Recovery of Pseudomonas anguilliseptica from Diseased Striped Beakperch (Oplegnathus fasciatus) in Korea

  • In recent years, several southern coastal fish farms in Korea have experienced 2-30% mortality in striped beakperch suffering from bacterial infections during the spring. In this study, we identified a bacterium isolated from diseased striped beakperch as Pseudomonas anguilliseptica via a biochemical test and 16S rDNA sequence analysis. To evaluate the susceptibility of striped beakperch to P. anguilliseptica, $4.39\times10^7$ or $4.39\times10^5$ CFU/fish of bacteria were injected intraperitoneally at $18\pm1^{\circ}C$ into fish weighing 5.5 g. Cumulative mortalities reached 100% and 45% in the $4.39\times10^7$ and $4.39\times10^5$ CFU/fish infected groups, respectively. Experimentally infected fish showed cell-associated inflammation as well as bacteria in the kidneys and spleen. This study is the first report of striped beakperch mortality caused by P. anguilliseptica, which has pathogenicity in striped beakperch.

PCR-based identification of Pseudomonas fluorescens in diseased olive flounder, Paralichthys olivaceus, in Jeju Island, South Korea

  • Pseudomonas is currently causing increasing mortality in farmed olive flounder in Jeju Island. It was previously reported that P. anguilliseptica is the pathogen causing the mortality. It is not known whether other sub-species are involved or not. In this study, P. fluorescens was identified from diseased olive flounder by a PCR-based diagnosis. Based on genomic sequencing and BLAST analysis, 5 out of 6 samples were closer with P. fluorescens than P. anguilliseptica. Our finding suggests that P. fluorescens may be the dominant species causing the disease in farmed olive flounder in Jeju Island, South Korea.

저수온기의 제주도 양식 돌돔, Oplegnathus fasciatus, 병어로부터 분리한 병원세균의 특성 (Characterization of a bacterium isolated from diseased rock sea bream, Oplegnathus fasciatus, during low water temperature season in Jeju island)

  • 제주도에서 양식되는 돌돔에서 저수온기에 체표에 출혈성 패혈증 혹은 진무름 및 안구백탁이 수반되며 폐사하는 증상이 지속적으로 발견되었다. 이러한 증상의 원인균으로 추정되는 세균을 병어의 체표 및 장기에서 순수분리하였고 형태학적, 배양적 생화학적 조사 및 16S rDNA sequencing 분석 결과 Pseudomonas anguilliseptica로 동정되었다. 본 균주의 병원성 유무를 확인하기 위하여 인위 감염시험을 실시하였고, 그 결과 시험구에서 21일만에 실험어 20마리 모두가 자연감염된 병어와 유사한 증상으로 폐사하였다. 분리균주의 생육 최적 온도는 $\25^circ{C}$였고 생육 최적 pH는 7-8사이로 좁은 범위를 보였다. 낮은 염분농도에서 생육능이 우수하였으며 3%의 염분이 첨가된 배지에서도 생육능이 양호하였다. 약제내성 검사에서는 doxycycline, chloramphenicol, ciprofloxacin, erythromycin, flumequine, nalidixic acid, norfloxacin, ofloxacin에 감수성이 높게 나타났으나 페니실린계의 항생제에 높은 내성을 나타내었다.
