Genetic characterization and population structure of six brown layer pure lines using microsatellite markers

  • Objective: The first stage in both breeding and programs for the conservation of genetic resources are the identification of genetic diversity in the relevant population. The aim of the present study is to identify genetic diversity of six brown layer pure chicken lines (Rhode Island Red [RIRI, RIRII], Barred Rock [BARI, BARII], Columbian Rock [COL], and line 54 [L-54]) with microsatellite markers. Furthermore, the study aims to employ its findings to discuss the possibilities for the conservation and sustainable use of these lines that have been bred as closed populations for a long time. Methods: In the present study, a total number of 180 samples belonging to RIRI (n = 30), RIRII (n = 30), BARI (n = 30), BARII (n = 30), L-54 (n = 30), and COL (n = 30) lines were genotyped using 22 microsatellite loci. Microsatellite markers are extremely useful tools in the identification of genetic diversity since they are distributed throughout the eukaryotic genome in multitudes, demonstrate co-dominant inheritance and they feature a high rate of polymorphism and repeatability. Results: In this study, we found all loci to be polymorphic and identified the average number of alleles per locus to be in the range between 4.41 (BARI) and 5.45 (RIRI); the observed heterozygosity to be in the range between 0.31 (RIRII) and 0.50 (BARII); and $F_{IS}$ (inbreeding coefficient) values in the range between 0.16 (L-54) and 0.46 (RIRII). The $F_{IS}$ values obtained in this context points out to a deviation from Hardy-Weinberg equilibrium due to heterozygote deficiency in six different populations. The Neighbour-Joining tree, Factorial Correspondence Analysis and STRUCTURE clustering analyzes showed that six brown layer lines were separated according to their genetic origins. Conclusion: The results obtained from the study indicate a medium level of genetic diversity, high level inbreeding in chicken lines and high level genetic differentiation between chicken lines.

  • In order to prevent collision accidents of ships, which has been increasing recently, research on artificial intelligence-based autonomously operated ships (Maritime Autonomous Surface Ship, MASS) is underway. However, most of the studies related to autonomous ships mainly target medium-to-large ships due to the size and cost of the autonomous navigation system, and the sensors used here have a problem in that it is difficult to mount them on small ships. Therefore, this paper provides a path tracking system equipped with GPS and IMU sensors for autonomous operation of small ships. GPS and IMU sensors are utilized to determine the exact position of the vessel, which allows the proposed system to manually control the small vessel model to create a path and then when the small vessel travels the same path. Use the Pure Pursuit algorithm to follow the path. As a result, In this research, it is expected that a lightweight and low-cost sensor can be used to develop an autonomous operation system for small ships at low cost.

Sol-gel synthesis, computational chemistry, and applications of Cao nanoparticles for the remediation of methyl orange contaminated water

  • Nanoparticles are known for their outstanding properties such as particle size, surface area, optical and electrical properties. These properties have significantly boasted their applications in various surface phenomena. In this work, calcium oxide nanoparticles were synthesized from periwinkle shells as an approach towards waste management through resource recovery. The sol gel method was used for the synthesis. The nanoparticles were characterized using X-Ray diffractometer (XRD), Fourier Transformed Infra-Red Spectrophotometer (FTIR), Brunauer Emmett Teller (BET), scanning electron microscopy (SEM), transmission electron microscopy (TEM) and ultra violet visible spectrophotometer (UV-visible). While DLS and SEM underestimate the particle diameter, the BET analysis reveals surface area of 138.998 m2/g, pore volume = 0.167 m3/g and pore diameter of 2.47 nm. The nanoparticles were also employed as an adsorbent for the purification of dye (methyl orange) contaminated water. The adsorbent showed excellent removal efficiency (up to 97 %) for the dye through the mechanism of physical adsorption. The adsorption of the dye fitted the Langmuir and Temkin models. Analysis of FTIR spectrum after adsorption complemented with computational chemistry modelling to reveal the imine nitrogen group as the site for the adsorption of the dye unto the nanomaterials. The synthesized nanomaterials have an average particle size of 24 nm, showed a unique XRD peak and is thermally and mechanically stable within the investigated temperature range (30 to 70 ℃).

Production and Properties of Ag Metallic Nanoparticle Fluid by Electrical Explosion of Wire in Liquid (유체 내 전기선폭발법에 의한 은 나노입자 유체의 제조 및 특성)

  • This paper presents a novel single-step method to prepare the Ag nanometallic particle dispersed fluid (nanofluid) by electrical explosion of wire in liquid, deionized water (DI water). X-ray diffraction (XRD), field emission scanning electron microscope (FE-SEM) and transmission electron microscope (TEM) were used to investigate the characteristics of the Ag nanofluids. Zeta potential was also used to measure the dispersion properties of the as-prepared Ag nanofluid. Pure Ag phase was detected in the nanofluids using water. FE-SEM analysis shows that the size of the particles formed in DI water was about 88 nm and Zeta potential value was about -43.68 without any physical and chemical treatments. Thermal conductivity of the as-prepared Ag particle dispersed nanofluid shows much higher value than that of pure DI water.

Ethanol Fermentation of the Enzymatic Hydrolysates from the Products Pretreated using [EMIM]Ac and Its Co-Solvents with DMF

  • Ethanol fermentation of the enzymatic hydrolysates from the products pretreated using 1-ethyl-3-methyl-imidazolium acetate ([EMIM]Ac) and its co-solvents with dimethylformamide (DMF) was conducted using Saccharomyces cerevisiae (D452-2). The optical density change due to the yeast cell growth, the consumption amount of monosugars (glucose, xylose), the concentration of acetate, and ethanol production yield were investigated. The co-solvent system lowered inhibition of the growth of the cells. The highest concentration of glucose (7.8 g/L) and xylose (3.6 g/L) was obtained from the enzymatic hydrolysates of the pretreated product by pure [EMIM]Ac. The initial concentration of both monosugars in the enzymatic hydrolysates was decreased with increasing fermentation time. Ethanol of Approximately 3 g/L was produced from the enzymatic hydrolysates by pure [EMIM]Ac and co-solvent with less than 50% DMF.

The Effect of Different Sources of Urease Enzyme on the Nutritive Value of Wheat Straw Treated with Urea as a Source of Ammonia

  • Wheat straw samples (3-4 cm) were sprayed with solutions of urea (U) alone or with a dry addition of garden soil (GS), midden soil (MS), soya bean meal (SM) or jack bean meal (JM) as crude urease sources and with a pure urease (UR) enzyme. Each of the urease sources was included at two levels: 30 and 60 g/kg except pure urease, which was added at a level of 2.5 & 5.0 g/kg treated straw dry matter. Untreated straw without urease source was used as a control. After treatment, samples were sealed in polythene bags and stored for 2, 7, 14, 21 and 35 days at $19{^{\circ}C}$. The urease sources, their levels and treatment time produced significant effects on ammonia production (p<0.01). The addition of urease offered more flexibility in hydrolyzing urea in the shortest possible time. Incorporation of soya bean and jack bean meal was effective in reducing the modified acid detergent fiber (MADF) content of straw and the same time increasing organic matter (OM) digestibility. Overall effect, addition of soya bean to urea at a ratio of 1:1 appeared to be the most satisfactory urease source for the treatment of urea and wheat straw.

Optimum Condition of Extracting Collagen from Chicken Feet and its Characetristics

  • The objective of this research was to evaluate alternative treatments for the best extraction condition for collagen from chicken feet. Various properties such as chemical composition, amino acid, pH, swelling percentage, yield and pure collagen, collagen loss, color (Hunter L, a and b) and electrophoresis of collagen from chicken feet treated by 5% acids (acetic acid, citric acid. hydrochloric acid and lactic acid) and soaking times (12, 24, 36 and 48 h) were evaluated. The crude protein, fat, ash and moisture contents of chicken feet was 17.42, 12.04, 5.98 and 62.05%, respectively. Amino acid composition of collagen from chicken feet indicated that the protein of collagen was markedly hydrolized by the hydrochloric acid treatment. The result of electrophoresis also supported this phenomenon. Both the swelling percentage of lactic acid and citric acid treatments were significantly higher than that of acetic acid and HC1 treatment. The pH of the acid treatments ranged from 2.43-3.62. According to the result of yield, pure collagen and loss of collagen, the best condition of extracting collagen from chicken feet was soaked in 5% lactic acid for 36 h. However, a brighter yellow color of collagen from all treatments was observed with a longer soaking time.

  • Structures of dimers of the acetonitrile and DMF diluted by carbon-tetrachloride and xenon matrices at 80K were studied with infrared absorption spectroscopy. Sample and matrix materials were codeposited onto a KBr cold trapping window. Infrared absorption spectra of pure samples and diluted samples were compared to explain the structures of dimers. The acetonitrile dimer showed the antiparallel shape and the DMF dimer showed the linear shape. After annealing, the infrared spectrum of the diluted sample showed the same shape with that of the pure sample.

  • When $^{14}C-2$, 6-Diethylaniline (2, 6-DEA) was incubated aerobically in soil, $^{14}CO_2$ evolved from non-sterile soil A and B was 6.5 and 10.1%, respectively, in the 21st week. Methanol could extract 3.1 and 13.5% of the radioactivity from soil A and B, respectively, 2, 6-Diethylacetanilide was detected as a degradation product in soil. Chaetomium globosum produced 2, 6-diethyl-p-benzoquinone as a degradation product in pure culture. A possible pathway was proposed to include p-hydroxylation of 2, 6-DEA, formation of quinoneimine, and the subsequent hydrolysis with the release of ammonia.

Development of 500W Inverter with Pure Sine Wave Output (500W급 순수사인파 인버터 설계)

  • This paper is elaborates on the pure sine wave single phase inverter of 500W level which is driven by battery. The inverter is structured to raise the battery voltage to 400V by using the push-pull topology, and by using H-bridge with the high voltage, it outputs common 220VAC. This topology is utilized to use the power semiconductor device in order to design and manufacture the inverter of 500W at input voltage rating of 12VDC. As a result, the efficiency exceeds 90% in partial sections, but on average, the efficiency is approximately 89.5%, and as for output voltage, frequency fluctuation range and THD, the result can be less than ${\pm}5%$.
