In Vitro Antitumor Properties of an Isolate from Leaves of Cassia alata L

  • Leaf extracts of Cassia alata L (akapulko), traditionally used for treatment of a variety of diseases, were evaluated for their potential antitumor properties in vitro. MTT assays were used to examine the cytotoxic effects of crude extracts on five human cancer cell lines, namely MCF-7, derived from a breast carcinoma, SK-BR-3, another breast carcinoma, T24 a bladder carcinoma, Col 2, a colorectal carcinoma, and A549, a nonsmall cell lung adenocarcinoma. Hexane extracts showed remarkable cytotoxicity against MCF-7, T24, and Col 2 in a dose-dependent manner. This observation was confirmed by morphological investigation using light microscopy. Further bioassay-directed fractionation of the cytotoxic extract led to the isolation of a TLC-pure isolate labeled as f6l. Isolate f6l was further evaluated using MTT assay and morphological and biochemical investigations, which likewise showed selectivity to MCF-7, T24, and Col 2 cells with $IC_{50}$ values of 16, 17, and 17 ${\mu}g/ml$, respectively. Isolate f6l, however, showed no cytotoxicity towards the non-cancer Chinese hamster ovarian cell line (CHO-AA8). Cytochemical investigation using DAPI staining and biochemical investigation using terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL)-a method used to detect DNA fragmentation-together with caspase assay, demonstrated apoptotic cell death. Spectral characterization of isolate f6l revealed that it contained polyunsaturated fatty acid esters. Considering the cytotoxicity profile and its mode of action, f6l might represent a new promising compound with potential for development as an anticancer drug with low or no toxicity to non-cancer cells used in this study.

  • In this study, hygroelastic behavior of thermosetting epoxy is predicted by molecular dynamics simulations. Since consistent exposures to humid environments lead to macroscopic degradation of polymer composite, computational simulation study of the hygroscopically aged epoxy cell is essential for long-time durability. Therefore, we modeled amorphous epoxy molecular unit cell structures at a crosslinking ratio of 30, 90% and with the moisture weight fraction of 0, 4 wt% respectively. Diglycidyl ether of bisphenol F (EPON862) and triethylenetetramine (TETA) are chosen as resin and curing agent respectively. Incorporating equilibrium and non-equilibrium ensemble simulation with a classical interatomic potential, various hygroelastic properties including diffusion coefficient of water, coefficient of moisture expansion (CME), stress-strain curve and elastic modulus are predicted. To establish the structural property relationship of pure epoxy, free volume and internal non-bond potential energy of epoxy are examined.

In vitro Biofumigation of Brassica Tissues Against Potato Stem Rot Caused by Sclerotinia sclerotiorum

  • Sclerotinia sclerotiorum is a serious pathogen which causes yield loss in many dicotyledonous crops including potato. The objective of this study was to assess the potential of biofumigation using three Brassica crops including Brassica napus, B. juncea and B. campestris against potato stem rot caused by S. sclerotiorum by in vitro tests. Both macerated and irradiated dried tissues were able to reduce radial growth and sclerotia formation of five pathogen isolates on PDA, but macerated live tissues were more effective. Compared with other tested crops, B. juncea showed more inhibitory effect against the pathogen. The volatile compounds produced from macerated tissues were identified using a gas chromatograph-mass spectrometer. The main identified compounds were methyl, allyl and butyl isothiocyanates. Different concentrations of these compounds inhibited mycelial growth of the pathogen in vitro when applied as the vapor of pure chemicals. A negative relationship was observed between chemicals concentrations and growth inhibition percentage. In this study, it became clear that the tissues of local Brassica crops release glucosinolates and have a good potential to be used against the pathogen in field examinations.

  • Electrochemical migration characteristics of Pb-free solder alloys are quantitatively correlated with corrosion characteristics in harsh environment conditions. In-situ water drop test and corrosion resistance test for Sn-3.0Ag-0.5Cu solder alloys were carried out in NaBr and NaF solutions to obtain the electrochemical migration lifetime and pitting potential, respectively. Sn-3.0Ag-0.5Cu solder alloy shows similar ionization and electrochemical migration behavior with pure Sn because of Ag and Cu do not migrate due to the formation of resistant intermetallic compounds inside solder itself. Electrochemical migration lifetime in NaBr is longer than in NaF, which seems to be closely related to higher pitting potential in NaBr than NaF solution. Therefore, it was revealed that electrochemical migration lifetime of Sn-3.0Ag-0.5Cu solder alloys showed good correlation to the corrosion resistance, and also the initial ionization step at anode side is believed to be the rate-determining step during electrochemical migration of Pb-free solders in these environments.

  • Kinetic studies of nucleophilic substitution reaction of substituted benzoyl cyanides and benzoyl chlorides with pyridines were conducted at 25$^{\circ}C$ in pure acetone solvent. Results showed that (ⅰ) magnitudes of $_{\rho}_S$, $_{\rho}_N$ and ${\beta}$ associated with a change of substituent in the nucleophile indicate relatively advanced bond-formation in the transition state, (ⅱ) the potential energy surface model is able to predict the reaction mechanism, but it is unable to predict the transition state variation to a more product-like transition state, where bond-formation is much more progressed than bond breaking, upon changing the leaving group to that with better leaving ability (ⅲ) the quantum mechanical model predicted the product-like transition state and slightly better leaving ability of CN- as compared with Cl-.

Biodegradation Study of Gasoline Oxygenates by Butane-Utilizing Microorganisms (부탄 분해 미생물을 이용한 휘발유 첨가제의 분해특성)

  • In this study, potential degradation of MTBE and other gasoline oxygenates by pure culture ENV425 and mixed culture isolated from gasoline contaminated soil using butane as the sources of carbon and energy was examined and compared. Butane monooxygenases(BMO) of butane-grown ENV425 and mixed culture generated 1-butanol as a major metabolite of butane oxidation and addition of acetylene, specific inhibitor of monooxygenase, inhibited both butane oxidation and 1-butanol production. The results described in this study suggest that alkanes including propane, pentane, and butane are effectively utilized as a growth substrate to oxidize MTBE cometabolically. And also BTEX compounds could be the potential substrate of the MTBE cometabolism. Cell density also affected on the MTBE degradation and transformation capacity(Tc). Increasing cell density caused increasing MTBE degradation but decreased transformation capacity. Other result demonstrated that MTBE and other gasoline oxygenates, ETBE and TAME, were degraded by butane-grown microorganism.

Calcium Current in the Unfertilized Egg of the Hamster

  • The presence of a calcium current $(i_{Ca^{2+}})$ passed via a specific channel was examined in the unfertilized hamster egg using the whole-cell voltage clamp technique. Pure inward current was isolated using a $Ca^{2+}-rich$ pipette solution containing 10 mM TEA. This current was independent of external $Na^+$ and was highly sensitive to the $Ca^{2+}$ concentration in the bathing solution, indicating that the inward current is carried by $Ca^{2+}$. The maximal amplitude was $-4.12{\pm}0.58nA\;(n=12)$ with 10mM $Ca^{2+}$ at -3OmV from a holding potential of -8OmV. This current reached its maximum within 20ms beyond -3OmV and decayed rapidly with an inactivation time constant $({\tau})$ of 15ms. Activation and inactivation of this $i_{Ca^{2+}}$ was steeply dependent on the membrane potential. The $i_{Ca^{2+}}$ began to activate at the lower voltage of -55 mV and reached its peak at -35 mV, being completely inactivated at potentials more positive than -40 mV. These result suggest that $i_{Ca^{2+}}$ in hamster eggs passes through channels with electrical properties similar to low voltage-activated T-type channels. Other results from the present study support this suggestion; First, the inhibitory effect of $Ni^{2+}\;(IC_{50}=13.7\;{\mu}M)$ was more potent than $Cd^{2+}\;(IC_{50}=123\;{\mu}M)$. Second, $Ba^{2+}$ conductance was equal to or below that of $Ca^{2+}$. Third, $i_{Ca^{2+}}$ in hamster eggs was relatively insensitive to nifedipine $(IC_{50}=96.6\;{\mu}M)$, known to be a specific t-type blocker. The physiological role of $i_{Ca^{2+}}$ in the unfertilized hamster eggs remains unclear. Analysis from steady-state inactivation activation curves reveals that only a small amount of this current will pass in the voltage range $(-70{\sim}-30\;mV)$ which partially overlaps with the resting membrane potential. This current has the property that it can be easily activated by a weak depolarization, thus it may trigger a certain kind of a intracellular event following fertilization which may cause oscillations in the membrane potential.

Partial Purification of Factors for Differential Transcription of the rrnD Promoters for Ribosomal RNA Synthesis in Streptomyces coelicolor

  • The Streptomyces coelicolor A3(2) genome contains six operons (rrnA to F) for ribosomal RNA synthesis. Transcription from rrnD occurs from four promoters (p1 to p4). We found that transcripts from the p1 and p3 promoters were most abundant in vivo in the early exponential phase. However, at later phases of exponential and stationary growth, transcripts from the p1 promoter decreased drastically, with the p3 and p4 transcripts constituting the major forms. Partially purified RNA polymerase supported transcription from the p3 and p4 promoters, whereas pure reconstituted RNA polymerase with core enzyme (E) and the major vegetative sigma factor ${\sigma}^{HrdB}$ ($E{\cdot}{\sigma}^{HrdB}$) did not. In order to assess any potential requirement for additional factor(s) that allow transcription from the p3 and p4 promoters, we fractionated a partially purified RNA polymerase preparation by denaturing gel filtration chromatography. We found that transcription from the p3 and p4 promoters required factor(s) of about 30-35 kDa in addition to RNAP holoenzyme ($E{\cdot}{\sigma}^{HrdB}$). Therefore, transcription from the p3 and p4 promoters, which contain a consensus -10 region but no -35 for ${\sigma}^{HrdB}$ recognition, are likely to be regulated by transcription factor(s) that modulate RNA polymerase holoenzyme activity in S. coelicolor.

Mechanical Properties and Corrosion Resistance of CP-Ti and Ti Alloy for Dental Implants (인공치근용 CP-Ti과 Ti 합금의 기계적 성질 및 내식성)

  • Commercially pure titanium(cp-Ti) and Ti-15wt%Zr-4wt%Nb-4wt%Ta alloy were melted in vacuum induction furnace. According to the chemical analysis, the content of carbon was above ASTM standard in the cast ingots because of using graphite crucible. The TEM micostructures of cp-Ti and Ti alloy shows that chemically stable TiC precipitates distribute in ${\alpha}-Ti$ matrix. In order to examine the properties of cp-Ti and Ti-Zr-Nb-Ta alloy for dental applications, mechanical properties and corrosion resistance were investigated. The anodic polarization properties of Ti-Zr-Nb-Zr alloy were almost same as that of cp-Ti in 1% lactic acid. However, as the results of the anodic polarization test in 5% HCl, it was known that Ti-Zr-Nb-Zr alloy showed a rapid decrease in current density at higher potential in comparison with cp-Ti. The yield stress and tensile strengh in Ti-Zr-Nb-Ta were ${\sigma}_{0.2}=623\;MPa$, ${\sigma}_{T.S.}=708\;MPa$ and these results showed 30% increase in yield stress in comparison with cp-Ti.

Genetic Characterization, Morphometrics and Gonad Development of Induced Interspecific Hybrids between Yellowtail Flounder, Pleuronectes ferrugineus (Storer) and Winter Flounder, Pleuronectes americanus (Walbaum)

  • Viable interspecific hybrids between yellowtail flounder (Pleuronectes ferrugineus, Store.) and winter flounder (Pleuronectes americanus, Walbaum) were produced by artificial insemination of yellowtail flounder eggs with winter flounder sperm. However, mean fertilization rate, hatching success and early survival up to 3 weeks post hatch were significantly lower than those of parental pure cross controls (P<0.01). Overall, cytogenetic traits (karyological analysis and estimation of cellular DNA contents using flow cytometry) of hybrid flounder were intermediate between the two parental species. Microsatellite assay was used to distinguish the parental genomes in the hybrids; in most cases, one allele was specific to each of the parents. Morphometrics assessed by body proportions indicated that hybrids generally displayed a morphology intermediate between the maternal and paternal species. Interspecific hybrids exhibited abnormal and retarded gonad development in both sexes based on histological analysis of gonads from adult fish. The sterility of the hybrids presents a significant advantage for their use in aquaculture, as potential escapees would not be capable of reproducing in the wild and contaminating natural stocks.
