Isolation and Characterization of an Eosinophilic GH 16 β-Agarase (AgaDL6) from an Agar-Degrading Marine Bacterium Flammeovirga sp. HQM9

  • A special eosinophilic agarase exo-type ${\beta}$-agarase gene, AgaDL6, was cloned from a marine agar-degrading bacterium, Flammeovirga sp. HQM9. The gene comprised 1,383-bp nucleotides encoding a putative agarase AgaDL6 of 461 amino acids with a calculated molecular mass of 52.8 kDa. Sequence analysis revealed a ${\beta}$-agarase domain that belongs to the glycoside hydrolase family (GH) 16 and a carbohydrate-binding module (CBM_4_9) unique to agarases. AgaDL6 was heterologously expressed in Escherichia coli BL21 (DE3). Enzyme activity analysis of the purified protein showed that the optimal temperature and pH of AgaDL6 were $50^{\circ}C$ and 3.0, respectively. AgaDL6 showed thermal stability by retaining more than 98% of activity after incubation for 2 h at $50^{\circ}C$, a feature quite different from other agarases. AgaDL6 also exhibited outstanding acid stability, retaining 100% of activity after incubation for 24 h at pH 2.0 to 5.0, a property distinct from other agarases. This is the first agarase characterized to have such high acid stability. In addition, we observed no obvious stimulation or inhibition of AgaDL6 in the presence of various metal ions and denaturants. AgaDL6 is an exo-type ${\beta}$-1,4 agarase that cleaved agarose into neoagarotetraose and neoagarohexaose as the final products. These characteristics make AgaDL6 a potentially valuable enzyme in the cosmetic, food, and pharmaceutical industries.

Investigation of the Insulin-like Growth Factor System in Breast Muscle during Embryonic and Postnatal Development in Langshan and Arbor Acres Chickens Subjected to Different Feeding Regimens

  • Nutrient availability may control muscle growth directly and indirectly through its influence on regulatory factors. We analyzed the effects of nutrient availability on the breast muscle insulin-like growth factor system. Real time RT-PCR was used to quantify the level of transcription in breast muscle from Langshan (LS) layer and Arbor Acres (AA) broiler chickens subjected to different feeding regimens during embryonic and postnatal development. The AA chickens were fed AA diet (AA, control group) while the LS chickens were either fed LS diet (LL) or AA diet (LA). According to our results, insulin-like growth factor (IGF)-II (embryonic day 16 (E16) - postnatal day 42 (P42)), IGF-I receptor (IGF-IR, E18-P42), and IGF binding protein (IGFBP)-2 (E18-P42), -5 (E16-P14), -7 (E12-P0), and -3 (E12-P0) were positively correlated with IGF-I, while IGFBP-3 (P0-P28) was negatively correlated with IGF-I. In comparison, IGF-IR (E18-P42), IGFBP-2 (E18-P42), IGFBP-5 (E14-P0), and IGFBP-3 (E16-P0) were positively correlated with IGF-II, while IGF-IR (E10-E16) and IGFBP-3 (P0-P28) were negatively correlated with IGF-II. Moreover, IGFBP-2 (E16-P42), -7 (E10-E16), and -3 (E10-E16) were positively correlated with IGF-IR, while IGFBP-3 (P0-P28) was negatively correlated with IGF-IR. Finally, IGFBP-7 (E12-P0) was positively correlated with IGFBP-3, while IGFBP-2 (P0-P28) and -7 (P0-P42) were negatively correlated with IGFBP-3. Overall, the AA chickens exhibited higher levels of IGF-I, IGF-IR, and IGFBP-2 mRNA expression than the LL chickens, while the opposite was true for IGFBP-7. No strain differences in IGF-I, IGF-IR, and IGFBP-7 mRNA expression were detected between LA and AA chickens; however, a strain difference was observed for IGFBP-2. LA chickens exhibited higher levels of IGFBP-2 than LL chickens, while the opposite was true for IGFBP-7. Our data show the first evidence that certain genes may be correlated during specific developmental periods and that strain differences in the expression of those genes in LS and AA chickens are due to differential responses to the same diet.

  • Background: Conventional treatment for mesothelioma is largely ineffective. We evaluated the novel approach of adenoviral gene transfection of PTEN gene in mesothelioma cancer cell lines, inflammatory and epithelial subtype, which are sensitive to adenoviral p53. Material and Method: Binary adenoviral PTEN and LacZ (Ad/GT-LacZ and Ad/GV16) vectors were used for transduction of the mesothelioma cell lines, REN (p53 sensitive). Protein levels were determined by Western blotting assay. Apoptosis was assessed by fluorescence-activated cell sorter analysis of subdiploid populations. Cell viability was determined with the XTT assay. Statistical analysis was performed with analysis of variance and the Student t test. Result: 72 hours after the treatment of adenoviral PTEN gene, cell killing were 32.9% for REN compared to control cell (2.5%) at MOI of 20. Also we observed the over-expression of proapoptotic protein, bax and decreased expression of bcl-2 protein in REN cells. But the expression of BCL-xl, Bak, Bad proteins were not altered. Conclusion: Adenovirus Pten-mediated overexpression of the Bax gene induces apoptosis and decreased cellular viability in p53-sensitive mesothelioma cells. These data suggest that the transfection of PTEN gene may represent a alternative gene therapy strategy to treat mesothelioma.

  • Ameloblasotma is slowly growing, locally invasive neoplasm with a potentially destructive behavior. The epithelium of ameloblastoma is thought to have an intrinsic growth potential and has been shown to present a higher rate of proliferation as compared to odontogenic cysts with low local recurrence rate. The molecular mechanisms that regulate the cell growth and invasion of ameloblastoma cells are unknown. Bcl-2 protein, which prevent apoptosis, is expressed in immortalized ameloblastoma cell line(AM-1)(Harada et al 1998). Expression of bcl-2 protein occurs in tooth germs, whose epithelial component may act as the histogenic precursor of ameloblastoma. Bax is considered as a main effector of apoptosis. Bax forms homodimers and also heterodimers with bcl-2. p53 tumor supressor gene participates not only in cell proliferation control but also in induction of apoptosis. The objective of the present study was to evaluate the apoptosis related protein expression in odontogenic cyst and ameloblastoma. A total of 10 dentigerous cysts and 16 ameloblastomas were used in the present study. Dentigerous cyst showed negative or slight positive for p53 and bcl-2 but strongly positive for bax, ameloblastoma, on the other hand, strongly positive for p53 and bcl-2 but weekly positive for bax. Bcl-2 was expressed for ameloblastoma mainly in outer layer or whole layer of epithelium and for dentigerous cyst mainly in basal layer. The difference in expression of apoptosis related protein in dentigerous cyst and ameloblastoma might explain the peculiar aggressive growth pattern of ameloblastoma.

Effects of Seeding Dates on Yield and Feed Value of Italian Ryegrass in Paddy Field Cultivation

  • This study was carried out to investigate the influence of the seeding dates on the yield and feed value in cultivating Italian rye grass on paddy soil. The field experiment was designed in a randomized block design of 3 repetitions with seeding dates of Oct. 19 (T1), Oct. 26 (T2), Nov. 2 (T3), Nov. 9 (T4), and Nov. 16 (T5) treatments. Plant length, fresh yield, dry matter yield and TDN yield were increased with early seeding dates (p<0.05). Crude protein, Crude fat and crude ash were increased with early seeding dates (p<0.05). On the other hand, NDF, ADF and Crude fiber were decreased with early seeding dates (p<0.05). Total mineral contents were higher in the order of T4 > T3 > T5 > T1 > T2 (p<0.05). The total composition amino acid content was decreased significantly (p<0.05) as the seeding dates delayed. Total free sugar content was highest in T1, whereas the lowest in T5 treatment (p<0.05). Nutrition yields (crude protein, crude fat, amino acid and free sugar) were increased significantly with early seeding dates (p<0.05). The above result indicated that it is favorable to seed soon after rice harvest to increase dry matter yield and nutrition yield of Italian ryegrass in the midlands.

Effects of Hydrostatic Pressure Treatment on the Physicochemical, Morphological, and Textural Properties of Bovine Semitendinosus Muscle

  • The effects of hydrostatic pressure (HP) treatment on the physicochemical, morphological, and textural properties of bovine semitendinosus (ST) muscle were assessed. Based on SDS-PAGE, the decrease in HP-treated ST muscle protein solubility in 0.1 M KCl buffer (pH 7.0) was attributable to a reduction in the levels of sarcoplasmic protein, and the protein solubility decrease observed in 0.6 M KCl buffer (PH 7.0) was attributable to a reduction in the levels of myosin heavy-chain and actin. Scanning electron microscope (SEM) observations showed that muscle fibers became finer and more compact with increasing pressures. The shear force and hardness of ST muscle pressurized to 300 MPa decreased significantly (p<0.05), however samples pressurized at 100 and 500 MPa exhibited a significant increase in both attributes relative to the control sample (p<0.05).

Anorexia, Nausea and Vomiting, and Food Intake Patterns in Patients on Chemotherapy (항암화학요법환자의 식욕부진, 오심 및 구토와 음식섭취 양상과의 관계)

  • Purpose: The purpose of this study was to identify the relationship between anorexia, nausea, and vomiting (ANV) and food intake patterns in patients with cancer on chemotherapy. Method: Ninety-one patients from a university hospital in Seoul were recruited and were asked to record food intake and ANV during one cycle of chemotherapy. Results: Caloric and protein intake decreased significantly. The mean caloric intake was 808.75 Kcal (SD=177.54), 48.2% of the recommended intake and the mean of protein intake was 28.44 g (SD=16.44) only 34.7% of the recommended intake. The mean score for dietary diversity was 3.57 (SD=.73) indicating that the patients had taken 3 or more of the 5 food groups. ANV had a significant negative correlation with caloric and protein intake and dietary diversity ($r=-.29{\sim}-.56$, p<.05) and dietary diversity had a significant positive correlation with caloric and protein intake (r=.46 and .57, p=.000). Conclusion: Patients receiving chemotherapy had a very poor intake that could lead to malnutrition and a compromised immune system. These findings suggest the need to develop interventions that encourage for food intake.

Effects of Brisk Walking and Brisk Walking Plus Diet on C-reactive Protein in Obese Women with Hypertriglycemia (건강걷기, 건강걷기와 식이 교육이 고중성지방혈증 비만 여성의 C-reactive protein에 미치는 영향)

  • Purpose: The purpose of this study was to investigate the effects of 12-week brisk walking and brisk walking plus diet program on C-Reactive Protein(CRP) in middle-aged obese hypertriglycemic($triglyceride{\geq}150mg/d{\ell}$) Korean women. Method: The subjects were 16 obese ($BMI{\geq}25$) hypertriglycemic middle-aged women (7 for brisk walking group, 9 for brisk walking plus diet group) who participated in a health promotion program at one public health center. Initially the brisk walking intervention consisted of walking for 20 minutes/day at an intensity of 40 to 50% of heart rate reserve(HRR) for 3 days/week and progressed to 50 minutes/day, 60 to 70% of their HRR, and 6 days/week. The diet intervention consisted of 60 minutes of group education and 20 to 30 minutes of individual counseling with a nutritionist every week. Data were analyzed with SPSS PC program. Results: There was no significant reduction in CRP levels in both brisk walking (Z=-1.70, p=0.088) and brisk walking plus diet group(Z=-0.31, p=0.752). In brisk walking only group, CRP levels were increased after the intervention. Conclusion: Brisk walking could increase the level of CRP when it is in the course of progression and diet could decrease the acute phase inflammatory response.

Effect of Genotype and Dietary Protein Level on Growth Performance and Carcass Characteristics of Fattening Pigs in Central Vietnam

  • This study aimed to determine the optimum dietary crude protein level in a typical diet for fattening pigs fed ad libitum under normal climate conditions in Central Vietnam. One hundred and ninety two gilts of Mong Cai local breed (MC), $F_1$ Large White${\times}$Mong Cai and $F_2$ crossbreds of (Landrace${\times}$Mong Cai)${\times}$Large White were used. At the start of the experiment, Mong Cai pigs weighed 12 kg at 11 weeks of age, $F_1$ pigs 12.1 kg at 8 weeks of age and $F_2$ pigs 12.2 kg at 8 weeks of age. Four diets differing in crude protein (CP) content (10.1, 13.1, 16.1 and 18.9% in DM) were formulated from rice bran, corn meal, cassava meal and fish meal. Calculated digestible energy content of the diets ranged from 13.5 to 13.8 MJ per kg DM. Pigs were housed individually in pens of $2.5\;m^2$ each and had ad libitum access to feed in a trough as well as water in bowls. The final weights after a growing period of 150 days were 66, 86 and 96 kg for MC, $F_1$ and $F_2$, respectively. Feed intake of MC pigs was highest at 13.1% CP while $F_1$ and $F_2$ had the highest feed intake at 16.1% CP. The results showed that for MC the maximum gain was obtained at levels between 13 to 16% CP. For the $F_1$ the maximum gain was at dietary protein levels of 16-17%. For $F_2$ the max gain was obtained at CP levels of 16 to 18%. Feed conversion was highest in MC pigs (~4.0) followed by $F_1$ (~3.3) and $F_2$ (~3.1), and within genotypes was lowest at the optimum CP level (p<0.05). Back fat thickness in MC (33.1 mm), $F_1$ (23.0 mm) and $F_2$ (20.5 mm) pigs was different and within genotypes was the lowest at intermediate CP levels. In conclusion, increasing the dietary crude protein contents in practical diets for pigs in Vietnam can increase production on small holder farms. Optimal performance for MC, $F_1$ and $F_2$ pigs is achieved at different dietary crude protein contents.

  • The study was designed to examine the of effects of level of protein intake on Cu metabolism in 6 healthy young adult women. The subjects were given 4 levels of protein diet, 0.45g (period I), 0.60g (period III), 0.75g (period II) and 0.90g (period IV) of protein per Kg of body weight and 0.86mg 0.70mg, 1.86mg and 2.34mg of Cu per day for 5 days respectively. During the experimental period, urine sample were collected everyday and fecal sample were collected for last 2 days of each dietary period. The samples were analyzed for Cu contents. Mean daily urinary Cu excretion were $0.12{\pm}0.03mg$ for period I, $0.16{\pm}0.02mg$ for period III, $0.35{\pm}0.08mg$ for period II and $0.11{\pm}0.02mg$ for period IV, and the mean daily urinary excretion of Cu was not affected significant difference by the level of protein intake. Mean daily fecal Cu excretion were $0.12{\pm}0.03mg$ for period I, $0.16{\pm}0.02mg$ for period III, $0.35{\pm}0.08mg$ for period II and $0.11{\pm}0.02mg$ for period IV, and the mean daily urinary excretion of Cu was not affected significant difference by the level of protein intake. Mean dailey Mean dailey fecal Cu excretion were $1.23{\pm}0.16mg$ for period I, $1.28{\pm}0.25mg$ for period III, $0.99{\pm}0.01mg$ for period II and $1.85{\pm}0.19mg$ for period IV, and the difference of the 2 periods I vs IV was significant(p<0.05). Mean dailey cu balance were $0.48{\pm}0.14mg$ for period I, $0.74{\pm}0.26mg$ for period III, $0.52{\pm}0.12mg$ for period II and $0.38{\pm}0.20mg$ for period IV, and the difference of the 2 periods I vs IV was significant(p<0.05).
