Detection, isolation, and characterization of the cucumber mosaic virus in Pseudostellaria heterophylla from Korea

  • Weeds play an important role in the survival of viruses and are potential inoculum sources of viral diseases for crop plants. In this study, specimens of Pseudostellaria heterophylla exhibiting symptoms of the cucumber mosaic virus (CMV) were collected in Bonghwa, Korea. The characteristics of the disease were described and leaf RNA was extracted and sequenced to identify the virus. Three CMV contigs were obtained and PCR was performed using specific primer pairs. RNA from positive samples exhibiting CMV leaf symptoms was amplified to determine the coat protein. A sequence comparison of the coat protein gene from the CMV BH isolate shared the highest nucleotide identity (99.2%) with the CMV ZM isolate. Phylogenetic analysis showed that CMV-BH belonged to subgroup IA and that the most closely-related isolate was CMV-ZM. All test plants used for the biological assay were successfully infected with CMV and exhibited CMV disease symptoms such as blistering, mosaic, and vein yellowing. To our knowledge, this is the first report of CMV infection in P. heterophylla from Korea.

  • The root of Pseudostellaria heterophylla Pax et Hoffm. (Caryophyllaceae) is a herbal medicine called Taejasam(太子蔘) and used as a good tonic in China that strengthens the functions of the lung and the stomach like Ginseng in Korea. It has been known to have the antitussive activity for tuberculosis, appetizing effect and antifatigue activity, and so on. Especially it has been known to generate body fluids for the severe thirst and the shortage of water in body after a febrile disease for a long time. Recently some components of cyclic peptides, pseudostellarins A,B,C,D,E,F,G, that inhibit the activity of tyrosinase, were separated from this root. In this report several biological activities including acute toxicity, analgesic activity, antifatigue activity, bile juice increasing activity, hepatoprotective activity and antiinflammatory activity were investigated. These results showed as such; $LD_{50}$ of methanol extracts was above 10,000 mg/kg. The analgesic effect was revealed in the writhing test using rats. The BuOH fractions showed a prominent antifatigue effect against immobilized stress, significant hepatoprotective activities against $CCl_4$ intoxication and inhibitory effect on carrageenin-induced edema in rat's paw.

Interaction of Pseudostellaria heterophylla with Quorum Sensing and Quorum Quenching Bacteria Mediated by Root Exudates in a Consecutive Monoculture System

  • Many plant-pathogenic bacteria are dependent on quorum sensing (QS) to evoke disease. In this study, the population of QS and quorum quenching (QQ) bacteria was analyzed in a consecutive monoculture system of Pseudostellaria heterophylla. The isolated QS strains were identified as Serratia marcescens with SwrIR-type QS system and exhibited a significant increase over the years of monoculture. Only one QQ strain was isolated from newly planted soil sample and was identified as Bacillus thuringiensis, which secreted lactonase to degrade QS signal molecules. Inoculation of S. marcescens to P. heterophylla root could rapidly cause wilt disease, which was alleviated by B. thuringiensis. Furthermore, the expression of lactonase encoded by the aiiA gene in S. marcescens resulted in reduction of its pathogenicity, implying that the toxic effect of S. marcescens on the seedlings was QS-regulated. Meanwhile, excess lactonase in S. marcescens led to reduction in antibacterial substances, exoenzymes, and swarming motility, which might contribute to pathogensis on the seedlings. Root exudates and root tuber extracts of P. heterophylla significantly promoted the growth of S. marcescens, whereas a slight increase of B. thuringiensis was observed in both samples. These results demonstrated that QS-regulated behaviors in S. marcescens mediated by root exudates played an important role in replanting diseases of P. heterophylla.

Study on Practical Prescription and It's Drug Composition for the Treatment of Diabetic Nephropathy (당뇨병성신병증(糖尿病性腎病證)의 활용 처방 및 약물에 관한 연구)

  • Based on the theories and clinical data of oriental medicine, traditional medicinal prescriptions known to be effective to diabetic nephropathy were collected. The prescriptions were carefully examined and analyzed in order to be used as fundamental material for experiments and in clinicals. More than 40 publications related to diabetes were arranged and analyzed. Of the complications, diabetic nephropathy part of the publications were specifically focused during analysis. Data were analyzed and classified according to the quantity, prescription, differentiation of symptoms, signs and addition and subtraction of each medicine. Frequently used medicines were statistically analyzed. The most frequently used prescription was Yukmigihwang-tang(六味地黃湯) based medicine and prescriptions of its addition and subtraction, which was 15% of the total prescriptions cited. The most frequently used medicine was Astragali Radix, mentioned 192 times in the prescriptions. Hoelen was next with 180 citations, followed by Dioscoreae Rhizoma with 147 times, Rehmanniae Radix with 140, Corni Fructus with 131, Salviae Miltiorrhizae Radix with 106, Angelicae Gigantis Radix with 101, Alismatis Rhizoma with 95, Rhei Rhizoma with 90, Atractylodis Rhizoma Alba with 84, Rehmanniae Radix Preparata with 78, Leonuri Herba with 74, Moutan Cortex Radicis with 66, Aconiti Lateralis Preparata Radix with 66 Cnidii Rhizoma with 65, Pseudostellaria heterophylla with 62, Liriopis Tuber with 55, Lycii Fructus with 52, Rhei Rhizoma with 49, Codonopsis Pilosulae Radix with 44, Paeoniae Radix Rubra with 44, Schisandrae Fructus with 42, Polyporus with 42, Achyranthis Radix with 41 and Euryales Semen with 40. Medicines prescribed more than 30 grams a day included Astragali Radix, Imperatae Rhizoma, Benincasae Pericarpium, Leonuri Herba, Dioscoreae Rhizoma, Plantaginis Semen, Polyporus, Hoelen, Halloysitum Rubrum, Achyranthis Radix, Arecae Pericarpium, Phaseoli Angularis Semen, Coicis Semen, Rhei Rhizoma, Salviae Miltiorrhizae Radix, Sargassum, Ostreae Concha, Pseudostellaria heterophylla, Epimedii Herba, Rehmanniae Radix, Scrophulariae Radix and Polygonati Rhizoma. No scientific reports on the traditional medicinal aspects of diabetic nephropathy was searched. This analysis report would be able to provide the basis of developing new drug candidates for the treatment of diabetic nephropathy, as well as securing the EBM for the traditional medicines already being prescribed to the patients.

Correlations between the Distribution of Major Native Herb Species and Site Factors at Natural Broadleaved Forest in Mt. Gariwang and Mt. Jungwang, Pyeungchang-gun (천연 활엽수림에서 주요 자생 초본식물의 분포와 입지인자와의 상관 - 평창군 가리왕산, 중왕산 지역 -)

  • To obtain the basic data for the conservation and cultivation of major native herb species under the trees, distribution of native herb species, and corrrlations between native herb species and site factors were studied on natural broadleaved forest in Mt. Gariwang and Mt. Jungwang, Pyeunchang-gun. 136 native herb species were distributed in studied area, and the importance percentage of Pseudostellaria heterophylla was proved the highest, 10.8%. The other major herb species were the following oders, Meehania urticifolia, Corydalis remota, Pimpinella brachycarpa, Carex siderosticta, Pseudostellaria palibiniana, Erythronium japonicum, Ligularia fischeri, Asarum heterotropoides var. mandshuricum, Hylomecon vernalis and Astilbe rubra var. rubra. Highly positive correlations were proved Hepatica asiatica and Ainsliaea acerifolia, Pimpinella brachycarpa and Aconitum jaluense subsp. jaluense, Aconitum jaluense subsp. jaluense and Smilacina japonica var. japonica, Meehania urticifolia and Pseudostellaria palibiniana. Seven groups of native herb species were divided by cluster analysis. Preferring site factors for each native herb species were determined. Ligularia fischeri, Ainsliaea acerifolia, Parasenecio auriculata var. matsumurana, Erythronium japonicum and Pimpinella brachycarpa were distributed mainly on the higher sites, Ligularia fischeri were ditributed more frequently in deep soil sites. These results might be useful measures in the conservation and cultivation of native edible and medicinal herb species on the forest floor in the natual broadleaved forest, Pyeungchang-gun.

A taxonomic study of the genus Pseudostellaria in Korea (한국산 개별꽃속의 분류학적 연구)

  • The genus Pseudostellaria (Caryophyllaceae) in Korea was investigated in order to clarify the taxonomic positions and scientific names among the species in this genus based on morphological characteristics. P. ${\times}$ bohyeonsanensis and P. ${\times}$ seoraksanensis share several characteristics (trichome on the sepal margin and midvein, five petals, and stem reclining to the ground after flowering) with P. davidii. P. ${\times}$ bohyeonsanensis, resulting from natural hybridization of P. davidii and P. okamotoi var. longipedicellata, has sterile fruit and a long pedicel (2.2-6.0 cm). P. ${\times}$ seoraksanensis, a natural hybrid of P. davidii and P. palibiniana var. palibiniana, has sterile fruit and a short pedicel (1.1-1.8 cm). P. okamotoi var. longipedicellata shares several characteristics (long pedicels of chasmogamic flowers that elongate to the ground when bearing fruit and cleistogamic flowers with a long pedicel) with the related taxa P. okamotoi var. okamotoi. P. okamotoi var. longipedicellata has slender root tubers, glabrous pedicels, and 5-9 petals. P. palibiniana var. gageodoensis is described from Gageodo Island. It is similar to P. palibiniana var. palibiniana having chasmogamic flowers at the stem terminal, 5-7 petals, and a short pedicel, but P. palibiniana var. gageodoensis is distinguished by 1-2 rows of trichomes on the pedicel, a large sepal size, a large petal size, and a branched stem at its base. P. ${\times}$ biseulsanensis, from a natural hybrid of P. heterophylla and P. okamotoi var. longipedicellata, has a solitary aerial stem, a chasmogamic flower at the stem terminal, a shallow marginate or bitten petal apex, one row of trichomes on the pedicel, and sterile fruit. P. ${\times}$ segeolsanensis, from a natural hybrid of P. palibiniana var. palibiniana and P. okamotoi var. okamotoi, is described from Segeolsan Mt. It has numerous aerial stems, a chasmogamic flower at the stem terminal, 1-2 rows of trichomes on the pedicel, and fertile fruit. Consequently, the genus Pseudostellaria in Korea has been classified into eight species, four hybrids, two varieties, and five forms.

Ecological Characteristics of Viola websteri Hemsley Habitats (왕제비꽃 자생지의 생태특성)

  • This study intended to investigate the environmental factors including soil and vegetation in order to understand the environmental and ecological characteristics of seven different habitats of Viola websteri. These habitats, according to investigations, are mostly located on the slope of mountains facing north at an altitude of 343 m to 991 m above sea level with angle of inclination from 1 degree to 33 degrees. The type of soil is mostly sandy loam and the average field capacity of soil is 28.97%. Their average organic matter is 16.63%, soil pH 5.62, and available phosphorus is 14.75%. A total of 133 vascular plants are identified in 18 quadrates of seven habitats. Dominant species of woody plants in seven habitats are represented as Acer pictum subsp. mono and Quercus mongolica in tree layer, and Acer pictum subsp. mono in subtree layer. Importance value of Viola websteri is 9.66%, as regards the herbaceous layer, and five highly ranked species such as Meehania urticifolia(8.53%), Pseudostellaria heterophylla(6.51%), Hylomecon vernalis(5.15%), Oxalis obtriangulata (4.52%), and Pseudostellaria davidii(4.15%) are considered to be an affinity with Viola websteri in their habitats. The degree of their average species diversity is 1.32, and that of dominance and evenness are 0.08 and 0.89, respectively. Correlation coefficients analysis based on environmental factors, vegetation and soil analysis shows that the coverage of Viola websteri is correlated with silt and sand ratio, and coverage of tree layers are correlated with species richness and altitude. Cluster analysis based on vegetation structure of each habitats are forms a three groups.

Growth Characters and Vegetation Survey in Natural Habitat of Dicentra spectabilis (L.) Lem (금낭화의 생육특성 및 자생지 식생조사)

  • This study was carried out to obtain the optimum cultivation conditions of wild ornamental plant resources, Dicentra spectabilis L. The altitude of the habitat was ranged from 630m to 690m in Mt. Gujeol. Most of the habitat faced toward the northeast aspect and the gradient was ranged from 25 to 33 degrees. The light intensity of Dicentra community was 14,000lux on the average, and it was tendency to decrease by foliation of other woody plants. The appearance number of Dicentra per quadrat was 30 individuals on the average, and total appearance species in all quadrat surveyed was 52 taxa. Among them, Quercus mongolica, Lindera obtusiloba, Corylus heterophylla, Cornus controversa, woody plants, Corydalis speciosa, Pseudostellaria heterophylla, Artemisia keiskeana, Dryopteris crassirhizoma, Arisaema amurense var. serratum, Carex siroumensis, and Chloranthus japonicus, herbaceous plants, were showed as dominant species. Soil acidity in the habitat was 5.15 to 5.96, and average content of soil moisture was 32.6%. The contents of average organic matter, $P_2O_5$, K, Ca, and Mg were 1.99%, 14mg/kg, 0.55me/100g, 15.2me/100g, and 3.3me/100g, respectively. Electron conductivity was ranged from 0.50 to 0.76dS/m. Also, the average air temperature of May and June was 14.2$^{\circ}C$ and 19.4$^{\circ}C$, respectively.

Forest Community Classification and Vegetation Structure in National Yonghyeon Natural Recreation Forest (용현자연휴양림 일대의 산림군락분류와 식생구조)

  • This study was carried out to investigate the forest community classification and vegetation structure in NYNRF(National Yonghyeon Natural Recreation Forest). We studied by 26 plots($20{\times}20m$) from April to September 2014. NYNRF were classified into the Quercus mongolica Community, Quercus variabilis community and Acer pictum community in the 3 community units. The Q. mongolica Community was subdivided into the Pinus densiflora group and Typical group and A. pictum community was also subdivided into the Euonymus sachalinensis group and Typical group. NYNRF were classified into 3 communities and 4 groups and 5 VU(vegetation units). According to the result of importance value analysis between vegetation units, VU 1 P. densiflora(65.1%), VU 2 Q. mongolica(73.6%), VU 3 Q. variabilis(75.1%), VU 4 E. sachalinensis(33.3%) and VU 5 Lindera erythrocarpa(27.1%) were highly showed in tree layer and VU 1 Rhododendron mucronulatum (19.3%), VU 2 Calamagrostis arundinacea (16.9%), VU 3 Indigofera kirilowii (75.1%), VU 4 and VU 5 Pseudostellaria heterophylla(each other 14.8% and 24.7%) were highly showed in herb layer. In INSPAN(INdicator SPecies ANalysis) of vegetation units, 33 species were analyzed significantly(p<0.05).
