Screening of Oriental Herbal Medicines for Antibacterial Activities

  • The water extracts of oriental herbal medicines which have been clinically used to treat bacterial infections in Korea were screened for in vitro antibacterial activity by the paper disc assay method. Two Gram positive bacteria, Staphylococcus aureus SG511, Bacillus subtilis ATCC 6633 and two Gram negative bacteria, Escherichia coli 055, Pseudomonas aeruginosa 9027 were used as test organisms. Among 83 of the extracts tested, 25 were active against Staphylococcus aureus SG511, 9 were active against Bacillus subtilis ATCC 6633, while none showed inhibitory activity against Eschelichia coli 055 and Pseudomonas aeruginosa 9027. Among them, Hwangyonhaedoktang plus hwangyon, Chongwisan, and Ssangbaksan showed remarkably potent antibacterial activity.

The Effect of Coptidis Rhizoma, Feaxini Cortex, Jinpisan(秦皮散) on Experimental Pseudomonas aeruginosa Keratitis. (黃連, 秦皮, 秦皮散이 綠膿菌性 角膜炎에 미치는 效果에 관한 實驗的 硏究)

  • Coptidis Rhizoma, Fraxini Cortex, Jinpisan(秦皮散) have been as eye washes of inflammatory eye disease in the oriental medicine. Especially Jinpisan(秦皮散) has been used for the disease which is similar to Peudomonas aeruginisa keratitis. This research was attempted to investigate the effect of Coptidis Rhizoma, Fraxini Cortex, Jinpisan(秦皮散), on Peudoronas aeruginisa keratitis. Pseudomonas aeruginosa keratitis causes a deep rapid intense ulceration which often leads to perforation of the cornea within 48 hours. In this research, we induced keratits in the rabbits by inoculating Pesudomonas aeruginosa(9027) and observed the effect on the keratitis and the irritation against the external eye. Also we mesured the minimum inhibitory consentration(MIC) of Coptidis Rhizoma, Fraxini Cortex, Jinpisan(秦皮散) by agar diliution method and the anti-bacterial activites by disk method. The tested bacteria were as follows : a) Pseudomonas aeruginosa (9027), b) Streptococcus pneumoniae(6303), c) Staphylococcus epidermidis(12228), d) Staphylococcus aureus(6538P). The results were as follows ; 1. The groups which were applied eye washes of Fraxini Cortex, Jinpisan reavealed a significant effect, but the group applied eye wash of Coptidis Rhizoma reveaded no effect on Pseudomonas aeruginosa keratitis. 2. Applying eye washes of Coptidis Rhizoma, Fraxini Cortex, Jinisan revealed an irritation against external eyes. 3. Coptidis Rhizoma showed an anti-bacterial activity on Pseudomonas aeruginosa, Streptococcus pneumoniae, Staphylococcus epidermidis, Staphylucoccus aureus by agar diliution method 4. Coptidis Rhizoma showed an anti-bacterial activity on Pseudomonas aeruginosa, Streptococcus pneumoniae, Staphylococcus epidermidis, Staphylococcus aureus by disk method. 5. Fraxini Cortex showed an anti-bacterial activity on Pseudomonas aeruginosa, Streptococcus pneumoniae, Staphylococcus epidermidis, Staphylococcus aureus by agar diliution method 6. Fraxini Cortex showed an anti-bacterial activity on Pseudomonas aeruginosa, Sireptococcus pneumoniae, Staphylococcus epidermidis, Staphy1ococcus aureus by disk method. 7. Jinpisan showed an anti-bacterial activity on Pseudomonas aeruginosa, Streptococcus pneumoniae, Staphylococcus epidermidis, Staphylococcus aureus by agar diliution method. 8. Jinpisan showed an anti-bacterial activity on Pseudomonas aeruginosa, Streptococcus pneumoniae, Staphylococcus epidermidis, Staphylococcus aureus by disk method. According to the above results, Fraxini Cortex, Jinpisan(秦皮散) are recognized to have an effective treatment on the Pesudomonas aeruginosa keratitis, so this experiment is thought to be a basic ingredient in proving the effect of Fraxini Cortex, Jinpisan which is applied many in documents and clinical medicine. In the comparison of anti-bacterial activity and results of treatment on the Pesudomonas aeruginosa keratitis, Jinpisan(秦皮散) was more effective than Coptidis Rhizoma, Fraxini Cortex.

Two Gram positive bacteria, Staphylococcus aureus SG 511, Bacillus subtilis ATCC 6633, and two Gram negative bacteria, Escherichia coli 055, Pseudomonas aeruginosa 9027 were used as test organisms. Among the extracts tested, MeOH extract of Baenongtang showed remarkably potent antibacterial activity. Activity-guided chromatographic fractionations of the $CH_2Cl_2$ extract of Baenongtang afforded seven antibacterial compounds.

A Study on Remediation of Diesel-Contaminated Soil by Biosurfactant- Enhanced Soil Washing (생물계면활성제를 이용한 디이젤 오염토양세척기술에 관한 연구)

  • Soil washing by surfactants is a technology to enhance mobilization and subsequent degradation of oil pollutants by reducing the surface tension of pollutants which is combined with soil. In this study, biosurfactant, rhamnolipid was produced from Pseudomonas aemginosa ATCC 9027 which had an excellent biodegradable activity in soil without causing secondary pollution. Effects of chemical surfactants on the removal of diesel from diesel-contaminated soil were compared to those of biosurfactants including rhamnolipid. Diesel removal efficiency by rhamnolipid extracted from P. aeruginosa culture broth was over 95% in both batch and column washing test in 5,000ppm diesel-contaminated soil with 1% surfactants after washing for 24 hours. On the contrary, the results of chemical surfactants were below 50∼80%, The chemical surfactants with HLB value(8∼15) showed more then 75% efficiency of diesel removal. But, when the HLB values were below 8 or over 15. their efficiency were observed as less then 60% of diesel removal. Rhamnolipid, biologically produced surfactants, may also be promising agent for enhancing diesel removal from contaminated soil.

Selection and Characteristics of Bacteriocin-Producing Microorganism to Utilize in Anti-Bacterial Rice Brain Protein Film Production (항균성 미강 단백질 필름 개발을 위한 Bacteriocin 생성균주의 선별 및 특성)

  • This study was conducted to select the bacteriocin-producing microoreanism cultivated in the rice bran culture and to characterize the produced bacteriocin for the further purpose of economical and anti-bacterial rice bran protein film. Pseudomonas putida 21025 was cultivated from rice bran and identified as a producer of a bacteriocin which showed bactericidal activity against Pseudomonas aeruginosa 9027. Bacteriocin produced by Pseudomonas putida 21025 showed a broad spectrum of activity against spoilage and soil bacteria. The activity of the bacteriocin produced by Pseudomonas putida 21025 decreased after 1 hr of staying at the temperature of $50^{\circ}C$, and with the presence of some organic solvents, except hexane and ethanol. However, the bacteriocin activity was stable throughout the pH ranges of 6-9 for 2 hrs, at the temperature lower than $50^{\circ}C$, and with the presence of ethanol for 3 hrs. The bacteriocin was partially purified by 50% ammonium sulfate precipitation followed by subsequent dialysis. Direct detection of the partially purified bacteriocin on SDS-PAGE suggested that it had an apparent molecular mass of about 21.6 kDa.

Studies on the Synthesis of Pterdine Substituted Pyridonecarboxylic Acids as Potential Antibacterial Agents and their Antimicrobial Activities (항균제로서 Pteridine이 치환된 Pyridonecarboxylic Acids의 합성 및 항균 작용에 관한 연구)

  • In order to synthesize a new antibacterial and antitumor agents, we have prepared new analogues pteroic acid(13a, 13b), which means C-9 position of pteroic acid has been replaced by norfloxacin(8) or ciprofloxacin(9) and amino group of C-2 position by $CH_3$. These derivatives were synthesized coupling at N-4 piperazine of norfloxacin and ciprofloxacin with 2-amino-3-cyano-5-chloromethylpyrazine(20) provided 1-alkyl(ethyl, cyclopropyl)-6-fluoro-1,4-dihydro-4-oxo-7-[[4-N-(2-amino-3-cyanopyrazin-5-yl)methyl]piperazin-1-yl]-3-quinoline-carboxylic acid(12a, 12b). It was then cyclized with acetamidine. HCI to obtain new analogues of C-2 desaminomethylpteroic acid(13a, 13b) in yield of 76.2% and 82.8 % respectively. These compounds were tested in vitro on antibacterial activity against Gram-positive and Gram-negative bacteria including Pseudomonas aeruginosa ATCC9027. In general, these synthesized compounds(13a, 13b) showed less potent activities than those of norfloxacin.

The Antimicrobial Effects of Natural Aromas for Substitution of Parabens (합성 항균제를 대체하기 위한 천연물질의 항균 효과)

  • Aroma oils extracted from the natural material have antibacterial, antivirus, antiinflammatory, and preservative effect. The preserve efficacy testing between aroma oils and parabens as an artificial preservative had been performed and then it had been suggested that aroma oil was possibile to apply to the cosmetics. Aroma oils were pine, rosemary, lemon and eucalyptus, and parabens were methylparaben, blitylparaben. Antiseptic concentrations of aroma oils and parabens having 0.0, 0.1, 0.2, 0.4, 0.8, 1.0wt% were tested respectively. Escherichia coil(ATCC No.8739), Pseudomonas aeruginosa(ATCC No. 9027) which are gram-negative and Staphylococcus aureus (ATCC No. 6538), Bacillus subtilis(ATCC No. 6633) which are gram-positive were used as the test organisms. Disk paper and broth dilution methods were used as the methods of preservative efficacy testing. The antibacterial activity of aroma oils and parabens for gram-positive were better than that for gram-negative. For the antibacterial activity aroma oils were better than parabens. Among the aroma oils, rosemary and pine having superior antibacterial activity were selected and blended to illuminate if there is any synergy, There was synergical effect and optimum ratio of aroma blend is 3 : 1(rosemary pine) in this study.

Preservation of Cosmetics by Ethanol Extract of Scutellaria baicalensis $G_{EORGE}$ (황금(黃芩)의 에탄올추출물에 의한 화장품 방부효과)

  • Ethanol extract (1.0 wt%) of Scutellaria baicalensis $G_{EORGE}$ practically satisfied CTFA (The cosmetics, Toiletry, and Fragrance Association) standard in its antimicrobial activity against Staphylococcus aureus, Pseudomonas aeruginosa and Candida albicans which were inoculated in skin toner and skin lotion. At concentrations of the extract less than 1.0 wt% the survival rate was the best for S. aureus and the worst for P. aeruginosa. The antimicrobial effect seems to be due to the damage to the bacterial cell wall as evidenced by the images of a Field Emission-Scanning Electron Microscope. The extract also showed a high anti-oxidant effect, and superoxide dismutase (SOD)-like activity reached 80% at 1,000 ppm. These findings suggest that the extract of Scutellaria baicalensis $G_{EORGE}$ is applicable to cosmetics as a natural preservative and an anti-oxidant.

Antibacterial Activities of Phenolic Components from Camellia sinensis L. on Pathogenic Microorganisms

  • Antibacterial activities of the major phenolic components from Camellia sinensis L. were investigated against several pathogenic microorganisms including Gram-positive strains like Staphylococcus aureus ATCC 29213 and Streptococcus pyogens 308A; and Gram-negative strains like Escherichia coli ATCC 25922, Escherichia coli 078, Pseudomonas aeruginosa 9027, and Enterobacter cloacae 1321E. The MIC values demonstrate that both (-)-epicatechin and (-)-epigallocatechin were more considerably toxic against Staphylococcus aureus ATCC 29213 than the other two catechins like (-)-epicatechingallate and (-)-epigallocatechin-3-gallate. (-)-Epicatechingallate and (-)-epigallocatechin-3-gallate were most inhibitory against Escherichia coli ATCC 25922. As a result, (-)-epicatechin showed predominant antibacterial activities among tea varieties. The contents of major polyphenolic components such as four catechins, theaflavin, and quercetin were different according to fermentation processes. The total contents of four catechins were ranged from 13.81 to 1.33%, with (-)-epigallocatechin-3-gallate being dominant among tea varieties; theaflavin was found the characteristic pigment in fully-fermented black tea.

Bioactivity of Metabolites from Actinomycetes Isolates from Red Sea, Egypt

  • Actinomycetes isolated from marine habitats represent a promising source of bioactive substances. Here, we report on the isolation, identification, productivity enhancement and application of the bioactive compounds of Streptomyces qinglanensis H4. Eighteen marine actinomycetes were isolated and tested for resistance to seven bacterial diseases. Using 16S rRNA sequencing analysis (GenBank accession number MW563772), the most powerful isolate was identified as S. qinglanensis. Although the strain produced active compound(s) against a number of Gram-negative and Gram-positive bacteria, it failed to inhibit pathogenic fungi. The obtained inhibition zones were 22.0 ± 1.5, 20.0 ± 1, 16.0 ± 1, 12.0 ± 1, 22.0 ± 1 and 24.0 ± 1 mm against Bacillus subtilis ATCC 6633, Escherichia coli ATCC 19404, Enterococcus faecalis ATCC 29212, Pseudomonas aeruginosa ATCC 9027, Candida albicans ATCC 10231 and Staphylococcus aureus ATCC6538, respectively. To maximize bioactive compound synthesis, the Plackett-Burman design was used. The productivity increased up to 0.93-fold, when S. qinglanensis was grown in optimized medium composed of: (g/l) starch 30; KNO3 0.5; K2HPO4 0.25; MgSO4 0.25; FeSO4·7H2O, 0.01; sea water concentration (%) 100; pH 8.0, and an incubation period of 9 days. Moreover, the anticancer activity of S. qinglanensis was tested against two different cell lines: HepG2 and CACO. The inhibition activities were 42.96 and 57.14%, respectively. Our findings suggest that the marine S. qinglanensis strain, which grows well on tailored medium, might be a source of bioactive substances for healthcare companies.
