Objective: The impact of forage feeding strategy on growth performance, ruminal fermentation and nutrient digestibility in post-weaning calves was investigated. Methods: Forty-five female Holstein calves (body weight [BW] = $79.79{\pm}0.38kg$) were enrolled in the 35-d study at one week after weaning and randomly assigned to one of three dietary treatments. All diets were fed as total mixed ration containing 60% (dry matter [DM] basis) of basal starter feed and 40% (DM basis) of forage, but varied in composition of forage source including i) alfalfa (40% DM, AH); ii) alfalfa hay (26.7% DM)+oat hay (13.3% DM; OH); iii) alfalfa hay (26.7% DM)+corn silage (13.3% DM; WS). Results: Dry matter intake was not different among treatment groups (p>0.05). However, BW (p<0.05) and average daily gain (p<0.05) of calves fed AH and OH were greater than WS-fed calves, whereas heart girth was greater in OH-fed calves than those fed AH and WS (p<0.05). Ruminal fermentation parameters including proportion of butyric acid, acetated-to-propionate ratio, concentration of total volatile fatty acid, protozoal protein, bacterial protein, and microbial protein in rumen were the highest in OH (p<0.05) and the lowest in WS. Compared with the AH and WS, feeding oat hay to postweaning calves increased crude protein digestibility (p<0.05), and decreased duration of diarrhea (p<0.05) and fecal index (p<0.05). Conclusion: Our results suggested that partially replacing alfalfa hay with oat hay improved ruminal fermentation, nitrogen utilization, and reduced incidence of diarrhea in post-weaning dairy calves.
The aim of this study was to develop muffins with whey protein concentrate (WPC) substituted for fat at the content of 10%, 20%, 40% or 80%. The quality characteristics were compared with those of a full-fat counterpart. With increasing WPC content, moisture, protein, and ash contents increased, fat content decreased, volume and specific volume of muffin decreased, but weight was unaffected. Crust lightness and yellowness of muffins increased, but redness decreased with increasing amount of WPC. Hardness, cohesiveness, springiness, gumminess and brittleness were the highest in muffin substituted with 80% WPC. Results of sensory evaluation indicated that muffin with up to 40% of the butter substituted by WPC was considered to be as acceptable as the control muffin prepared without WPC.
The objective of this study is to investigate the effects of binding agents (carrageenan, transglutaminase, isolated soy protein, and wheat fiber) on the physicochemical and sensory characteristics of pork patties. One percent of each pork patty formulation was prepared with one of the following carrageenan, transglutaminase, isolated soy protein, or wheat fiber. The lightness and redness values of raw and cooked pork patties with carrageenan, isolated soy protein, and wheat fiber were significantly higher than the control (p<0.05). The water holding capacity, cooking yield, and moisture content of pork patties containing carrageenan, isolated soy protein, and wheat fiber were significantly higher than the control (p<0.05). However, the reduction in diameter and thickness was lower than the control (p<0.05). The protein and fat content of the pork patties were not significantly different between the control and patties with binding agent addition. The shear forces of the pork patties with transglutaminase, isolated soy protein, and wheat fiber were significantly higher than the control (p<0.05), while the pork patties with carrageenan were significantly lower than control (p<0.05). Among the sensory characteristics, tenderness, juiciness, and overall acceptability of pork patties containing carrageenan, transglutaminase, isolated soy protein, and wheat fiber were slightly higher, although there was no significant difference. Therefore, pork patties containing binding agents are useful in making new ground meat products with desirable quality characteristics.
This study was conducted to validate the theoretical feasibility of a technique to identify biomarkers in Korean native black pig (KNP) and a commercial Landrace breed. Using two-dimensional electrophoresis, we found six proteins (NADH dehydrogenase Fe-S protein 1, an unnamed protein product, similar to T-complex protein 1, annexin V = CaBP33 isoform, fatty acid-binding protein, and catechol O-methyltransferase), which appeared in KNP alone. We raised polyclonal antibodies (used as the primary antibody) for Western blotting to confirm the characteristics of the six KNP proteins. As a result, catechol O-methyltransferase, annexin V = CaBP33 isoform, and the unnamed protein product presented thicker bands in KNP than those in Landrace. Moreover, catechol O-methyltransferase was shown to be more feasible as a biomarker for KNP. However, cross-reactivity was observed with the polyclonal antibodies for KNP and the other three proteins (NADH dehydrogenase, a protein similar to T-complex protein 1, and fatty acid-binding protein). This study only showed limited results from a limited number of animals; however, our research suggests possibilities for future studies.
To assess the quality of nutrient intake by area of Korean adults, a dietary survey with the 3-day record method was obtained from 324 subjects aged 40 years and older but younger than 70 (52.4 $\pm$ 8.7) living in a rural area (Ansung) and suburban area of a middle-sized city (Ansan). The quality of nutrient intake was assessed by analyzing Nutrient Adequacy Ratio (NAR), Mean Adequacy Ratio (MAR) and Index of Nutritional Quality (INQ). The average daily mean energy intakes were 1,832 ㎉ for Ansung and 1,842 ㎉ for Ansan, respectively. Daily intakes of fat for Ansung and Ansan subjects were 40.9 and 40.3 g, and those for protein were 75.1 and 73.1 g, respectively. The overall calorie: protein: fat ratio (CPF) of energy intake was 63 : 17 : 20. Daily mean intakes of protein, fat, calcium, phosphorus, iron, potassium, carotene, sodium, thiamin, and niacin were significantly higher in Ansung residents than in Ansan subjects (p < .05). The average intakes of energy, calcium, vitamin A were lower than Recommend Dietary Allowance (RDA) in both areas. Note, over 30% of the study subjects had less than 75% of RDA of calcium, vitamin A and riboflavin. The MAR was higher in Ansung than Ansan residents (0.86 and 0.85, respectively; p < .05). INQs were over 1 for most nutrients except calcium (0.87), and that of calcium and phosphorus was each significantly higher in Ansung than Ansan subjects. Based on these results, nutrient intake quality of subjects aged 40 to 69 years living in the surveyed rural area is comparable to that of semi-industrialized suburban area in Korea. Dietary deficiency in all of calcium, vitamin A, and riboflavin, however, was a common problem for both rural and suburban residents.
To evaluate the effects of early postmortem vacuum packaging (VP) on meat quality during postmortem chilled storage, hot-boned lamb was vacuum-packaged at 1, 6, 12, 24, and 48 h postmortem and stored around 2℃ until 168 h postmortem, with lamb packaged in plastic wrap as the control (aerobic packaging). Intramuscular pH decline was delayed when lamb was vacuum packaged at 1, 6, and 12 h postmortem (p<0.05). The lamb vacuum-packaged at 1 h postmortem (VP-1h group) had significantly lower shear force values and purge losses accompanied by lower free thiol group values than other treatments during postmortem storage and was also higher in extractable calpain-1 activity by 6 h postmortem (p<0.05). Free thiol group concentrations were significantly higher after VP at 6 and 12 h postmortem (p<0.05). Packaging lamb under vacuum very early postmortem produced the lowest shear force and purge loss, likely by slowing heat loss and muscle temperature decline, implying that lamb quality is improved by VP when applied very early postmortem. This was at the expense of protein oxidation, which was unrelated to other meat quality measurements, most likely because potential contracture during hot boning confounded its impact. Further research is required to understand the implications of the interaction between protein oxidation, VP, and hot boning on the acceptability of lamb.
Objective: The study was to investigate the effects of alanyl-glutamine (Ala-Gln) and glutamine (Gln) supplementation on the intestinal mucosa barrier in piglets. Methods: A total of 180 barrows with initial weight $10.01{\pm}0.03kg$ were randomly allocated to three treatments, and each treatment consisted of three pens and twenty pigs per pen. The piglets of three groups were fed with control diet [0.62% alanine (Ala)], Ala-Gln diet (0.5% Ala-Gln), Gln diet (0.34% Gln and 0.21% Ala), respectively. Results: The results showed that in comparison with control diet, dietary Ala-Gln supplementation increased the height of villi in duodenum and jejunum (p<0.05), Gln supplementation increased the villi height of jejunum (p<0.05), Ala-Gln supplementation up-regulated the mRNA expressions of epidermal growth factor receptor and insulin-like growth factor 1 receptor in jejunal mucosa (p<0.05), raised the mRNA expressions of Claudin-1, Occludin, zonula occludens protein-1 (ZO-1) and the protein levels of Occludin, ZO-1 in jejunal mucosa (p<0.05), Ala-Gln supplementation enlarged the number of goblet cells in duodenal and ileal epithelium (p<0.05), Gln increased the number of goblet cells in duodenal epithelium (p<0.05) and Ala-Gln supplementation improved the concentrations of secretory immunoglobulin A and immunoglobulin G in the jejunal mucosa (p<0.05). Conclusion: These results demonstrated that dietary Ala-Gln supplementation could maintain the integrity of small intestine and promote the functions of intestinal mucosa barriers in piglets.

Parkin, known as an E3 ubiquitin ligase, has essential role in protein quality control, and its severe dysfunction leads to neurodegenerative disorders. Human Parkin was excessively degraded when expressed in Escherichia coli under the conventional induction condition ($37^{\circ}C$ culture condition with 0.5 mM IPTG). To optimize the induction and culture conditions for recombinant human Parkin and develop a rapid method for the Parkin purification, we expressed Parkin by using PCEX system at the different culture temperatures and IPTC concentrations. The intact Parkin protein was purified to approximately $90\%$ purity with suitable amounts of protein under the optimal culture condition ($25^{\circ}C$E with 0.01 mM IPTG). Additionally, we constructed various parkin plasmids with different tagging systems and investigated their expression patterns in HEK293 cells. We found that the proteolytically sensitive site is localized within a ubiquitin-like domain of Parkin. This study developes a method for generating useful reagents to investigate biochemical properties of Parkin.
In this study the rheological properties of Jeung-pyun prepared with soybean protein isolate (SPI) were investigated. SPI Jeung-pyun samples were manufactured with 3% whole protein, 7S protein, or 11S protein (w/w). In terms of moisture content the Jeung-pyun samples prepared with soybean flour had greater moisture contents than the control group. With the addition of SPI water binding capacity solubility and swelling power increased. Dough pH decreased during the fermentation, but increased after steaming and the SPI Jeung-pyun samples presented higher pH levels han the control group. Foaming ability was significantly strong in the 7S, 11S and whole protein groups. The surface structures of the 7S, 11S and whole protein Jeung-pyun samples displayed small uniform pores when examined by SEM. Overall, the results suggest that SPI can contribute to quality improvements in Jeung-pyun through effects on dough fermentation.

This experiment was conducted with total 960 brown layers, consisted of 8 treatment to investigate the effects of dietary chromium as chromium picolinate on egg production, egg quality, nutrient utilizability, serum traits and mortality in brown layers. Layers were fed diets with two levels of dietary protein (14% and 16%) and supplemented with 0, 200, 400, 800 ppb/kg of chromium as chromium picolinate, respectively. The highest egg production, egg weight and egg mass were found in 800 ppb chromium picolinate supplementation group with high protein level (16%) (p < 0.05). Although there was no significant difference, layers receiving 400 ppb of chromium picolinate with high protein (16%) represented the lowest broken eggs. The utilization of energy, dry matter and crude protein of 400 ppb chromium picolinate group with low protein level (14%) were significantly higher than those of control or other chromium picolinate group (p < 0.05). 400 ppb chromium picolinate with low protein level (14%) showed the lowest serum glucose concentration. But serum glucose concentrations in all treatments showed no significant differences. Present date revealed that the lowest serum cholesterol concentration of layers was found at 400 ppb chromium picolinate group with high protein level (16%) (p < 0.05). Crude protein content in yolk was significantly higher in eggs of layers received 800 ppb chromium picolinate and the lowest in eggs from layers received 400 ppb chromium picolinate among chromium picolinate levels (p < 0.05). Mortality was remarkably decreased by chromium picolinate supplementation and the lowest mortality value was found in layres receiving 800 ppb chromium picolinate with high protein level.
