천식모델을 이용한 호도, 파고지 및 두 배합약물의 실험적 연구 (The Synergistic Effect of Juglans sinensis, Psoralea corylifolia, and Their Combination on the Asthmatic Murine Model)

  • Objectives : To clarify the possible effect of JS (Juglans sinensis), PCF (Psoralea corylifolia L.), and J+P(JS+PCF), we examined their influence on the development of pulmonary eosinophilic inflammation in the asthmatic murine model. Methods : All mice were immunized on two different days (21 days and 7 days before inhalational exposure) by intraperitonial injections of 0.2 ml alum-precipitated Ag containing 100 ${\mu}$g of OVA bound to 4 mg of aluminum hydroxide in PBS. Seven days after the second sensitization, mice were exposed to aerosolized ovalbumin for 30 minutes/day on 3 days/week for 8 weeks (at a flow rate of 250 L/min, 2.5% ovalbumin in normal saline) and, JS, PCF and J+P (200 mg/kg, 400 mg/kg) were orally administered 3 times per week for 8 weeks. Results : The suppressive effects of JS, PCF, and J+P were demonstrated by the accumulation of eosinophils into airways, with the reduction of eosinophils and lung leukocytes. These were correlated with the marked reduction of IL-4, IL-5, IL-13 levels in the BALF and serum. OVA-specific IgE levels were also decreased in serum and BAL from these mice. And also JS, PCF, and J+P decreased eosinophilic CCR3 and CD11b expression in lung tissue. Conclusions : These results indicate that JS, PCF, and J+P have deep inhibitory effects on airway inflammation and hyper-responsiveness in the asthmatic murine model. The suppression of IL-5, IgE, and eosinophilils and the increase of IFN-${\gamma}$ production in BALF seem to contribute to these effects. Specially, esosinophils and TNF-a in J+P combination group were significantly reduced in BALF and lung tissue. Hence, the results indicated that JS, PCF, and J+P could act as an immuno-modulator which possesses anti-inflammatory and anti-asthmatic property by modulating the imbalance between Th1 and Th2 cytokines.

  • Psoralen(7H-Furo[3.2-g] [1]benzopyran-7-one) and angelicin(2-Oxo-[2H]- furo[2.3-h]-1-benzopyran) are angular furocoumarin with diverse photobiological effects. They are major components of Psoralea corylifolia L.(破古紙). Psoralea corylifolia L. is used for a tonic and nursing one's energy. It can be also used for loss of virility. vitiligo. a skin disease, etc.. But a well known and often appreciated side effects f psoralens is the hyperpigmentation caused by this treatment. A women who used the herbal drug-mixed-medicine named'sobaeksu' to treat her vitiligo made a complaint against the orinetal medical doctor. She complained that her skin got burned to 2nd degree by the liquid. 'sobaeksu' through a medical celtficate. So we analyzed the components of that liquid with gas chromatography and gas chromatography/mass spectometry. It has 57.3% ethyl alcohol and two kinds of psoralens. Psoralens were psoralen and angelicin and each one of their contained quantity was 0.128mg/ml and 0.123mg/ml.

계혈등, 반지련, 보골지, 봉출, 오미자, 현호색 헥산 분획층의 항산화활성 (Antioxidative Activity of the n-Hexane Fractions from Spatholobus suberectus (SS), Scutellsria barbata (SB), Psoralea corylifolia (PC), Curcuma zedoaria (CZ), Schisandra chinensis (SC), and Corydalis turtschaninovii (CT))

  • 약용식물 6종을 메탄올로 추출한 다음, n-hexane층을 DMSO에 녹여 항산화 활성을 측정 비교하여 기능성 소재인 천연 항산화제로써의 활용 가능성을 조사하였다. DPPH를 이용한 라디칼 소거능 실험에서는 250 ${\mu}g/mL$ 농도에서 보골지가 대조군인 ascorbic acid에 비해 높은 유리기 소거능에 의한 항산화 활성을 나타냈다. 농도 500 ${\mu}g/mL$에서는 ascorbic acid가 가장 높은 항산화 활성을 보였다. ABTS 유리기 소거능 실험에서는 농도 증가에 따른 항산화 활성이 DPPH assay와 상반된 결과를 나타냈고, 특히 보골지의 항산화 활성이 대조군인 ascorbic acid보다 유의적으로 높게 나타났다. DPPH 라디칼, ABTS 유리기 소거능 실험 결과 공통적으로 우수한 것으로 조사 된 약용 식물은 반지련, 보골지이며 500 ${\mu}g/mL$ 이상의 농도에서 ascorbic acid와 유사한 90% 활성을 보였으며 750 ${\mu}g/mL$ 농도에서는 ascorbic acid 보다 우수한 유리기 소거 활성을 보였다. 따라서, 반지련과 보골지는 항산화 활성이 우수하므로 천연 식물소재로 이용하기에 적합하며, 천연물의 추출 부위, 용매, 추출 조건에 따라 다양한 항산화 활성 능력을 기대 할 수 있으므로 천연 항산화제로서의 응용 가능성 또한 있을 것으로 사료된다.

산수유(山茱萸)와 보골지(補骨脂) 복합추출물의 Sprague-Dawley 랫드를 이용한 13 주 반복경구투여 독성시험 및 4 주 회복시험 (A 13-Week Repeated Oral Dose Toxicity Test and a 4-Week Recovery Test of Standardized Cornus officinalis and Psoralea corylifolia L . in Sprague-Dawley Rats)

  • Objectives : In the current study, we performed the 13-week repeated oral dose toxicity test and a 4-week recovery test of standardized Cornus officinalis Sieb. et Zucc. and Psoralea corylifolia L. 30 % ethanol extract (SCP) in Sprague-Dawley (SD) rats owing to aims for verifying no observed adverse effect level (NOAEL). Methods : The animal study was performed according to OECD guidelines for the testing of chemicals section 4 health effects test No.408 repeated dose 90-day oral toxicity study in rodents (03 October 2008). In the repeated dose toxicity study, SCP was orally administered to female and male rats at dose levels of 1,000, 2,000, and 4,000 mg/kg/day for 13-week. The control group and high dose (4,000 mg/kg/day) group were then monitored for 4 extra weeks to determine recovery time after the study period. 1) Results : Compared with the control group, there were no treatment-related adverse effects in clinical signs, body weight, hematology, serum biochemistry (Aspartate aminotransferase, Alanine aminotransferase, Alkaline phosphatase, 𝛾-Glutamyl transpeptidase, Blood urea nitrogen, Creatinine, Glucose, Total cholesterol, Total protein, Creatine phosphokinase, Albumin, Total bilirubin, Triglyceride, Inorganic phosphorus, Albumin/Globulin ratio, Calcium ion, Sodium ion, Potassium ion, Chloride ion), necropsy findings and organ weight (Ovary, Adrenal gland, Pituitary, Thymus, Prostate, Testis, Epididymis, Spleen, Kidney, Heart, Lung, Brain, Liver) at any dose tested. Conclusions : Taken together, these results suggest that the NOAEL of SCP in both genders was considered as over 4,000 mg/kg. Results from this study provide scientific evidence for the safety of SCP.

파고지 추출물의 렛트에 대한 단회 경구 투여 독성 및 복귀돌연변이능 평가 (Single-Dose Oral Toxicity in Rat and Bacterial Reverse Mutation Assay of Psoralea corylifolia L. Extracts)

  • 천연생리활성물질 소재로서 파고지 추출물의 안전성을 검증하기 위하여 렛트에 대한 단회 경구투여 독성 평가와 복귀돌연변이평가를 실시하였다. 파고지 에탄올 추출분말 2 g/kg, 5 mL를 암${\cdot}$수 각각 5마리씩 경구 투여한 후 14일 동안의 일반증상, 체중변화 및 부검시의 육안적 소견을 관찰한 결과, 사망례, 일반증상 및 체중변화는 관찰되지 않았고 부검결과에서도 특이할 만한 육안적 이상소견은 관찰되지 않아, 파고지 추출물은 $LD_{50}$값이 렛트 체중 kg당 2g 이상이며 단회 경구 투여에 대한 독성을 가지는 않는 것으로 평가되었다. 파고지 추출분말의 복귀돌연변이 유발성 여부는, 히스티딘 요구성 Salmonella Typhimurium TA98, TA100, TA1535, TA1537 균주 및 트립토판 요구성 Escherichia coli WP2uvrA(pKM101) 균주를 이용하여 S9 mix 첨가 여부에 따라 실험하였다. 용량설정시험 결과, S9 mix 첨가 여부에 관계없이 사용한 모든 시험균주에서 생육저해가 관찰되었다. 시험물질의 석출은 S9 mix 첨가 여부와 상관없이 사용한 모든 시험균주의 1,250, 2,500 및 5,000 ${\mu}g/plate$에서 관찰되었지만 콜로니 계수에는 영향이 없었다. 따라서, 본 시험의 용량은 S9 mix 미첨가 TA100, TA1537 균주 및 S9 mix 첨가 RA1535 균주의 경우에는 생육저해가 관찰된 625 ${\mu}g/plate$를, S9 mix 첨가 여부에 관계없이 TA98 균주 및 S9 mix 첨가TA100, TA1537, WP2uvrA(pKM101) 균주의 경우에는 생육저해가 관찰된 1,250 ${\mu}g/plate$를, S9 mix 미첨가의 TA1535 및 WP2uvrA(pKM101) 균주의 경우에는 2,500 ${\mu}g/plate$를 최고용량으로 하였고, 그 이하 공비 2로 최고용량을 포함한 5용량의 시험물질군과 음성대조군 및 양성대조군으로 하여 실험을 실시하였다. 그 결과, 모든 시험균주에서 시험물질에 의한 복귀변이 콜로니수는 S9 mix의 첨가 여부에 관계없이 용량의존적으로 증가되지 않았으며, 음성대조군과 비교하여 2배 이상의 증가를 보이지 않았다. 시험물질에 의한 균주의 생육저해는 S9 mix첨가 여부에 관계없이 사용한 모든 시험균주의 최고용량에서 관찰되었다. 시험물질의 석출은 S9 mix 첨가 여부에 관계없이 1,250 및 2,500 ${\mu}g/plate$에서 관찰되었지만 콜로니 계수에는 영향이 없는 것으로 나타났다.

경동맥 혈전 유발 동물모델을 이용한 한약재 추출물의 효능 검색 (Screening of Anti-thrombotic Activity of Herbal Extracts in Ferric Chloride-induced Thrombosis Model)

  • Objectives : The aim of this study was to examine anti-thrombotic effect of traditional herbal extracts in a rat model of ferric chloride ($FeCl_3$)-induced carotid arterial thrombosis. Methods : Thirty minutes prior to a 35% $FeCl_3$ application, Sprague Dawley(SD) rats were injected with the 10 types of traditional herbal extracts (100mg/kg, intraperitoneal injection), respectively. The effect of these herbal extracts was examined for time to occlusion(TTO) using the Laser doppler flow meter and measured for thrombus weight (TW) in $FeCl_3$-induced thrombosis model. Results : In the TTO, Salvia miltiorrhiza (Sm, $2.30{\pm}0.28$ min, p<0.001) and Santalum album (Sa, $2.19{\pm}0.19$ min, p<0.001) showed significantly delayed TTO more than twice compared with Saline-treated group. Cnidium officinale (Co), Psoralea corylifolia (Pc), Scutellatia baicalensis (Sba), Panax notoginseng (Pn), Angelica tenuissima (At), Scrophularia buergeriana (Sbu), Rhus verniciflua (Rv) and Picrasma quassioides (Pq), except for Rhus verniciflua (Rv) also meaningfully impeded TTO more than one fold. In addition, Salvia miltiorrhiza, Santalum album, Cnidium officinale, Psoralea corylifolia and Scutellatia baicalensis significantly reduced TW more than 10% compared with Saline-treated group. Especially, Salvia miltiorrhiza and Santalum album showed the most excellent anti-thrombotic effect among the 10 herbal extracts tested on the restoration of altered TTO and TW. Conclusions : These results suggest that Sm and Sa extracts have outstanding anti-thrombotic effect in $FeCl_3$-induced thrombosis model and is potentially useful as herbal medicines for the treatment and prevention of thrombosis.

Quantitative Determination of Psoralen and Angelicin from Some Medicinal Herbs by High Performance Liquid Chromatography

  • A reversed-phase high performance liquid chromatographic method was developed to determine the contents of psoralen and angelicin from some medicinal herbs. The optimum eluent for chromatography was 20 v/v% acetonitrile in water on a Zorbax 300SB $C_{18}$ column. The identification was carried out by comparing the retention time and mass spectra of the relevant peaks with their standards. The variation of the concentration of psoralen and angelicin was wide between different species. The seeds of Psoralea corylifolia showed the highest contents of psoralen (7.8 mg/g) and angelicin (2.3 mg/g) among the tested herbs.
