Simple Purification of Bromelain from Pineapple

  • Bromelain(EC 3.4.22.4), the collective name for the proteolytic enzymes found in tissues of the plant family Bromeliaceae(pineapple), has been used as a tenderizing agent in food processing, and as an antiinflammatory agent in pharmaceuticals. In this paper, we describe the simple purification method of bromelain using Korean pineapple fruit. After removing contaminants at 30% saturation of ammonium sulfate, the supernatant obtained was treated again with ammonium sulfate to 80% saturation. Wit the above salt fractionation, partially purified bromelain could be obtained. To get highly purified bromelain, the previous 30% to 80% ammonium sulfate treated precipitate was dialyzed against 25mM sodium acetate buffer(pH 5.0) followed by passing through a CM- cellulose cation exchange column. Fruit bromelain was eluted as a major peak at 0.5~0.8M NaCI gradient. The present method is simpler with high wield than the traditional purification method-acetone treatment and several consecutive chromatographic processes.

A Novel Purification Process for Homoharringtonine from Celphalotaxus koreana

  • An effective purification method was developed for producing Homoharringtonine (HHT), to guarantee high purity and yield from Cephalotaxus koreana. This process was a simple and efficient procedure, for the isolation and purification of HHT form the biomass of Cephalotaxus koreana, consisting of extract, adsorbent treatment, precipitation and followed by a chromatography. The extraction, adsorbent treatment and precipitation in pre-purification process allows for rapid and efficient separation of HHT from many compound and dramatically increases the yield and purity of crude HHT for HPLC purification steps compared to alternative processes. This purification processes serves to minimize solvent usage, size, and complexity of the operations for HHT purification.

  • 식물세포배양으로부터 항암제 paclitaxel의 생산을 위해 회수 및 정제는 산업적 공정에 있어서 필수적이다. 본 총설은 식물세포배양으로부터 고순도, 고수율의 paclitaxel 생산을 위한 대량 회수 및 정제 방법을 기술하고자 한다. 또한 이러한 분리 및 정제 공정은 추출, 전 처리, 정제, 제품화 단계를 총괄하여 최종 제품의 요구조건들 즉, 순도, 잔류용매, 제품형태, 불순물 함량, 엔도톡신 함량 등을 충족시킬 수 있도록 최적화되어야 한다. 이러한 관점에서 본 총설은 산업화 단계에서의 의약품 생산 및 품질관리에 상당히 유용하게 활용될 수 있을 것으로 판단된다.

  • To determinate self-purification constants of pollutants loaded in the ecosystems, the self- purification process was formulated, and a measurement method of the self-purification constants was derived. $C=C_0e^{-st}$ When $C_0$ is the initial pollutant amounts loaded in a ecosystem, and C is the rest pollutant amounts after the time, t, the equation of the self-purification, s, is $s=\frac{P}{C}$ When in aquatic ecosystem, $C_0$ is the initial polluant amounts loaded in water body, and Cis the rest pollutation amounts after the time, t, the self-purification constant, s, is $s=(\frac{\ln C_0-\ln C}{t}$ Self-purification constants of pine and oak forests at kwangneung in kyonggido were 0.07 and 10 respectively, of BOD in gokneung stream in kyonggido was 0.51, and of glucose and phosphate in pools on the stone in mt.jiri were 0.49 and 15.19 respectively.

Identification and Characterization of Homoharringtonine from Cephalotaxus koreana

  • 한국산 개비자나무로부터 새로운 개념의 homoharringtonine 분리 및 정제 공정을 개발하였다. 메탄올을 사용한 4회 반복 추출에 의해 biomass로부터 대부분(>99%)의 homoharringtonine을 회수 할 수 있었다. 흡착 공정에서는 활성 백토(active clay)를 사용하여 추출물에 포함되어 있는 식물유래 타르, 왁스 성분을 효과적으로 제거하였다. Silica gel low-pressure chromatography공정을 통해서 순도 52% 이상의 homoharringtonine을 얻었으며, HPLC 공정을 통해 고순도 및 고수율의 homharringtonine을 정제할 수 있었다. 정제된 homoharringtonine의 분자량 및 구조분석을 수행한 결과 표준물질(Homharringtonine)과 동일 물질임을 확인할 수 있었다.

가다랭이 안와조직으로부터 DHA의 추출 및 정제 (Isolation and Purification of DHA from Skipjack Orbital Tissue Oil)

  • 가다랭이 안와유로부터 docosahexaenoic acid(DHA)를 정제하기 위해 기존의 방법들을 적용하여 비교 검토하고, DHA의 효과적인 정제를 위해 조작방법을 개량하였다. 가다랭이 안와조직의 총지질은 $55.4\%$였으며, 이 중 DHA는 $23.7\%$였다. 저온분별결정법과 요소결정법을 적용한 결과 순도에서 각각 약 $46\%$$61\%$의 DHA가 얻어졌다. 이들 방법들은 순도면에서는 다소 떨어지나, 정제조작이 단순하여 다량의 DHA 분리에 적합하였다. 질산은 수용액법은 상기 2가지 방법에 비하여 순도면에서는 약간 개선되었으나, 회수율이 대단히 낮았다($10\%$ 이하). 질산은 함침 실리카 칼럼 크로마토그래피법은 고순도 DHA의 정제방법으로써 적합하였다(순도 $98\%$ 이상, 회수율 $90\%$ 이상). 결과적으로 저온분별결정법과 질산은 함침 실리카 칼럼 크로마토그래피법을 조합한 개량법(2단계 정제법)이 가다랭이 안와유로부터 고순도 DHA($99.9\%$)의 정제를 위한 가장 효과적인 방법으로 평가되었다.

One-Step Purification of Melittin Derived from Apis mellifera Bee Venom

  • The concern over the use of melittin in honey bee venom due to its adverse reaction caused by allergens such as phospholipase A2 ($PLA_2$) and hyaluronidase (HYA) has been an obstacle towards its usage. We developed a novel single-step method for melittin purification and the removal of $PLA_2$ and HYA. This study explores the influence of pH, buffer compositions, salt concentration, and types of cation-exchange chromatography resins on the recovery of melittin and the removal of both HYA and $PLA_2$. Melittin was readily purified with a strong cation-exchange resin at pH 6.0 with sodium phosphate buffer. It resulted in a recovery yield of melittin up to 93% (5.87 mg from a total of 6.32 mg of initial melittin in crude bee venom), which is higher than any previously reported studies on melittin purification. $PLA_2$ (99%) and HYA (96%) were also successfully removed. Our study generates a single-step purification method for melittin with a high removal rate of $PLA_2$ and HYA, enabling melittin to be fully utilized for its therapeutic purposes.

극저온 정제시스템의 품질관리를 위한 SF6 가스 분석방법 개선 (Improved sulfur hexafluoride(SF6) gas analysis method for quality management of cryogenic refinement system)

  • Because sulfur hexafluoride(SF6) is classified as one of the six major greenhouse gases, SF6 handling in power plant such as recovery, purification, and reuse is considered to be important. KEPCO has focused to develop the advanced recovery and purification technology of SF6 reuse. SF6 analysis includes the on-site analyses and on-line analyzer; i.e., (1) on-site analysis has an error rate of ±0.5% and (2) on-line analysis has an error rate of ±0.1%, which is possible to adjust operating conditions and to make the work more conveniently by analyzing SF6 concentration before and after purification step. This paper presents an online analysis method in the SF6 purification and reuse system. In addition, the analysis results and quality guarantees for each section of the analysis system were presented.

적대적 공격을 방어하기 위한 StarGAN 기반의 탐지 및 정화 연구 (StarGAN-Based Detection and Purification Studies to Defend against Adversarial Attacks)

  • 인공지능은 빅데이터와 딥러닝 기술을 이용해 다양한 분야에서 삶의 편리함을 주고 있다. 하지만, 딥러닝 기술은 적대적 예제에 매우 취약하여 적대적 예제가 분류 모델의 오분류를 유도한다. 본 연구는 StarGAN을 활용해 다양한 적대적 공격을 탐지 및 정화하는 방법을 제안한다. 제안 방법은 Categorical Entropy loss를 추가한 StarGAN 모델에 다양한 공격 방법으로 생성된 적대적 예제를 학습시켜 판별자는 적대적 예제를 탐지하고, 생성자는 적대적 예제를 정화한다. CIFAR-10 데이터셋을 통해 실험한 결과 평균 탐지 성능은 약 68.77%, 평균정화성능은 약 72.20%를 보였으며 정화 및 탐지 성능으로 도출되는 평균 방어 성능은 약 93.11%를 보였다.

반응표면 분석법을 이용한 천연마섬유보강 순환굵은골재 콘크리트의 성능 평가 (Performance Evaluation of Natural Jute Fiber Reinforced Recycled Coarse Aggregate Concrete Using Response Surface Method)

  • In this study, evaluated ware the strength and durability of the vegetated water purification channel concrete to which recycled aggregates, hawang-toh and jute were applied. Box-Behnken method of response surface analysis in statistics was applied to the experimental design. Experimental variables are as follows, recycled coarse aggregates, hawang-toh, blast-furnace slag and jute fiber. In the experiment, conducted were the tests of compressive strength, chloride ion penetration, abrasion resistance and impact resistance the replacement rate effects of the recycled aggregates, blast-furnace slag and hwang-toh on the performance of vegetated water purification channel concrete were analyzed by using the response surface analysis method on the basis of the experimental results. In addition, an optimum mixing ratio of vegetated water purification channel concrete was determined by using the experimental results. The optimum mixing ratio was determined to be in 10.0% recycled coarse aggregates, 60.0% blast-furnace slag, 10.1% hwang-toh and 0.16% jute fiber. The compressive strength, chloride ion penetration, abrasion rate, and impact number of fracture test results of the optimum mixing ratio were 24.1 MPa, 999 coulombs, 10.30 g/mm3, and 20 number, respectively.
