Studies on the Glycolipid Biosurfactant(2);The Rhamnolipid Production and Isolation by Pseudomonas sp. 13 (당 지질계 미생물 계면활성제에 관한 연구(제2보);Pseudomonas sp. 13에 의한 Rhamnolipid의 생성 및 분리)

  • A microorganism, isolated from soil and designated Pseudomonas sp13, produced two kinds of rhamnolipid in the medium containing glucose as carbon source. There were both rhamnolipid contain L-rhamnose and ${\beta}$-hydroxydecanoic acid. Coumpound A and B elucted chloroform-methanol mixed solution of silicic acid column chromatography and recrystallized from a mixture of ether and n-hexane. Studies on the structure of these products reveled that compound A is L-rhamnopyranosyl-${\beta}$-hydroxydecanoyl-${\beta}$-hydroxydecanoic acid and compound B is L-rhamnopyranosyl-L-rhamnopyranosyl-${\beta}$-hydroxydecanoyl-${\beta}$-hydroxydecanoic acid.

  • Fro the purpose of producing glouse from cellobiose or oligo saccharide and obtaining genetic information of beta-1,4-glucosidase gene, alpha beta-1,4-glucosidase gene of Pseudomonas sp. LBC505, potent cellulase complex and xylanase producing strain, was cloned in Esherichia coli and Bacillus subtilis into pUC19 and pBD64, respectively. Recombinant plasmid pGL1 contained 1.2kb EcoRI fragment was isolated from transformants forming blue color around colony on LB agar plate containing 20 ng/ml of 5-bromo-4-chloro-3-indolyl-beta-D-glucopyranoside(X-glu) and ampicillin.

  • Pseudomonas sp. EL-G527 was grown to produce a biosurfactant on 2% n-hexadecane as the energy and carbon source. This biosurfactant significantly reduced the surface tension of water from 72 to 28 dyne/cm at a critical micelle concentration(CMC) of 140 mg/l at pH 2.0. As the pH value decreased, the reduction in the surface tension due to the biosurfactant increased. The surface activity of the biosurfactant was unaffected when the NaCl concentration was increased to 5% and the calcium ion concentration increased to 100 mM, plus it remained stable at 10$0^{\circ}C$ for 180 min.

Inhibitory Activity of Bacterial Isolate Pseudomonas sp. KTB61 against Tobacco Mosaic Virus(TMV) Infection to Tobacco Plants (세균 분리주 KTB61의 담배 모자이크 바이러스(TMV) 감염 억제 효과)

  • During the screening or antiviral substances having inhibitory effect on tobacco mosaic virus(TMV) infection to tobacco plants, we found that a bacterial isolate, KTB61, which was identified as a Pseudomonas sp., strongly inhibited the formation of TMV local lesions. When the culture filtrate from KTB61 was applied on the upper surface of leaves of N. tabaccum Xanthi-nc tobacco at the same time of or 24 hours before TMV inoculation, almost complete inhibition was achieved. Incidence of systemic TMV infection to the susceptible tobacco cultivar, NC82, was reduced by 95% when TMV was inoculated onto the upper surface of leaves 24 hours after spraying the culture filtrate. Also 75∼80% of inhibitory effect was obtained by the inoculation of TMV onto the under surface of the leaves treated with culture filtrate 24 hours beforehand. In field trials, the TMV infection was reduced by 96.5% when the tobacco seedlings, N. tabaccum cv. NC82, were soaked with culture filtrate before transplanting.

  • The degradative pathway of homoprotocatechuate (HPC) is the bacterial routhe wherby 3,4-dihydrox-yphenylactic acid is catabolized to pyruvate and succinate by a series of sequential reactions . The HPC is catalzed by homoprotocatechuate 2, 3-dioxygenase(HPC-2,3O) to from 5-carboxymethy1-2-hydroxy-muco semialdehyde. In this study, the hha D gene encoding. HPC, 2, 3O was Cloned from the chromo-somal DNA of Pseudomonas sp. DJ-12 and its nucleotide sequence was analyzed. The open reding frame of hpaD gene was found to be composed of 864 nucleotide pairs and to encode a poypetide with 287 amino acide residues. The deduced amino acid sequence of the HPC-2,3O from Pseudomonas. sp. DJ-12 exhibited 60~64% homology with those of the corresponding enzymes from E. coli. Salmonella enterica, and Klebsiella pneumoniae.

  • Rescently we are interested in the biosurfactant. Biosurfactant have a low toxcity and easily biodegradable compound. Pseudomonas sp. 13 was isolated from soil. This microorganism produced biosurfactant that consists of glycolipid R-1 and R-2. A time course study of fermentation indicated that the appearance of glycolipid in the fermentation broth the commencement of the stationary phase with the respect to biomass. The effect of variation of the media components such as amount of glucose, nitrogen, phosphate and metal ions has been investigated. The following values found to be optimum for biosurfactant production (glucose, $20g/{\ell}$; carbon to nitrogen ratio, 40; carbon to phosphate, 18; $FeSO_4{\cdot}7H_2O\;20mg/{\ell}$).

  • Formaldehyde, an indoor volatile organic compound, is considered toxic due to its carcinogenic risk. Recently, we isolated a formaldehyde-degrading bacterium Pseudomonas sp. YK-32. A crude enzyme prepared from YK-32 also degraded formaldehyde, suggesting that YK-32 cells have formaldehyde hydrogenase activity which is one of the important factors in formaldehyde degradation. The formaldehyde hydrogenase activity was increased 1.25 fold by adding 0.1 % glucose and formaldehyde to the culture medium. In addition, treatment with 1 mM EDTA as a permeabilizer promoted the degradation of formaldehyde and increased the enzymatic activity.

Isolation and Characterization of Benzene-degrading Bacteria. (Benzene 분해 세균의 분리와 특성연구)

  • To evaluate the treatability of activated sludge induced by benzene with microorganisms, isolation and characterization of benzene-degrading microorganisms were carried out. Six bacterial isolates from the activated sludge were identified ; Pseudomonas fluorescens, Enterobacter agglomerans, Enterobacter cloacae, Klebsiella oxytoca, Citrobacter freundii and Klebsiella pneumoniae. P. fluorescens degraded 55% of benzene contained in the medium as a sole carbon source, E. cloacae 24%, E. agglomerans 41%, and K. oxytoca 32%. Optimal temperature, pH and benzene concentration for growth of P. fluorescens appeared to be 31$^{\circ}C$, pH 7.0, and 300mg benzene per liter. When the P. fluorescens was dominant in the activated sludge induced by benzene, the indicator protozoa was Aspidisca sp. When concentration of benzene was about 387mg per liter, the growths of Aspidisca sp. and Litonotus sp. were high. Protozoa, Litonotus sp. and Vorticella sp. did not grow over 1600mg of benzene per liter.

  • Cellulose-degrading bacteria were isolated and identified from cocopeat which has a good quality as a bulking agent in composting. Various bacteria from different sourecs of cocopeat were detected on CMC agar media, and these were found to be Burkholderi2a sp., Bacillu subtilis, Sphingomonas sp., Rhodotorula sp. & Pseudomonas sp. etc. Among these, four bacteria were further selected and analyzed for their biochemical characteristics and CMCase activities. CMCase activities of four bacteria, P. aeruginosa, P. stutzeri, B. subtilis, and P. luteola were found to be 83%, 40%, 8%, 6%, respectively, compared with that of the standard strain Cellulomonas sp.

Isolation of Plasmids from the Moderately Halophilic Bacteria (Moderate 호염성 세균의 Plasmid 유전자 분리)

  • Moderately halophilic bacteria were collected from solar salt with Larsen medium containing $10\%$ NaCl. A total of 56 strains were isolated and tested for the presence of plasmid DNA by agarose gel electrophoresis. Twelve isolates ($21\%$) carried at least one kind of plasmid. Six different isolates among them were selected to study the molecular weight of plasmids and the morphological and physiological characters. Vibrio sp. 14, Alcaligenes sp. 63, Pseudomonas sp. 11, Flavobacterium sp. 38, Bacillus sp. 16, and Alcaligenes sp. 52 carried at least one plasmid of about 7.2 kbp, 6.4 kbp, 6.85 kbp, 8.5 kbp, 8.75 kbp, and 6.8 kbp respectively.
