Correlation of Marbling Characteristics with Meat Quality and Histochemical Characteristics in Longissimus Thoracis Muscle from Hanwoo Steers

  • The objective of this study was to investigate the correlations of marbling characteristics, including marbling score, intramuscular fat (IMF) content, and fleck traits, with meat quality traits and histochemical characteristics of the longissimus thoracis muscle from Hanwoo steers. Marbling fleck characteristics, especially area, number, and fineness (F) index, measured by computerized image analysis were strongly correlated with marbling score and IMF content (p<0.05). However, coarseness (C) index and F/C ratio were somewhat limited relationships with marbling score. In contrast, the IMF content and the number of smaller white flecks increased with increasing lightness value (p<0.05). Moreover, beef with higher marbling scores showed lower cooking loss and Warner-Bratzler shear force value compared to beef with lower marbling scores (p<0.05). Regarding the muscle bundle traits, as number of bundle increased, number of marbling flecks increased (p<0.05), although most marbling characteristics did not have significant correlation with muscle fiber or bundle characteristics.

Evaluation of Crossbreeding Effects for Wool Traits in Sheep

  • Crossbreeding effects for wool quality traits viz. greasy fleece weight (kg), staple length (cm), average fibre diameter (${\mu}$) and medulation percentage were estimated using the Dickerson's and Kinghorn's models. The data analyzed involved 15 genetic groups including Nali purebred, $F_1$'s of two and three breeds, $F_2$'s and reciprocal crossbred obtained from the crossing of Nali (N), Merino (M) and Corriedale (C) breeds during 1980-96. Nali and Corriedale breeds had non-significant negative additive genetic effects (Dickerson's model) on greasy fleece weight, while effects of Corriedale were negative for staple length only from both models. In general additive genetic effects of all three breeds were non-significant for all the wool traits except medulation percentage. Non significant heterotic and recombination effects (epistatic loss) were estimated from both models. However, the estimates of crossbreeding effects varied between the models both in magnitude as well as in direction barring few exceptions. Undesirable positive heterosis was found on medulation percentage for all types of combinations involving three breeds. Comparison of least squares means of various genetic groups revealed that both two breed and three breed crosses were superior to the Nali breed for all wool quality traits. Fibre diameter of MN crossbreds was significantly less than CN crossbreds. Results also indicated that as the inheritance of Nali breed in a cross is decreased, the medulation percentage decreases which is desirable. Inter se mating of crossbreds (two breed, three breed) has not resulted in a decline in the wool quality traits. These results indicate that the synthetic population derived from three breeds can be stabilized easily for wool traits as there may not be epistatic loss on subsequent inter se mating of crossbreds.

Novel SNP in the coding region of the FTO gene is associated with marbling score in Hanwoo (Korean cattle)

  • The fat mass and obesity associated (FTO) gene plays an important role in the regulation of energy homeostasis, fat deposition and obesity. For this reason, the FTO gene is a physiological and functional candidate gene for carcass and meat quality traits in beef cattle. The objectives of this study were to identify SNPs in the exonic regions of FTO gene and to evaluate the association of these SNPs with carcass traits in Hanwoo (Korean cattle). In this study, we newly identified two exonic SNPs in Hanwoo population. The g.125550A > T SNP was located in exon 3 and the g.175675C > T SNP was located in exon 6. Genotyping of the two SNP markers was carried out using PCR-RFLP analysis in Hanwoo steers to evaluate their association with carcass traits. As a result, g.125550A > T SNP genotype was significantly associated with effects on marbling score. Animals with the AA and TT homozygous genotypes had a significantly higher marbling score (p < 0.001) than those with AT heterozygous genotype, and this was significant after Bonferroni correction of the significance threshold (p = 0.003). Dominance effect was also observed for the marbling score (P < 0.05) with higher marbling score of homozygous animals. However, no significant associations with meat quality traits were observed for the g.175675C > T SNP. Our results suggest that the exonic SNP g.125550A > T in the FTO gene may be used as a DNA marker for the selection of Hanwoo with higher marbling.

Relationship of Slaughter Age and Carcass Traits of Hanwoo Cattle

  • Two experiments were carried out to investigate the relationship of slaughter age and carcass traits of Hanwoo cattle. A total number of 11,284 heads of cattle (4,624 bulls, 6,120 steers, and 540 females) were used in the experiment (Exp.) 1. In the Exp. 2, 3,887 heads of steers slaughtered at age of 24-36 mon were used. After 24 h post-slaughter chilling, the carcasses were weighed and evaluated by an official grader of carcass traits according to the Korean carcass grading standard. Backfat thickness was the main variable in yield index and grade whereas marbling score was the main variable in carcass quality grade. Meat color was an important factor affecting carcass quality grade of bulls; fat color was an important factor affecting those of steers and females; and maturity was an important factor affecting that of females. After 24-mon-old, extension of slaughter age did not automatically increase the quantity and quality of meat. However, 29 mon of age can be regarded as the optimum time to slaughter steers to attain the highest take home money for farmers.

Effect of a c-MYC Gene Polymorphism (g.3350G>C) on Meat Quality Traits in Berkshire

  • c-MYC (v-myelocytomatosis viral oncogene homologue) is a transcription factor that plays important role in many biological process including cell growth and differentiation, such as myogenesis and adipogenesis. In this study, we aimed to detect MYC gene polymorphisms, their genotype frequencies and to determine associations between these polymorphisms and meat quality traits in Berkshire pigs. We identified a single nucleotide polymorphism (SNP) in intron 2 of MYC gene by Sanger sequencing, i.e., g.3350G>C (rs321898326), that is only found in Berkshire pigs, but not in other breeds including Duroc, Landrace, and Yorkshire pigs that were used in this study. Genotypes of total 378 Berkshire pigs (138 sows and 240 boars) were determined using Hha I restriction enzyme digestion after polymerase chain reaction. Observed allele frequencies of GG, GC, and CC genotypes were 0.399, 0.508, and 0.093 respectively. Statistical analysis indicated that the g.3350G>C polymorphism was significantly associated with $pH_{45min}$ and cooking loss (p<0.05), suggesting that g.3350G>C SNP can be used for pre-selection of $pH_{45min}$ and cooking loss traits in Berkshire pigs.

Confirmation of SSR Markers and QTLs Associated with Seed Size and Water Absorbability in Soybean (Glycine max) Cultivars for Fermented Product, Saengcheonggukjang

  • Saengcheonggukjang, known as Natto in Japan, is a Korean fermented soybean food that has various bioactive compounds for antioxidant and antidiabetic activity. The development of soybean (Glycine max L.) cultivars for saengcheonggukjang production relies on the selection of seed traits that influence the quality and sensory properties. One of the important traits for cultivars is seed characteristics such as seed hardness and size. In order to select the lines for breeding good quality saengcheonggukjang varieties, several simple sequence repeat (SSR) markers and quantitative trait loci (QTLs) related to seed quality of Korean cultivars, Pungsannamulkong, Socheongja, Pungwon, Heawon, and Hoseo, were analyzed. Based on the many studies to detect stable QTLs for seed traits, we tested several QTLs related to seed size and water absorbability using SSR markers on Korean cultivars. The results showed that two regions for water absorbability of Pungsannamulkong and one region for seed size traits of Haewon and Hoseo were identified in this study. These results could have applications to soybean breeding for seed size and hardness and it is necessary to narrow it down through further study.

Effects of Dietary Supplemental $Megazone^{(R)}$ on Growth Performance, Nutrients Digestibility, Blood Characteristics, Meat Quality and Carcass Traits in Weaning-to-Finishing Pigs

  • This study was conducted to investigate the effects of $Megazone^{(R)}$ (a mineral mix) supplementation on growth performance, nutrients digestibility, blood characteristics, meat quality and carcass traits in weaning-to-finishing pigs. A total of 48 crossbred $(Landrace{\times}Yorkshire{\times}Duroc)$ pigs with initial body weight (BW) of $4.46{\pm}0.18kg$ were used in a 21 wks trial. Pigs were blocked by weight and allotted to two dietary treatments in a completly randomized design. There were 6 pens per treatment. Dietary treatments included: 1) Control (CON: basal diet) and 2) MT (basal diet+0.8% $Megazone^{(R)}$). Through the entire experimental period, there were no effects of dietary $Megazone^{(R)}$ supplementation on growth performance nutrients digestibility, blood characteristics and meat quality traits (p>0.05). Market weight and backfat thickness also had no differences between the two treatments (p>0.05). However, carcass weight and carcass ratio in MT treatment were improved significantly compared with CON treatment (p<0.05). In conclusion, supplmentation of $Megazone^{(R)}$ can increase carcass weight and carcass ratio in weaning-to-finishing pigs, however, it has no effects on growth performance, nutrients digestibility, blood characteristics and meat quality traits.

소의 도체, 육질형질과 CSRP3, ACOX1 유전자들과의 상관관계 (Association of Bovine CSRP3 and ACOX1 Genes with Carcass and Meat Quality Traits)

  • There is no investigation has yet been conducted for ACOX1 and CSRP3 gene polymorphisms in Korean cattle (Hanwoo), and their associations with carcass and meat quality traits. In this study, SNPs in ACOX1 and CSRP3 genes were identified and their associations with carcass and meat quality traits were investigated in 227 Hanwoo animals. Two SNPs (g.224G> A and g.19491G>A) in ACOX1 gene and one SNP (g.14859C>T) in CSRP3 gene were identified in Hanwoo and sequence analysis indicated that these SNPs were located in the coding regions. The allele frequencies of ACOX1 g.224G>A and g.19491G>A SNPs were 0.57, 0.43, and 0.56 and 0.44, respectively, For CSRP3 g.14859C>T polymorphism, the C and T allele frequencies were 0.64 and 0.36, respectively. The Hanwoo cattle were used to detect PCR-RFLP patterns for estimating the allele frequencies. Single marker association analyses were performed between genotype of each SNP, and carcass and meat quality association traits to evaluate the relationships in Hanwoo. The g.224G>A SNP genotypes of ACOX1 gene, which was significantly associated with meat quantity grade at slaughter (P<0.03) and backfat thickness tended to be greater (P=0.06) in Hanwoo. The previously identified g.14859C>T SNP was used in this study and the obtained genotype and allele frequencies are almost similar with the previous results reported by Bhuiyan et al. (2007). However, no significant association was found between g.19491G>A SNP in the ACOX1 and g.14859C>T SNP genotypes of CSRP3 gene and considered carcass and meat quality traits. In conclusion, the information on the identified SNPs in CSRP3 and ACOX1 genes could be useful for further association study and haplotype analysis for the development of carcass and meat quality traits in Hanwoo.

Effect of Hybridization on Carcass Traits and Meat Quality of Erlang Mountainous Chickens

  • Native chickens hold a significant share of the market in China. In response to the huge demand from the market, the productivity of Chinese native chickens needs to be improved. Cross breeding is an effective method to increase productivity, although it might affect meat quality. In this study, two pure lines (SD02 and SD03) of Erlang mountainous chickens were hybridized with a yellow feather and faster growing line (SD01). The effect of hybridization on carcass and meat quality (physiochemical and textural traits) was measured in the $F_1$ population at d 91 of age. The hybrids exhibited higher body weight and dressed weight, and amount of semi-eviscerated, eviscerated, breast muscle and abdominal fat (p<0.05). Abdominal fat yield also increased (p<0.05) compared to the offspring of the two pure-lines. Meanwhile, there was no significant difference in meat quality traits except for the myofiber diameter and density and the shear force of the breast muscle. Overall, the offspring of cross-lines were similar to pure lines in meat color, pH value, inosinic acid, crude protein, crude fat, dry matter, moisture content and amino acid composition in the breast muscle. These results suggest that productivity can be improved via cross-breeding while maintaining meat quality of the Erlang mountainous chicken.

Meat Quality Traits of Longissimus dorsi Muscle from Carcasses of Hanwoo Steers at Different Yield Grades

  • The strategy for increasing the palatability of Hanwoo beef through fattening could lead to a decline in yield grade. The aim of this study was to examine the meat quality traits of Longissimus dorsi (LD) muscle from carcasses of Hanwoo steers at different yield grades. A total of 246 Hanwoo steers was divided into the following yield grades: A (n=77), B (n=76) and C (n=93). Meat quality traits, including proximate composition, cholesterol content, nucleotide content, dipeptide content, creatine and creatinine, free amino acid content, fatty acid composition, instrumental meat color, pH, water holding capacity, drip loss, cooking loss, and sensory qualities of the LD muscle from the 3 yield grades of Hanwoo carcasses were measured. The decline in yield grade from A to C resulted in an increase in crude fat and cholesterol content as well as a decrease in inosine 5'-monophosphate and aspartic acid in the LD muscle (p<0.05). In terms of fatty acid composition, the LD muscle from yield grade A had higher SFA and PUFA and lower MUFA content than that from yield grade C (p<0.05). However, the ratio of PUFA/SFA and n-6/n-3 did not differ among LD muscles from the 3 yield grades. There were no significant differences among other meat quality traits in relation to the yield grade. In conclusion, we suggest that the changes of substances related with health and flavor can be considered in order to obtain better quality Hanwoo beef.
