Characteristics of Wood Tar Produced as Byproduct from Two Types of The Kiln in The Manufacture of Oak Charcoal

  • This study investigated the characteristics of wood tar, produced as a byproduct during the production of charcoal using oak wood by the modified traditional kiln and mechanical steel kiln. The wood tar was analyzed with a number of techniques, including Py-GC/MS, NMR, MALDI-TOF, FT-IR, TG and DSC. The Py-GC/MS analysis indicated that modified traditional kiln generated a higher hydrocarbon ratio in the wood tar than that of mechanical steel kiln. On the other hand, mechanical steel kiln resulted in a higher proportion of phenolic and aromatic hydrocarbon components than that of modified traditional kiln. Those results were also confirmed by NMR analysis. The MALDI-TOF analysis suggested that the wood tar produced in the mechanical steel kiln had a slightly higher molecular weight than the wood tar produced in the modified traditional kiln. In addition, the FT-IR analysis showed characteristic peak of symmetrical stretching vibration of $CH_3$ from the modified traditional kiln while characteristic peaks of the C-C and C-O stretching vibration were observed from the mechanical steel kiln. Moreover, TG and DSC analysis suggested that the mechanical steel kiln is more thermally stable than that of modified traditional kiln. Those findings clearly showed that the method of making charcoal greatly affects the properties of wood tar.

Purification and Characterization of Angiotensin I-Converting Enzyme Inhibitor from Porphyra yezoensis (김으로부터 분리한 Angiotensin-I Converting Enzyme 저해제의 정제 및 특성)

  • This study focused on the purification and characterization of ACE inhibitor from Porphyra yezoensis. The dried Porphyra yezoensis was ground and hydrolyzed with 2.5 N HCl, followed by neutralization and centrifugation. Then, the subsequential purification of ACE inhibitor was carried out by Amberlite XAD 8, DEAE-Toyopearl 650C, Sephadex LH-20 column chromatography and reverse phase HPLC with C18 column. The purified ACE inhibitor was peptide which consisted of glycine (24.5%), arginine (56.8%) and proline (18.8%). Also, it showed the competitive inhibition pattern to ACE. The apparent molecular mass of purified peptide was 580 dalton, and an IC50 value of ACE inhibitor was 10.6 $\mu\textrm{g}$.

Hybrid All-Reduce Strategy with Layer Overlapping for Reducing Communication Overhead in Distributed Deep Learning (분산 딥러닝에서 통신 오버헤드를 줄이기 위해 레이어를 오버래핑하는 하이브리드 올-리듀스 기법)

  • Since the size of training dataset become large and the model is getting deeper to achieve high accuracy in deep learning, the deep neural network training requires a lot of computation and it takes too much time with a single node. Therefore, distributed deep learning is proposed to reduce the training time by distributing computation across multiple nodes. In this study, we propose hybrid allreduce strategy that considers the characteristics of each layer and communication and computational overlapping technique for synchronization of distributed deep learning. Since the convolution layer has fewer parameters than the fully-connected layer as well as it is located at the upper, only short overlapping time is allowed. Thus, butterfly allreduce is used to synchronize the convolution layer. On the other hand, fully-connecter layer is synchronized using ring all-reduce. The empirical experiment results on PyTorch with our proposed scheme shows that the proposed method reduced the training time by up to 33% compared to the baseline PyTorch.

Comparison of genome-wide association and genomic prediction methods for milk production traits in Korean Holstein cattle

  • Objective: The objectives of this study were to compare identified informative regions through two genome-wide association study (GWAS) approaches and determine the accuracy and bias of the direct genomic value (DGV) for milk production traits in Korean Holstein cattle, using two genomic prediction approaches: single-step genomic best linear unbiased prediction (ss-GBLUP) and Bayesian Bayes-B. Methods: Records on production traits such as adjusted 305-day milk (MY305), fat (FY305), and protein (PY305) yields were collected from 265,271 first parity cows. After quality control, 50,765 single-nucleotide polymorphic genotypes were available for analysis. In GWAS for ss-GBLUP (ssGWAS) and Bayes-B (BayesGWAS), the proportion of genetic variance for each 1-Mb genomic window was calculated and used to identify informative genomic regions. Accuracy of the DGV was estimated by a five-fold cross-validation with random clustering. As a measure of accuracy for DGV, we also assessed the correlation between DGV and deregressed-estimated breeding value (DEBV). The bias of DGV for each method was obtained by determining regression coefficients. Results: A total of nine and five significant windows (1 Mb) were identified for MY305 using ssGWAS and BayesGWAS, respectively. Using ssGWAS and BayesGWAS, we also detected multiple significant regions for FY305 (12 and 7) and PY305 (14 and 2), respectively. Both single-step DGV and Bayes DGV also showed somewhat moderate accuracy ranges for MY305 (0.32 to 0.34), FY305 (0.37 to 0.39), and PY305 (0.35 to 0.36) traits, respectively. The mean biases of DGVs determined using the single-step and Bayesian methods were $1.50{\pm}0.21$ and $1.18{\pm}0.26$ for MY305, $1.75{\pm}0.33$ and $1.14{\pm}0.20$ for FY305, and $1.59{\pm}0.20$ and $1.14{\pm}0.15$ for PY305, respectively. Conclusion: From the bias perspective, we believe that genomic selection based on the application of Bayesian approaches would be more suitable than application of ss-GBLUP in Korean Holstein populations.

  • The paramagnetic organoruthenium(III) complexes $({\eta}^5-C_5Me_5)RuCl_2(PR_3) (PR_3 = PMe_3,\;PEt_3,\;PiPr_3,\;PCy_3,\;PMe_2Ph,\;PMePh_2,\;PPh_3,\;P(p-C_6H_4CH_3)_3$, DPPE, DPPB, Py) (2a∼2k) were synthesized by the reaction of $[({\eta}^5-C_5Me_5)RuCl_2]_2$ (1) with 1 equivalent of the corresponding phosphines $(PR_3)$. The effective magnetic moment ((${\mu}_{eff} = 1.65∼2.07 B.M.$)) derived from the magnetic susceptibility measurements of the complexes (2a∼2k) were consistent with the presence of a "single" unpaired electron in the molecule. Treatment of dichlororuthenium (III) complex ({\eta}^5-C_5Me_5)RuCl_2(PR_3)$ (2) (i) with KBr in acetone afforded the dibromoruthenium (III) complex $({\eta}^5-C_5Me_5)RuBr_2(PR_3) (PR_3 = PPh_3)$, (ii) with sodium amalgam in diethylether led to the bis(phosphine) derivatives $({eta}^5-C_5Me_5)RuCl(PR_3)_2 (PR_3 = PMe_3,\;PMePh_2)$, and (iii) with carbonmonoxide gave to the carbonyl derivatives $({\eta}^5-C_5Me_5)RuCl(PR_3)(CO) (PR_3 = PMe_3,\;PPh_3)$.

Instrumental Analysis of Deposits on Paper Machine and Holes/Spots in Paper (제지공정 침착이물질 및 종이내 불순물 성분의 기기분석)

  • The constituents of deposits on paper machine and holes/spots in paper have been analyzed by a combination of analytical techniques, such as FTIR, Py-GC-MS, and EDS. FTIR spectroscopy was used prior to Py-GC-MS and EDS analysis, as a preliminary analysis. The analysis of organic components was carried out with a pyrolysis unit connected to a GC-MS, and inorganic components in ash were analyzed by SEM equipped with an EDS analyzer after pyrolysis at $590^{\circ}C$. The deposits on the dryer section were complex pitch, which was the mixture of the organic components of fatty acid ester and starch, and the inorganic components of talc, clay, and calcium carbonate. The complex pitch was estimated to come from the coated broke. We knew the deposits on the metering rod of sym-sizer were associated with the interaction of unstable alkyl keten dimer(AKD) and $CaCO_3$. The compositions of holes or spots varied considerably and were associated with chemical interaction within the system. The holes, spots, and blotches in the finished paper were PE and PP from pulp sources, complex pitch that were caused by the interaction of the different additives in the system, polymer such as flexible PVC that was used for the prop of palette, and hot melt as adhesives that came from the inadequate handling of broke. In addition, we identified that poly(caprolactam) which is used for forming fabrics or press felts, could be mixed with the raw materials by accident and results in streaks on coating.

Classification of body types of male wheelchair users (휠체어를 사용하는 성인 남성의 체형분류)

  • The purpose of this study was to analyze the physical characteristic of the wheelchair users by directly measuring the disabled men who use a wheelchair and to classify body types. The subjects were 178 male wheelchair users 20-69 years range of age. The results of this study were as follows. The cause of disability was classified into four groups; Poliomyelitis(P), Spinal Cord Injury(S), Cerebral Palsy(C), Amputee(A). There was a remarkable difference in the physical characteristic of the wheelchair users due to their cause of disability. A have greater values in all the dimensions. P have the largest drop values and smaller values in the lower body dimensions. S have greater values in the height of upper body trunk, back interscye length, and chest circumference, whereas smaller values in waist front length and thigh circumference. C have smaller values in most body dimensions except circumference of lower limb. By the factor analysis on the anthropometric data came out the result to be eight factors. The result of cluster analysis using factor scores shows the body types of wheelchair users can be divided into four body types; PY, RBB, TBP, RA. PY type is short and has the largest drop values, whereas RBB type tends to have the smallest drop values. TBP type is tall and the torso tends to be larger than those of the other types. RA type has smaller values in front body dimensions and moderate values in other dimensions. The diversity in body types should be considered among the wheelchair users.

A cold-active acidophilic endoglucanase of Paenibacillus sp. Y2 isolated from soil in an alpine region

  • A cellulolytic strain Y2 was isolated from soil obtained in the Canadian Alpine region. The isolate was identified as Paenibacillus sp. Y2 by 16S rRNA sequencing. When grown in LB medium supplemented with carboxymethyl-cellulose (CMC), CMCase production increased to 122.0% of that observed in LB without CMC. Culture supernatant was concentrated by ultrafiltration and 80% ammonium sulfate precipitates were separated by Hi-Trap Q and CHT-II chromatography. The purified enzyme (EG-PY2) showed a homogeneous single band and the molecular mass was estimated to be 38 kDa by SDS-PAGE. Optimum pH and temperature of the enzyme were 4.5 and $30^{\circ}C$, respectively. The half-life of enzyme activity at 50 was 140.7 min, but the enzyme was drastically inactivated within 5 min at $55^{\circ}C$. The enzyme was highly activated to 135.7 and 126.7% by 5.0 mM of $Cu^{2+}$ or $Mg^{2+}$ ions, respectively, and moderately activated by $Ba^{2+}$ and $Ca^{2+}$ ions, whereas it was inhibited to 76.8% by $Fe^{2+}$, and to ${\leq}50%$ by $Mn^{2+}$, $Co^{2+}$, $Zn^{2+}$, and EDTA. The enzyme was activated to 211.5% in the presence of 0.5 M of NaCl and greatly tolerant to 3.15M of NaCl. The enzyme showed 2.98 times higher ${\beta}$-glucanase activity than CMCase activity. Based on these results, it can be concluded that EG-PY2 is an acidophilic, cold-active, and halotolerant endoglucanase. The authors suggest it is considered to be useful for various industrial applications, such as, fruit juice clarification, acidic deinking processes, high-salt food processing, textile and pulp industries, and for biofuel production from seaweeds.

Characterization and optimum production condition of extracellular protease from Pseudoalteromonas donghaensis HJ51 (Pseudoalteromonas donghaensis HJ51의 체외 단백질 분해효소 특성과 생산 조건)

  • Pseudoalteromonas donghaensis HJ51, isolated from the East Sea, has been reported as a novel strain to produce extracellular protease. Crude supernatant was used to determine optimal activity and optimal production conditions for the enzyme. It was found that the optimal temperature and pH of the protease were $40^{\circ}C$ and pH 7.5-10.5, respectively. The enzyme activity was kept to 88% at the pH 11. In metal requirement analysis, the enzyme exhibited the highest activity when 10 mM $Fe^{3+}$ was supplied. While supplementation of additional carbon sources used in study showed no positive effect on cell growth and enzyme activity, the addition of beef extract, tryptone, or casamino acids instead of peptone of PY-ASW containing 1% glucose increased enzyme production to 21, 7, 4%, respectively. Taken together these properties, the enzyme produced from P. donghaensis HJ51 can be applied to the industries that require protease activity under alkaline pH and low temperature.
