모유영양아와 인공영양아의 성장과 에너지 및 단백질대사에 관한 종단적 연구 (Longitudinal Study of Growth, Energy and Protein Metabolism of Korean Breast Fed and Formula Fed Infants from 1 to 3 Postpartum Months)

  • This study was carried out to investigate growth performance, energy and protein metabolism of breast and formula fed infants from birth to 3 months postpartum. There were four groups : breast fed(BF) and three formula fed groups(FFM, FFN and FFP). There was no significant difference in the height of infants according to feeding method and formula brands. However, mean weight of FFM was significantly higher than that of FFP and BF at 1 and 3 months postpartum, respectively. Mean head circumference of FFN was lowest among groups. The average intake of breast milk was $781.4{\;}{pm}{\;}119.3m{\ell}/day$ and that of formula was $848.6{\;}{pm}{\;}118.5m{\ell}/day$. Mean apparent digestible energy intakes of formula-fed infants and breast-fed infants during 3months were 568.9 $\pm$146.9 kca1/day and 657.9$\pm$212.8 kca1/day, respectively. The average protein intake of brest-fed and formula-fed infants were 8.3$\pm$1.6g/day and 14.1$\pm$14.1 g/day, respectively. The protein intakes of formula-fed infants were significantly higher than those of breast-fed infants at 1, 2, 3 months. The apparent protein digestibility of breast-fed infants was singnificantly higher compared to formula fed infant. These data suggest that RDA for infants be established and breast feeding be encouraged.

Coordinated alteration of mRNA-microRNA transcriptomes associated with exosomes and fatty acid metabolism in adipose tissue and skeletal muscle in grazing cattle

  • Objective: On the hypothesis that grazing of cattle prompts organs to secrete or internalize circulating microRNAs (c-miRNAs) in parallel with changes in energy metabolism, we aimed to clarify biological events in adipose, skeletal muscle, and liver tissues in grazing Japanese Shorthorn (JSH) steers by a transcriptomic approach. Methods: The subcutaneous fat (SCF), biceps femoris muscle (BFM), and liver in JSH steers after three months of grazing or housing were analyzed using microarray and quantitative polymerase chain reaction (qPCR), followed by gene ontology (GO) and functional annotation analyses. Results: The results of transcriptomics indicated that SCF was highly responsive to grazing compared to BFM and liver tissues. The 'Exosome', 'Carbohydrate metabolism' and 'Lipid metabolism' were extracted as the relevant GO terms in SCF and BFM, and/or liver from the >1.5-fold-altered mRNAs in grazing steers. The qPCR analyses showed a trend of upregulated gene expression related to exosome secretion and internalization (charged multivesicular body protein 4A, vacuolar protein sorting-associated protein 4B, vesicle associated membrane protein 7, caveolin 1) in the BFM and SCF, as well as upregulation of lipolysis-associated mRNAs (carnitine palmitoyltransferase 1A, hormone-sensitive lipase, perilipin 1, adipose triglyceride lipase, fatty acid binding protein 4) and most of the microRNAs (miRNAs) in SCF. Moreover, gene expression related to fatty acid uptake and inter-organ signaling (solute carrier family 27 member 4 and angiopoietin-like 4) was upregulated in BFM, suggesting activation of SCF-BFM organ crosstalk for energy metabolism. Meanwhile, expression of plasma exosomal miR-16a, miR-19b, miR-21-5p, and miR-142-5p was reduced. According to bioinformatic analyses, the c-miRNA target genes are associated with the terms 'Endosome', 'Caveola', 'Endocytosis', 'Carbohydrate metabolism', and with pathways related to environmental information processing and the endocrine system. Conclusion: Exosome and fatty acid metabolism-related gene expression was altered in SCF of grazing cattle, which could be regulated by miRNA such as miR-142-5p. These changes occurred coordinately in both the SCF and BFM, suggesting involvement of exosome in the SCF-BFM organ crosstalk to modulate energy metabolism.

Energy Requirement of Rhode Island Red Hens for Maintenance by Slaughter Technique

  • Energy requirement of Rhode Island Red (RIR) hens was studied by comparative slaughter technique. Seventeen hens above 72 weeks of age were slaughtered in batches. Batch I consisted of 5 hens which were slaughtered initially. Batch II comprised of six hens, which were fed ad libitum broken rice (BR)-based diet for 18 days. Record of feed intake, number of eggs laid and egg weight during the period was kept. These hens were slaughtered and body energy content was determined. Egg energy was consisted as energy deposited. Batch III consisting of six hens which were fed varying quantity of diet for 15 days, were slaughtered similarly as hens of batch II. Regression equation (body weight to body energy) developed on batch I was applied to batch II and developed on batch II was applied to batch III hens, to find out initial body energy content of hens. Egg energy (EE) was calculated according to formula: EE (kcal) = -19.7 + 1.81 egg weight (g). Regressing metabolisable energy (ME) intake on energy balance (body energy change + egg energy), maintenance ME requirement of hens was found to be $119.8kcal/kg\;W^{0.75}/d$. Multiple regression of ME required for production on energy retained as protein and fat (body plus egg energy) indicated that RIR hens synthesize proteins with an efficiency of 85.5 and fat with an efficiency exceeding 100 percent on BR based diet.

  • Objectives: The prevalence of obesity and metabolic syndrome is increasing worldwide. Regulation of cellular energy metabolis has the potential to be manipulated therapeutically to serve as a target for obesity and insulin resistance. Skeletal muscle is regarded as a target for regulation of energy metabolism and insulin resistance. In this study, the authors investigated the regulatory effect of (Cinnamomum cassia Blume, CCB), Aconitum carmichaeli Debx (ACD), and Benth (Pueraria lobata Benth, PLB) on energy and glucose metabolism in C2C12 myotubes. Methods: The water extracts of CCB, ACD, and PLB (0.5 mg/ml) were treated in differentiated C2C12 myotubes. The expressions of adenosine monophosphate-activated protein kinase (AMPK) and phosphorylation AMPK were detected with western blotting. Glucose metabolism was investigated with glucose uptake assay and glucose consumption assay, total adenosine triphosphate (ATP) content was also analyzed. Results: CCB, ACD, and PLB activated the phophorylation of AMPK, they also increased the glucose metabolism and total ATP contents in C2C12 myotubes. Conclusions: This study suggests that CCB, ACD, and PLB have the potential to increase energy and glucose metabolism in skeletal muscle.

Characterization of Beef Transcripts Correlated with Tenderness and Moisture

  • To identify transcriptional markers for beef traits related to meat tenderness and moisture, we measured the transcriptome of the Longissimus dorsi skeletal muscle in 10 Korean native cattle (KNC). We analyzed the correlation between the beef transcriptome and measurements of four different beef traits, shear force (SF), water holding capacity (WHC), cooking loss (CL), and loin eye area (LEA). We obtained non-overlapping and unique panels of genes showing strong correlations (${\mid}r{\mid}$ > 0.8) with SF, WHC, CL, and LEA, respectively. Functional studies of these genes indicated that SF was mainly related to energy metabolism, and LEA to rRNA processing. Interestingly, our data suggested that WHC is influenced by protein metabolism. Overall, the skeletal muscle transcriptome pointed to the importance of energy and protein metabolism in determining meat quality after the aging process. The panels of transcripts for beef traits may be useful for predicting meat tenderness and moisture.

Metabolic Components of Energy Expenditure in Growing Beef Cattle - Review -

  • A large portion of total energy expenditure associated with ruminant livestock production goes towards maintenance. Approximately 55% of whole body energy use is consumed by visceral tissues (including internal organs) with the majority of this going to the liver and gastrointestinal tract. Muscle and adipose tissues consume about 27% of total body energy expenditure. Metabolic components within the viscera responsible for the majority of energy consumption include ion transport, protein turnover, substrate cycling, and urea synthesis (liver). Within muscle tissue of growing animals ion transport and protein turnover account for most of the energy expenditure. Protein synthesis consumes approximately 23% of whole body energy use and visceral tissues account for proportionally more of whole body protein synthesis than skeletal muscle. Research efforts focused on improving energetic efficiency of the tissues and metabolic mechanisms responsible for the majority of whole animal energy expenditure should provide information leading to more efficient production of an edible product.

EFFECTS OF CIMATEROL ON THE GROWTH PERFORMANCE, CARCASS CHARACTERISTICS AND TISSUE METABOLISM IN BROILER CHICKS FED DIFFERENT DIETARY ENERGY

  • This study was a sequential experiment consisting if feeding trial and in vitro culture studies. Feeding was conducted by $2{\times}2{\times}2$ factorial design with two cimaterol levels (0, 0.25 mg/kg), two energy levels (3,200, 2,900 ME kcal/kg) and two sexes. In starting period (0-21 days) broilers were fed diets containing two energy level without dietary supplementation of cimaterol. During finishing period (21-42 days) cimaterol groups were fed cimaterol supplemented diets. In vitro cultures were carried out to study the cellular metabolism of protein and fat in liver and adipose tissues prepared from chicks used in feeding trials. Body weight gain was significantly improved by the administration of cimaterol to experimental diets by 2.4% (p < 0.05). Feed intake was reduced by cimaterol administration at the high energy level, but this trend was reversed at low energy level. Feed efficiency was improved by cimaterol administration and at high energy level the difference (5.7%) was significant(p < 0.05). The administration of cimaterol had no effects on percentage of abdominal fat content, giblet and neck. There was little difference in carcass yield between control and cimaterol treated group. The administration of cimaterol had no effects on nutrient metabolizability or carcass composition. The results of in vitro studies with liver tissues showed that cimaterol increased the lipolytic activities (p < 0.05) and decreased lipogenic activities (p < 0.05). In in vitro studies with acinar cell of liver tissues. cimaterol increased the amount of retained protein and decreased secreted protein at high energy level. but the trend was opposite at low energy level.

The effect of red ginseng and ginseng leaves on the substance and energy metabolism in hypothyroidism rats

  • Background: Recent studies have revealed that the properties Traditional Chinese Medicine is mostly associated with are substance and energy metabolism. Our study aimed to compare the effect of red ginseng (RG) (warm property) and ginseng leaves (GL; cold property) on the substance and energy metabolism of rats with hypothyroidism. Materials and methods: Rats were administered propylthiouracil intraperitoneally for 20 d to cause hypothyroidism. The reference group was orally administered Aconiti Lateralis Radix Praeparaia [FZ (Fuzi in Chinese)], while both the RG and GL groups were orally administrated crude drugs. The rectal, tail, toe, and axilla temperature of the rats were assayed every 3 d. Oxygen consumption, carbon dioxide production, heat production, and energy expenditure were measured via TSE phenoMaster/LabMaster animal monitoring system. Adenosine monophosphate-activated protein kinase, $Na^+-K^+$-ATPase, fumarase, pyruvic acid and cyclic adenosine monophosphate/cyclic guanosine monophosphate were determined. Results: The lower levels of triiodothyronine, tetraiodothyronine, and thyrotropin-releasing hormone and the higher level of thyroid stimulating hormone revealed the successful establishment of a hypothyroidism model. Oxygen consumption, carbon dioxide production, heat production, and energy expenditure in the FZ and RG groups were obviously increased. The activity of $Na^+-K^+$-ATPase and fumarase in the FZ and RG groups was significantly increased. The cyclic adenosine monophosphate/cyclic guanosine monophosphate level in the FZ and RG groups was increased, while the GL group showed the opposite. Conclusion: Our research provides a new way to explore the efficiency of Chinese medicine on the basis of the relationship between drug property and effects on substance and energy metabolism.

벼메뚜기(Oxya chinensis sinuosa Mistshenko) 분말 섭취와 유산소성 운동훈련에 의한 마우스의 에너지 대사 변화 (Effects of Grasshopper (Oxya chinensis sinuosa Mistshenko) Powder and Aerobic Exercise on Energy Metabolism in ICR Mice)

  • 본 연구의 목적은 벼메뚜기(Oxya chinensis sinuosa) 분말 섭취가 유산소성 운동훈련(트레드밀 달리기)의 병행 유무에 의해 ICR 생쥐의 에너지 대사를 증가시키는지를 알아보고자 하는 것이다. 이 목적을 달성하기 위하여, 28 마리의 ICR 생쥐를 보통식 대조군(CON), 보통식 대조군으로서 운동훈련 병행군(COEX), 벼메뚜기 분말이 혼합된 사료 섭취군(GH), 그리고 벼메뚜기 분말이 혼합된 사료를 섭취함과 동시에 운동훈련을 병행군(GHEX)으로 구분하였다. 벼메뚜기 분말 사료섭취 및 운동은 6주간 진행하였다. 체중증가율은 유의하지 않았다. 지방량은 GH와 GHEX에서 유의하게 감소하였다. 혈중 glutamic oxaloacetic transaminase와 glutamic pyruvic transaminase 수준은 처치 집단간 변화가 없었다. 제2형 당수송체 및 제4형 당수송체는 처치 집단간 유의한 차이가 없었다. GHEX의 fibronectin type III domain-containing protein 5 단백질 발현량이 가장 높았다. AMP-activated protein kinase 단백질 수준은 GHEX에서 유의하게 증가하였다. Glycogen synthase kinase 3 beta 단백질 발현량은 GHEX가 CON과 비교할 때 감소하였다. 이러한 결과들은 벼메뚜기 분말을 섭취하면서 지구성 운동훈련을 하는 경우에 에너지 대사에 영향을 준다는 것을 제시하고 있다.

식이 단백질의 종류와 함황아미노산 함량이 성장기 쥐의 골밀도에 미치는 영향 (The Effect of Dietary Protein Source and Sulfur Amino acid Content on bone Metabolism in Growing Rats)

  • This study was performed to evaluate the effect of dietary protein source and sulfur amino acid content on bone metabolism in ra. Thirty male rats (body weight 145$\pm$2g) were divided into three groups. The rats in the first group were fed on casein 20% diet as animal protein source and those in the second group were fed on soy 20% diet as plant protein source. Sulfur amino acid ratio of these group was 1.07:1. The rats in the third group were fed on soy 20% diet and the sulfur amino acid were supplemented with the amount contained as much in the soy 20% diet. All rats were fed on experimental diet and deionized water ad libitum for 9 weeks, The total body, spine, femur bone mineral density and bone mineral content were measured using Dual Energy X-ray Absorptiometry Calcium, phosphate, pyridinoline, creatinine in urine and calcium, phosphate, alkaline phosphatase, osteocalcin in serum were measured. During the experimental period, plant protein (soy protein) group had a lower urinary Ca excretion, urine pyridinoline & crosslinks value and had a higher Ca efficiency in total bone and femur bone mineral density than animal protein (casein) group. There were no significant differences in serum calcium, phosphate, alkaline phosphatase and osteocalcin among the three groups of the rats. The findings from this study demonstrated that plant protein (soy protein) is beneficial of bone mineral density because it had a higher Ca efficiency in total bone and femur bone mineral density than animal protein (casein). However, the supplementation of sulfur amino acid on soy results were consistent with prior studies that dietary sulfur amino acid load had a negative effect on calcium balance. The rats fed sulfur amino acid supplementation diet increased urinary calcium excretion and decreased calcium efficiency for total and femur mineral density. Therefore, dietary protein source and sulfur amino acid content influence bone metabolism. (Korean J Nutrition 37(2): 100-107, 2004)
