Thermodynamic Incompatibility of Food Macromolecules (식품 거대분자의 열역학적 비혼합성)

  • Proteins and polysaccharides are major food macromolecules. Generally, the mixture of these macromolecules can be separated into two phases because of their thermodynamic incompatibility. Phase separ-ation is explained by equilibrium phase diagram, which comprises binodal curve, critical point, phase separation threshold, tie-line and rectilinear diameter. Phase separation of protein-polysacc-haride solution is affected by pH, temperature, ionic strength, molecular weight, molecular structure, etc. Membraneless osmosis has been developed to concentrate protein solutions, using the phase diagram constituted by proteins and polysaccharides. Protein-polysaccharide mixtures are very promising fat mimetics because solution of mixtures forms water-continuous system with two phase-separated gels, which give plastic texture and a fatty mouthfeel.

Comparison of Protein Binding Polysaccharide from Agaricus blazei Murill Prepared by Ultrafiltration and Spray-Drying Process

  • Chemical properties of spray-dried powders separated based on molecular weight from crude protein binding polysaccharide (CP-SD) of Agraricus blazei were examined. Contents of ${\beta}$-glucan in SD-1, SD-2 and SD-3 were 18.67%, 48.24%, and 37.15% respectively, and SD-2 (10-150 kDa) showed the highest molecular weight. Obtained ${\beta}$-glucans were not pure glucan, but was determined to be an acidic proteo-heteroglycan with a large amount of glucose (74.46-80.05%), galactose (8.91-15.2%), and mannose (4.9-5.46%). Composition of their amino acids was mainly aspartic and glutamic acids. FT-IR spectrum revealed SD-1, SD-2 and SD-3 were structures of ${\beta}$-1,3-glucans and ${\alpha}$-1,6-glucans at 890 and 930 $cm^{-1}$, respectively, signals of ${\alpha}$-1,6-glucans for CP-SD was not found. Useful CP-SD was recovered from A. blazei for preparation of three powder types as food materials.

Antitumor Components of Cryptoporus volvatus (한입버섯의 항암성분(抗癌成分)에 관한 연구(硏究))

  • The carpophores of Cryptoporus volvatus collected in Gyeong-gi Province of Korea were extracted with water and a protein-polysaccharide fraction was obtained after dialysis and lyophilization. The antitumor activity of this fraction was tested against sarcoma 180 implanted in A-strain mice. The tumor inhibition ratio was 80.4% in case of the high dose group (50mg/kg, ip, 10 days) and 70.3% in the low dose group (20mg/kg, ip, 10 days). The protein­polysaccharide fraction was chemically analyzed and was found to be a complex of a protein which was 18.2% of the fraction when determined by Lowry-Folin method, and a polysaccharide which was 55.3% of ther fraction when determined by Anthrone method. Their subunits were identified as four monosaccharides and 18 amino acids by gas-liquid chromatography and amino acid autoanalysis.

  • In the course of searching immunomodulators from natural sources, the protein-bound polysaccharide, CM-Ala, has been isolated from the water extract of Chelidonium majus L. (Papaveraceae). The immunostimulatory characteristics have been investigated in several experiments such as generation of activated killer (AK) cells, proliferation of splenocytes, activation of macrophages and granulocyte macrophage-colony forming cell (GM-CFC) assay. Of the fractions obtained using Sephacryl S200 column chromatography, CM-Ala was the most effective fraction that augmented the cytotoxicity against Yac-1 tumor cells from 0.88% to 34.18% by culturing with splenocytes for 5 days. CM-Ala also enhanced nitric oxide production by two fold in peritoneal macrophages and exhibited antitumor activity. It showed mitogenic activity on both spleen cells and bone marrow cells. CM-Ala induced proliferation of splenocytes by 84 fold and increased GM-CFC numbers by 1.48 fold over than the non-treated. On the contrary, CM-Ala had cytotoxic activity to a diverse group of tumor cells. From the above results, we proposed that CM-Ala has a possibility of an effective antitumor immunostimulator.

  • To find pharmacologically active components of Agrocybe cylin(Iracea, its basidiocarps were extracted with water. The extracts were separated by DEAE cellulose column chromatography, Sepharose CL-4B gel filtration, and Concanavalin A-Sepharose 4B affinity chromatography. Among the obtained fractions from A, cylinclracea, fraction IN which was the neutral proteinbound-polysaccharide fraction exhibited a marked antitumor activity and it was tentatively named "Cylindan". It showed about 70% of tumor inhibition against the solid form of sarcoma 180 when a dose of 30 mg/kg/day was intraperitoneally injected into ICR mice. When each fraction was examined by chemical analysis, Cylindan consisted of 85% polysaccharide, 3% protein and 1% hexosamine. Its polysaccharide moiety contained glucose, mannose, fucose and galactose and its protein moiety contained the comparatively large amounts of aspartic acid and glycine, and other 11 amino acids.

  • To examine physico-chemical properties of the polysaccharide extracted from liquid-cultured mycelia of Schizophyllum commune, each the polysaccharide was extracted with hot water treatment and then fractionated with ethanol, alkaline solution and ultrafiltration. And we determined carbohydrate contents, composition of amino acids, infra-red spectrum and viscosity. Carbohydrate contents of polysaccharide treated with ethanol and ultrafiltration were 72.0% and 62.3%, and proteins content were 15.3% and 32.0% respectively. The carbohydrate consisted of four monosaccharides and the protein contained 16 amino acids. The polysaccharide obtained from ultrafiltration was shown an absorption band characteristic of the ${\beta}$-glycosidic linkage by infra red spectra. These results suggest that the polysaccharide extracted from Schizophyllum commune showed the characteristics of proteinbounded polysaccharide, and it was ${\beta}$-glycosidic linkage with strong viscosity.

Isolation of an Algal Growth-enhancer Polysaccharide from the Chlorophyta Monostroma nitidum

  • A micro algal growth-enhancing polysaccharide compound was isolated from the green alga Monostroma nitidum using water extraction, molecular fractionation, a DEAE-cellulose column, and fast protein liquid chromatography using a Superose-12 column. The yield of the compound from the seaweed powder was 8.3$\times$l0$^{-3}$%. At 2 mg/mL concentration, the polysaccharide enhanced Tetraselmis suecica cell growth in f/2 medium by approximately 160%.

  • Polysaccharide fractions were prepared from Forsythia Corea by several different extraction schedules. The fractions obtained were designated as ForCo A, ForCo B and ForCo C respectively. ForCo C was further purified through Sephadex G 200 column chromatography and obtianed two subfractions (Coreana A, Coreana B). ForCo showed marked antitumor activity against sarooma 180 and its activity was dose-dependent. Coreana A, purified from ForCo C, showed somewhat higher antitumor activity. ForCo C increased the number of circulating leucocytes and peritoneal exudate cells, but didn't show any significant effects on the phagocytic activity and total serum protein level. Chemical analysis showed that ForCo C was composed of glucose, galactose, xylose and arabinose. A close relationship between antitumor activities and polysaccharide contents waw observed. These results indicate strongly that antitumor active principle of Forsythia Corea is polysaccharide.

An Antitumor Constituent of the Cultured Mycelia of Hydnum repandum (턱수염 버섯 배양(培養) 균사(菌絲)의 항종양(抗腫瘍) 성분(成分))

  • To investigate antitumor constituents of Hydnum repandum, which belongs to edible basidiomycetes, the mycelia separated from the carpophore were shake-cultured and subjected to hot water extraction. The extract was concentrated under vacuum and mixed with a three-fold volume of 95% ethanol to yield precipitates. The water soluble fraction of the precipitates was dialyzed and then lyophilized to yield a water soluble protein-polysaccharide fraction (= WPPF). This exerted antitumor activity against sarcoma 180 implanted in ICR mice. When administered i.p. at the dose level of 20mg/kg once daily for ten consecutive days, it showed an inhibition ratio of 54.3%. WPPF was found to be composed of a polysaccharide moiety (42% of WPPF) and a protein moiety (28% of WPPF) when determined by colorimetric method using anthrone reagent and Folin's phenol reagent. The polysaccharide moiety of WPPF was found to contain glucose (57.4%), mannose(19.3%), galactose (10.8%), xylose (6.8%), and fucose (5.7%), when the methanolysate of WPPF was analysed by GLC method.

Effects of Protein-Bound Polysacharide Isolated from Acanthopananx senticosus in Reducing the Toxic Effects of Cisplatin (가시오가피 추출물의 독성경감 및 면역증강효과)

  • Protein-bound polysaccharide is derived from Acanthopananx senticosus by the cold water extraction. The aim of the study was to evaluate the effects of PS against weight loss and hematological change as a indication of toxicity produced by the treatment of cisplatin. PS protected the weight loss caused by cisplatin (6 mg/kg) and significantly recovered hematological change. Treatment of PS showed the recovery on the weight loss and hematological change as indicators of toxicity of cisplatin treatment. By increasing lymphocyte proliferation and cytokine production, PS may be highly effective in protecting against cisplatin-induced toxicity. The results suggest PS might have a role in reducing toxicity or permitting larger dose of cisplatin to be given.
