We hypothesized that the extract from steam exploded Q. variabilis might be cytoprotective for tenofibroblasts cells during oxidative stress. In the present study, the extracts obtained from steam exploded (severity log Ro 4.68) Q. variabilis contained high quantities of phenolics and flavonoids contents. Also, the extracts appear to have, on these tenofibroblasts, a protective effect against oxidative stress. Tenofibroblasts cells incubated with the extracts and stressed with $H_2O_2$ showed an increase in cell viability by MTT assay. The extracts is found to inhibit $H_2O_2$-induced apoptosis in tenofibroblasts cells, as shown by Annexin V/PI double staining analysis. Western blot data showed that in the extracts/$H_2O_2$-treated cells, the extracts inhibited the $H_2O_2$-dependent phosphorylation of ERK and p38. From these results, it is suggested that the extracts showed the protective effect on $H_2O_2$-mediated oxidative stress. The main chemical compounds of the extract was identified as 1,8-cineole by GC-MS analysis. The anti-apoptosis activity is accordingly believed to be attributable to the 1,8-cineole.
Objective: This study aimed to determine the protective efficacy of Buddha's Temple (BT) extract against tert-butyl hydroperoxide (t-BHP)-induced oxidative stress in Gallus gallus chicken embryo fibroblast cell line (DF-1) and its effects on the cell lipid metabolism. Methods: In this experimental study, Gallus gallus DF-1 fibroblast cells were pretreated with BT 10-7 for 24 hours, followed by their six-hour exposure to t-BHP (100 μM). Water-soluble tetrazolium salt-8 (WST-8) assays were performed, and the growth curve was computed. The intracellular gene expression changes caused by BT extract were confirmed through quantitative polymerase chain reaction (qPCR). Flow cytometry, oil red O staining experiment, and thin-layer chromatography were performed for the detection of intracellular metabolic mechanism changes. Results: The WST-8 assay results showed that the BT pretreatment of Gallus gallus DF-1 fibroblast cell increased their cell survival rate by 1.08%±0.04%, decreased the reactive oxygen species (ROS) level by 0.93%±0.12% even after exposure to oxidants, and stabilized mitochondrial activity by 1.37%±0.36%. In addition, qPCR results confirmed that the gene expression levels of tumor necrosis factor α (TNFα), TIR domain-containing adapter inducing IFN-beta (TICAM1), and glucose-regulated protein 78 (GRP78) were regulated, which contributed to cell stabilization. Thin-layer chromatography and oil red O analyses showed a clear decrease in the contents of lipid metabolites such as triacylglycerol and free fatty acids. Conclusion: In this study, we confirmed that the examined BT extract exerted selective protective effects on Gallus gallus DF-1 fibroblast cells against cell damage caused by t-BHP, which is a strong oxidative inducer. Furthermore, we established that this extract significantly reduced the intracellular ROS accumulation due to oxidative stress, which contributes to an increase in poultry production and higher incomes.
Purpose: Although automatic quantification software of myocardial perfusion SPECT provides highly objective and reproducible quantitative measurements, there is still some limitation in the direct use of quantitative measurements. In this study we derived parameters using normal variation of perfusion measurements, and tried to test the usefulness of these parameters. Materials and Methods: In order to calculate normal variation of perfusion measurements on myocardial perfusion SPECT, 55 patients (M:F = 28:27) of low-likelihood for coronary artery disease were enrolled and $^{201}TI$ rest/$^{99m}Tc$-MIBI stress SPECT studies were performed. Using 20-segment model, mean (m) and standard deviation (SD) of perfusion were calculated in each segment. As a myocardial viability assessment group, another 48 patients with known coronary artery disease, who underwent coronary artery bypass graft surgery (CABG) were enrolled. $^{201}TI$ rest/$^{99m}Tc$-MIBI stress / $^{201}TI$ 24-hr delayed SPECT was performed before CABG and SPECT was followed up 3 months after CABG. From the preoperative 24-hr delayed SPECT, $Q_{delay}$ (perfusion measurement), ${\Delta}_{delay}$ ($Q_{delay}$ - m) and $Z_{delay}$ (($Q_{delay}$ - m)/SD) were defined and diagnostic performances of them for myocardial viability were evaluated using area under curve (AUC) on receiver operating characteristic (ROC) curve analysis. Results: Segmental perfusion measurements showed considerable normal variations among segments. In men, the lowest segmental perfusion measurement was $51.8{\pm}6.5$ and the highest segmental perfusion was $87.0{\pm}5.9$, and they are $58.7{\pm}8.1$ and $87.3{\pm}6.0$, respectively in women. In the viability assessment $Q_{delay}$ showed AUC of 0.633, while those for ${\Delta}_{delay}$ and $Z_{delay}$ were 0.735 and 0.716, respectively. The AUCs of ${\Delta}_{delay}$ and $Z_{delay}$ were significantly higher than that of $Q_{delay}$ (p = 0.001 and 0.018, respectively). The diagnostic performance of ${\Delta}_{delay}$, which showed highest AUC, was 85% of sensitivity and 53% of specificity at the optimal cutoff of -24.7. Conclusion: On automatic quantification of myocardial perfusion SPECT, the normal variation of perfusion measurements were considerable among segments. In the viability assessment, the parameters considering normal variation showed better diagnostic performance than the direct perfusion measurement. This study suggests that consideration of normal variation is important in the analysis of measurements on quantitative myocardial perfusion SPECT.
Micromechanical facture models can be used to predict ductile fracture in steel structures. In order to calibrate the parameters in the micromechanical models for the largely used Q345 steel in China, uniaxial tensile tests, smooth notched tensile tests, cyclic notched bar tests, scanning electron microscope tests and finite element analyses were conducted in this paper. The test specimens were made from base metal, deposit metal and heat affected zone of Q345 steel to investigate crack initiation in welded steel connections. The calibrated parameters for the three different locations of Q345 steel were compared with that of the other seven varieties of structural steels. It indicates that the toughness index parameters in the stress modified critical strain (SMCS) model and the void growth model (VGM) are connected with ductility of the material but have no correlation with the yield strength, ultimate strength or the ratio of ultimate strength to yield strength. While the damage degraded parameters in the degraded significant plastic strain (DSPS) model and the cyclic void growth model (CVGM) and the characteristic length parameter are irrelevant with any properties of the material. The results of this paper can be applied to predict ductile fracture in welded steel connections.

Purpose: This paper is a basic study done to establish spiritual health concepts according to Korean culture. The focus was on identifying perceived concepts of spiritual health in Korean people. Methods: The Q method, which is effective in measuring individual subjectivity, was used in this study. Results: Perception of the concept of spiritual health was found to have 3 independent types, Type I is a self-directed, present life centered type. This is a group stressing the importance of planning self-directed life and the people of this group consider the values of the present life as important. Type II is a faith-oriented, afterlife centered type. This is a group whose essence of spiritual health is religious devotion like faith in god and the people of this group do not sympathize with the oriental world view. Type III is an oriental, value-sharing type. This is a group with an oriental world view and the people of this group stress importance in sharing values with communities or others. Conclusion: These results not only become the basis for understanding the concept of spiritual health among Koreans, but also suggest the necessity of comprehensive education for spiritual health promotion.
Surface hardening treatments, such as using the high-frequency induction hardening method, are widely used to increase the fatigue life and prevent the failure of materials by locally increasing the surface hardness. This method, in particular, brings an improvement in static strength by compressive residual surface stress due to the hardening. In this study, the mechanical properties of high-frequency induction hardened SCM440 steel were investigated. These results were also compared with those for base metal and a Q/T (tempering after quenching) treatment specimen. The test results showed that partially high-frequency induction hardened SCM440 steel specimens were more improved in static strength, surface hardness, fatigue limit, and anti-wear than the base metal and Q/T treatment specimens. In particular, the fatigue limit of the high-frequency induction hardened SCM440 steel increased by more than about 52% compared to that of base metal and by about 25% compared to that of the Q/T specimen.
FFP(Fitness For Purpose) type defect assessment methodologies based on ECA(Engineering Critical Analysis) have been established and are in use for the structural integrity evaluation of gas pipelines. ECA usually includes the fracture mechanics analysis, and it assumes that J-integral uniquely characterizes crack-tip stress-strain fields. However, it has been shown that it is not sufficient to characterize the crack-tip field under low levels of constraint with a single parameter. Since pipeline structures are made of ductile material, locally loaded in tension, cracks may experience low level of constraint, and therefore, J-dominance will be lost. For this reason, the level of constraint must be quantified to establish a precise assessment procedure for pipeline defects. The objective of this paper is to Investigate the fracture behavior of a crack in gas pipeline by quantifying the level of constraint. For this purpose, tensile tests and CTOD tests were performed at room temperature$(24^{\circ}C)$ and low temperature$(-40^{\circ}C)$ to obtain the material properties. J-Q analyses were performed for SENB and SENT specimens based on 2-D finite element analyses, in order to investigate the in-plane constraint effects on pipeline defects.

The creep crack growth rate (da/dt) of the Cr-Mo steels tested by pre-crack and the voltage (or resistance) variables were related into fracture parameter (Ct), crack growth coefficient (H), and an exponent (q) in the parts of Base, weld and HAZ. The fracture parameter (Ct) has various variables relating to the specimen and crack shape, applied stress, and creep strain curve. The H and q was inferred by OLS regression (ordinary least square method), and the H values were solved in statistics and probability assessment, which were attained fromPDF's distributions (probability density function). The HAZ part has the highest value of q by OLS regression and the widest distribution of H by PDF of WEIBULL, which means that the crack sensitivity of HAZ should be cautioned against the creep crack growth and failure.
This paper deals with the modal analysis of rotational shell structures by means of the numerical solution technique known as the Generalized Differential Quadrature (G. D. Q.) method. The treatment is conducted within the Reissner first order shear deformation theory (F. S. D. T.) for linearly elastic isotropic shells. Starting from a non-linear formulation, the compatibility equations via Principle of Virtual Works are obtained, for the general shell structure, given the internal equilibrium equations in terms of stress resultants and couples. These equations are subsequently linearized and specialized for the rotational geometry, expanding all problem variables in a partial Fourier series, with respect to the longitudinal coordinate. The procedure leads to the fundamental system of dynamic equilibrium equations in terms of the reference surface kinematic harmonic components. Finally, a one-dimensional problem, by means of a set of five ordinary differential equations, in which the only spatial coordinate appearing is the one along meridians, is obtained. This can be conveniently solved using an appropriate G. D. Q. method in meridional direction, yielding accurate results with an extremely low computational cost and not using the so-called "delta-point" technique.

Peroxiredoxins (Prxs) are ubiquitously expressed peroxidases that reduce hydrogen peroxide or alkyl peroxide production in cells. Prxs are released from cells in response to various stress conditions, and they function as damage-associated molecular pattern molecules. However, the secretory mechanism of Prxs and their roles have not been elucidated. Thus, we aimed to determine whether inflammasome activation is a secretory mechanism of Prxs and subsequently identify the effect of the secreted Prxs on activation of the classical complement pathway. Using J774A.1, a murine macrophage cell line, we demonstrated that NLRP3 inflammasome activation induces Prx1, Prx2, Prx5, and Prx6 secretion in a caspase-1 dependent manner. Using HEK293T cells with a transfection system, we revealed that the release of Prx1 and Prx2 relies on gasdermin-D (GSDMD)-mediated secretion. Next, we confirmed the binding of both Prx1 and Prx2 to C1q; however, only Prx2 could induce the C1q-mediated classical complement pathway activation. Collectively, our results suggest that inflammasome activation is a secretory mechanism of Prxs and that GSDMD is a mediator of their secretion. Moreover, secreted Prx1 and Prx2 bind with C1q, but only Prx2 mediates the classical complement pathway activation.
