Recently, there is growing interest in ultraviolet (UV) irradiation as a disinfection technic in drinking water production due to its effectiveness to inactivate microorganisms such as Crytosporidium parvum without forming disinfection byproducts. However, UV disinfection is known for its drawback such as photoreactivation. Despite many works concerning the photoreactivation, most of works were focused on indicator or non pathogenic microorganisms. The objective of this study is to examine the photoreactivation of Pseudomonas aeruginosa which is an opportunistic pathogen as UV radiation by LP and MP UV lamp was applied. The result showed that P. aeruginosa had high photo repair efficiency regardless of the type of UV irradiation. Both of the effective log repair values of LP and MP UV irradiation were found approximately 2.6 log. In addition, photo repaired P. aeruginosa was not significantly different in forming biofilm in comparison with non treated P. aeruginosa.
Temperature and pH conditions were studied for an effective biosurfactant production by Pseudomonas aeruginosa YPJ-80. Efficient methods of biosurfactant separation were also investigated. pH-uncontrolled experiments at 35$^{\circ}C$ and an initial pH of 8 resulted in the best cell growth (3.6 g/L) and biosurfactant production (0.073 g biosurfactant/g cell). Biosurfactant separation was most efficient using solvent extraction with chloroform/methanol (2:1 vol%) followed by acidification using 1N HCl.
광역학 요법(photodynamic therapy)은 특정 파장의 빛에 의해 활성화되는 광민감제(photosensitizer)를 사용하여 세포 내 산소를 활성화시키는 치료 방법으로, 항생제 내성균에 의한 상처 감염의 치료에 유망한 접근 방법이다. 일반적으로 건강한 사람에게 비병원성인 녹농균(Pseudomonas aeruginosa, P. aeruginosa)은 특정 병원성 징후를 보이지 않지만, 피부 손상이나 면역력이 저하된 사람들에서는 패혈증과 같은 심각한 질병을 유발할 수 있다. 항생제는 P. aeruginosa 감염에 대한 전통적인 치료법이나 약물 오용으로 인한 항생제 내성의 증가는 이러한 감염을 관리하는 데 큰 어려움을 준다. 본 연구에서는 P. aeruginosa의 억제제로서 광민감제(PhotoMed, Methyl pheophorbide A, Radachlorin®)와 다이오드 레이저를 이용한 광역학 치료 효과를 조사하는 것을 목표로 했다. P. aeruginosa 현탁액과 광민감제(PhotoMed, Methyl pheophorbide A, Radachlorin®)를 페트리 접시에 접종하여 30분 후 다이오드 레이저를 사용하여 3 J/cm2의 에너지 밀도로 조사했다. 그 결과, P. aeruginosa는 PhotoMed에서 79.65%, Methyl pheophorbide A에서 47.36%, Radachlorin®에서 40.91%의 사멸률을 보였다. 이번 연구는 P. aeruginosa를 억제하기 위한 가장 효과적인 접근법이 PhotoMed와 다이오드 레이저를 이용한 광역학 치료임을 보여준다.
The aim of this study was to investigate the antimicrobial resistance among Pseudomonas (P.) aeruginosa isolated from dogs and cats. A total of 45 (6.2%) P. aeruginosa was isolated from 710 dogs and 21 cats with clinical signs. Resistance to one or more of the antimicrobials tested was observed in 26 (57.8%) P. aeruginosa. Resistance to cefepime was the most frequent (44.4%), followed by ofloxacin (22.2%), levofloxacin (17.8%), norfloxacin (8.9%), ciprofloxacin (6.7%), ceftazidime, aztreonam, colistin, polymixin B and gentamicin (4.4%, respectively), while resistance to piperacillin/tazobactam, imipenem, tobramycin and amikacin was 2.2%, respectively. All isolates were susceptibility to doripenem and meropenem. Antimicrobial susceptibility testing should be a crucial step in selection of appropriate antimicrobial therapy in veterinary medicine. Also, the prudent use of antimicrobials and continuous monitoring for companion animals are required.
본 실험은 생분해가 어려운 절삭유를 단일 균주에 의해 생물학적 처리를 하는 데에 목적이 있다. 절삭유, 절삭폐유로부터 호기 균주 81개를 분리하여 이중 절삭유 분해능이 가장 높은 균주로, 48시간 내에 90.4%를 제거한(초기농도 699.1 mg/L) KS47을 선별하였다. KS47은 형태학적, 생리 화학적, 16S rDNA 염기서열, 그리고 지방산 분석을 통해 Pseudomonas aeruginosa로 동정되었다. P. aeruginosa KS47은 절삭유를 탄소원으로 사용하여 성장 할 수 있었으며, 절삭유 분해시, 최적 분해 조건은 1.5 g/L(wet weight), pH 7.0, $30^{\circ}C$이었다. 최적 조건 하에서 절삭유의 제거능을 본 결과, 1,060 mg/L의 절삭유를 12시간 내에 83.7% 제거함을 확인하였다.
사람의 감염증 치료에 사용되는 carbapenem계 약제에 대한 내성균의 출현 및 확산은 감염증 치료를 제한할 뿐만 아니라 집단 발병의 원인이 될 수 있다. 이에 본 연구에서는 ${\beta}$-lactam 약제에 내성을 갖는 Pseudomonas aeruginosa (P. aeruginosa)를 대상으로 metallo-beta-lactamase (MBL)의 유전형을 규명함으로써 내성세균의 감염증 치료지침 및 확산방지책 마련에 기초 자료를 제공하고자 하였다. 본 연구의 대상이 된 254개의 임상 검체 중에서 42주의 P. aeruginosa 를 분리하여 imipenem 혹은 meropenem에 내성을 나타내는 Hodge 변법과 EDST에서 각각 28주와 23주가 양성반응을 보였다. DNA의 염기서열 분석결과 $bla_{IMP-6}$ 유전자 보유균이 8주, $bla_{VIM-2}$ 유전자 보유균이 17주로 59.5%(25/42)가 MBL을 생성하는 것으로 나타났다. $bla_{IMP-6}$의 유전자 환경은 $bla_{IMP-6}$-qac-aacA4-$bla_{OXA-1}$-aadA1 유전자 배열을 지니고 있었다. 또한 ERIC PCR 결과 IMP-6과 VIM-2를 생성하는 일부 균주에서 역학적 연관성이 있음이 확인되었다. 본 연구에서 분리한 carbapenem계 항균제 내성 P. aeruginosa가 보유한 $bla_{IMP-6}$ 유전자는 대구지역에서 발병이 보고된 유전자의 gene cassette와 일치하는 것으로 확인되었다. 따라서 이들 세균이 지역사회에 정착하고 있고 이들을 보유한 세균에 의한 감염증 치료시 치료약제에 대한 선택압을 증가시킬 것으로 우려된다. 그러므로 항균제 내성 검사를 통하여 적절한 항균제를 선택하고, 항균제 내성균들의 출현과 확산을 막는 연구가 계속되어야 할 것으로 생각된다.
Objectives: The most prominent microorganisms that cause hospital infections and acquire antibiotic resistance are Staphylococcus aureus and Pseudomonas aeruginosa. The present study aimed to compare the phenolic and flavonoid compounds of various Allium ampeloprasum and Allium porrum extracts and evaluate the antibacterial effects of these extracts against these two microorganisms. Methods: The total phenolic and flavonoid contents of the acetone, methanol, aqueous, and hexane leeks extracts from A. ampeloprasum and A. porrum were measured. The antibacterial activity of these extracts against S. aureus and P. aeruginosa was tested using the disk diffusion method for 24, 48, and 72 hours. Further, the minimum inhibitory concentrations and the minimum bactericidal concentrations of these extracts for these two bacteria were evaluated and compared with those of common antibiotics. Results: The aqueous extracts showed the highest phenolic and flavonoid contents and at concentrations of 35 and 40 mg per disk, showed the most antibacterial activity against S. aureus and P. aeruginosa; P. aeruginosa showed more sensitivity to the aqueous extracts than S. aureus. Conclusion: Aqueous A. ampeloprasum and A. porrum extracts may prevent the growth of hospital pathogens, especially P. aeruginosa; our findings will aid the discovery of new antimicrobial substances against antibiotic-resistant bacteria.
The 543 microbial strains were isolated from the rhizosphere in ginseng root rot-suppressive soil. From these, isolate KGM-100 was finally selected for the screening of powerful antagonistic bacterium for the soil-born Fusarium solani causing root rot of many important crops. The isolate KGM-100 was identified as Pseudomonas aeruginosa. Antibiotics produced by the P. aeruginosa KGM-100 were partially characterized as heat-stable and low-molecular weight antibiotics. The strain also produced siderophore, which was assumed to be pseudobactin. Pot test showed approximately 30-50% biological control effects when Fusarium solani-infected soil was treated with the P. aeruginosa KGM-100.
This study presents a fluorescent mechanism for two-step amplification by combining two widely used techniques, exponential amplification reaction (EXPAR) and catalytic hairpin assembly (CHA). Pseudomonas aeruginosa (P. aeruginosa) engaged in competition with the complementary DNA in order to attach to the aptamer that had been fixed on the magnetic beads. The unbound complementary strand in the liquid above was utilized as a trigger sequence to initiate the protective-EXPAR (p-EXPAR) process, resulting in the generation of a substantial quantity of short single-stranded DNA (ssDNA). The amplified ssDNA can initiate the second CHA amplification process, resulting in the generation of many double-stranded DNA (dsDNA) products. The CHA reaction was initiated by the target/trigger DNA, resulting in the release of G-quadruplex sequences. These sequences have the ability to bond with the fluorescent amyloid dye thioflavin T (ThT), generating fluorescence signals. The method employed in this study demonstrated a detection limit of 16 CFU/ml and exhibited a strong linear correlation within the concentration range of 50 CFU/ml to 105 CFU/ml. This method of signal amplification has been effectively utilized to create a fluorescent sensing platform without the need for labels, enabling the detection of P. aeruginosa with high sensitivity.
In order to develop an effective means to treat P. aeruginosa infections, we have purified P. aeruginosa outer membrane proteins (OMPs)-specific human IgG antibody. In this study, we investigated the protective activity of the purified anti-OMPs IgC against P. aeruginosa infection in a rabbit endophthalmitis model. Rabbits were inoculated by an intravitreal injection with P. aeruginosa, and treated with a single dose of 1 mg anti-P. aeruginosa OMPs IgG. All the control rabbits predominantly developed edematous responses and opacity in the eyes, but the rabbits treated with the antibody showed only very limited degree of edema. Aliquots of the vitreous humor were extracted and analyzed for the number of viable bacteria and endotoxin level. The results showed that the anti-OMPs IgC significantly reduced the bacterial count compared with the control group, and that the endotoxin level of the vitreous from the IgG-treated rabbits was more than 70-fold lower 6 h after the administration than the control animals. These data suggested that the anti-P. aeruginosa OMPs IgG is effective in inhibiting the bacterial growth and thereby in reducing endotoxin levels in the vitreous, warranting further development of the anti-P. aeruginosa OMPs IgG as a therapeutic means for treating Pseudomonas endophthalmitis.
