Improving Water Quality and Bacterial Characteristics during Water Treatment Process Using Biological Activated Carbons on Downstream of the Nakdong River (낙동강 하류 상수원수의 생물활성탄에 의한 수질개선 및 세균분포 특성)

  • Improvement of water quality and Investigation of bacterial characteristics have been conducted in a pilot plant using biological activated carbon (BAC) in water treatment process at the downstream of the Nakdong River. Most of water control parameters were highly improved after passing through BAC. Approximately 54% of dissolved organic carbon was removed in coal-based BAC process. Bacterial biomass and bacterial production appeared $9.8{\times}10^8 CFU/g and 7.1mg-C/m^3$.hr in coal-based BAC, respectively. Predominant bacteria species grown in BAC were identified as Pseudomonas, Flavobacterium, Alcaligenes, Acinetobacter and Aeromonas species. Particularly Pseudomonas vesicularis was dominant in both coal-based and coconut-based BACs, while Pseudomonas cepacia was dominant in wood-based BAC.

  • Isolated the heterotrophic aerobic bacteria in sandfiltered water on NA and TSBA solid medium, selected 8 dominant species and identified by Sherlock System. Each samples are irradiated 0, 5, 16, 40 and $60\;mJ/cm^2$ using on CBD (Collimated Beam Device) Medium Pressure UV lamp after these identified bacterium did liquid culture how to make $10^6{\sim}10^7\;cells/mL$ suspended in dilution water. Then cultured bacteria are estimated inactivation rate on plate media. Identified Gram positive group are Bacillus Subtilus, Bacillus megaterium, Rhodococcus erythropolis and Microbacterium laevaniformans; Gram negative group are Pseudomonas vesicularis, Pseudomonas pseudoflava, Alcaligenes paradoxus and Zooglea ramigera. These isolation of bacterium are more stronger reference strain and high resistance of MP UV irradiation, Besides Gram negative bacterium are more sensitive Gram positive bacterium on MP UV dose. Now we are estimating to $60{\sim}100\;mJ/cm^2$ MP UV dose for efficient disinfection in water treatment plant.

Characteristics of Biodegradation of Geosmin using BAC Attached Bacteria in Batch Bioreactor (정수처리용 생물활성탄(BAC) 부착 박테리아를 이용한 회분식 반응기에서의 Geosmin 생분해 특성)

  • In this study, three different biological activated carbons (BACs) were prepared from activated carbons made of each coal (F400, Calgon), coconut (Samchully) and wood(Pica, Picabiol) which were run for two and half years in the pilot plant. The attached bio-film microorganisms in and on the BACs were isolated and identified. The results showed that nine different bacteria species (Chryseomonas luteola, Stenotrophomonas maltophilia, Pseudomonas vesicularis, Aeromonas hydrophila, Spingomonas paucimobilis, Agrobacterium radiobacter, Pseudomonas fluorescens, Spirillum spp., and Pasteurella haemolytica) were isolated and identified, the dominant species was Pseudomonas sp. that had occupied 56.5%. More specifically, it was observed that the populations of the microorganisms deceased in the order: Pasteurella haemolytica (18.9%) > Chryseomonas luteola (4.0%) > Agrobacterium radiobacter (3.5%) > Aeromonas hydrophila (2.0%) in and on the BACs. After isolating of 9 species of biofilm microorganisms, the growth curve for the biomass was investigated. During 24~96 hours, the biomass has the highest concentration, and activity of the biomass was the best to uptake geosmin as carbon resources. The operation temperatures for investigating the biodegradation of geosmin were set at $4^{\circ}C$ and $25^{\circ}C$. Pseudomonas vesicularis, Pseudomonas fluorescens, Agrobacterium radiobacter and Stenotrophomonas maltophilia played a maior role in removing the target compound as geosmin. However, geosmin was not biodegraded well by Chryseomonas luteola, Spingomonas paucimobilis, and Spirillum spp.. It is also interesting to evaluate kinetics of biodegradability of geosmin. The first-order rate constants for biodegradability of geosmin at $4^{\circ}C$ and $25^{\circ}C$ were $0.00006{\sim}0.0002\;hr^{-1}$ and $0.0043{\sim}0.0046\;hr^{-1}$ respectively. Higher water temperature produced better geosmin removal rates. When concentrations of geosmin increased from 10 to 10,000 ng/L, the rate constants for biodegradability of geosmin increased from 0.0003 to $0.0882\;hr^{-1}$. As described earlier, higher geosmin concentration in the reactor produced higher rate constant.

  • Monthly distribution of marine bacteria and phytoplankton in Suyeong Bay were investigated with laboratory experiment of dominant algal species and bacterial flora. During the periods of study, the highest density of phytoplankton and bacteria occurred in May with the number of $3.3{\times}10^6cells/l$ and $1.93{\times}10^8cells/ml$, respectively. 10 genera of bacteria and 22 genera of phytoplankton were isolated and identified. In May when phytoplankton bloom occurred, dominant species of bacteria were Acinetobacter calcoaceticus($29.1\%$) and Bacillus subtilis($22.9\%$), and dominant species of phytoplankton were Chaetoceros spp.($62.8\%$) and Skeletonema spp. ($19.4\%$). Pseudomonas spp., which was the most abundant bacterial species during the study periods, were rapidly decreased in May. In laboratory studies of culturing bacteria and phytoplankton isloated in May, the growth of Pseudomonas vesicularis seems to be influenced by the concentrations of excretion matter of Chaetoceros spp. To examine the result colsely, the problem of pure isolation for phytoplankton must be solved and more experimental process have to be conducted.

Characteristics of Bacterial Community for Biological Activated Carbon(BAC) by Culturable and Unculturable Methods. (배양적 및 비배양적 방법에 의한 생물활성탄 부착세균 군집 특성)

  • The Biological Activated Carbon (BAC) process in the water treatments represents a kind of biofiltration process which capabilities of bacteria to remove organic matters are maximized. It enables to eliminate organic matters and effectively reduce microbial regrowth potentials. As attached bacteria employ natural organic matter as a substrate, they are significantly dependent on indigenous microorganisms. In this study, characteristics of bacterial community by culturable and unculturable Methods have been conducted in a pilot plant using SAC in water treatment process at the downstream of the Nakdong River. Based on the results, HPC and bacterial- production for coal-based activated carbon material were $1.20{\sim}56.2{\times}l0^7$ cfu/g and $1.2{\sim}3.7\;mgC/m^{3}h$, respectively, in the SAC process. The highest level of attached bacteria biomass and organic carbon removal efficiency was found in the coal-based activated carbon. The genera Pseudomonas, Flavobacterium, Alcaligenes, Acilzetobacter, and Spingomonas were identified for each activated carbon material. Pseudomonas vesicularis was the dominant species in the coconut- and coal-based materials, where as Pseudomonas cepacia was the dominant species in the wood-based material. The Scanning Electron Microscope (SEM) observation of the activated carbon surface also found the widespread distribution of rod form and coccus. The community of attached bacteria was investigated by performing Fluorescent in situ hybridization (FISH) analysis. a group was dominant in coal, wood and coccunt-based materials, ${\alpha},\;{\beta}\;and\;{\gamma}$ group ranged from 27.0 ${\sim}$ 43.0%, 7.1 ${\sim}$ 22.0%, 11.3 ${\sim}$ 28.6%, respectively. These results suggest that a group bacterial community appears to be regulated removal efficiency of organic material in water treatment process.

Environmental Factors Affecting on Shrimp Cultivation and Bacterial Examination in Shrimp Aquaculture

  • Cultivation environment for effective production of shrimp in shrimp aquaculture, Institute of Marine & Fishery, Soonchunhyang University located at Tae-An, Chung-Nam Province, with the ultimate aim of probiotics development, was monitored. Several environmental factors including dissolved oxygen, water body temperature, pH, salinity, $NH_4-N$, $NO_2-N$, $NO_3-N$, chlorophyll a affecting on the growth of shrimp were examined. Concentrations of $NH_4-N$, $NO_2-N$, and $NO_3-N$ dissolved in water samples were determined to 0.024-0.034 mg/L, 0.03-0.02, and 0.004-0.009 mg/L, respectively. Chlorophyll a content was examined in the range of 0.002-0.118 $ug/m^3$. In order to understand the distribution of different bacteria in water samples collected in shrimp aquaculture, bacteria were isolated and enumerated on the marine agar plates. Total number of bacteria were increased to approximately $6.5\;{\times}\;10^4$. Thirteen predomonant bacteria were isolated and identified. As the results of BIOLOG test of the isolates, these bacteria were identified as Corynebacterium nirilophilus, Clavibacter agropyri, Sphingomonas adhaesiva, Brevundimonas vesicularis, Vibrio parahaemolyticus, Pseudomonas bathycetes, Vibrio tubiashii, Sphingomonas macrogoltabidus, Rhodococcus, rhodochrous, Burkholderia glumae, Corynebacterium urealyticus, Rhodococcus fascians, Psychrobacter immobilis, respectively. Further work will stimulate the elucidation of pathogenicity, corresponding bacteria related to environment and probiotics, providing good information for effective production of shrimp.

Isolation of Dye-degrading Microbes for the Treatment of Dyeing Wastewater and Dye Decoloring (염색폐수 처리를 위한 미생물 선별 및 염료 탈색)

  • 87 microbes were isolated from dyeing wastewater collected at Dongducheon and Banweol industrial complex. Five microbes showed excellent ability of color removal and were identified as Shewanella putrefaciens, Aeromonas salmonicida(3 different strains), and Pseudomonas vesicularis. Five identified strains had optimal pH and optimal temperature as 7.0 and 30$^{\circ}C$ for cultivation, and showed morphological characteristics of Gram negative, oxidase negative, rod shape, and non-motility, but their biochemical characteristics were distinguishable. Each single strain of five microbes were tested in the 500 mL flask to treat dyeing wastewater, and achieved about 35% color removal efficiency in average. When two strains were selected and applied to the treatment at same time, color removal efficiency was increased up to 65%. While three or more associations of each strain did not show the improvement of color removal. Inhibition effects by $Mn^{2+}\;and\;Fe^{3+}$ on the dye degradation were tested and resulted in no effect under 70 ppm concentration.

Influence of Pipe Materials on Corrosion and Bacteria Regrowth in A Model Home Plumbing System (급수관에서 관재에 따른 부식특성과 미생물 재성장 고찰)

  • This study performed in order to evaluate the effects of pipe materials on corrosion and bacteria regrowth using a laboratory scale batch test. Two varieties of feed water with different microbial conditions were selected: tap water, surface river water (Han River water), and five pipe materials; carbon steel, copper, galvanized iron, stainless steel, and PVC, Carbon steel and galvanized iron pipes showed higher corrosion rates than other materials. In terms of attached bacterial growth, pipes with PVC and stainless steel showed higher bacteria concentration compared to other materials. Pseudomonas vesicularis was the predominant bacteria found on biofilm. The behavior of bacterial growth in the pipes was observed using a scanning electron microscope.

  • This study was aimed to isolation and identification of attached bacteria on the clothes excavated from Deajeon area dating on the 16th century. From the observation with colony shape, 17 bacterial strains were isolated, and then 7 bacterial strains were identified with morphological and biochemical characteristics. Streptococcus sp., Alcaligenes faecalis, Gemella sp., Acinetobacter sp., Pseudomonas vesicularis, Aeromonas sain. salmonicida, Moraxella spp. In observation of the bacterial strains by the sort of textile, more bacterial strains were found in silk, cotton, and cotton batt than in ramie and hemp. It is suggest that hemp has antibacterial characteristics due to the presence of lignin. In the comparison washed samples with unwashed ones, there were more kinds of bacterial strains in washed samples. In the cellulase activity tests, all isolated bacteria had low level cellulase activity.

Distribution and Characteristics of Acidotolerant Heterotrophic and Naphthalene­Degrading Bacteria in Acidic Soils (산성토양에서 내산성 종속영양세균과 나프탈렌분해세균의 분포 및 특성)

  • The distribution and characteristics of acidotolerant heterotrophic and naphthalene-degrading bacteria were investigated in two forest areas, one near Ulsan petrochemical industrial complex (Sunam) and the other in countryside (Daeam). Average values of soil pH at Sunam and Daeam were 3.8 and 4.6, respectively. When het­erotrophic and naphthalene-degrading bacteria were enumerated by most probable number (MPN) procedures at Sunam, the median values of heterotrophs growing at pH 7.0 and pH 4.0 were $5.3{\times}10^7\;and\;3.3{times}10^7$ MPN/g, whereas those of naphthalene-degraders were $5.6{\times}10^4\;and\;4.0{times}10^5$ MPN/g, respectively. While the medians of heterotrophs at Daeam were larger than those at Sunam, the concentrations of naphthalene-degraders were higher at Sunam compared to those at Daeam. From the MPN tubes and enrichment cultures, we obtained 17 isolates of naphthalene-degraders which were identified as Sphingomonas paucimobilis, Brevundimonas vesic­ularis, Burkholderia cepacia, Ralstonia pickettii, Pseudomanas fluorescens, and Chryseomonas luteola. Among them, 6 isolates showed higher naphthalene-degrading activity on minimal media of pH 4 compared to pH 7, whereas the extent of growth was not greater at pH 4 than at pH 7 when they were inoculated on nutrient-rich media. It is plausible that the pH may affect naphthalene-degrading activity of the isolates by changing fatty acid composition of bacterial membrane.
