Current status and clinical application of patient-derived tumor organoid model in kidney and prostate cancers

  • Urological cancers such as kidney, bladder, prostate, and testicular cancers are the most common types of cancers worldwide with high mortality and morbidity. To date, traditional cell lines and animal models have been broadly used to study pre-clinical applications and underlying molecular mechanisms of urological cancers. However, they cannot reflect biological phenotypes of real tissues and clinical diversities of urological cancers in vitro system. In vitro models cannot be utilized to reflect the tumor microenvironment or heterogeneity. Cancer organoids in three-dimensional culture have emerged as a promising platform for simulating tumor microenvironment and revealing heterogeneity. In this review, we summarize recent advances in prostate and kidney cancer organoids regarding culture conditions, advantages, and applications of these cancer organoids.

Meta-analysis of the Relation Between the VDR Gene TaqIpolymorphism and Genetic Susceptibility to Prostate Cancer in Asian Populations

  • Background: Polymorphisms of the Taq I gene have been associated with prostate cancer risk. Methods:We applied a fixed-effects model to combine odds ratios (ORs) and 95% confidence intervals (95% CI). The Egger's test was carried out to evaluate potential publication bias. Results: A total of 10 case-control studies enrolling 1,141 prostate cancer patients and 1,685 controls were included in this meta-analysis. Compared with the T allele, the OR for the C allele was 0.81 (0.70-0.94). The ORs for CT and CC+CT genotypes were 0.86 (0.74-1.01) and 0.84 (0.73-0.97) compared to wide type genotype (homozygote TT). Conclusions: The present meta-analysis suggests that the TF gene Taq I polymorphism may reduce the prostate cancer risk in Asian populations.

Inhibitory Effects of Dunning Rat Prostate Tumor Fluid on Proliferation of the Metastatic MAT-LyLu Cell Line

  • Tumor fluid accumulation occurs in both human cancer and experimental tumor models. Solid tumors show a tendency to tumor fluid accumulation because of their anatomical and physiological features and this may be influenced by molecular factors. Fluid accumulation in the peri-tumor area also occurs in the Dunning model of rat prostate cancer as the tumor grows. In this study, the effects of tumor fluids that were obtained from Dunning prostate tumor-bearing Copenhagen rats on the strongly metastatic MAT-LyLu cell line were investigatedby examining the cell's migration and tumor fluid's toxicity and the kinetic parameters such as cell proliferation, mitotic index, and labelling index. In this research, tumor fluids were obtained from rats injected with $2{\times}10^5$ MAT-LyLu cells and treated with saline solution, and 200 nM tetrodotoxin (TTX), highly specific sodium channel blocker was used. Sterilized tumor fluids were added to medium of MAT-LyLu cells with the proportion of 20% in vitro. Consequently, it was demonstrated that Dunning rat prostate tumor fluid significantly inhibited proliferation (up to 50%), mitotic index, and labeling index of MAT-LyLu cells (up to 75%) (p<0.05) but stimulated the motility of the cells in vitro.

Diabetes Mellitus and Prostate Cancer Risk in Asian Countries: a Meta-analysis

  • Background/Aims: Diabetes mellitus (DM) is widely considered to be associated with risk of cancer, but studies investigating the association between DM and prostate cancer in Asian countries have reported inconsistent findings. We examined this association by conducting a detailed meta-analysis of studies published on the subject. Methods: Cohort or case-control studies were identified by searching Pubmed, Embase and Wanfang databases through May 30, 2012. Pooled relative risk (RR) with its corresponding 95% confidence interval (95% CI) were calculated using the random-effects model. Subgroup analyses were performed by the study type. Results: Finally, we identified 7 studies (four cohort studies and three case-control studies) with a total of 1,751,274 subjects from Asians. DM was associated with an increased risk of prostate cancer in Asians (unadjusted RR= 2.82, 95% CI 1.73.4.58, P < 0.001; adjusted RR= 1.31, 95% CI 1.12.1.54, P = 0.001). Subgroup analyses by study design further confirmed an obvious association. Conclusion: Findings from this meta-analysis strongly support that diabetes is associated with an increased risk of prostate cancer in Asians.

복분자 미숙과 추출물이 전립선암 세포주와 전립선비대 백서모델에 미치는 영향 (Effects of Unripe Black Raspberry Extracts on Prostate Cancer Cell Line and Rat Model of Benign Prostatic Hyperplasia)

  • 본 연구는 전립선암세포주인 LNCaP 세포주와 테스토스테론으로 유도된 전립선비대 백서모델에서 복분자 추출물의 전립선비대 억제 효과를 조사하였다. 첫째, 전립선암 세포주(LNCaP)에서 복분자 미숙과 추출물의 안드로겐 관련 전립선비대 유전자 억제 효과를 조사한 결과 미숙과 50% 에탄올 추출물은 안드로겐 수용체(AR)뿐만 아니라 전립선특이항원(PSA)과 5-알파 환원 효소 type 2(5AR2)의 발현을 가장 높게 억제하였다. 또한 LNCaP 세포에 DHT로 안드로겐 관련 유전자를 유도한 후 복분자 미숙과 50% 에탄올 추출물을 처리한 결과 AR과 PSA mRNA의 발현이 억제됨을 확인하였다. 둘째, 테스토스테론을 이용하여 전립선비대를 유도한 동물모델에서 복분자 미숙과 50% 에탄올 추출물을 6주간 투여한 후 전립선비대 개선 효과를 조사한 결과, 전립선비대유발군에 복분자 미숙과 추출물을 투여한 군에서 전립선 무게, 전립선 소포의 상피세포 두께 및 면적이 감소함을 확인할 수 있었고, 전립선비대 유발 호르몬인 DHT level이 감소함을 확인할 수 있었다.

3차원 의료영상진단기기를 이용한 가상 전립선 용적 측정 (Measurement of Prostate Phantom Volume Using Three-Dimensional Medical Imaging Modalities)

  • Recently, advance on various modalities of diagnosing, prostate volume estimation became possible not only by the existing two-dimension medical images data but also by the three-dimensional medical images data. In this study, magnetic resonance image (MRI), computer tomography (CT) and ultrasound (US) were employed to evaluate prostate phantom volume measurements for estimation, comparison and analysis. For the prostate phantoms aimed at estimating the volume, total of 17 models were developed by using devils-tongue jelly and changing each of the 5ml of capacity from 20ml to 100ml. For the volume estimation through 2D US, the calculation of the diameter with C9-5Mhz transducer was conducted by ellipsoid formula. For the volume estimation through 3D US, the Qlab software (Philips Medical) was used to calculate the volume data estimated by 3D9-3Mhz transducer. Moreover, the images by 16 channels CT and 1.5 Tesla MRI were added by the method of continuous cross-section addition and each of imaginary prostate model's volume was yielded. In the statistical analysis for comparing the availability of volume estimation, the correlation coefficient (r) was more than 0.9 for all indicating that there were highly correlated, and there were not statistically significant difference between each of the correlation coefficient (p=0.001). Therefore, the estimation of prostate phantom volume using three-dimensional modalities of diagnosing was quite closed to the actual estimation.

Citrus Fruits Intake and Prostate Cancer Risk: A Quantitative Systematic Review

  • Objectives : The purpose of this systematic review was to investigate the association between dietary intake of citrus fruits and prostate cancer risk. Methods : Authors searched electronic databases and the reference lists of publications of diet and prostate cancer studies until August 2007. All of the epidemiological studies that obtained individual data on dietary intake of citrus fruits and presented risk estimates of the association between intake of citrus fruits and risk of prostate cancer were identified and included. Using general variance-based methods, study-specific odds ratios (OR)/ relative risk (RR) and associated confidence interval (CI)/ standard error (SE) for highest versus lowest intake of citrus fruits level were extracted from each paper. Results : Eleven articles including six case-control studies, one nested case-control study and four cohort studies, proved eligible. Overall summary OR using random effect model did not show an association in risk of prostate caner with intake of citrus fruits (summary OR=1.03, 95% CI=0.89-1.19) with large heterogeneity across studies that we were unable to explain ($I^2$=67.88%). The summary ORs in case-control studies and cohort studies were 1.10 (95% CI=0.97-1.22) and 1.05 (95% CI=0.96-1.14), respectively. Conclusions : Pooled results from observational studies did not show an association between intake of citrus fruits and the risk of prostate cancer, although results vary substantially across studies.

Relationship of Prostate-Specific Antigen Level With Obesity Indices in Korean Middle-Aged Population

  • Purpose: We evaluated the relationship of prostate-specific antigen (PSA) and obesity indices (weight, body mass index [BMI] and waist circumference [WC]) in Korean middle-aged men. Materials and Methods: From February to September 2013, 1,900 police men under 60 years old who participated in a prostate health screening program were included this cross-sectional study. All subjects underwent clinical examinations including weight, height, BMI, WC, fasting blood sugar, lipid profiles, estimated glomerular filtration rate (GFR), and PSA. Total prostate volume (TPV) was assessed clinically. Spearman correlation and multiple linear regression tests were performed to evaluate the obesity indices and PSA relationships. Results: The mean age was $52.0{\pm}4.7years$, and the mean PSA was $0.97{\pm}0.99ng/mL$. The PSA showed a significant positive correlation with the age (r=0.108, p<0.01), TPV (r=0.349, p<0.01), height (r=-0.052, p<0.05), weight (r=0.186, p<0.05), low-density lipoprotein cholesterol (r=0.056, p<0.05), and GFR (r=-0.096, p<0.01). All obesity indices including weight, BMI, and WC showed negative correlations with PSA (beta=-0.013, p<0.001; beta=-0.039, p<0.001; and beta=-0.010, p=0.005; respectively) in age and TPV-adjusted model. Conclusions: Common obesity indices (weight, BMI, and WC) were associated with lower PSA in Korean middle-aged population. Thus, an individual's degree of obesity should be considered when PSA is checked in the first prostate cancer screening of life.

Corn silk extract improves benign prostatic hyperplasia in experimental rat model

  • BACKGROUND/OBJECTIVES: This study was conducted to investigate the effect of a corn silk extract on improving benign prostatic hyperplasia (BPH). MATERIALS/METHODS: The experimental animals, 6-week-old male Wistar rats, were divided into sham-operated control (Sham) and experimental groups. The experimental group, which underwent orchiectomy and received subcutaneous injection of 10 mg/kg of testosterone propionate to induce BPH, was divided into a Testo Only group that received only testosterone, a Testo+Fina group that received testosterone and 5 mg/kg finasteride, a Testo+CSE10 group that received testosterone and 10 mg/kg of corn silk extract, and a Testo+CSE100 group that received testosterone and 100 mg/kg of corn silk extract. Prostate weight and concentrations of dihydrotestosterone (DHT), $5{\alpha}$- reductase $2(5{\alpha}-R2)$, and prostate specific antigen (PSA) in serum or prostate tissue were determined. The mRNA expressions of $(5{\alpha}-R2)$ and proliferating cell nuclear antigen (PCNA) in prostate tissue were also measured. RESULTS: Compared to the Sham group, prostate weight was significantly higher in the Testo Only group and decreased significantly in the Testo+Fina, Testo+CSE10, and Testo+CSE100 groups (P < 0.05), results that were consistent with those for serum DHT concentrations. The concentrations of $(5{\alpha}-R2)$ in serum and prostate as well as the mRNA expression of $(5{\alpha}-R2)$ in prostate were significantly lower in the Testo+Fina, Testo+CSE10, and Testo+CSE100 groups than that in the Testo Only group (P < 0.05). Similarly, the concentrations of PSA in serum and prostate were significantly lower in the Testo+Fina, Testo+CSE10, and Testo+CSE100 groups (P < 0.05) than in the Testo Only group. The mRNA expression of PCNA in prostate dose-independently decreased in the Testo+CSE-treated groups (P < 0.05). CONCLUSIONS: BPH was induced through injection of testosterone, and corn silk extract treatment improved BPH symptoms by inhibiting the mRNA expression of $(5{\alpha}-R2)$ and decreasing the amount of $(5{\alpha}-R2)$, DHT, and PSA in serum and prostate tissue.

Variants on ESR1 and their Association with Prostate Cancer Risk: A Meta-analysis

  • Background: Epidemiological studies evaluating the association of two variants rs9340799 and rs2234693 on estrogen receptor 1 (ESR1) with prostate risk have generated inconsistent results. Methods: A meta-analysis was here conducted to systematically evaluate the relationship of these two variants with prostate cancer susceptibility. Results: For rs9340799, heterozygosity of T/C carriers showed a significant increased prostate cancer risk with a pooled odds ratio (OR) of 1.34 (95% CI = 1.06-1.69) while homozygote C/C carriers showed an increased but not statistically significant association with prostate cancer risk (pooled OR = 1.29, 95% CI = 0.94-1.79). Compared to the homozygous TT carriers, the allele C carriers showed a 31% increased risk for prostate cancer (pooled OR = 1.31, 95% CI = 1.06-1.63). No significant association between the rs2234693 and prostate cancer risk was found with the pooled OR of 1.15 (95% CI = 0.97-1.39, T/C and C/C vs. T/T) under the dominant genetic model. Compared to the homozygote T/T carriers, the heterozygous T/C carriers did not show any significantly different risk of prostate cancer (pooled OR = 1.13, 95% CI = 0.94-1.36) and the homozygous C/C carriers also did not show a significant change for prostate cancer risk compared to the wide-type T/T carriers (pooled OR = 1.26, 95% CI = 0.98-1.62). Conclusion: These data suggested that variant rs9340799, but not rs2234693, on ESR1 confers an elevated risk of prostate cancer.
