BACKGROUND: Green manure and graminaceousmanure crops have several benefits, such as improving soil physical and chemical properties and utilizing excessive greenhouse nutrients that they have a potential to be a water pollutant source. METHODS AND RESULTS: The objective of this study was to investigate nitrogen (N) supplying capabilities of green manure and graminaceous manure crops for lettuce (Lactuca sativa L.) and Chinese cabbage (Brassica campestris L.) grown under greenhouse conditions. For this two leguminous manures (Crotalaria juncea (Cr.) and Sesbaniaexaltata (Se.)) and two graminaceous manures (Sorghum bicolor; Haussolgo(Ha.) and Sudangrass (Sg.)) in the greenhouse were grown, cut, and incorporated into the greenhouse soil before planting. Chemical nitrogen (N) fertilizer rate was estimated based on N recommendation for lettuce and Chinese cabbage. 100% of the N recommended rates (1N) were 70 kg N $ha^{-1}$ for lettuce and 60 kg N $ha^{-1}$ for Chinese cabbage and 50% of the N recommendation rates (0.5N) were 35 kg N $ha^{-1}$ for lettuce and 30 kg N $ha^{-1}$ for Chinese cabbage. Nitrogen treatments were control (0N), Cr., Se., Cr + 0.5 N, Se + 0.5 N, Ha + 0.5 N, Sg + 0.5 N, and N recommendation rate (1N). Incorporated N from green manure and graminaceous manure crops were 130, 116, 93, and 87 kg N $ha^{-1}$ for Cr., Se., Ha., and Sg., respectively. Lettuce and Chinese cabbage were grown after incorporated green manure crops into the greenhouse soil. There was no significant difference in lettuce and Chinese cabbage yields under N treatments except control (0 kg/ha). Nitrogen use efficiency (NUE)was from 44% to 73% and the highest NUE was under Se. treatment. Although yields were not statistically different under N treatments except control, actual yield increase ranged from 170 to 1,100 kg/ha for lettuce and ranged from 2,770 to 5,210 kg/ha for Chinese cabbage compared to yield under N recommendation rate. Estimated economic benefit from this would be higher approximately between \2,770,000 and \5,210,000/ha under N treatments except control than the N recommendation rate. CONCLUSION: These results suggest that incorporating green manure crops, such as Cr. and SeSe. into soil or adding 0.5 N after incorporation of them can be beneficial in many ways in that it increases economic return because of yield increase, reduces the use of chemical N, and decreases the negative environmental impact on water quality because excessive N in the greenhouse soil can be used by green manure crops during the fallow.

Total of fifty accessions of Brassica rapa with various morphological characteristics were used for production of double haploid plants though microspore culture in Brassica rapa. Among them, only 30 accessions induced embryos from microspores. The highest efficiency of embryo induction of 1.194 per bud was obtained from IT135449 of turnip type, while 3 accessions of sarson (winter oil) type did not generate embryo. The effect of heat shock periods for embryogenesis was also investigated with 4 accessions (IT135449; Turnip type, IT199710; Chinese cabbage type, IT212886; Pak choi type, IT218043; Summer oil type). The high productions of embryos were observed in IT135449, IT199710 and IT212886 when microspores were pre-cultured to $32^{\circ}C$ for 2 days. In IT218043, high embryogenesis was observed at the 3 days of heat shock treatment. The optimal condition of shoot regeneration for IT199710 was observed in MS medium supplemented with NAA $0.5mg{\cdot}L^{-1}$ and BAP $1mg{\cdot}L^{-1}$. In contrast, the IT135449 and IT212886 were observed high regeneration frequency in MS medium without plant growth regulators. All the plantlets regenerated from microspore-derived embryos have been successfully transplanted to soil, and bud self-pollinated seeds were produced from doubled haploid plants. This indicated that double-haploid genotype was likely generated naturally during embryogenesis process.
Larvae of the silkworm (Bombyx mori L.) were reared during the 5th instar on the four kinds of artificial diets on the basis of the different amounts of soybean meal used as the protein source. In this experiment it was shown that the various levels of protein in the diet affected not only the growth and silk production but the digestibility of the diet. haemolymph protein and uric acid excretion. The results obtained are summarized as follows; 1. By an increase of the level of protein in the diet the apparant digestibility was increased. but the protein digestibility was comparatively decreased. 2. Larval body weight increment was not observed by the 3rd day of the 5th instar, but was increased from the 4th day as the level of protein was increased in the diet. 3. After the 3rd day of the 5th instar, protein content in the hemolymph was increased steeply by an increase of the protein content in the diet. However, the percentage of hemolymph protein to the ingested protein was decreased from the 2nd day of the 5th instar and increased more or less after the 4th day. 4. An increase of the uric acid excretion was observed as the content of protein in the diet was increased but the pattern of the uric acid excretion was different between high and low-protein diet. However, the percentage of the uric acid excretion to the ingested protein and to the hemolymph protein were both decreased steeply after the 2nd day of the 5th instar. 5. It was also evident that the high-protein diet increased the cocoon productivity. 6. It showed that the feed efficiency for body weight increment and silk formation was high by an increase of the level of protein in the diet, but the protein efficiency was not.

This study was carried out to investigate the effect of feeding whole crop barley silage (WBS) on the growth performance, gain, feed requirement, carcass grade and beef quality of Hanwoo steers in 1999 to 2001. Twenty one steers were allocated, and divided into three treatment groups which fed rice straw only (control), WBS only, and fresh rice straw silage plus WBS during overall period ad libitum. Concentrates were fed at 1.5% level of live-body weight during growing stages, 4~12 months of age, at 1.8% level of live-body weight during early fattening stages, 13~18 months of age, and ad libitum after late fattening stages, 19~27months of age. The production yields of WBS were 17,135 kg as a silage, and 6,011 kg as a dry matter per ha, and the quality of WBS was 2~3 grade, while that of rice straw silage was 4grade. The daily gain with feeding of WBS (0.70 kg) during experimental period was higher than that of control group (0.65 kg), and the concentrate requirement (7.31 kg) per kg gain in WBS was lower than that of control (8.95 kg). The effects of gain and feed efficiency were very distinct during growing stage. i.e., the daily gain of WBS (0.84 kg) was increased by 65% compared to control (0.51 kg), and the concentrate requirement (4.16 kg) per kg gain in feeding of WBS was decreased by 35%, compared to control (6.39 kg). Meat quality with WBS was higher than that of control. Frequency rate of 1 and/or $1^+$ grade, and marbling score of feeding of WBS were 62.5% and 4.38, while those of control were 37.5% and 2.75, respectively. The results of the sensory evaluation showed that Hanwoo beef fed with WBS were better than that of control. In conclusion, feeding of WBS was desirable to improve daily gain, reduction of concentrates, feed efficiency and meat quality of Hanwoo steers.
In this study, we constructed and tested retrovirus vectors designed to express the human thrombopoietin gene under the control of the tetracycline-inducible promoters. To increase the hTPO gene expression at him-on state, WPRE sequence was also introduced into retrovirus vector at downstream region of either the hTPO gene or the sequence encoding reverse tetracycline-controlled transactivator (rtTA). Primary culture cells (PFF, porcine fetal fibroblast; CEF, chicken embryonic fibroblast) infected with the recombinant retrovirus were cultured in the medium supplemented with or without doxycycline for 48hr, and induction efficiency was measured by comparing the hTPO gene expression level using RT-PCR, western blot and ELISA. Higher hPTO expression and tighter expression control were observed from the vector in which the WPRE sequence was placed at downstream of the hTPO (in CEF) or rtTA(in PFF) gene. This resulting tetracycline inducible vector system may be helpful in solving serious physiological disturbance problems which have been a major obstacle in successful production of transgenic animals.
We investigated growth, clove development, and photosystem II activity in garlic (Allium sativum L.) grown under different day/night temperature regimes using Soil-Plant-Atmosphere - Research (SPAR) chambers to determine the optimum cultivation temperature and to assess the impact of temperature stress on garlic. In the early stages of growth, plant growth increased markedly with temperature. At harvest time, however, the pseudostem diameter decreased significantly under a relatively low day/night temperature range ($14/10-17/12^{\circ}C$), suggesting that these temperature conditions favor regular bulb growth. At harvest time, the bulb diameter and height were great at $14/10-23/18^{\circ}C$, whereas the bulb fresh weight and number of cloves per bulb were greatest at $17/12-20/15^{\circ}C$. However, the number of regularly developed cloves per bulb was highest at the relatively low temperature range of $14/10-17/12^{\circ}C$, as were the clove length and fresh weight. The photochemical efficiency ($F_v/F_m$) and potential photochemical efficiency ($F_v/F_o$) of photosystem II in the leaves of garlic plants were higher at $14/10-20/15^{\circ}C$ and lower at temperatures below $14/10^{\circ}C$ or above $20/15^{\circ}C$, implying that the $14/10-20/15^{\circ}C$ temperature range is favorable, whereas temperatures outside this range are stressful for garlic growth. Furthermore, at temperatures above $20/15^{\circ}C$, secondary growth of garlic, defined as lateral bud differentiation into secondary plants, continuous growth of the cloves of the primary plants, or the growth of bulbil buds into secondary plants, was enhanced. Therefore, to achieve commercial production of fresh scapes and bulbs of garlic, it may be better to grow garlic at relatively low temperature ranges of $14/10-17/12^{\circ}C$.
As information technology fusion is accelerated, the researches to improve the quality and productivity of crops inside a plant factory actively progress. Advanced growth environment management technology that can provide thermal environment and air flow suited to the growth of crops and considering the characteristics inside a facility is necessary to maximize productivity inside a plant factory. Currently running plant factories are designed to rely on experience or personal judgment; hence, design and operation technology specific to plant factories are not established, inherently producing problems such as uneven crop production due to the deviation of temperature and air flow and additional increases in energy consumption after prolonged cultivation. The optimization process has to be set up in advance for the arrangement of air flow devices and operation technology using computational fluid dynamics (CFD) during the design stage of a facility for plant factories to resolve the problems. In this study, the optimum arrangement and air flow of air circulation fans were investigated to save energy while minimizing temperature deviation at each point inside a plant factory using CFD. The condition for simulation was categorized into a total of 12 types according to installation location, quantity, and air flow changes in air circulation fans. Also, the variables of boundary conditions for simulation were set in the same level. The analysis results for each case showed that an average temperature of 296.33K matching with a set temperature and average air flow velocity of 0.51m/s suiting plant growth were well-maintained under Case 4 condition wherein two sets of air circulation fans were installed at the upper part of plant cultivation beds. Further, control of air circulation fan set under Case D yielded the most excellent results from Case D-3 conditions wherein air velocity at the outlet was adjusted to 2.9m/s.

In this study, the sludge that occurs in the initial operation of coagulation system developed for the treatment of CSOs were returned to the flocculation reactor. The purposes of this study were to analyze the Characteristics of flocs that are generated through the recycling sludge and settling characteristics of sludge, and to evaluate the possibility that high concentrations of particulate matter in the initial inflow of CSOs could be used as an weighted coagulant additive. As a result, the concentration of treated CSOs pollutants at the beginning of the CSOs influent with a large amount of particulate matter over 20 ${\mu}m$ was low, after gradually increasing the concentrations of them. The flocs generated from the sludge return were similar in size compared to flocs generated through injection of micro sands, and settling velocity in case of return sludge injection was decreased from 55.1 cm/min to 21.5 cm/min. SVI value of the sludge accumulated at the bottom of the sedimentation tank was 72, and settled sludge volume decreased rapidly due to the consolidation of sludge to the time it takes to 10 minutes. these mean that sludge used for recycling has good settling characteristic. A condition of returned sludge which is 0.1% return of 0.3% extraction was formed in the balance of settlement and extraction. In this case, This condition was to be adequate to maintain the proper concentration such as 100~200 mg/L of TS and 50~100 mg/L of VS in the flocculation reactor. The usage of the return sludge containing particulate matters of CSOs as an weighted coagulant additive was able to secure a stable treated water quality despite the change of influent water quality dynamically. Furthermore, it can be expected to reduce the alum dosage along with the sludge production.

In this study, granular sludge used in an anaerobic process treating brewery waste was inoculated in a laboratory scale of reactor to induce anaerobic ammonium oxidation(ANAMMOX). The reactor was operated with synthetic wastewater, which prepared at 1:1 ratio of $NH_4^+-N$ over $NO_2^--N$. Changes in nitrogen concentration, COD, alkalinity and gas production were analyzed. There are 3 phases of spanning in experimental period according to influent nitrogen concentration. In the Phase 1, each of the concentration of $NH_4^+-N$ and $NO_2^--N$ were increased from 1.91 $gN/m^3{\cdot}d$ to 14.29 $gN/m^3{\cdot}d$. Ammonium nitrogen loading(same as nitrite nitrogen) was 23.81 $gN/m^3{\cdot}d$ in the Phase 2 and 19.05 $gN/m^3{\cdot}d$ in the Phase 3, respectively $NO_2^--N$ has been removed up to 99% during whole period while the removal efficiency of $NH_4^+-N$ was significantly varied. In Phase 2, $NH_4^+-N$ was removed up to 75%. Microorganisms varied temporally through three phases were characterized by 16s rDNA analysis methods. ANAMMOX bacteria were dominantly found in phase 2 when the removal rate of $NO_2^--N$and $NH_4^+-N$ was the highest up to 99% and 75%, respectively. Due to erroneous exposed to air, the removal efficiency of $NH_4^+-N$ was unexpectedly lowered, but ANAMMOX bacteria still existed.

The performance of upflow anaerobic bioelectrochemical reactor (UABE), equipped with electrodes (anode and cathode) inside the upflow anaerobic reactor, was compared to that of upflow anaerobic sludge blanket (UASB) reactor for the treatment of acidic distillery wastewater. The UASB was stable in pH, alkalinity and VFAs until the organic loading rate (OLR) of 4.0 g COD/L.d, but it became unstable over 4.0 g COD/L.d. As a response to the abrupt doubling in OLR, the perturbation in the state variables for the UABE was smaller, compared to the UASB, and quickly recovered. The UABE stability was better than the UASB at higher OLR of 4.0-8.0 g COD/L.d, and the UABE showed better performance in specific methane production rate (2,076mL $CH_4/L.d$), methane content in biogas (66.8%), and COD removal efficiency (82.3%) at 8.0 g COD/L.d than the UASB. The maximum methane yield in UABE was about 407mL/g $COD_r$ at 4.0 g COD/L.d, which was considerably higher than about $282mL/g\;COD_r$ in UASB. The rate limiting step for the bioelectrochemical reaction in UABE was the oxidation of organic matter on the anode surface, and the electrode reactions were considerably affected by the pH at 8.0 g COD/L.d of high OLR. The maximum energy efficiency of UABE was 99.5%, at 4.0 g COD/L.d of OLR. The UABE can be an advanced high rate anaerobic process for the treatment of acidic distillery wastewater.
