Growth Performance and Meat Quality of Broiler Chickens Supplemented with Bacillus licheniformis in Drinking Water

  • A feeding trial was conducted to investigate effects of Bacillus licheniformis on growth performance and meat quality of broilers. Nine hundred one-d-old broiler chicks were randomly assigned to 3 experimental groups with three replicate pens of 100 broiler chicks. Three treatments were i) control, ii) basal diets supplemented with 1 ml of B. licheniformis for each in feed water per day iii) basal diets supplemented with 2 ml of B. licheniformis per chick in feed water per day. The supplementation of B. licheniformis significantly increased body weight in grower chickens (p<0.05), and significantly improved the feed conversion in 3 to 6 and 0 to 6 wk feeding period compared with the control group (p<0.05). Additionally, the supplement also resulted in increased protein and free amino acid contents, and decreased fat content in chicken breast fillet (p<0.05). Furthermore, improvement in sensory attributes was observed in broilers fed with the probiotic. In conclusion, B. licheniformis treatments resulted in a significant increase (p<0.05) in broiler productivity based on an index taking into account daily weight gain and feed conversion rate. Meanwhile, the probiotic contributed towards an improvement of the chemical, nutritional and sensorial characteristics of breast fillet. Overall, the study indicates that B. licheniformis can be used as a growth promoter and meat quality enhancer in broiler poultry.

오적산 (五積散)의 고지방식이 마우스 지방축적억제 및 3T3-L1지방세포에서의 비만 조절 기전에 관한 연구 (Inhibitory effect of by Ojeok-san lipid accumulation in high fat diet-induced obesity mice and 3T3-L1 adipocytes)

  • Objectives : Ojeok-san (OJS), an oriental herbal formula, has been used in Asian countries including Korea, China and Japan to treat the common cold and illnesses including fatigue and gastrointestinal disorders. The purpose of this study was to examine the anti-obesity effect and molecular mechanism of OJS, on adipogenesis in 3T3-L1 cells. Also, the effects of OJS in obese mice fed a high-fat diet on adiposity were examined.Methods : Preferentially, we analyzed the component of OJS and measured the stability of its component in OJS according to study periods using high performance liquid chromatography (HPLC). In vitro, 3T3-L1 cells were treated with OJS (50 to 200 μg/mL) during differentiation for 8 days. The accumulation of lipid droplets was determined by Oil Red O staining. The expressions of genes related to adipogenesis were measured by RT-PCR and Western blot analyses. For anti-obesty effect in vivo, we experimented for 8 weeks with four group (normal diet (CON), high-fat diet (HF), high-fat diet with OJS (HF+OJS) and high-fat diet with Bang-pung-tong-sung-san (HF+BTS) in comparison group HF+OJS).Results : OJS showed inhibitory activity on adipocyte differentiation at 3T3-L1 preadipocytes without affect cell toxicity as assessed by measuring fat accumulation and adipogenesis. In addition, OJS significantly reduced the expression levels of several adipocyte marker genes including proliferator activated receptor-γ(PPAR-γ) and CCAAT/enhancer-binding protein-α(C/EBP-α). Also OJS-administered mice showed significant inhibitory of body weights and abdominal adipose tissue weights.Conclusions : This study showed that traditional medicine OJS has an anti-obesity effect in vitro and in vivo. Thus, OJS could be developed as a supplement for reduction of body weight gain induced by an obesity.

꽃게액비 엽면시비가 추부 들깻잎의 영양성분에 미치는 영향 (Effects of Liquid Fertilizer of Portunus trituberculatus Foliar Application on the Proximate Components of Chubu Perilla frutescens Leaves)

  • The purpose of this study was to investigate the effects of fertilizer application on the content of functional materials, such as fatty acids and organic acids in perilla leaves. (1) As compared to the controlled with Ammonium sulfate fertilizer, the yield of Perilla frutescens leaves increased by 7.3% and 12.8% of biomass at 2000 and 1000 times liquid respectively, but decreased by 7.6% at 500 times. The lipid and protein contents of perilla leaves were higher than those of Portunus trituberculatus liquid at 1000 times. The average contents of minerals (mg/100g) were as follows; K (638.4) > Ca (561.6) > P (145.4) > Mg (133.5) > Fe (36.2) > Zn (1.9) > Mn (1.6) > Na (1.4 mg). However, the correlation between the controlled with P. trituberculatus liquid was relatively low. (2) The fat-soluble vitamin E content of P. frutescens leaves was 3.4, 3.9, 3.3, and 3.9 mg in the controlled with liquid by 500, and 1000, 2000 times respectively; Vitamin A contents were 6.4, 8.9, 10.9, and 8.5 mg respectively, which was more than twice as much as the corresponding vitamin E content. The water soluble vitamin C contents were 177.9, 172.6, 195.2, and 163.5 mg respectively. (3) Amino acid contents of P. frutescens leaves in 100 g of fresh weight were 3821.7 mg in the controlled with Ammonium sulfate fertilizer and 3918.8, 4054.0, and 4005.4 mg in the controlled with P. trituberculatus liquid at 2,000, 1000, 500 times respectively. Amino acid contents of each controlled group with P. trituberculatus liquid above were as small as 2.5~6.1%, and these contents of amino acid were as follows: Glutaminic acid > Aspartic acid > Leucine > Arginine > Phenylalanine. In further study, it is necessary to develop an effective microorganism and a variety of amino acid fertilizer to supplement the study on new manufacturing.

Bacillus subtilis BK-17 유래 혈전용해효소(Bacillokinase)의 고체배양 (Solid Cultivation of Fibrinolytic Enzyme (Bacillokinase) from Bacilis subtilis BK-17)

  • 곡물을 소재로 혈행개선에 효과 있는 물질을 개발하기 위하여 Bacillus subtilis BK-17을 이용하여 고체 배양을 시도하였다. 고체배양에 접종하는 미생물은 영양세포보다 포자 접종이 혈전용해효소 생산에 효과적이었다. 포자 생산을 위한 배지는 SFM (0.8% nutrient broth, 0.05% yeast extract, $10^{-1}$ M $MgCl_2$, $10^{-3}$ M $FeCl_3$, $10^{-4}$ $MnCl_2$, $10^{-5}$ M dipicolic acid, pH 6.5)이 최적이었고, 포자 생성에 대한 최적 pH 와 온도는 각각 pH6과 $30^{\circ}C$였으며 포자는 배양 후 60 시간에서 가장 많은 생산을 보였다. 각종 곡물을 기질로 하였을 때 녹두가 혈전용해능이 가장 높았으며 강낭콩, 검정콩, 대두, 옥수수는 녹두의 약 90% 수준의 활성으로 높은 혈전용해능을 보였다. 반면에 보리와 밀과 같은 탄수화물 곡물은 낮은 활성을 보였다. 고체 배양한 발효 대두는 단백질과 지질 함량에 있어 발효하지 않은 대두보다 약 10% 이상의 높은 수치를 보였다. 또한 필수 아미노산과 일반 아미노산의 함량 역시 발효 대두 쪽이 약 5 - 20% 높은 것으로 보아 혈행개선기능성 뿐만 아니라 식품으로 실용 가능성이 높은 것으로 사료된다.

1,2-Dimethylhydrazine에 의해 유발된 Colonic Aberrant Crypt Foci에 대한 마늘추출물의 암예방효과 (Chemopreventive Effects of Garlic Extracts on Rat Colonic Aberrant Crypt Foci Induced by 1,2-Dimethylhydrazine)

  • Garlic (Allium sativum L.) with the food supplement material and medicine was used traditionally in Asia and Europe. Epidemiological studies revealed that the intake of garlic reduced incidences of various cancer including digestive system. The present study was designed to investigate the effect of garlic ethanol extract on the development of colonic aberrant crypt foci (ACF) induced by 1,2-dimethylhydrazine (DMH) in male F344 rats. Five-week-old rats were given four times for two weeks to subcutaneous injections by DMH (30 mg/kg body weight) to induce ACF. The animals were divided into groups that fed diet containing garlic ethanol extract at five different doses (0.1, 0.2, 0.5, 2, 5%), respectively, animals were evaluated for the total number of ACF and total aberrant crypts (AC) per colon detected from methylene blue-stained rat colon. ACF were formed in animals in DMH-treated group. The feeding suppressed potently the appearance ACF in the colon of rats. Especially, fed diet containing garlic ethanol extract at intermediate dose (0.5%) significantly reduced the number of ACF and AC per colon (p < 0.05). Garlic ethanol extract inhibited DMH-induced overexpression of Activator Protein-1 (AP-1) and ${\beta}-catenin$ genes related to cell proliferation that also upregulated the expression of p21Waf1/Cip1 mRNA, a cell cycle-regulating gene. These results suggested that garlic ethanol extract may inhibit ACF formation, ${\beta}-catenin$ gene as the early preneoplastic marker of malignant potential in the process of colon carcinogenesis.

이란산 흑석류 농축액과 그 제품의 성분 및 함유된 Phyto 에스트로겐류에 관한 연구 (Proximate Compositions and Selected Phytoestrogens of Iranian Black Pomegranate Extract and Its Products)

  • 천연 호르몬 보충 제제의 개발을 위한 기초 연구로 이란산 흑석류 과즙 농축액과 이를 이용한 제품의 화학성분들을 분석하였다. 석류 농축액의 일반성분은 수분 39.3%, 조지방 0.4%, 조단백질 0.9%, 조회분 1.4%, 그리고 탄수화물은 42.0%이었다. 아미노산 함량은 글루탐산이 1310.0ppm, 아스파르트산이 896.2ppm, 아르기닌이 877.7ppm, 페닐알라닌이 57.5ppm순으로 무기 성분들은 철분 6640.0ppm, 염소 3464.0ppm, 칼륨 2550.8ppm, 인 150.0ppm, 칼슘 80.0ppm순으로 많이 함유되어 있었다. 비타민은 5가지의 수용성 비타민이 함유되어 있었으며 그 중 비타민 C(20mg/100g)를 제외한 나머지 비타민들이 아주 적은 양이 있었다. 주요 지방산은 palmitic acid(8.3%)와 stearic acid(69.4%)로 전체 지방산의 약 60~80%를 차지하고 있었다. 또한 6종의 Phyto 및 에스트로겐류가 들어 있었으며 각각 daidzein 23.72ppm, quercetin 9,75ppm, catechin 1.48ppm, genistein 0.29ppm, 2,3-di-MeO-estradiol이 0.04ppm, 그리고 17$\beta$-에스트라디올이 0.15ppm이 함유되어 있었다. 이상의 결과들을 석류 농축액으로 제조한 제품 포에버 120과 칡과 대두 isoflavon 농축분말과 비교하였다.

Anti-Helicobacter pylori IgY의 안전성 및 임상적 효과 (The Safety and Clinical Test of Anti-Helicobacter pylori IgY)

  • 본 연구는 H. pylori를 산란계에 과면역시켜서 얻은 anti-H pylori IgY난황 분말의 안전성과 위염 환자에 있어서 H. pylori에 대한 억제 효과를 알아보기 위하여 실시하였다. Anti-H. pylori IgY난황 분말의 안전성 실험은 급성 독성실험과 아급성 독성실험으로 알아보았다. 급성 독성 실험시 식이섭취나 성장 모두 정상적이었고, 마우스의 체중(kg)당 400mg까지 투여했을 때 이상을 관찰할 수 없었다. 아급성 독성실험에서 외관상의 이상현상은 관찰할 수 없었고, 혈청의 CBC 검사에서 백혈구 수치도 정상적이었다. 그리고 소화기관과 간조직의 병리학적 이상은 관찰되지 않았다. 이 결과로 미루어 anti-H pylori IgY난황분말은 독성이 없음이 확인되었다. 임상실험에서는 WSF 분말 급이군의 UBT 수치는 58.65에서 35.33으로 약 23.31 감소하였으며, anti-H pylori IgY 난황분말 혼합 급이군의 UBT 수치는 47.77에서 28.81로 약 17.95 감소 효과를 보였다. WSF 분말 급이군은 anti-H. pylori IgY 난황분말 혼합 급이군보다 UBT 수치의 감소율이 더 크게 나타났다. 이와 같이 임상실험에서 anti-H, pylori IgY는 H. pylori의 억제 효과가 있음이 확인되었다.

Nutritional Evaluation of Lentil (Lens culinaris) Straw and Urea Treated Wheat Straw in Goats and Lactating Buffaloes

  • The nutritive value of lentil straw (Lens culinaris) relative to 4% urea treated wheat straw (w/w) was assessed in adult bucks and milking buffaloes on-station and on-farm, respectively. A digestion-cum-nitrogen balance experiment was conducted with three bucks (24.35 kg) fed ad libitum lentil straw (LS), urea treated wheat straw (UTS) and a 1:1 (w/w DM basis) mixture of LS and UTS (LS:UTS) in a 3$\times$3 Latin square design. Though daily dry matter intake (g kg $W^{0.75}$ or % LW) of bucks fed LS and UTS did not differ significantly, that of LS:UTS was significantly (p<0.05) low. Similarly, digestibility of nutrients (DM, OM and CP) and nitrogen balance (g/day) did not differ significantly for bucks given LS or UTS although a significant positive improvement was evident in these parameters for LS:UTS. The digestibility of fibre fractions (NDF and ADF) was significantly (p<0.05) lower in LS as compared to UTS or LS:UTS. Except for the significantly (p<0.05) lower intake of DCP (g kg$W^{0.75}$), the bucks fed LS had statistically similar nutrient intake as compared to UTS. Feeding of LS:UTS resulted in significantly higher nutrient (DCP, TDN) intake by bucks. The important blood-biochemical parameters (haemoglobin,packed cell volume, glucose, urea nitrogen, total protein) and activity of serum enzymes (alanine aminotransferase and aspartate aminotransferase) did not differ significantly in goats irrespective of dietary treatment. In the onfarm study, eighteen multiparous milking buffaloes were randomly assigned into three equal groups to assess the relative effect of feeding untreated wheat straw (WS), UTS and LS:UTS ad libitum as basal feed with a supplement. The study continued for 3 months and revealed that daily dry matter intake and average milk production (liters/day) by buffaloes offered LS:UTS and UTS was statistically similar but it was significantly higher compare to WS. However, milk composition did not differ significantly among the dietary treatments. It may be concluded that the nutritive value of lentil straw appeared to be no different to UTS, however, a positive synergistic effect was evident by feeding a mixture of LS and UTS on performance of goats and lactating buffaloes.

Supplemental Enzymes, Yeast Culture and Effective Micro-organism Cultureto Enhance the Performance of Rabbits Fed Diets Containing High Levels of Rice Bran

  • An experiment was carried out to study the effects of exogenous enzymes (cellulases and proteases), yeast culture and effective micro-organism (EM) culture on feed digestibility and the performance of rabbits fed rice bran rich diets over a period of ten weeks. Twenty four, 8 to 9 weeks old male and female New Zealand White rabbits were allotted to 4 dietary treatments; a basal (control) feed containing 43% rice bran, basal feed supplemented with either enzymes, yeast culture or EM. Individual feed intake, body weight gain, nutrient digestibility, carcass characteristics and feed cost were studied. Sex of the rabbits had no significant (p<0.05) influence on the parameters studied. The control group showed the lowest daily feed intake (104.8 g), body weight gain (12.8 g) and the highest feed/gain ratio (8.20 g/g). The highest daily feed intake (114.3 g), body weight gain (20.42 g) and the lowest feed/gain ratio (5.60) were observed with enzymes. Compared to the control, yeast significantly (p<0.05) improved the feed intake, body weight gain and feed/gain ratio by 4.9, 34.4 and 22.0%, respectively, while EM improved (p<0.05) them by 4.0, 32.6 and 21.6%, respectively. All the additives improved (p<0.05) the digestibility of dry matter, crude protein, crude fiber and energy by 4.9-8.7, 3.6-10.7, 5.9-8.3 and 4.3-6.4%, respectively. Higher weights of pancreas (by 38.5-56.4%) and caecum (by 13.1-26.8%, compared to the control) were recorded with all additives but liver weight was increased only by yeast (24.5%) and enzymes (26.7%). Significantly (p<0.05) higher carcass recovery percentages were observed with enzymes (60.55), yeast (60.47) and EM (56.60) as compared to the control (48.52). Enzymes, yeast and EM reduced (p<0.05) the feed cost per kg live weight by 23.8, 15.9 and 15.5%, respectively. Results revealed that enzymes, yeast culture and EM can be used to improve the feeding value of agro-industrial by-products for rabbits in Sri Lanka and thereby to reduce the feed cost. Under the present feeding system, enzyme supplement was the best.

Use of Leucaena leucocephala and Gliricidia sepium as Nitrogen Sources in Supplementary Concentrates for Dairy Goats Offered Rhodes Grass Hay

  • A study was conducted to evaluate the replacement value of Leucaena leucocephala and Gliricidia sepium as nitrogen sources in commercial type supplements for dairy goats. Six crossbred $(Toggenburg{\times}Saanen)$ goats at late stage of lactation were allocated to three dietary treatments in a double $3{\times}3$ Latin square design. The animals were offered rhodes grass (Chloris gayana) hay ad libitum and supplemented with either Leucaena-based concentrate (LBC), Gliricidia-based concentrate (GBC) or commercial based concentrate (CC). Voluntary food intake, milk yield and composition and changes in live weight were measured. The total dry matter (DM) intake was higher (p<0.05) in goats fed GBC than CC (1385 vs 1331 g/d). The DM intake for LBC (1343 g/d) was similar to CC (1331 g/d). The DM intake of hay was also higher (p<0.05) in goats fed GBC (834 g/d) than those receiving LBC or CC (789, 782 g/d, respectively). Animals supplemented with GBC recorded positive (11 g/d) weight gain while the other groups lost weight (13, 19 g/d) for LBC and CC respectively, although these differences were not statistically significant (p>0.05). The composition of milk were: butterfat 58, 49 and 55 g/kg; crude protein 37.0, 35.4 and 36.1 g/kg; lactose 33, 29 and 30 g/kg; Ash 8.5, 8.5 and 7.9 g/kg and total solids 136.5, 121.9 and 129.0 g/kg, for goats fed LBC, GBC and CC respectively. There were no differences in the composition of milk due to these dietary treatments. At the end of performance trial, a digestibility trial was conducted using 6 female goats allocated to the three treatments (LBC, GBC and CC) in an incomplete randomized block design and each goat received a different supplement in each of two successive periods. There were no differences in nutrient digestibility except for DM, which was higher (p<0.05) in CC compared to the other treatments (615, 622, 720 g/kg for LBC, GBC and CC, respectively). Economic analysis showed that CC diet was more expensive (0.20 US$/kg) and had a lower margin over supplementation (0.11 US$) compared to LBC and GBC (0.13 vs 0.12 US$/kg and 01.5 vs 0.12 US$, respectively). It is concluded that the Leucaena and Gliricidia could contribute as nitrogen sources in compounded diet supplements without any detrimental effects on production in dairy goats.
