Effect of Vehicles and Penetration Enhancers onthe Percutaneous Absorption of Ketorolac Tromethamine across Hairless Mouse Skin

  • The effects of vehicles and penetration enhancers on the in vitro permeation of ketorolac tromethamine (KT) across excised hairless mouse skins were investigated. Among pure vehicles examined, propylene glycol monolaurate (PGML) showed the highest permeation flux, which was 94.3${\pm}$17.3 mg/cm$^2$/hr. Even though propylene glycol monocaprylate (PGMC) alone did not show high permeation rate, the skin permeability of DT was markedly increased by the addition of diethylene glycol monoethyl ether (DGME); the enhancement factors were 19.0 and 17.1 at 20 and 40% of DGME, respectively. 수종 용제 중 퀘르세틴의 용해성 및 안정성 (Solubility and Physicochemical Stability of Quercetin in Various Vehicles)

  • The solubility and stability of quercetin in various vehicles were determined. The solubility of quercetin at $28^{\circ}C$ increased in the rank order of isopropyl myristate < oleyl alcohol < propylene glycol monolaurate < oleoyl macrogol­6 glycerides < linoleoyl macrogol-6 glycerides < propylene glycol laurate (PGL) < propylene glycol monocaprylate (PGMC) < polyethylene glycol-8 glyceryl linoleate < caprylocaproyl macrogol-6 glycerides < diethylene glycol mono ethyl ether (DGME). The addition of DGME to non-aqueous vehicles such as PGL ad PGMC markedly increased the solubility of quercetin. From the stability studies, it was found that quercetin was unstable due to rapid oxidation by dissoved oxygen. The addition of a combination of ascorbic acid and edetic acid (EDTA) at 0.1 % markedly decreased the degradation rates of quercetin in 40% polyethylene glycol 400 in saline. Quercetin was relatively unstable in non-aqueous vehicles such as PGL and PGMC alone, and PGL-PGMC co-solvent The degradation of quercetin in such non-aqueous vehicles was fast, depending on temperature. The addition of butylated, hydroxytoluene, butylated hydroxyanisole, citric acid and/or EDTA at 0.1 % was effective in retarding the degradation of quercetin.

Effect of Vehicles and Enhancers on the In Vitro Permeation of Melatonin through Hairless Mouse Skin

  • The effects of vehicles and penetration enhancers on the in vitro permeation of melatonin through dorsal hairless mouse skin were investigated. Propylene glycol laurate (PGL), isopropyl myristate (IPM), propylene glycol monolaurate (PGML) and propylene glycol monocaprylate (PGMC) showed high permeation fluxes and PGL, PGML and PGMC decreased lag time significantly. In both of the binary co-solvents of diethylene glycol monoethyl ether (DGME)-PGL and DGME-IPM, the highest fluxes were achieved at 20% of DGME, which were $10.5{\pm}1.5$ and $9.1{\pm}2.4{\;}{\mu\textrm{g}}/cm^2/h$, respectively. Among fatty acids used as a permeation enhancer, capric acid and oleic acid in DGME-PGL (80:20 v/v) showed relatively high enhancing effects. Capric acid also shortened the lag time of melatonin from $2.4{\pm}0.7{\;}to{\;}1.3{\pm}0.2{\;}h$. Oleic acid, however, failed to shorten the lag time. Therefore, for effective solution formulations in terms of permeation flux and lag time, capric acid-containing DGME-PGL (80 : 20 v/v) could be used to enhance the skin permeation of melatonin.

  • The solubility and stability of ondansetron hydrochloride (OS) in various vehicles were determined. The effect of cyclodextrins (CD) on the solubility of OS in water was determined by equilibrium solubility method. The solubility of OS at $32^{\circ}C$ increased in the rank order of isopropyl myristate (IPM) < propylene glycol laurate (PGL) ${\ll}$ propylene glycol monolaurate < propylene glycol monocaprylate (PGMC) < poly(ethylene glycol) 400 < diethylene glycol mono ethyl ether (DGME) < ethanol < poly(ethylene glycol) 300 < water (36.1 mg/ml) ${\ll}$ propylene glycol (PG) (283 mg/ml). The addition of PG or DGME to non-aqueous vehicles such as IPM, PGL and PGMC markedly increased the solubility of OS. The addition of CDs in water increased the solubility. Apparent stability constant for the CD complexation with OS was calculated to be $25.5\;M^{-1}$ for $2-hydroxypropyl-{\beta}-CD\;(2HP{\beta}CD)$. Twenty mM ${\beta}-CD$, 69.4 mM sulfobutyl ether ${\beta}-CD$ and 115.4 mM $2HP{\beta}CD$ increased the aqueous solubilty of OS 1.27, 2.18 and 1.85 times, respectively. In Vitro Percutaneous Absorption of Tenoxicam from Pressure-sensitive Adhesive Matrices across the Hairless Mouse Skin

  • To investigate the feasibility of developing a new tenoxicam plaster, the effects of vehicles and penetration enhancers on the in vitro permeation of tenoxicam from a pressure-sensititre adhesive (PSA) matrices across the dorsal hairless mouse skin were studied. Vehicles employed in this study were propylene glycol (PC)-oleyl alcohol (OAI), PG-oleic acid (OA), and diethylene glycol monoethyl ether (DCMI)-propylene glycol monolaurate (PCML) cosolvents with/without fatty acids. In this studys amines such as triethanolamine (TEA) and tromethamine (TM) were additionally used as a solubilized. Among PSAs used, $Duro-Tak^{\circledR}$87-2510 showed much higher release rate than either $Duro-Tak^{\circledR}$ 87-2100 or $Duro-Tak^{\circledR}$87-2196. The relatively high flux rate was obtained with the formulation of DCMI-PCML (40:60, v/v) with 3% OA and 5% TM, and the flux increased as a function of the dose;the initial flux up to 12 h was $4.98{\pm}1.38{\;}{\mu\textrm{g}}/{\textrm{cm}^2}/h$ at the tenoxicam dose of $50{\;} mg/70{\;}{\textrm{cm}^2}$. This flux was much higher than that of a commercial piroxicam patch ($Trast^{\circledR}$) ($1.24{\pm}0.73{\;}{\mu\textrm{g}}/$\textrm{cm}^2/hr$) with almost only one-third that of the commercial patch. Therefore, these observations indicated that these composition of tenoxicam plaster may be practically applicable.

이온토포레시스를 이용한 프로스타글란딘 $E_1$의 경피흡수 (lontophoretic Delivery of Prostaglandin $E_1$)

  • We have studied the transdermal flux of prostaglandin $E_1$ $(PGE_1)$ from a hydrogel patch through hairless mouse skin, to test the possibility of developing a transdermal delivery system. Karaya gum patch containing $PGE_1$ was prepared by casting method. $PGE_1$ was stable in the patch for 10 weeks. The effect of current application, enhancer (propylene glycol monolaurate : PGML), adhesive and patch thickness on the flux was studied using side-by-side diffusion cell. Passive flux of $PGE_1$ was negligible. Cathodal delivery increased the flux about 20 fold. As the concentrations of PGML increased, flux increased. When 5% PGML was used as the enhancer, maximum flux by cathodal iontophoresis was $55\;{\mu}g/cm^2\;hr$. It increased about 2 folds to $100\;{\mu}g/cm^2\;hr$, when the amount of PGML used was 9%. Large increase in flux and the decrease in time to reach maximum flux were observed when the skin was pretreated with neat PGML (maximum flux obtained was about $200\;{\mu}g/cm^2\;hr$). Use of adhesive decreased the flux significantly. To the contrary of our expectation, increase in current density decreased the flux. These flux data together with the stability data indicate that, though the onset of sufficient delivery occur after 1-2 hours of application, therapeutic amount of $PGE_1$ can be delivered through skin using iontophoresis and penetration enhancer.

프로스타글란딘 $E_1$의 안정성 및 경피흡수 (Stability and Percutaneous Transport of Prostaglandin $E_1$)

  • We have studied the stability and transdennal flux of prostaglandin $E_1\;(PGE_1)$ from various donor solutions through hairless mouse skin. Stability in HEPES buffer or in propylene glycol (PG) solution where enhancer (oleic acid (OA), propylene glycol monolaurate (PGML), transcutol (TC), ethanol (EtOH))s dissolved was investigated. $$PGE_1 was not stable in HEPES buffer. The concentration of $$PGE_1 decreased continuously for 7 days, and the degradation rate constant was $0.0028\;h^{-1}$, assuming first order reaction. The effect of current or penetration enhancer on the degradation was minimal. Percutaneous transport from HEPES buffer by passive or iontophoretic delivery without enhancer was close to nil. When OA or PGML was used together with PG, both passive and iontophoretic flux increased. PGML showed better enhancing effect than OA. Flux by cathodal delivery was about 2 times larger than that by passive delivery. Flux by anodal delivery was lower than that by passive delivery. TC and EtOH also increased the transdermal flux, but the effect was not as good as that observed when OA or PGML was used. These stability and flux data provide important information on how to formulate the patch, which will be the next step of this work, and on the polarity of current to use during iontophoresis.

성장호르몬방출펩타이드-6 (GHRP-6)의 경피투과 (Iontophoretic Transport of GHRP-6)

  • The purpose of this study is to characterize the iontophoretic transport of growth hormone releasing peptides (GHRP-6) through hairless mouse skin from aqueous solution. The effect of various factors, such as pH, poloarity, current profile, current density, current duration, ionic strength, drug concentration, and enhancer application was studied to obtain basic knowledge on the transport. We have also studied the stability of GHRP-6 in solution with/without current. The donor chamber was filled with phosphate buffer solution containing GHRP-6 and the receptor chamber was filled with phosphate buffer solution (pH 7.4). Ag/AgCl electrode was used for their stability and reversibility. At a predetermined time interval, sampling was made and the concentration of drug was analysed using HPLC system. The results showed that, compared to passive flux, the total amount of drug transported increased markedly by the application of anodal current. Cathodal flux was similar to passive flux. Flux increased with the current density, the duration of current application and drug concentration. The effect of enhancers on the flux was studied using hydrophilic (5% N-methyl pyrrolidone) and hydrophobic (5% propylene glycol monolaurate, 5% oleic acid) enhancers. Application of enhancer also increased the flux.

이온토포레시스를 이용한 케토프로펜의 경피전달 (Iontophoretic Transport of Ketoprofen)

  • We have studied the effect of polarity, current density, current duration, crosslinking density, swelling ratio, and permeation enhancers on the transdermal flux of ketoprofen from acrylamide hydrogel. Hydrogel was prepared by free radical crosslinking polymerization of acrylamide. Drug loading was made just before transport experiment by soaking the hydrogel in solution containing drug. In vitro flux study using hairless mouse skin was performed at $36.5^{\circ}C$ using side-by-side diffusion cell, and the drug was analysed using HPLC/UV system. The result showed that, compared to passive flux, the total amount of drug transported increased about 18 folds by the application of $0.4\;mA/cm^2$ cathodal current. Anodal delivery with same current density also increased the total amount of drug transported about 13 folds. It seemed that the increase in flux was due to the electrorepulsion and the increase in passive permeability of the skin by the current application. Flux increased as current density, the duration of current application and loading amount (swelling duration) increased. As the cross linking density of the hydrogel increased, flux clearly decreased. The effect of hydrophilic enhancers (urea, N-methyl pyrrolidone, Tween 20) and some hydrophobic enhancers (propylene glycol monolaurate and isopropyl myristate) was minimal. However, about 3 folds increase in flux was observed when 5% oleic acid was used. Overall, these results provide some useful information on the design of an optimized iontophoretic delivery system of ketoprofen.

Interlaboratory Validation Study of In Vitro Alternatives to the Draize Eye Irritation Test : HET-CAM Test and Cytotoxicity Test for 20 Cosmetic Ingredients

  • 피부 전용 제재 개발을 위해 요구되는 동물 대체 시험법 중 가장 적극적으로 연구가 행하여지곤 실제 실용화가 예상되는 것은 안점막 자극 시험으로 지금까지 여러 가지 방법이 개발되었지만 그 중 계란 유정란의 응모요막(CAM)을 이용한 방법이 현재 가장 활발히 진행되고 있다. 이 방법이 일부 국가에서 이미 안점막 자극 시험 동물 대체 시험법으로 공인되었으며 현재까지도 validation 연구를 활발히 진행하고 있다. 본 연구에서도 국내에 적합한 안점막 자극 시험 동물 대체 시험법의 공인 시험법 개발 및 validation study를 목표로 계란 유정란의 응모요막을 이용한 방법 중 HET-CAM 방법을 시행하였으며 안점막 동물 대체 시험법으로 확립하고자 하였다. 틴ET-CAM 방법의 보완을 위해 배양된 세포를 통해 자극도를 측정할 수 있는 방법인 Cytotoxicity test를 도입하여 시행하였으며 두 방법의 data들을 분석하여 validation study를 수행하였다. 국내 유수의 6개 장업사가 본 연구에 참가하여 20가지의 화장품 전용제재를 대상으로 1차, 2차 validation study 를 진행하였다. HET-CAM test, Draize eye irritation test, Cytotoxicity test 측정 결과 HET-CAM 의 “Q” 수치는 대부분 강자극 수치인 2 이상이었고 10% sodium hydroxide가 가장 높은 수치를 보였으며 Tween 20(sorbitanpolyoxyethylene monolaurate) 100%가 가장 낮은 수치를 보였다. In vi패의 경우 10% sodium hydroxide가 가장 높은 수치를 보였으며 30군 propylene glycol 이 가장 낮은 자극수치를 보였다. HET-CAM test 와 Draize eye irritation test, Cytotoxicity test 간의 상관성 분석은 linear correlation coefficient 와 rank correlation coefficient를 구하여 비교하였으며 6개 장업사(A-F)의 실험실에서의 HET-CAM test 결과를 취합하여 각각 두 실험실간의 상관관계(linear correlation)를 분석하였다. Linear correlation coefficient 분석 결과를 보면 전반적으로 상관관계가 0.589 - 0.954의 범위였으며, 특히 A사와 B사 사이의 경우 0.954이었으며, E사와 D사 사이의 경우 0.942로 높은 상관관계를 보였다. 그 외에도 A사와 D사 사이의 경우(0.589)와 B사와 D사 사이의 경우(0.638)를 제외하고는 대체로 높은 상관관계를 나타내었다.
