Characteristics of Biodegradation of Geosmin using BAC Attached Bacteria in Batch Bioreactor (정수처리용 생물활성탄(BAC) 부착 박테리아를 이용한 회분식 반응기에서의 Geosmin 생분해 특성)

  • In this study, three different biological activated carbons (BACs) were prepared from activated carbons made of each coal (F400, Calgon), coconut (Samchully) and wood(Pica, Picabiol) which were run for two and half years in the pilot plant. The attached bio-film microorganisms in and on the BACs were isolated and identified. The results showed that nine different bacteria species (Chryseomonas luteola, Stenotrophomonas maltophilia, Pseudomonas vesicularis, Aeromonas hydrophila, Spingomonas paucimobilis, Agrobacterium radiobacter, Pseudomonas fluorescens, Spirillum spp., and Pasteurella haemolytica) were isolated and identified, the dominant species was Pseudomonas sp. that had occupied 56.5%. More specifically, it was observed that the populations of the microorganisms deceased in the order: Pasteurella haemolytica (18.9%) > Chryseomonas luteola (4.0%) > Agrobacterium radiobacter (3.5%) > Aeromonas hydrophila (2.0%) in and on the BACs. After isolating of 9 species of biofilm microorganisms, the growth curve for the biomass was investigated. During 24~96 hours, the biomass has the highest concentration, and activity of the biomass was the best to uptake geosmin as carbon resources. The operation temperatures for investigating the biodegradation of geosmin were set at $4^{\circ}C$ and $25^{\circ}C$. Pseudomonas vesicularis, Pseudomonas fluorescens, Agrobacterium radiobacter and Stenotrophomonas maltophilia played a maior role in removing the target compound as geosmin. However, geosmin was not biodegraded well by Chryseomonas luteola, Spingomonas paucimobilis, and Spirillum spp.. It is also interesting to evaluate kinetics of biodegradability of geosmin. The first-order rate constants for biodegradability of geosmin at $4^{\circ}C$ and $25^{\circ}C$ were $0.00006{\sim}0.0002\;hr^{-1}$ and $0.0043{\sim}0.0046\;hr^{-1}$ respectively. Higher water temperature produced better geosmin removal rates. When concentrations of geosmin increased from 10 to 10,000 ng/L, the rate constants for biodegradability of geosmin increased from 0.0003 to $0.0882\;hr^{-1}$. As described earlier, higher geosmin concentration in the reactor produced higher rate constant.

Identification of Bacteria by Sequence Analysis of 16S rRNA in Testes of Jeju Horses (제주마 고환내 세균의 16S rRNA 염기서열 분석을 이용한 동정)

  • Many bacteria colonized in the horse semen affect quality of the sperm and some may cause infection in the mare reproductive tract and infertility of susceptible mare. This study was initiated to determine the prevalence of bacteria in testes of Jeju horses by determining rRNA sequence. The samples were swabed from the testes of nine Jeju horses (aged from 8 to 12 months after birth). Bacteria isolated from testes were identified by 16S rDNA sequencing. 1.6-kbp PCR products for 16S rRNA coding region were obtained using the universal primers. The PCR products were further purified and sequenced. Maximum similar species were found by BLAST search in the GenBank DNA database. BLAST results showed that the sequences were similar to those of Acinetobacter sp (A. schindleri, A. ursingii)., Bacillus cereus, Corynebacterium glutamicum, Escherichia coli, Gamma proteobacterium, Micrococcus luteus, Pseudomonas mendocina, Shigella sonnei, Sphingomonas sp., Staphylococcus sp (S. cohnii, S. saprophyticus, S. xylosus)., and Stenotrophomonas maltophilia. DNA sequences for 16S rRNA is provided useful informations for species identification of pathogenic microorganisms for the reproductive organs in horses.

  • Safety of yellow pigment produced by Monascus sp. CS-2 was evaluated. Acute oral toxicity, pyrogen test, and histamine test, as well as antimicrobial activity were determined. The results obtained were; LD oral in mice was 132.5 mg/20 g, pyrogen test in rabbit was 5 mg/kg, and histamine test in cat was 10 mg/kg. Also the pigment was particularly sensitive to Bacillus subtilis (ATCC 6633), Sarcina lutea (ATCC 9341) and Staphylococcus aureus (ATCC 6538 P), whereas not sensitive to Pseudomonas pyosyanea (ACTC 10490), Bacillus var. mycoides (ATCC 11778), Bordetella bronchiseptica (ATCC 4617) and Staphylococcus epidermidis(ATCC 12228).

Characterization of TCE-Degrading Bacteria and Their Application to Wastewater Treatment

  • Two bacterial strains capable of degrading trichloroethylene (TCE), isolated form soils contaminated with various chlorinated alkenes, were identified as Alcaligenes odorous N6 and Nocardia sp. Hl7. In addition, four KCTC strains, including three strains of Pseudomonas putida and one strain of Sphingomonas chlorophenolica, exhibited an ability to degrade toluene. A. odorans N6 and Nocardia sp. H17 degraded 84% of the initial amount of TCE in a basal salts medium (BSM), containing 0.2 mM TCE as the sole source of carbon and energy, in a day. The optimal pH for growth was within a range of 7.0-8.0. A mixed culture of the four toluene-degrading isolates degraded 95% of 0.2 mM TCE with 1.5 mM toluene as an inducer, whereas no TCE was degraded by the same mixture without an inducer. When a mixed culture of all 6 isolates was used, the degradation efficiency of 0.2 mM TCE was 72% without an inducer, in a day, and 82% with toluene as an inducer. In a continuous treatment, 1,000 mg/1 of TCE in an artificial wastewater was completely removed within 18 h when an activated sludge was used along with the microbial mixture, which was 27 h laster than when only an activated sludge was used. Accordingly, it would appear that such a microbial mixture could be effectively applied to the biological treatment of wastewater containing TCE with or without an inducer.

  • Water samples collected from marine fish culture system in Korea were compared for their capability to reduce the infectivity titers of HRV (rhabdovirus olivaceus), FBV(flounder birnavirus) and RVS(retrovirus of salmonid). In addition, interaction between viruses and microorganisms present in the rearing seawater was examined. The titer of HRV and RVS were reduced at $15^{\circ}C$ to less than detectable limits within 3 to 5 days using untreated samples of seawater. No reduction of infectivity was noted in bacteria-free water treated by filtration or autoclaving. Bacteria (Pseudomonas and Vibrio sp.) isolated from the water collected from a flounder culture system showed the inactivation activity of HRV.

  • An antifungal substance was purified from culture broth of Pseudomonas flulorescens 2112 that showed a broad-spectrum antagonistic activity against various phytopathogenic fungi including capsici. The substance was identified as 2,4-diacetylphloro-glucinol basd on NMR analysis. The 2,4-diacetylphloroglcinol showed antibiotic activity in broad acidic range from pH 1.0 to pH 9.0. About 83% of initial activity was remained after incubation for 30min ar $60^{\circ}C$, however, the activity was dropped up to 50% after 30 min incubation in $80^{\circ}C$. When the nucleotides of P. capsici treated with 2,4-diacetylphloroglucinol were labeled with[$^{3}$ H]-Adenin, the newly synthesized and radioactive-labeled RNA was significantly reduced than those of untreated P. capsici. indicating that the 2,4-diacetylphloroglucinol inhibits RNA synthesis.

  • AvrRpt2 protein triggers hypersensitive response (HR) and strong disease resistance when it is translocated from a bacterial pathogen Pseudomonas sp. to host plant cells containing a cognate RPS2 resistance protein through Type III Secretion System (TTSS). However, AvrRpt2 protein can function as the effector that suppresses a basal defense and enhances the disease symptom when functional RPS2 resistance protein is absent in the infected plant cells. Using Affymetrix Arabidopsis DNA chip, we found that many genes were specifically regulated by AvrRpt2 protein in the rps2 Arabidopsis mutant. Here, we showed that expression of AtERF11 that is known as a member of B1a subcluster of AP2/ERF transcription factor family was down regulated specifically by AvrRpt2. To determine its function in plant resistance, we also generated the Arabidopsis thaliana transgenic plants constitutively overexpressing AtERF11 under CaMV 355 promoter, which conferred an enhanced resistance against a bacterial pathogen, Pseudomonas syringae pv. tomato DC3000. Thus, these results collectively suggest that AtERF11 plays a role as a positive regulator for disease resistance against biotrophic bacterial pathogen in plant.

The Effects of Anticarcinogenic Activity of Solanum tuberosum Peel Fractions (감자 껍질 분획성분의 항발암 효과)

  • This study was peformed to determine the anticarcinogenic activity of the Solanum tuberosum Peel (SP) on several microorganisms and human cancer cell lines. Among the various solvent fractions of SP, the ethylether Partition layer (SPMEE) showed the strongest antimicrobial activity, ethylacetate partition layer (SPMEA) and butanol partition layer (SPMB) resulted in good antimicrobial activity. We also determined the effect of SP extract and fractions on cytotoxicity, and chemopreventive effect on human cancer cells. The experiment was conducted to determine cytotoxicity of SP partition layers on HepG2, HeLa and MCF-7 cells by MTT assay. Among the various partition layers of SP, SPMEE and SPW were showed the strongest cytotoxic effects on all cancer cell lines. The quinone reductase induced activities of HepG2 cell, the butanol partition layer (SPMB) at a does of 40 $\mu\textrm{g}$/mL was 8.49 times more effective compared to the control value of 1.0. This value was significantly higher than that of previous results using the other materials. Therefore, based on these studies, SP may be developed into a potentially useful antimicrobial and anticarcinogenic agents.

Characterization of antimicrobial proteins produced by Bacillus sp. N32 (Bacillus sp. N32 균주가 생산하는 항균 단백질 특성)

  • An antagonistic bacterial isolate, that inhibits the growth of plant pathogens, was selected and identified from 5,000 isolates screened from the rhizosphere of various crop plants. An isolate Bacillus sp. N32, tested against Colletotrichum gloeosporioides causing anthracnose disease in hot pepper, produced both a heat resistant antifungal protein and a heat sensitive antifungal protein. The heat resistant protein was partially purified by Ammonium sulfate fractionation and gel filtration chromatography. The bioautography showed that the proteins possessed high antifungal activity. The biosynthetic gene cluster responsible for the heat resistant antifungal protein was cloned from cosmid library using DNA probe obtained from PCR product with the primers targeting the conserved nucleotide sequence of the synthetic genes reported earlier, Most of the clones obtained showed higher homology to fengycin antibiotic synthetic gene family reported earlier. On the other hand, the heat sensitive protein was isolated from SDS-PAGE and electroblotting to determine the N-terminal amino acid sequences. The heat sensitive antifungal protein gene was cloned from the ${\lambda}-ZAP$ libraries using a DNA probe based on the N-terminal amino acid sequences of the heat sensitive protein. We are contemplating to clone and sequence the whole gene cluster encoding the heat sensitive protein for further analysis.

Effects of the Presence of Nonionic surfactants on Diesel Biodegradation (비이온계 계면활성제가 미생물의 디이젤 분해에 미치는 영향)

  • The effects of the presence of commercial non-ionic surfactants on the cell growth and diesel degradation by Pseudomonas sp. OSD were studied. Most surfactants inhibited the diesel biodegradation at high concentration(1000mg/1). However, some surfactants showed no inhibition at lower concentrations. Tween 20, Brij 58, Brij 78 were not inhibitory to the diesel biodegradation even at high concentration. These chosen surfactants has relatively high HLB values. There exists complicated relationship for diesel bioremediation between cell hydrophobicity, surfactant HLB, contaminants, an soil.
