Effects of Persimmon (Diospros kaki L.) Vinegar as a Dietary Supplement on Feed Intake, Digestibility, and Ruminal Fermentation Indices in Sheep

  • This study estimated the effect of fermented persimmon (Diospros kaki L.) extract (FPE) supplement on feed intake, digestibility, nitrogen (N) balance, and rumen fermentation characteristics in sheep. Five male sheep (Corriedale${\times}$Polwarth) with average body weight of $48.6{\pm}1.3\;kg$ were housed in metabolism crates and assigned to a 5${\times}$5 Latin square design with five consecutive 20-d periods which consisted of 14-d adaptation and 6-d data collection. The sheep were fed ad libitum a diet containing concentrate and rice straw (3:7). The five treatments were FPE supplemented at 0 (Control), 5, 10, 20, and 30 g/kg of concentrate. Intakes of dry matter (DM, p<0.01), organic matter (OM, p<0.01), neutral detergent fiber (NDF, p<0.05), acid detergent fiber (ADF, p<0.05), and nitrogen-free extract (NFE, p<0.01) increased quadratically with increasing intake of FPE supplement and maximized (p<0.05) at 10 g/kg FPE. The digestibilities of DM (p<0.05), OM (p<0.05), crude protein (p<0.01), and NFE (p<0.01) increased quadratically with increasing amount of FPE supplement, and sheep fed 5 and 10 g/kg diets had greater (p<0.05) DM, OM, and NFE digestibilites than the Control treatment. By increasing FPE supplement concentration, N intake (p<0.01) and fecal N (p<0.05) increased linearly, whereas retained N (p<0.05) and retained N ratio (p<0.05) increased quadratically. The retained N was maximized (p<0.05) in sheep fed 5 and 10 g/kg diets. The mean rumen pH was not affected by FPE supplement, but there was a quadratic increase (p<0.05) of mean rumen ammonia N concentration and a linear increase (p<0.01) in mean rumen total volatile fatty acid (VFA) and acetate concentrations. The mean concentration of rumen propionate in sheep fed all FPE supplemented diets was greater (p<0.05) than the Control, but the mean ratios of rumen acetate to propionate in sheep fed 5 and 10 g/kg diets were lower (p<0.05) than that of Control sheep. In conclusion, FPE supplemented at 5-10 g/kg of concentrate improved feed intake, the digestibilites of OM and NFE, N metabolism, and rumen fermentation indices of sheep.

Effects of Feeding Rancid Rice Bran on Growth Performance and Chicken Meat Quality in Broiler Chicks

  • A total of 225 day-old broiler chicks (43.08 g initial body weight) were allotted to three dietary treatments for a 6-week feeding trial. The treatments were 1) Control (defatted rice bran; DFRB), 2) fresh rice bran (FRB) and 3) rancid rice bran (RRB). Rice brans were intentionally spoiled by two degrees of rancidity by the values of free fatty acids (FFA): 7.6% (FRB) and 16.3% (RRB). Diets were prepared on an isonutrient basis, and defatted or rancid rice brans were included 5 and 10% for starter (0-3 week) and finisher (3-6 week), respectively. At the end of the feeding trial, six chicks per treatment were sacrificed, and thigh meats were ground and stored at $1^{\circ}C$ for thiobarbituric acid reactive substances (TBARS) and peroxide value (POV) analyses. For a digestibility, 48 growing chicks (4 weeks old) were employed in cages (3 replicates/treatment, 2 birds/cage) according to the experimental design: FRB, RRB, pelleted and extruded rice bran. Some of the FRB were pelleted ($70^{\circ}C$) or extruded ($110^{\circ}C$). There was no significant difference in growth performance during the starter period, but chicks fed a diet containing DFRB grew faster (p<0.05) with increased feed intake (p<0.05) than those fed diets containing rice brans, FRB or RRB, during the finisher period. Feed conversion ratio in the RRB was inferior (p<0.05) to the DFRB. Between rice bran groups, weight gain was higher (pco.os) in FRB than in RRB during finisher period. There was a similar trend in growth performance of chicks for the overall period (0-6 week) as the finisher period. Dry matter and energy digestibilities were higher (p<0.05) in extruded than in RRB group. Protein digestibility was improved (p<0.05) when rice bran was extruded, but not pelleted. The chicken meats from RRB showed higher (p<0.05) TBARS than those from FRB during storage for 4 weeks at $1^{\circ}C$. In conclusion, it would appear that feeding rancid rice bran gave negative effects on growth performance and lipid stability of meat in broiler chicks.

Effects of Green Tea Catechin on Mixed Function Oxidase System and Antioxidative Defense System in Rat Lung Exposed to Microwave

  • The purpose of this study was to investigate the effects of green tea catechin on mixed function oxidase system (MFO), lipofuscin contents, carbonyl value, oxidative damage and the antioxidative defense system in lung of microwave exposed rats. Experimental groups were divided to normal group and microwave exposed group. The microwave exposed groups were subdivided into three groups: catechin free diet (MW-0C) group, 0.25% catechin (MW-0.25C) group and 0.5 % catechin (MW-0.5C) group according to the levels of dietary catechin supplementation. The rats were irradiated with microwave at frequency of 2.45 GHz for 15 min. Experimental animals were sacrificed at 6th day after microwave irradiation. The contents of cytochrome P$_{450}$ contents in MW-0C group was increased to 95% , compared with normal group. MW-0.25C and MW-0.5C groups were reduced to 16% and 31%, respectively, compared with MW-0C group. The activity of NADPH-cytochrome P$_{450}$ reductase in MW-0C group was increased to 44%, compared with normal group. MW-0.25C and MW-0.5C groups were reduced to 12% and 17%, respectively, compared with MW-0C group. The activity of superoxide dismutase (SOD) in MW-0C group was decreased to 21 %, compared with normal group. MW-0.25C and MW-0.5C group were significantly (p < 0.05) increased, compared with MW-0C group. The activity of glutathione peroxidase (GSHpx) in MW-0C group was significantly decreased, compared with normal group. MW-0.25C and MW-0.5C groups were recovered to the level of normal group. The thiobarbituric acid reactive substances (TBARS) content in MW-0C group was increased to 34 %, compared with normal group. Catechin supplementation groups were maintained the level of normal group. The levels of caybonyl value in MW-0C group was increased to 21 %, compared with normal group. MW-0.25C and MW-0.5C groups were reduced to 14% and 12%, respectively, compared with MW-0C group. The lipofuscin contents in MW-0C group were increased to 23.4 %, compared with normal group. That of MW-0.5C group was significantly reduced, compared with MW-0C group. In conclusion, MFO system was activated and the formation of oxidized protein, lipofuscin was increased and antioxidative defense system was weakened of lung tissue in microwave exposed rats, thus oxidative damage was increased. But it was rapidly recovered to normal level by green tea catechin supplementation.n.

유산균 발효 와송이 알코올성 유발 지방간 흰쥐의 조직 과산화지질 및 혈중 지질농도에 미치는 영향 (Effect of Orostachys malacophyllus by Fermented Lactic Acid Bacteria on Plasma Levels of Lipid and Lipid Peroxidation in Alcohol Feeding Rats)

  • 와송(OM) 분말과 3종의 유산균 혼합물로 발효하여 얻은 발효 와송(FOM)을 식이 중에 각각 2.5%와 5%의 농도로 첨가하여 알코올 투여와 함께 4주동안 흰쥐에 급여한 후 조직의 과산화지질 농도와 혈중 지질수준에 미치는 영향에 대하여 검토하였다. 혈중 지질과 혈중 콜레스테롤의 농도는 알코올 투여 대조군(C)에서 높아졌지만, OM군 FOM군 모두 낮은 수치를 보였고, 특히 5FOM군에서 유의적으로 낮은 수치를 나타내었다. 5FOM군에서 혈중 총 콜레스테롤 수치는 감소하는데 반해 HDL-콜레스테롤 수치는 높아 동맥경화지수 수치가 낮았다. 또한, 간장 및 혈중 중성지질 농도는 알코올 투여 대조군(C)에서 증가하였고, OM군 FOM군 모두 농도 의존적으로 감소하였으며, 5FOM군에서 정상 수준의 감소를 보여 정상군과 유의한 수치를 나타내었다. 그리고 5 FOM군에서 혈중 총단백질 함량의 유의적인 차이는 인정되지 않았지만, 혈중 알부민 함량은 증가하였다. 간장, 혈청, 고환, 신장, 비장, 심장 조직의 TBARS 함량은 모두 알코올 투여 대조군(C)에서 증가하였고, OM군 FOM군 모두 알코올 투여 대조군에 비해 감소하였으나 5FOM군에서 양성 대조군(SM)의 감소 경향과 비슷한 결과를 나타내었다. 간장 및 혈청의 glutathione 농도 또한 5FOM군에서 증가 경향을 나타내었다. 이상의 결과로 알코올 유발 흰쥐에서 FOM투여는 OM 투여보다 지질 개선에 더 긍정적인 영향을 나타내었고, 생체 내 스트레스를 감소시켜 체내 조직의 항산화 증진에 관여할 것으로 판단되어진다.

장생도라지 생리활성물질이 Streptozotocin으로 유발된 당뇨쥐에 미치는 영향 (Effect of Physiologically Active Compounds Isolated from Platycodon grandiflorum on Streptozotocin-Induced Diabetic Rats)

  • Streptozotocin으로 유발된 당뇨쥐에 있어서 장생도라지로부터 분기된 생리활성물질(이눌린,사포닌, 올리고당) 및 열수추출물을 5주동안 투여한 결과는 다음과 같다. 실험동물의 체중은 이눌린(IN)과 올리고당(OS) 투여군에서는 다소 유의적인 증가를 보였으며, 혈당은 시료투여군에서는 다소 감소하는 경향을 보였는데, 특히 장생도라지 이눌린 투여군(IN)과 총추출물 투여군(WE)에서는 각각 21.3%, 21.2%로 유의적인 감소효과를 보였다. 시료투여군의 혈청 총콜레스테롤은 당뇨대조군에 비해 감소하였는데, 특히 이눌린 투여군(IN)은 정상군 수준을 나타내었고, HDL 콜레스테롤은 당뇨유발군이 정상군보다 높은 수준이었으며, 시료투여군 간의 유의적인 차이는 보이지 않았다. 중성지방의 함량은 장생도라지의 이눌린 및 이눌린$.$사포닌 혼합 투여군에서 감소효과가 가장 높게 나타났다. 간, 신장, 심장, 비장 및 고환의 평균무게를 측정한 결과, 시료투여군에서 전반적으로 간장 및 심장의 무게가 낮아지는 경향을 보였으며, 신장은 당뇨유발군 모두에서 무게가 증가하였다. 혈청 단백질과 알부민 농도는 정상군에 비하여 당뇨대조군 및 시료투여군 모두에서 유의하게 감소되었으며, 당뇨유발군의 혈청 인슐린 농도는 이눌린 투여군(IN)에서 다소 높은 경향을 보였다. 뇨중 에너지 대사물의 조성을 관찰한 결과, 뇨단백은 이눌린(IN) 및 총추출물(WE) 투여군에서 정상군 수준으로 감소되었고, 당뇨유발군에서는 비교적 높은 뇨당이 검출되었으나 이눌린 투여군(IN)에서 검출되지 않았다.

황 및 유황 급이에 따른 흰점박이꽃무지 유충의 체성분 변화 (Changes in Body Composition of Protaetia brevitarsis seulensis Larvae Fed Sulfur and Selenium)

  • 본 연구는 유황 및 셀레니움을 급이하였을 때 굼벵이 체 성분의 변화를 관찰하기 위해 수행되었다. 일반 성분 중에서 단백질의 증가는 황 급이구에서 나타난 반면에, 지방산 증가는 셀레니움 및 황/셀레니움 급이구에서 보였다. 구조아미노산은 대조구에 비교하여 전반적으로 상승되는 경향성을 보였지만, 아미노산 중에서는 특이적으로 변화가 유발되는 것은 관찰되지 않았다. 유리아미노산은 셀레니움 및 황/셀레니움 급이구에서 증가되었다. 비필수아미노산 중 GABA, Pro가 황/셀레니움 급이구에서 증가되었다. 유리아미노산의 필수아미노산 중에서는 Met, Trp, Val이 셀레니움 및 황/셀레니움 그룹에서 증가되는 경향성을 보였다. 지방산은 황/셀레니움 급이구에서 monounsaturated fatty acid가 감소한 반면에, saturated and polyunsaturated fatty acids가 증가되는 경향성을 보였다. 특히, oleic acid가 전혀 검출되지 않은 반면에, palmitic acid, palmitoleic acid, linoleic acid가 증가되는 양상을 보였다. 황 및 셀레니움의 급이에 따른 체 성분 변화를 관찰해 본 결과, 황/셀레니움 급이구는 체 성분의 급격한 변화가 유발될 가능성이 있는 것으로 추정된다. 이와 같은 변화 적절하게 조절하여 기능성을 강화한 굼벵이를 제조하는 것이 가능한 것으로 제의된다.

Effects of fermented soybean meal with Bacillus velezensis, Lactobacillus spp. or their combination on broiler performance, gut antioxidant activity and microflora

  • Objective: A series of experiment were conducted to evaluate the effects of replacing a part of soybean meal (SBM) at 6% of broiler diets with fermented soybean meal (FSBM) obtained by single or two-stage fermentation by measuring growth performance, antioxidant activity in the jejunum and distal intestinal microflora. Methods: Soybean meal samples were prepared by single-stage fermentation using Bacillus velezensis (Bv) (FSBMB), or Lactobacillus spp. (as commercial control) (FSBML). Additional SBM sample was prepared by two-stage fermentation using Bv and subsequently using Lactobacillus brevis ATCC 367 (Lb) (FSBMB+L). Enzyme activity, chemical composition, trichloroethanoic acid-nitrogen solubility index (TCA-NSI) and antioxidant activity were measured. Then, in an in vivo study, 320 Ross308 broilers were divided into four groups with ad libitum supply of feed and water. Four groups were fed either a corn-soybean meal diet (SBM), or one of fermented SBM diets (FSBMB+L, FSBMB, and FSBML). Growth, serum characteristics, microflora, and the mRNA expression of selected genes were measured. Results: Compared to SBM, FSBMB+L contained lower galacto-oligosaccharide, allergic protein, and trypsin inhibitor, and higher TCA-NSI by about three times (p<0.05). Reducing power and 1,1-diphenyl-2-picrylhydrazyl free radical scavenging ability correlated positively with the TCA-NSI content in FSBM. Growth performances were not significantly different among four groups. In jejunum of 35-day-old broilers, partial replacement of SBM by FSBMB+L increased the activity of superoxide dismutase and catalase (CAT), and the FSBMB group had the highest catalase activity (p<0.05). Partial replacement of SBM by FSBM increased relative mRNA expressions of nuclear factor erythroid 2-related factor 2 (Nrf2), heme oxygenase-1 (HO-1), and peptide transporter 1 (PepT1) (p<0.05); however, FSBMB+L increased CAT mRNA level to 5 times of the control (p<0.05). Conclusion: Using Bv- and Lb-processed SBM through two-stage fermentation to partially replace 6% of diets will improve the gut's antioxidant activity under commercial breeding in broilers.

Comparison of amino acid digestibility of soybean meal, cottonseed meal, and low-gossypol cottonseed meal between broilers and laying hens

  • Objective: This study aimed to determine and compare the apparent ileal digestibility (AID) and the standardized ileal digestibility (SID) of amino acids (AA) in soybean meal (SBM), cottonseed meal (CSM), and low-gossypol cottonseed meal (LCSM) fed to broiler chickens and laying hens. Methods: Three semi-purified diets containing the identical crude protein concentration at 20% were formulated to contain SBM, CSM, or LCSM as the sole source of N. A N-free diet was also formulated to estimate the basal ileal endogenous losses of AA for broilers and hens. A total of 300 male Ross 308 chicks at one-day-old and 144 Hy-Line Brown laying hens at 30-week-old with initial egg production rate of 88.3%±1.0% were randomly allocated into 1 of 4 dietary treatments, respectively. Results: CSM and LCSM showed more Arg and Cys+Met while less Lys, Ile, Leu, and Thr relative to SBM. Significant interactions existed between species and experimental diets for AID (except for Arg, Asp, Glu, Gly, and Pro) and SID (except for Arg, His, and Phe) of most AA. Most AA in diets showed higher AID (except for Lys) and SID (except for Lys, Met, and Ser) in broilers relative to laying hens. The AID and SID of all AA were significantly different between the three diets. In broilers, the AID and SID of most indispensable AA except for Arg in SBM and LCSM was higher than CSM. In laying hens, the AID and SID of most indispensable AA except for Arg, Met+Cys, and Phe in SBM was higher than CSM and LCSM. Conclusion: The accurate determination of AID and SID of AA in CSM and LCSM for broilers and layers benefits the application of CSM and LCSM in chicken diets. The cottonseed by-products CSM or LCSM showed the species-specific AA digestibility values for broilers and layers.

Determination and prediction of amino acid digestibility in brown rice for growing-finishing pigs

  • Objective: The experiment aimed to determine the standardized ileal digestibility (SID) of crude protein (CP) and amino acids (AA) in 10 brown rice samples fed to pigs, and to construct predictive models for SID of CP and AA based on the physical characteristics and chemical composition of brown rice. Methods: Twenty-two cannulated pigs (initial body weight: 42.0±1.2 kg) were assigned to a replicated 11×3 incomplete Latin square design, including an N-free diet and 10 brown rice diets. Each period included 5 d adaptation and 2 d ileal digesta collection. Chromic oxide was added at 0.3% to all the diets as an indigestible marker for calculating the ileal CP and AA digestibility. Results: The coefficients of variation of all detected indices for physical characteristics and chemical composition, except for bulk weight, dry matter (DM) and gross energy, in 10 brown rice samples were greater than 10%. The SID of CP, lysine (Lys), methionine, threonine (Thr), and tryptophan (Trp) in brown rice was 77.2% (62.6% to 85.5%), 87.5% (80.3% to 94.3%), 89.2% (78.9% to 98.9%), 55.4% (46.1% to 67.6%) and 92.5% (86.3% to 96.3%), respectively. The best prediction equations for the SID of CP, Lys, Thr, and Trp were as following, SIDCP = -664.181+8.484×DM (R2 = 0.40), SIDLys = 53.126+6.031×ether extract (EE)+0.893×thousand-kernel volume (R2 = 0.66), SIDThr = 39.916+7.843×EE (R2 = 0.41), and SIDTrp = -361.588+4.891×DM+0.387×total starch (R2 = 0.85). Conclusion: Overall, a great variation exists among 10 sources of brown rice, and the thousand-grain volume, DM, EE, and total starch can be used as the key predictors for SID of CP and AA.

  • 조피볼락의 필수지방산인 n-3HUFA가 어체에 미치는 생화학적 변화를 알아보기 위하여, n-3HUFA 함량이 $0.0\~1.5\%$가 되도록 조정한 6종의 실험사료로 조피볼락 치어를 10주간 사육하여 혈액성상, 혈청성분 및 간세포 성상의 변화를 검토하였다. 혈액성상(RBC, Hb, Ht, MCV, MCHC 및 MCH)은 사료의 n-3HUFA 함량에 따라 유의적인 차이를 나타내지는 않았다. 혈청단백질 농도$(2.76\~3.63g/100ml)$ 및 혈당량$(36\~71mg/100ml)$은 n-3HUFA $0.9\%$ 사료까지는 증가하다가 그 이상에서는 일정한 값을 유지하였다. 혈중 total cholesterol 농도$(137\~86mg/100ml)$와 free cholesterol 농도$(84.2\~63.8mg/100ml)$는 n-3HUFA $1.2\%$까지 점차 감소하는 경향을 보였으며, 그 이상에서는 일정 하였다. 혈청 GPT $(159.7\~47.0 Karmen\; unit)$, GOT $(28.0\~11.1\;Karmen\;unit)$ 및 LDH $(3888\~1539\;W-unit)$도 모두 n-3HUFA $1.2\%$까지는 감소 경향을 보이다가 그 이후에는 일정한 값을 유지하였다. n-3HUFA 무첨가 사료를 먹은 실험어의 간조직은 n-3HUFA $1.5\%$ 첨가 사료를 먹은 실험어에 비해 인포색의 구조 붕괴와 부분적인 세포 융해가 관찰되었다. 간 cytosol의 LDH 활성과 microsomal membrane의 $Ca^{2+}-ATPase$ 활성도 변화되어 사료의 n-3HUFA가 증가할수록 증가한 경향을 보였다.
