Effect of Precultural and Nutritional Parameters on Compactin Production by Solid-State Fermentation

  • In the present study, production of compactin by Penicillium brevicompactum WA 2315 was studied. In the first step, various precultural parameters were studied by substituting one factor at a time. Subsequently, the effect of maltodextrin DE 18 on compactin production was studied. The optimized parameters gave maximum compactin production of 850 ${\mu}g/gds$as compared with 678 ${\mu}g/gds$before optimization. Statistical study was performed to further improve the production and develop a robust model. An improved yield of 950 ${\mu}g/gds$was obtained using the conditions proposed by the experimental model. The present study emphasizes the importauce of precultural and nutritional parameters on the production of compactin, and further confirms the usefulness of solid-state fermentation for the production of industrially important secondary metabolites. It also confirms that complex nitrogen sources such as oil cakes can be used for the production of compactin.

A Study on the Effect and Mechanism of Gamikyejakjimogawusul-tang Herbal Acupuncture on Induced Rheumatoid Arthritis model of DBA/1 mice (계작지모가우슬탕(桂芍知母加牛膝湯) 약침이 류마티스 관절염 생쥐에 미치는 영향)

  • Objectives : The purpose of this study is to prove the effect and mechanism of Gamikyejakjimogawusul-tang(GKHA) herbal acupuncture on induced rheumatoid arthritis model of DBA/1 mice. Methods : We check effect of GKHA extract on the AST, ALT, Creatinine, BUN of serum and cell viability of GK extract in RAW 264.7 cells to test the stability of this study. In vitro, we measure total phenol contents, total flavonoid contents, DPPH free radical scavenging activity, ABTS cation radical scavenging activity of Gamikyejakjimogawusul-tang, effect of GK extract on ROS(Reactive Ooxygen Species) production to estimate a anti-oxidant capacity, and we also measure effect of GK extract on NO (Nitric Oxid), IL-$1{\beta}$, IL-6, IL-17, IL-21, TNF-${\alpha}$, MCP-1, GM-CSF production in RAW 264.7 cells to estimate a anti-inflammatory efficacy. In vivo, we compare a rheumatoid arthritis manifestation between control and experimental group and estimate a AI. Then we check effect of GKHA on the level of WBC, neutrophil, lympocyte, monocyte in the blood to see the effect of immune cells in blood. In addition we measure effect of GKHA on the level of hs-CRP, IgM, IgG, IL-$1{\beta}$, IL-6, IL-17, IL-21, TNF-${\alpha}$, MCP-1, GM-CSF in serum. We observe effects of GKHA on imaging of cartilage degeneration using micro CT-arthrography in paw hind. And we calculate effects of GKHA that reduced BV ratio, BS/BV ratio using 3D Micro-CT. Lastly we observe effects of GKHA histopathologic examination analysis. Results : 1. The toxicity on liver and kidney was disregardable and the cytotoxicity against RAW 264.7 cells was also disregardable. 1. Total phenol contents and total flavonoid contents in GK extract were in high level. 2. DPPH free radical scavenging activity and ABTS cation radical scavenging activity were increased according to concentration of GK extract 3. ROS production was significantly decreased in GK extract (at 10, $100{\mu}g/ml$). 4. NO, IL-6, TNF-${\alpha}$, MCP-1 production were significantly decreased in GK extract(at 10, $100{\mu}g/ml$). IL-17, GM-CSF production were significantly decreased in GK extract(at 1, 10, $100{\mu}g/ml$). IL-$1{\beta}$, IL-21 production were also decreased but there was no statistical significance. 5. 25x observation after H&E and M-T staining, infiltration of immune cells and subsidence of the cartilage and damage to the synovial cells were decreased. Conclusions : This study showed that GKHA extract had anti-oxidant capacity, anti-inflammatory efficacy. GKHA extract also had inhibiting effect on the process of rheumatoid arthritis and can protect joint and cartilage. So we expect that GKHA extract can be a meaningful treatment to rheumatoid arthritis patients.

Mechanism of Palmul- Tang on the Change of Cerebral Hemodynamics in Rats (팔물탕이 뇌혈유력학 변화에 미치는 작용기전)

  • This experimental study was designed to investigate the mechanism of Palmul-Tang(PMT) on the changes of cerebral hemodynamics in rats. The changes of cerebral hemodynamics in normal rats were as follows ; The PMT-induced increase in regional cerebral blood flow was significantly inhibited by pretreatment with indomethacin(1㎎/㎏, i.p.), an inhibitor of cyclooxygenase, and was inhibited by methylene blue(10㎍/㎏, i.p.), an inhibitor of guanylate cyclase. The PMT-induced dilation in pial arterial diameter was significantly inhibited by pretreatment with indomethacin and methylene blue. The PMT-induced increase in mean arterial blood pressure was significantly inhibited by pretreatment with indomethacin but was increased by methylene blue. This results were suggested that the mechanism of PMT was mediated by cyclooxygenase. The changes of cytokine production in cerebral ischemic rats were as follows ; In cytokine production of serum by drawing from femoral arterial blood after middle cerebral arterial occlusion 1hr, sample group was decreased IL-1β and TNF-α production compared with control group, IL-10 production of sample group was similar to that of control group, but sample group was significantly increased TGF-β production compared with control group. In cytokine production of serum by drawing from femoral arterial blood after reperfusion 1hr, sample group was significantly decreased IL-1β production compared with control group and decreased TNF-α production compared with control group. IL-10 production of sample group was similar to that of control group, but sample group was significantly increased TGF-β production compared with control group. In cytokine production of serum by drawing from femoral arterial blood after reperfusion 4 hrs, sample group was significantly decreased IL-1β production compared with control group, but IL-10 production of sample group was similar to that of control group. sample group was increased TNF-α and TGF-β production compared with control group. These results suggested that PMT had inhibitive effect on the brain damage by inhibiting IL-1β and TNF-α production, but by accelerating TGF-β production. The present author thought that PMT had an anti-ischemic effect through the improvement of cerebral hemodynamics and inhibitive effect on the brain damage.

Suppression Effect of Curcuma longa Rhizome-Derived Components against Nitric Oxide Synthase

  • The inhibitory effects of Curcuma longa rhizome-derived materials against nitric oxide (NO) production were assessed. The inhibitory effect (57%) on NO production was evidenced by the methanol extract of C. longa at $1\;{\mu}g/mL$. In the fractionation of the methanol extract, the ethyl acetate fraction evidenced an inhibitory effect greater than 62.1% at $1\;{\mu}g/mL$. The active constituent was identified as curcumin. Curcumin exerted potent inhibitory effects of 78.7 and 65.7% at concentrations of 1 and $0.5\;{\mu}g/mL$, respectively. Furthermore, the inhibitory effect of ar-turmerone was measured as 31.3 and 15.8% at 1 and $0.5\;{\mu}g/mL$, respectively. The iNOS expression-suppressive effects of curcumin were assessed via western blot analysis. Our results suggest that curcumin and ar-turmerone may prove useful in the development of new types of NO inhibitors.

Effect of Itaconate on Acid production of Saccharomycopsis lipolytica (Saccharomycopsis lipolytica의 산 생산에 미치는 Itaconate의 영향)

  • Effect of itaconate upon citrate and isocitrate production of Saccharomycopsis lipolytica were investigated. The percent inhibition of isocitrate lyase activity was about 80% at 0.8 mM itaconate concentration, with Ki value of 0.17 mM in the cleavage reaction. Inhibitory effect of itaconate at 20 mM on S. lipolytica growth was significant on n-hexadecane medium, whereas almost no inhibitory effect on glucose medium. Increasing itaconate concentration in n-hexadecane medium improved isocitrate production up to 80% but no difference was found in glucose medium.

Analysis of Marginal Productivity and Return to Scale Using Estimation of Production Function in Offshore Fisheries (근해어업 생산함수 추정을 이용한 규모수익 및 한계생산성 분석)

  • The production of Korean offshore fisheries has been gradually decreasing due to the severe depletion of offshore fisheries resources caused by excessive fishing efforts. The production of the offshore fisheries in 2016 was the lowest since 1975. So the federal and local governments in Korea adopted and implemented various fisheries management plans and policies in order to restore fisheries resources. However, these plans and polices have not been successful in re-establishing fisheries resources. Thus, in order to accurately diagnose the situation with regard to offshore fisheries, this study sought to estimate not only the return to scale by fishing gear of offshore fisheries, but marginal productivity of individual fishing gear based on production factors derived from offshore fisheries production functions. The study was organized in the following manner. First of all, this study estimates production functions of offshore fisheries. The Cobb-Douglas and the translog production functions are adopted as offshore fisheries production functions. Specifically, the functions are estimated by crew, vessels, and offshore resource as production factors. The offshore resource is estimated by the Clarke Yoshimoto Pooley model based on the surplus production model. Secondly, the fisheries production functions are extended to the fixed-effect model and the random-effect model with panel data. Thirdly, this study analyzes the return to scale of offshore fisheries and the marginal productivity of the production factors from the estimated offshore fisheries production function. In conclusion, this study suggests plans and countermeasures for productivity improvement by group (labor intensive or technology intensive) based on the characteristics of individual offshore fishing gear.

Training Effect on the Perception and Production of English Grapheme by Korean Learners of English (한국 학생들의 영어 철자 인지와 발화에 대한 훈련효과)

  • Given that English grapheme is realized as five different American English vowels [ʌ, ju, ʊ, u, ə], the purpose of the current study is to examine Korean learners' perception and production of English grapheme and training effect on words with . Thus, the current study conducted pretest, training, and posttest for 31 Korean university students on 24 English words with . The overall results showed that the participants' perception and production accuracy was significantly improved in the posttest, thus indicating training effect on both perception and production. However, it was not the case that all five different vowels demonstrated training effect. In perception the accuracy rates of [ʌ], [ju], and [ə] were improved after training whereas those of [ʊ] and [u] were not. In production [ʌ], [ʊ], and [u] did not show training effect. These results indicate that the Korean participants had difficulty distinguishing between tense [u] and lax [ʊ] both in perception and production. In particular, the Korean participants tended to replace lax [ʊ] with tense [u] in production. This is because tense [u] is the best match to Korean [u] in acoustic measurements, so that tense [u] is easy for the Korean participants to pronounce than lax [ʊ]. Also, English [ʌ] tended to be mispronounced as [u]-quality vowels such as [u] and [ju], which is due to the spelling . The Korean participants also showed errors which insert [j] after alveolars [t, d, n, s], which runs against yod-dropping in American English. They also deleted [j] after labials and velars, which is due to the absence of orthography in the target words. Finally, pedagogical implications were discussed based on the findings of the current study.

A Cross-Language Comparison of Speaking Rate Effects on the Production and Perception of English Word-final Stops

  • The primary goal of this study is to find out how the effect of speaking rate has some influence on the production and perception across languages. Through both experiments of production and perception, an English native speaker changes both production and perception simultaneously. Especially the production of the temporal features changes relatively fast. On the contrary, Chinese and Korean speakers changes their production rather than perception by following the speaking rate.

Association of Beta-lactoglobulin Polymorphism with Milk Production Traits in Cattle

  • The study was carried out in Sahiwal, Holstein Friesian, Jersey and crossbred cattle to find out the effect of genotype of beta-lactoglobulin gene on milk production traits. The polymorphism at beta-lactoglobulin gene was identified by conducting PCRRFLP studies. A 398 bp fragment of the gene was amplified and digested with Hae III restriction enzyme. The two alleles A and B and three genotypes AA, AB and BB were identified in all cattle breeds. The frequency of B allele was comparatively higher than that of A allele. The AA genotype produced significantly higher milk yield in Sahiwal cattle whereas BB genotype yielded higher milk in Holstein friesian cattle. In other cattle breeds the genotypic effect was non-significant. In conclusion it may be stated that the genotype with significantly higher milk yield may be favoured in the farm along with other conventional selection criteria to enhance the milk production of animals.

Contributions of Public Investment to Economic Growth and Productivity

  • Whereas a large variety of previous studies show mixed results regarding the relationship between public investment and economic outcome, several studies have been conducted on related issues in Korea. The present study deals with the effect of public investment in Korea on economic growth and productivity. Using administrative data, it exploits three different methodologies: the total factor productivity approach, production function approach, and stochastic frontier production function approach. The results of this study show that public investment has a statistically significant effect on economic growth. However, it contributes little to enhance productivity. It is explained that there exists inefficiency of production in the Korean economy. These findings indicate that public investment has played a central role in the direct input factor and not in indirect role in Korea. Thus, it is necessary for public investment policies to concentrate on enhancing the efficiency of the Korean economy.
