosorbent assay as confirmatory test. Questionnaire based data on age, gender, area, client's complaint, number of animals in herds, disease history, reproductive problems such as abnormal uterine discharge, abortion or previous abortion in sheep and goat and their reproductive diseases were recorded. A total of 200 sera samples were collected from 80 sheep and 120 goats. The prevalence of brucellosis in goat was 2.50% and 1.25% in sheep. Positive reactors were only detected in female of both goat and sheep. In this study, there existed a significant association among abortion and the prevalence of brucellosis (p < 0.01). The prevalence of brucellosis in sheep and goat in Bangladesh is not negligible, and it is therefore worth considering the adoption of preventive measures.

  • The most common symptoms of COVID-19 are high fever, cough, headache, and fever. These symptoms may vary from person to person, but checking for "fever" is the government's most basic measure. To confirm this, many facilities use thermographic cameras. Since the previously developed thermographic camera measures body temperature one by one, it takes a lot of time to measure body temperature in places where many people enter and exit, such as multi-use facilities. In order to prevent malfunctions and errors and to prevent sensitive personal information collection, this research team attempted to develop a facial recognition thermographic camera. The purpose of this study is to compensate for the shortcomings of existing thermographic cameras with disaster safety IoT integrated solution products and to provide quarantine systems using advanced facial recognition technologies. In addition, the captured image information should be protected as personal sensitive information, and a recent leak to China occurred. In order to prevent another case of personal information leakage, it is urgent to develop a thermographic camera that reflects this part. The thermal imaging camera system based on facial recognition technology developed in this study received two patents and one application as of January 2022. In the COVID-19 infectious disease disaster, 'quarantine' is an essential element that must be done at the preventive stage. Therefore, we hope that this development will be useful in the quarantine management field.

Incidence of Citrus vein enation virus in Citrus spp. and Poncirus trifoliata in Korea (국내 감귤류에 발생한Citrus vein enation virus 분포조사)

  • Citrus vein enation virus (CVEV), which was regulated as a quarantine virus in Korea, was firstly found on Jeju Island in 2017. In February 2018, a survey was carried out to determine the distribution of CVEV in the main commercial areas growing Citrus spp. and Poncirus trifoliata. The survey was performed at 203 groves in the southern Korean Peninsula and Jeju Island. CVEV infection was determined by reverse transcription polymerase chain reaction detection and sequencing. The coat protein (CP) gene sequences obtained from the CVEV-infected samples showed high similarities (more than 98%) to the previously reported CVEV CP sequences. In summary, CVEV was detected in 136 groves (67%), in which 85.4% of Citrus junos and 77.8% of Citrus unshiu were infected by CVEV. In Jeju Island, the infection rate of CVEV was relatively higher (90.6%). Our result revealed that CVEV has spread widely in Citrus and Poncirus in Korea. Based on the result, the Korean quarantine agency decide to exclude CVEV from quarantine in Korea.

Analysis of in vitro apoptosis induced by virulent Korean isolate of classical swine fever virus in peripheral blood B cell line

  • Classical swine fever (CSF) is a highly contagious disease among swine that has an important economic impact on worldwide. One clinical symptom of CSF is leukopenia, in particular lymphopenia, which is a characteristic event that occurs early in the course of CSF. Though lymphopenia associated with apoptosis, the pathogenic mechanism underlying the lymphopenia has not been well studied. To understand these mechanisms, we investigated the response of porcine B cell lines to infection with SW03, virulent strain isolated from swine tissue in Korea. This study demonstrated that SW03-infected L35 cell were induced apoptosis through the detection of activated caspase-3. In addition, SW03 infection leaded to alterations in pro-apoptotic, Bax, and anti-apoptotic, Bcl-xL proteins of Bcl-2 family. Our results would suggest that SW03-infected L35 cells induced apoptosis via intrinsic mitochondrial pathway.

Epidemiological analysis of Escherichia coli O157 : H7 by pulsed-field gel electrophoresis and multiplex polymerase chain reaction

  • Twenty three strains of Escherichia (E) coli O157 : H7 isolated from Korea, Japan, USA were analyzed by pulsed-field gel electrophoresis (PFGE) of XbaI-digested chromosomal DNA and multiplex polymerase chain reaction. Various PFGE patterns of E. coli O157 : H7 were found on the same farm. Most of the E, coli O157 : H7 strains had shiga-like toxin (slt) II gene only (43.5%) or both slt I and slt II genes(30.4%). eaeA gene was highly conserved in the E. coli O157 : H7. There was no correlation between PFGE and slt gene patterns. The results indicate that various genotypes of E. coli O157 : H7 have spread throughout the country and genomic DNA patterns generated by PFGE are highly specific for different strains and have significant value in epidemiologic investigations of infectious disease outbreaks.

Comparison of tissue tropism of Newcastle disease vaccine viruses by Immunohistochemistry techniques (면역조직화학기법을 이용한 뉴캣슬병 백신바이러스의 조직친화성 비교)

  • Mean death time of inoculated embryonated egg is one of the methods to determine the virulence of the Newcastle disease viruses (NDV). Evaluation of tissue tropism of NDV in the host has been proposed as an another way to determine the pathogenicity of NDV based on the principal site of viral replication. To evaluate the tissue tropism among NDV, an immunohistochemistry(IHC) technique using monoclonal antibody was applied in one-day-old SPF chickens inoculated with different ND vaccine strains such as Ulster 2C, VG/GA and B1 viruses by eye drop instillation. The tissues used for this comparison were trachea, intestine, Harderian gland and cecal tonsil, which were collected at 0.5, 1, 2, 3, 5, 8, 10, 14 days post inoculation. Among test groups, chickens inoculated with B1 viurs, which is known to replicate in the respiratory system, have IHC positive staining mainly in the trachea and those inoculated with Ulster 2C have IHC positive staining mainly in the intestine. However, chickens inoculated with VG/GA strain have IHC positive staining in both the trachea and intestine. Therefore, a differences in tissue tropism among ND vaccine strains has been proved by the IHC technique. Based on this results, the IHC staining technique could be used to classify the NDV or to study the pathogenesis of NDV in chickens.

Investigation of Prohibited Seed-Borne Plant Pathogenic Bacteria in Korea (국내 종자전염 규제 식물병원세균 조사)

  • The goal of this manuscript is to determine seed-borne plant pathogenic bacteria and phytoplasmas among quarantine pests in Korea. Four and two prohibited bacteria and phytoplasmas, respectively, and 35 and 17 restricted bacteria and phytoplasmas, respectively, were assessed whether they are seed-borne or not based on preliminary reports. As results, two species of prohibited bacteria, eighteen species of restricted bacteria, and one species of restricted phytoplasma have been determined as being seed-borne plant pathogenic bacteria. Thus, quarantine fields must account for these lists once inspection has been conducted on imported seeds and also use of these lists can help to reduce the production of new diseases that can spread from infected imported seeds.

Establishment of a diagnostic method for porcine proliferative enteropathy using polymerase chain reaction (중합효소연쇄반응을 이용한 돼지 증식성 장염 진단기법 확립)

  • Porcine Proliferative Enteropathy(PPE) is an infectious enteric disease and a major cause of economic loss in swine industry due to weight loss, poor growth and sudden death in growing and finishing pigs at 6 to 20 weeks of age. PPE has been diagnosed by clinical signs, syndrom and lesions in the intestine in Korea. However, the diagnostic method had several problems in the detection of infected or carrier pigs. Therefore, in this study, we established the polymerase chain reaction(PCR) which was a fast, specific and sensitive method for identification of Lawsonia intracellularis (L intracellularis). We designed and synthesized primer on the 16S rDNA and p78 gene encoding L intracellularis. Specificity of the method was confirmed by comparison of the PCR results using other enteric bacteria and the study has shown that PCR method was sensitive to detect 1ng of genomic DNA as a template. Identity of the PCR products was confirmed by comparison of pattern of restriction endonuclease analysis with restriction enzyme Hae III and Pst I. Also, the PCR method was applicable to the naturally affected pigs with PPE. Based on the results from this study, the PCR method could be used as a fast and specific diagnostic tool for PPE.
