Thermodynamic Parameters of Complexation of Lanthanides by L-proline (Lanthanides-L-proline 착물의 형성에 관한 열역학적 연구)

  • The stability constants for lanthanides complexes with optically active L-proline (1 : 1) were determined in aqueous solution in the ionic medium of 0.1 M $NaClO_4$ at 25$^{\circ}C$ using a pH titration method. The results show called "gadolinium break" between lighter and heavier lanthanides. The linear relation between the stability constant (log$\beta$1) and the pKa values of ligands indicates that L-proline acts as a bidentate ligand in the complexation. The thermodynamic parameters (${\Delta}H$ and ${\Delta}S$) were also determined using an enthalpy titration method at the same condition. The positive endothermic enthalpy change and positive entropy change clearly indicate that the driving force for the complexation is an entropy effect. The comparison of the thermodynamic parameters of L-proline complexes with anthranilate complexes supports the conclusion that the heterocyclic nitrogen atom and carboxylate of L-proline are involved in the chleate formation. The enthalpy values for L-proline are more positive than the ones for anthranilate complex. The difference in enthalpy change for the complex formation between L-proline complex and anthranilate complex is explained in terms of the basicity of the nitrogen donor atom in the ligand. The relatively large entropy change may be described by the extra dehydration related to the rigidity of L-proline ring.

Salt Tolerance Enhanced by Transformation of a P5CS Gene in Carrot

  • Proline is known as an osmoprotectant accumulating in response to salt and dehydration stresses. An increased level of proline is achieved by either an induced synthesis or a reduced degradation of proline. In an attempt to increase salt tolerance in carrot, a P5CS gene from mothbean was introduced via an Agrobacterium-mediated transformation. The resulting carrot cells and the regenerated plants containing the transgene showed increased levels of proline compared to nontransgenics. The transgenic cell line, Pj2 showed about 6 times increased degree of tolerance determined by relative growth after a treatment in 250 mM NaCl. In facts, due to the retarded growth shown in non-saline condition, Pj2 cells grow only about 1.2 times better than nontransgenic control under salt stress condition. Taken together, it appears that a P5CS is a key enzyme in proline biosynthesis and the increased accumulation of proline by overexpression of the enzyme is enough to enhance tolerance to salt stress in carrot.

Effects of L-proline on the Growth Performance, and Blood Parameters in Weaned Lipopolysaccharide (LPS)-challenged Pigs

  • This trail was conducted to study the effect of L-proline on the growth performance, and blood parameter in the weaned lipopolysaccharide (LPS)-challenged pigs. Thirty six pigs ($9.13{\pm}0.85$ kg) were assigned randomly to dietary treatments in a $2{\times}3$ factorial arrangement in a 20-d growth assay. Factors were intraperitoneal injection with saline or LPS, and three dietary L-proline supplement levels (0%, 0.5%, or 1.0%). On d 10, blood samples were collected at 3 h after LPS (100 ${\mu}g$ LPS/kg body weight [BW]) or saline injection. On d 20 of the trial, all pigs were orally administrated D-xylose (0.1 g/kg BW) at 2 h, and blood samples were collected at 3 h after LPS or saline injection. As a result, dietary supplementation with 0.5% proline had a tendency to increase average daily gain (ADG) in piglets during d 10 to 20 (p = 0.088). Without LPS challenge, dietary supplementation with 1.0% proline had no effect on growth hormone (GH) concentrations on d 10 (p>0.05), but decreased it after LPS challenge (p<0.05). There was LPS challenge${\times}$proline interaction for GH concentrations on d 10 (p<0.05). Dietary supplementation with 1.0% proline decreased glucagon concentration on d 10 after LPS challenge (p<0.05). In addition, dietary supplementation with proline increased superoxide dismutase (SOD) activity significantly on d 10 and 20 (p<0.05), and 1.0% proline increased heat shock proteins-70 concentration on d 10 (p<0.05). Moreover, proline supplementation increased diamine oxidase (DAO) concentrations after LPS challenge (p<0.05). There was LPS challenge${\times}$proline interaction for DAO (p<0.05). Furthermore, dietary supplementation with 1.0% proline increased the D-xylose level when no LPS challenge (p<0.05). These results indicate that proline supplementation could improve growth performance, increase SOD activities, and has a positive effect on the gastrointestinal tract digestibility in early weaned pigs.

Optical Resolution of Free Amino Acids with Addition of Copper(II) Chelates in a Reversed-Phase Liquid Chromatography (구리(II) 킬레이트의 첨가에 의한 자유아미노산 광학이성질체의 역상 액체크로마토그래피적 분리)

  • Separation of the optical isomers of free amino acids by a reversed phase high performance liquid chromatography has been studied by adding a copper(II) complex of L-proline or L-proline derivatives (hydroxy-L-proline, N-benzyl-L-proline, p-xylenyl-L-proline, p-xylenyl-hydroxy-L-proline) in the mobile phase. An OPA postcolumn detection system was used for the detection of amino acids. The chromatographic properties for the free amino acids were discussed in terms of the pH, the kinds and concentration of chelate or organic modifier. The retention behaviors of the free amino acids were considerably different from, those of DNS-amino acids or DABS-amino acids. The enantioselectivity of the free amino acids was better than that of derivatized amino acids. The enantioselectivity between the optical isomers observed by use of the Cu(II)-p-xylenyl-L-proline chiral cheleate was the best among the several copper(II) chelate. A separation mechanism could be illustrated not only by the hydrophobic interaction of the diastereomer with stationary phase but also by the steric effect of the ligand exchange reaction between the free-amino acids and copper chelate.

Effects of salicylic acid on growth and proline of cucumber seedlings

  • The effects of salicylic acid (SA) on growth and proline were investigated in cucumber seedlings. Exogenous application of SA(100 uM - 1mM) led to a noticeable decrease in root and shoot growth, and dry weights of seedlings. Anatomical observation on leaf anatomy of cucumber revealed that the thickness of all leaf tissue components decreased in SA-treated plants. The effect was most promounced on the width of the adaxial epidermis. In the separate and simultaneous effects of SA and water deficit induced by PEG on growth and proline accumulation, the water deficit treatments had greater effects on growth traits and proline content than SA. Combinations of SA and PEG decreased dry matter and root length, and resulted in higher proline in both shoots and roots than SA stress alone. Shoots had higher proline than roots.

Proline Accumulation and P5CS ($\Delta^1$-pyrroline-5-carboxylate synthetase) Gene Expression in Response to Salt Stress in Zoysiagrasses

  • Proline is known as an osmotrotectant to enhance tolerance against both salt and dehydration stresses. A P5CS ($\Delta^1$-pyrroline-5-carboxylate synthetase) plays a major role in regulation of synthesis of proline. An overexpression of the mothbean P5CS gene in transgenic tobacco plant increased the levels of proline and osmotolerance. In an attempt to look for the possibility to use content of proline as well as a level of P5CS gene expression as molecular markers for salt tolerance, the amounts of proline and transcript levels of P5CS were measured as functions of either concentration of NaCl or length of treatment period among different species of zoysiagrass. Hybridzoysia showed the highest level of proline ($329\mu\textrm{g}$/g.f.w.) among five different species of zoysiagrass at 250 mM NaCl in 24 hours. The level of P5CS transcript was also the highest in the hybridzoysia at 250 mM NaCl in 24 hours. The transcriptions of P5CS gene were induced at the rates of 1.2, 1.2, 1.8, and 1.8, upon treatment of 250 mM NaCl in Z. japonica, Z. matrella, Z. sinica and hybridzoysia respectively. Based on a correlation between the level of P5CS transcript and the proline content among different species of zoysiagrass, a comparative structural analysis of the gene for P5CS from either Z. sinica or hybridzoysia may lead to an understanding of mechanism for salt tolerance shown differently among zoysiagrasses.

Effects of Manganese Chloride on Chlorophyll, Free Proline and SOD Activity of Rice Seedling (염화망간 처리가 벼 유묘의 엽록소, 유리 Proline 및 SOD 활성에 미치는 영향)

  • The experiment was conducted to find the changes of early growth and chemical components such as chlorophyll content, superoxide dismutase(SOD) activity, free proline content on the different manganese chloride concentration(2,500, 3,500 and 4,500ppm) in rice seedling. Root growth was decreased in highest concentration, 4,500 ppm of Mn compared with the control and germination rate was also decreased 43% at 4,500 ppm of Mn. Chlorophyll content was decreased at Mn 4,500ppm with 1.16mg. Free proline content at 3 day after germination in Mn 4,500ppm was highest relative to the other manganese chloride concentrations. SOD activity was gradually increased as manganese chloride concentration was increased. As a result, it was suggested that an increment of free proline and SOD activity results from the higher manganese chloride concentration.

  • The continuous production of L -proline was studied using L-histidine auxotrophic mutant of Corynebacterium acetoacidophilum under various substrate limited conditions. Among the $NH_4\;^+$ $PO_4\;^3$ and L -histidine limited conditions, the highest production of L -proline was observed under the L-histidine limited condition. Under $NH_4\;^+$ and $PO_4\;^3$ limited conditions, no or poor L-proline production was observed, respectively. For the kinetic parameters under L -histidine limitation the specific rate of L -proline production was increased with dilution rate upto $0.1hr^{-1}$ but decreased above $0.1hr^{-1}$. The volumetric rate of L -proline production was showed similar pattern with specific rate. The dried cell weight was gradually increased according to decrease the dilution rate. Specific rate of glucose consumption was proportionally increased with dilution rate. The results of continuous culture (higher production of L-proline at dilution rate $0.1hr^{-1}$) will be used in fed-batch culture for the control of cell growth rate and mass production of L-proline.

Effect of Growth Rate on the Production of L-Proline in the Fed-batch Culture of Corynebacterium acetoacidophilum

  • Corynebacterium acetoacidophilum RYU3161 was cultivated in a L-histidine-limited fed-batch culture. To investigate the effect of cell growth on the L-proline production, 5 L fed-batch culture was performed using an exponential feeding rate to obtain the specific growth rates $(\mu)$ of 0.04, 0.06, 0.08, and 0.1 $h_1$. The results show that the highest production of L-proline was obtained at $\mu$ = 0.04 $h_1$. The specific L-proline production rate $(Q_p)$ increased pro-portionally as a function of the specific growth rate, but decreased after it revealed the maxi-mum value at $\mu$ = 0.08 $h_1$. Thus, the highest productivity of L-proline was 1.66 g $L^-^1 h^-^1$ at $\mu$ = 0.08 $h_1$. The results show that the production of L-proline in C. acetoacidophilum RYU3161 has mixed growth-associated characteristics.

Salt Tolerance of Vigna angularis during Germination and Early Seedling Growth

  • The present study was undertaken in investigate the response to salinity and effect of plant growth regulators and proline under salinity stress on the germination and seedling growth of Vigna angularis. The protective effect of external Ca2+ on root elongation under saline conditions was also investigated. The seed germination of Vigna angularis decreased with an increase in salinity. The growth regulators GA3 was more effective than kinetin. At a higher salinity, low concentrations of kinetin and high concentrations of GA3 were more effective. The external application of proline and betaine improved germination under saline conditions. At a low salinity proline and betaine alleviated the salinity-induced inhibition of germination, yet at higher NaCl concentrations, proline and betaine were both ineffective. Exposure to salinity during germination was accompanied by an increase in the proline content, thereby suggesting that one compatible solute in the germinating seed would seem to be proline. The inhibition of germination by high NaCl concentrations was relatively more severe in scarified seeds than in intact seeds, indicating that the seed coat acts as a partial barrier to an Na2+ ameliorated the adverse effect of salinity stress.
