Recently, ex-vivo gene therapy has emerged as a promising approach to enhance the therapeutic potential of mesenchymal stem cells (MSCs) by introducing functional genes in vitro. Here, we explored the need of using selection markers to increase the gene delivery efficiency and evaluated the potential risks associated with their use in the manufacturing process. We used MSCs/CD that carry the cytosine deaminase gene (CD) as a therapeutic gene and a puromycin resistance gene (PuroR) as a selection marker. We evaluated the correlation between the therapeutic efficacy and the purity of therapeutic MSCs/CD by examining their anti-cancer effect on co-cultured U87/GFP cells. To simulate in vivo horizontal transfer of the PuroR gene in vivo, we generated a puromycin-resistant E. coli (E. coli/PuroR) by introducing the PuroR gene and assessed its responsiveness to various antibiotics. We found that the anti-cancer effect of MSCs/CD was directly proportional to their purity, suggesting the crucial role of the PuroR gene in eliminating impure unmodified MSCs and enhancing the purity of MSCs/CD during the manufacturing process. Additionally, we found that clinically available antibiotics were effective in inhibiting the growth of hypothetical microorganism, E. coli/PuroR. In summary, our study highlights the potential benefits of using the PuroR gene as a selection marker to enhance the purity and efficacy of therapeutic cells in MSC-based gene therapy. Furthermore, our study suggests that the potential risk of horizontal transfer of antibiotics resistance genes in vivo can be effectively managed by clinically available antibiotics.
Reduction of risk plays a pivotal role in the development of medical instruments. A hyperbaric oxygen chamber, as a medical device, is known to help medical therapy for diversity of diseases through provision of high purity oxygen. The use of hyperbaric oxygen is expected to increase in the future and study to rigorously examine reliability and safety is needed. We have performed risk assessment for a newly developed hyperbaric oxygen chamber in this study. We first briefly discussed the system structure and concept of risk assessment for the study. Based on the hazards identified, we performed preliminary hazard analysis for the chamber.
Microsatellite 표지를 이용하여 국내에서 시판되고 있는 멜론 58품종의 식별과 멜론 육종 계통 '10H08'을 이용하여 $F_1$ 종자 순도를 평가하였다. 412개의 microsatellite 표지 중 다형성 정도가 높은 29개는 품종 그룹 내에서도 다양한 유전 변이를 나타내었으며 분자표지의 유전자형에 의해 모든 품종을 식별할 수 있었다. Microsatellite 표지의 대립유전자를 이용하여 멜론 58품종에 대한 계통도를 작성하였을 때 멜론의 형태적 특성과 일치하면서 2개의 대그룹으로 구분되었다. $F_1$ 종자의 순도 검정에 microsatellite 표지를 활용하기 위하여 29개의 표지를 '10H08' 계통의 양친에 대하여 검정하였을 때 5개의 프라이머가 다형성을 보였으며, 이 중 한 개의 프라이머 'CMGAN12'는 양친간에 뚜렷한 다형성 밴드를 나타내었다. 이 프라이머를 192개의 $F_1$ 종자에 대하여 검정하였을 때 자식주로 보이는 개체가 분석된 종자 내에서 명확하게 구분되었다. 본 연구 결과에서 선정된 멜론 품종 식별용 microsatellite 표지는 멜론 품종의 지문화뿐만 아니라 $F_1$ 종자의 순도 검정이 가능하여 종자회사에서 매우 유용하게 활용될 수 있는 것으로 나타났다.
EBR-II사용후핵연료의 파이로건식처리공정에 의해 발생된 우라늄의 순도에 대한 포괄적인 분석을 수행하였다. 분석 결과를 미국 아이다호 국립연구소 및 한국원자력 연구원의 협력과제 하에서 한국과 미국의 저준위 폐기물 기준으로 비교하였다. 미국의 저준위 폐기물 기준은 우라늄 등위원소를 포함하지 않으나, 한국의 경우는 포함하는 것으로 조사되었다. 분석 결과 EBR-II 우라늄 생성물 내에서 저준위 기준을 초과하는 유일한 알파 핵종은 우라늄 동위원소가 아니라 Pu-239였다. 생성물 내의 Pu 오염은 개량된 염증류공정을 통한 예비실험 결과 획기적으로 줄일 수 있음을 알 수 있었으며, 보다 공정을 개선시킨다면 제안된 기술을 이용하여 미국의 저준위 기준을 만족시킬 수 있을 것으로 판단된다.
반도체 산업용 9N 이상의 초고순도 N2, Ar 등 불활성 가스 제조를 위해 가스 정제공정에 사용되고 있는 Ni 촉매의 물성 평가 및 촉매적 특성을 확인하였다. 조성이 다른 원기둥 형태의 C1, 츄러스 형태의 C2의 두 가지 Ni 촉매에 대해 비교 평가를 진행하였다. Ni 촉매의 형상과 미세구조를 분석하기 위해 광학현미경과 FE-SEM을 이용하였으며, 조성 확인 및 물성을 분석하기 위해 EDS, XRD, 그리고 micro-Raman 분석을 이용하였다. 또한 Ni 촉매의 비표면적 및 촉매적 특성을 확인하기 위해 BET, Pulse Titration 분석을 진행하였다. 조성 분석결과, C1의 경우, 상대적으로 graphite가 불순물로 다량 포함되어 있는 것을 확인하였으며, C2는 C1에 비해 Ni의 함량이 높은 것을 확인하였다. 비표면적 분석 결과, C2의 비표면적이 C1보다 약 1.69배 정도 큰 것을 확인할 수 있었다. 촉매적 특성분석 결과, 상온에서 O2와 CO 불순물 제거 정도가 C2가 우수함을 확인하였다. 따라서 반도체 산업용 초고순도 불활성 기체 제조를 위한 Ni 촉매로는 불순물이 적고, 비표면적이 크며, 상온에서 O2와 CO 제거 성능이 우수한 C2가 적합함을 확인하였다.
상용화된 자동합성장치는 사용되는 유기용매의 종류가 다르고 합성수율에 차이를 보인다. 따라서 본 연구에서는 자동합성장치에 따른 $^{18}F$-FDG의 방사선분해에 관한 방사화학적순도 변화를 비교하였다. Cyclotron (PETtrace, GE Healthcare)을 사용하여 $^{18}F$를 생산하고, 자동합성장치(FASTlab, Tracerlab MX, GE Healthcare)를 이용하여 FDG로 합성하였다. 방사화화적순도는 Radio-TLC Scanner (AR 2000, Bioscan), GC(Gas Chromatography, Agilent 7890A)를 사용하여 $^{18}F$-FDG에 함유되어 있는 에탄올의 양을 측정하였다. 고정상은 실리카겔로 도포된 유리판($1{\times}10cm$), 이동상은 아세토니트릴과 물 19:1 혼합액을 사용하고, 각각의 합성장치에서 고농도와 저농도의 $^{18}F$-FDG를 생산 후 2시간 간격으로 방사화학적순도를 측정하였다. 저농도 (약 2.59 GBq/mL 이하)에서 순도변화는 Tracerlab MX에서는 99.26%, 98.69%, 98.25%, 98.09%, FASTlab에서는 99.09%, 97.83, 96.89%, 96.62%를 얻었다. 고농도(약 3.7 GBq/mL 이상)에서 순도변화는 Tracerlab MX에서는 평균 99.54%, 96.08%, 93.77%, 92.54%, FASTlab의 경우 99.53%, 95.65%, 92.39%, 89.82%를 얻었다. 그리고 FASTlab에서 생산한 $^{18}F$-FDG의 GC에서는 에탄올이 검출되지 않았으며, Tracerlab MX에서는 100~300 ppm의 에탄올이 검출되었다. 이러한 결과를 비추어 봤을 때 방사선 보호제인 에탄올의 유무보다 방사능농도가 방사선분해에 더 큰 영향을 미치기 때문에 고농도의 $^{18}F$-FDG 생산 후 무균 생리식염수로 희석하여 농도를 낮춘 후 사용해야 한다.

The use of hyperbaric oxygen chamber is expected to increase in the future and study to examine reliability and safety is needed. We have performed reliability assessment for a newly developed hyperbaric oxygen chamber in this study. We first briefly discussed the system structure and mechanism. We then performed FMEA (Failure Mode and Effect Analysis) for the chamber. We drew major failure modes affecting the system performance and performed in depth analysis for measuring the expected effects.
This study aims to estimate chlorophyll-a concentration in rivers using multi-spectral RapidEye imagery and Spectral Mixture Analysis (SMA) and assess the applicability of SMA for multi-temporal imagery analysis. Comparison between images (acquired on Oct. and Nov., 2013) predicted and ground reference chlorophyll-a concentration showed significant performance statistically with determination coefficients of 0.49 and 0.51, respectively. Two band (Red-RE) model for the October and November 2013 RapidEye images showed low performance with coefficient of determinations ($R^2$) of 0.26 and 0.16, respectively. Also Three band (Red-RE-NIR) model showed different performance with $R^2$ of 0.016 and 0.304, respectively. SMA derived Chlorophyll-a concentrations showed relatively higher accuracy than band ratio models based values. SMA was the most appropriate method to calculate Chlorophyll-a concentration using images which were acquired on period of low Chlorophyll-a concentrations. The results of SMA for multi-temporal imagery showed low performance because of the spatio-temporal variation of each end members. This approach provides the potential of providing a cost effective method of monitoring river water quality and management using multi-spectral imagery. In addition, the calculated Chlorophyll-a concentrations using multi-spectral RapidEye imagery can be applied to water quality modeling, enhancing the predicting accuracy.
Background: Food consumption is one of the most important routes for radionuclide intake for the public; therefore, there is the need to have a comprehensive understanding of the amount of radioactivity in food products. Consumption of radionuclide-contaminated food could increase potential health risks associated with exposure to radiation such as cancers. The present study aims to determine radioactivity levels in some food products (milk, rice, sugar, and wheat flour) consumed in Mali and to evaluate the radiological effect on the public health from these radionuclides. Materials and Methods: The health impact due to ingestion of radionuclides from these foods was evaluated by the determination of activity concentration of radionuclides 238U, 232Th, 40K, and 137Cs using gamma spectrometry system with high-purity germanium detector and radiological hazards index in 16 samples collected in some markets, mall, and shops of Bamako-Mali. Results and Discussion: The average activity concentrations were 9.8±0.6 Bq/kg for 238U, 8.7±0.5 Bq/kg for 232Th, 162.9±7.9 Bq/kg for 40K, and 0.0035±0.0005 Bq/kg for 137Cs. The mean values of radiological hazard parameters such as annual committed effective dose, internal hazard index, and risk assessment from this work were within the dose criteria limits given by international organizations (International Commission on Radiological Protection and United Nations Scientific Committee on the Effects of Atomic Radiation) and national standards. Conclusion: The results show low public exposure to radioactivity and associated radiological impact on public health. Nevertheless, this study stipulates vital data for future research and regulatory authorities in Mali.
This study applied the Neural Network and Fuzzy theory to show water-purity control and preventive measure in water quality forecasting of the future river. This study picked out NAJU and HAMPYUNG as the subject of investigation and used monthly the water quality and the outflow data of KWANGJU2, NAJU, YOUNGSANNPO and HAMPYUNG from 1995 to 1999 to forecast BOD, COD, T-N, T-P water density. The datum from 1995 to 1999 are used for study and that of 2000 are used for verification. To develop model of water quality forecasting, firstly, this research formed Neural Network model and divided Neural Network model into two case - the case of considering lag and not considering. And this study selected optimal Neural Network model through changing the number of hidden layer based on input layer(n) from n to 3n. Through forecasting result, the case without considering lag showed more precise simulated result. Accordingly, this study intended to compare, analyse that Fuzzy model using the method without considering lag with Neural Network model. As a result, this study found that the model without considering lag in Neural Network Network shows the most excellent outcome. Thus this study examined a forecasting accuracy, analyzed result and verified propriety through appling the method of water quality forecasting using Neural Network and Fuzzy Algorithms to the actual case.
