  • The part Ⅰ was for developing regression models to predict the moisture content, protein content and viscosity of brown and milled rice using Near Unfrared (NIR) Reflectance analyzer. The purpose of this study(part Ⅱ) is to measure fundamental data required for the prediction of rice quality , and to develop regression models to predict the protein content of brown and milled rice, brown rice yield from undreid paddy powder by using Near Infrared (NIR) Reflectance analyzer. The results of this study were summarized as follows . The predicted values of protein contents obtained from the undried paddy powder were will correlated to the measured values from brown and milled rice. The predicted yields of brown rice from undried paddy powder were not well correlated to be lab measured values from dried paddy. Continuous study in wavelength selection and of constituent relationship is necessary for practical application.

개 심실 형질막의 분리 및 그 방향성에 관한 연구 (Purification and Sidedness of Sarcolemma from Canine Ventricle)

  • Sarcolemmal membrane fraction from canine ventricle was isolated from the discarded pellet after the first homogenization in the isolation procedure of sarcoplasmic reticulum (Method 1) and the protein yield, purity, and sidedness of this preparation were compared to those of sarcolemmal fraction prepared by method of Lee et al. (Method 2) and a slight modification of original protocol of Jones et al. (Method 3). Method 1 differed from Method 2 essentially only in that vigorous homogenization was carried out by omnimixer and homogenization medium containing 30mM Tris-maleate was used in the first step. The sarcolemmal fraction was enriched from 45 to 50 and 29-fold in [$^3H$] ouabain, [$^3H$] DHA, [$^3H$] QNB binding and $Na^+$, $K^+$-ATPase activity, respectively, compared to homogenate. Total $Na^+$, $K^+$-ATPase activity of highly sarcolemma enriched fraction was 144.6$\pm$16.4$\mu\textrm{mol}$ Pi/mg protein/hr, which was about 85%, of total ATPase activity, and the yield of the preparation was 15.7 mg protein per 100g of starting ventricular tissue. The sarcolemmal preparation supported $^{45}Ca^{2+}$-uptake in the presence of ATP but this uptake was not dependent on oxalate. Sarcolemmal $Na^+$, $K^+$-ATPase activity and detectable [$^3H$] ouabain binding were increased about 32% and 35%, respectively, by pretreatment of sarcolemmal fraction with optimal concentration of sodium dodecylsulfate (0.3-0.4mg/mg protein), suggesting that this preparation contained about 24% of sealed rightside-out vesicles, 26% of sealed inside-out vesicles, and 5001o of freely permeable (leaky) form. This procedure showed the highest protein yield and leaky population, compared to Method 2 and 3. On the other hand, sarcolemmal fraction prepared by Method 2 and 3 showed low value in protein yield but comtained high population of inside-out (46%) and rightside-out (49%) vesicles, respectively, compared to present procedure (Method 1). The results indicate that vigorous homogenization decreases the population of sealed sarcolemmal vesicles but increases the sarcolemmal protein yield per gram tissue and that this procedure is available for further purification of sarcolemmal fraction and for the receptor binding study of sarcolemma.

Protein Aggregation and Adsorption upon In vitro Refolding of Recombinant Pseudomonas Lipase

  • Recombinant Pseudomonas lipase was used to study protein aggregation and adsorption upon in vitro refolding. Protein adsorption as well as aggregation was responsible for major side reactions upon in vitro refolding as a function of protein concentration. The optimal range of protein concentration was determined by the relative contribution of protein aggregation and adsorption. Above the optimal range, the yield of active lipase inversely correlated with protein aggregation, showing a competition between folding and aggregation. However, adsorption of protein rather than protein aggregation is thought to contribute as a major side reaction of the refolding process at sub-optimal concentrations at which the formation of aggregates should be more reduced. Protein aggregation was influenced by the amount of guanidine hydrochloride in the refolding solvent. The refolding temperature was a critical factor determining the extent of protein aggregation. The refolding yield was also affected by the dilution fold and dilution mode, which suggests that the refolding process might kinetically compete with the rate of mixing.

시비량, 재식밀도, 수확시기가 담배 잎단백질 생산에 미치는 영향 (Effect of the Amount of Fertilizer, Plant Density, and Halvesting Time on the Production of Tobacco leaf Protein)

  • Effect of the amount of fertilizer, plant density, and harvesting time on the production of tobacco leaf protein and fresh biomass was investigated. Flue-cured tobacco(M tabacum, L., cv. NC 82) seedlings were transplanted in the field dressed 200kg N per ha at 1$\times$105, 3$\times$105, 5$\times$105, 7$\times$105 plants per ha, and were harvested at the time when 6 and 10 weeks after transplanting, respectively. Harvest at 10 weeks after transplanting increased greatly number of leaves per plant and fresh weight of a plant, Precentage of senescent leaf weight, but significantly decreased fresh weight of a leaf and total protein contents g-1 fresh weight of leaf and stalk over the amount obtained from the harvest at 6 weeks after transplanting. Also, fresh leaf numbers of a plant, fresh weight of a leaf and of a plant, and total protein contents g-1 fresh weight of biomass were more decreased, but percentage of senescent leaf weight were remarkably increased under higher plant density. Therefore, it was seemed that harvesting at 6 weeks after transplanting under 1$\times$105 plant density per ha is more effective for producing higher yield of biomass and protein per plant than 10 weeks harvesting with 7$\times$105 population per ha. A trend was observed that biomass and protein yields per ha are positively correlated with plant population. Biomass yield per ha was the greatest at 7$\times$105 density(80.5t), but the peak of protein yield was at the near of 5$\times$105 population(2454kg as total protein) per ha on the regression curve. It was assumed that if tobacco plants are transplanted under 5$\times$105 plant density at the mid of May, and thereafter harvest at 6 weeks repeatedly during the growing season, it is possible to harvest 2~3 times per year, and to yield more 6.024kg of protein and over 140me1ric tons of fresh biomass ha 1 year 1 statistically in the korea tobacco growing regions.

Effect of nitrogen fertilize application levels on yield and quality of Korean wheat cultivars

  • This study was done to determine the effect of additional nitrogen fertilization on the yield and quality of the Korean wheat cultivars Keumkang, Jokyoung, Baegjoong, Sooan, Uri and Goso. Different levels of nitrogen applications (109, 82, 55, 41, and 27 kg/ha) were applied to six cultivars. The results show that the yield and protein contents were increased in all tested cultivars. The grain yields of the cultivars Keumkang, Jokyoung, Baegjoong and Sooan were greatly increased in the case of double fertilization treatments. Moreover, Uri and Goso had greatly increased yields by the additional fertilization at a 50% rate compared with korea wheat standard fertilization rate. A significantly higher yield was observed in Uri. Baegjoong was the highest yielding cultivar among the tested cultivars with the additional nitrogen fertilization. As the fertilization was increased up to double the fertilization treatment, the yield of Baegjoong also showed a constant increase. Positive correlations were found between the nitrogen fertilizer application levels and the protein contents of the grain in all the cultivars except for Uri, and among these, Jokyoung had a most significant correlation between the nitrogen fertilizer application level and the increase in its protein contents. Keumkang had the highest protein contents and highest increase in the protein content change according to the amount of nitrogen application. However, amylose, damaged starch and ash contents were not significantly changed by the different levels of nitrogen applications.

Effect of Nitrogen Fertilizer Application on Yield and Quality of Korean Soft Wheat Cultivar 'Goso'

  • Wheat flour can be categorized into bread, all-purpose, cake flour according to its protein content. Since optimal wheat flour protein content is different for each end use, it is necessary to diversify the nitrogen fertilizer methods depending on the end use and cultivar. Optimal wheat flour protein content of soft wheat (for cake flour) is lowest (<=10%) among all end use, it is necessary to develop nitrogen fertilizer methods for high yield and low protein content. In order to analyze the yield and quality changes of soft wheat as nitrogen fertilizer amount and splitting timing, soft wheat cultivar 'goso' was sown on paddy soil in jeunju, Republic of Korea ('21.10). the amount of nitrogen fertilizer was divided into 4 levels by adjusting 2kg/10a increments from 5.1 to ll.lkg/lOa, and in the N 7.1 and 9.1 kg/1 Oa(standard) treatment, N amount divided into sowing date:regrowing stage=3:7,4:6(standard), 5:5. In regrowing stage, Tiller number and N fertilizer amount at sowing date showed a correlation; y=-121.14x2+792.66x-525.41 (R2=0.77*, y: Tiller number/m2, x: N amount at sowing date(kg/10a)). Tiller number in regrowing stage was the highest when the nitrogen fertilizer amount at sowing date was 3.23kg/10a. spike number per m2 was the highest when N fertilizer was divided into sowing date:regrowing stage=3:7(N amount: 9.1kg/10a). If N fertilizer amount was fixed, grain yield was also the highest when N fertilizer was divided into sowing date :regrowing stage=3:7. Also, N amount at sowing date and grain yield showed no correlation, but N amount at regrowing stage and grain yield showed significant correlation. As N amount increased, protein content also showed a tendency to increase.

예취기시에 따른 참억새의 생육특성 , 성분함량 및 영양소수량의 변화 (Changes in the Growth , Chemical Composition and Nutritive Yield of Miscanthus sinensis at Different Cutting Dates)

  • This experiment was conducted to know what does the cutting date have an effect on the chemical composition and nutritive yield of Miscanthus sinensis, in order to determine the optimum cutting date of the plant when it is utilized as roughage. Miscanthus sinensis growing natively in the summit of Sam Ak mountain(550m above sea level) was cut at 10 days intervals from 25 July to 15 October, 1988. Plant length and dry matter contents were generally increased with delayed cutting date. Crude protein contents of the plant cut on 5, 15, and 25 of September were 5.5, 5.2 and 5.1%, respectively. Cmde fiber contents was similar with cutting treatment and averaged 35.6%. Cmde protein yield was higher in September than in other cutting date. Yield of crude fiber were 8.3, 9.3 and 9.3t/ha on 5, 15 and 25 September, respectively. Ratio of Cmde protein yield to Crude fiber yield on 5 September tended to be higher than on 15 September and 25 September. These results suggest that the optimum cutting date of the Miscanthus sinensis for good quality forage seems early September.

Effects of Rhizobium Inoculant, Nitrogen, Phosphorus, and Molybdenum on Nodulation, Yield, and Seed Protein in Pea

  • The effects of Rhizobium inoculant, nitrogen, phosphorus, and molybdenum on nodulation, dry matter production, yield attributes, pod and seed yields, protein and phosphorus contents in seed of pea (pisum sativum) var. IPSA Motorshuti-3 were assessed by a field experiment. Among the treatments Rhizobium inoculant in combination with 25kg P and 1.5kg Mo/ha performed best in recording number of nodules/plant, total dry matter yield, number of pods/plant, number of seeds/pod, 1000-seed weight, green pod yield, green and mature seed yields of pea. The highest green pod yield of 15.37 t/ha ($97.05\%$ increase over control) and green seed yield of 9.6t/ha ($69.31\%$ increase over control) were obtained by inoculating pea with Rhizobium inoculant in association with 25kg P and 1.5 Mo/ha. The effects of 60 or 120kg N/ha were comparable to Rhizobium inoculant in most cases. There were positive correlations among yield attributes, yield, protein and phosphorus contents in seeds of pea. From the viewpoint of yield attributes, yield, and seed quality, application of Rhizobium inoculant along with 25kg P and 1.5kg Mo/ha was considered to be the balanced combination of nutrients for achieving the maximum output from cultivation of pea in Shallow-Red Brown Terrace Soil of Bangladesh.

Holstein종 유우(乳牛)의 비유량(泌乳量) 및 유조성분(乳組成分)에 미치는 유전(遺傳) 및 환경(環境)의 효과(效果) (Genetic and Environmental Effects on the Lactation Yield and Milk Compositions in Holstein Cow)

  • 본(本) 연구(硏究)는 1990년부터 1991년까지 국립종축원에서 사육한 Holstein종 159두에서 조사된 성적을 근거로 최소자승법을 이용하여 종모우(種牡牛), 분만계절(分娩季節)및 산차(産差)가 비유량(泌乳量)인 유량(乳量) 및 유지방량과 유조성분(乳組成分)인 유지율(乳脂率), 유단백질율(乳蛋白質率), 무지고형분율(無脂固型紛率)및 총고형분율(總固型紛率)에 미치는 효과(效果)를 추정하였던바 그 결과(結果)를 요약하면 다음과 같다. 1. 비유량(泌乳量)에 있어서 유량(乳量)및 유지량(乳脂量)은 각각 $7,26.56{\pm}3,57.24$$230.95{\pm}43.56Kg$ 이었고, 유조성분(乳組成分)에 있어서는 유지율(乳脂率), 유단백질율(乳蛋白質率), 무지고형분율(無脂固型紛率) 및 총고형분율(總固型紛率)에 있어서 각각 $3.69{\pm}0.43$, $3.32{\pm}0.41$, $9.15{\pm}0.49$$12.75{\pm}0.96%$ 이었고, 변이계수는 유량, 유지량, 유지율 및 유단백질율에서 각각 18.68, 17.64, 11.88 및 12.34%이었다. 2. 종모우(種牡牛)의 효과(效果)에 있어서는 유량(乳量)과 유지량(乳脂量)및 유지율(乳脂率)에서 고도의 유의차 (P<0.01)가 인정되었으며, 유단백질율(乳蛋白質率) 및 총고형분율(總固型粉率)에서도 유의차 (P<0.05)가 인정되었다. 모우별(牡牛別)로는 유량(乳量)에서 B, L, 및 O 종모우(種牡牛)가 각각 7,571.22, 7,499.11 및 7,420.58Kg으로 가장 우수하였고, 유단백질율에서는 A, F 및 K 종모우가 각각 3.75, 3.64 및 3.65%로 다른 종모우에 비하여 높았다. 3. 산차(産差)의 효과(效果)에 있어서 유량(乳量)에서 고도의 유의차 (P<0.01)가 인정되었으며, 유지방량 및 유지율(乳脂率)에서는 유의차(P<0.05)가 인정되었고, 산차별(産差別)로는 유량(乳量)과 유지방량에서 3산차(産差)가 각각 7,634.54 및 274.98Kg로 가장 우수하였고, 유지율 및 유단백질율에서는 4산차 이상이 각각 3.90, 3.50%로 다른산차에 비하여 높았다. 4. 분만계절(分娩季節)의 효과(效果)에 있어서는 유량(乳量)에서 고도의 유의차 (P<0.01)가 인정되었으며 유지량(乳脂量), 유지율(乳脂率), 유단백질율(乳蛋白質率), 및 총고형분율(總固型紛率)에서 유의차(P<0.05)가 인정되었으며, 분만계절별(分娩季節別)로는 유량(乳量)에 있어서 봄과 겨울이 각각 7,310.31 및 7,364.57Kg로 우수하였고, 유단백질율에 있어서는 봄과 가을이 각각 3.68 및 3.52%로 우수하였다.

  • This experiment was tarried out to investigate the effect of plant spacing and amount of side dressing on the yield, quality and protein pattern in burley 21 and KB 101. The results obtained were summarized as follow. 1 Yield and alkaloid content are increased In high plant population and side dressing. 2 . Quality is not affected by plant population and side dressing. 3 . Nitrogen content is decreased by late growing stage. 4. The bands of burley 21 seed are fewer than KB 101 seed bands in protein pattern. 5. In protein pattern, the bands of KB 101s leave are thicker than that of burley21s leave between 18,000∼14,000 of molecular weight.
