Response of broilers to supplementation of branched-chain amino acids blends with different valine contents in the starter period under summer conditions

  • Objective: The objectives of this study were to compare the effects of normal and low protein content (PC) of starter diet supplemented or not with blends of branched-chain amino acids (BCAAs) on growth performance of broilers under summer conditions and to investigate whether these effects altered some quality traits and the characteristics of gastrointestinal tract. Methods: A total of 768 mixed-sex broiler chicks (Ross 308, one-d-old) with an average initial body weight (BW) of 47.6±1.03 g were allocated into six treatments with four replications in 2×3 factorial arrangement. Factors were: PC, normal (N, 22% to d 15); and low (L, 20% to d 15); and added BCAA blends, L-leucine, L-isoleucine, and L-valine at zero (0L:0I:0V); 1.0, 0.25, and 0.25 (4L:1I:1V); or 1.0, 0.25, 0.75 (4L:1I:3V) g/kg of diet. Hence, six dietary treatments were named as N0L:0I:0V, N4L:1I:1V, N4L:1I:3V, L0L:0I:0V, L4L:1I:1V, and L4L:1I:3V. Average indoor temperature and humidity were 32.8℃±1.7℃ and 61.1%±4.12%, respectively. Results: BW, feed conversion ratio (FCR) and carcass weight were not affected by PC, BCCA and their interaction (p>0.05). The L diets decreased the water holding capacity of the breast (p = 0.002) and thigh (p = 0.050) meats and dressing percentage (p = 0.005) compared to the N diets. The 4L:1I:1V diet decreased breast yield compared to the 0L:0I:0V diets (p = 0.041). The effect of PC on feed intake, mortality and gastrointestinal trait weight were depended on the L:I:V ratios under summer conditions due to interactions between factors (p<0.05). The FI and mortality of L4L:1I:1V broilers were lower than those of N4L:1I:1V birds (p<0.05). Conclusion: It was concluded that the blends of BCAAs used failed to improve performance and to promote breast yields, because diets with normal or with reduced protein supplemented or not with BCAAs up to d 15 produced a similar BW and FCR in broilers raised in hot-climate conditions.

현사시나무 원형질체에서 리보핵산단백질을 활용한 유전자 교정 방법 연구 (Genome editing of hybrid poplar (Populus alba × P. glandulosa) protoplasts using Cas9/gRNA ribonucleoprotein)

  • CRISPR/Cas9에 의한 유전자 교정 기술은 유용 형질을 갖는 작물 및 임목의 육성에 있어 널리 사용되고 있는 핵심 기술이다. 유전자 교정 임목 육성에는 아그로박테리움에 의한 형질전환 방법이 높은 효율로 시행된 연구가 많았고 따라서 형질전환에 사용된 플라스미드 서열이 식물 유전체 안에 존재한다는 문제가 남아 있었다. 본 연구에서는 CRISPR/Cas9을 사용하여 유전자 교정 임목을 육성하는 데 기존에 알려진 벡터 도입 기술이 아닌, 단일 가닥 가이드 RNA (sgRNA)와 Cas9 단백질을 혼합하여 만든 리보핵산단백질을 현사시나무 원형질체에 도입하는 방법을 기술하였다. 염 스트레스 내성 관련 인자 PagSAP1 유전자를 표적으로 하는 3종류의 sgRNA를 디자인하고, 각 sgRNA와 Cas9 단백질을 혼합하여 만든 리보핵산단백질을 원형질체에 도입하였다. 표적화 딥시퀀싱을 통해 리보핵산단백질 형성 시 sgRNA와 Cas9 단백질을 혼합하고 일정 시간 배양하여 안정화되는 시간이 필요한 것을 확인하였다. 또한 sgRNA3의 리보핵산단백질이 sgRNA1, sgRNA2의 리보핵산단백질보다 높은 교정 효율을 보이는 것을 확인하였다. 본 실험을 통해 리보핵산단백질을 이용한 유전자 교정 기술이 임목에도 적용될 수 있음이 확인되었고, 이는 외래 유전자 없이 유전자 교정 임목을 육성하는 데 활용할 수 있을 것으로 사료된다.

습지 전문종인 제비동자꽃(Lychnis wilfordii (Regel) Maxim.) 개체군의 내적동태 (Internal Dynamics of Wetland Specialist, Population of Lychnis wilfordii (Regel) Maxim.)

  • 제비동자꽃(Lychnis wilfordii (Regel) Maxim.)은 주로 고위도의 이탄습지에 분포하는 전문종이다. 우리나라에서는 2개 지역에 고립되어 분포한다. 본 연구에서는 생육지 특성, 생장특성 및 자가화합성 그리고 안정적인 개체군을 유지하고 있는 용늪습지보호지역에서 개체군의 지속에 관여하는 내적 동태를 평가하였다. 분포지의 식생환경은 용늪습지보호지역과 평창군 대관령면 분포지 사이에 뚜렷한 차이가 있었다. 화분매개충의 방문에 의해 종자의 생산이 촉진되기는 하지만 자가화합성을 함께 소유하였다. 토양의 유기물함량이 높은 조건에서 다수의 겨울눈을 생성하였고, 줄기 수, 열매 수가 최대에 달하였다. 그렇지만 유기물함량이 낮은 조건에서도 생장하고 개화하여 종자를 생산하였다. 용늪습지보호지역에서 제비동자꽃은 고층습원에는 분포하지 않았고 뚝사초가 형성하는 사초기둥이 발달한 저층습원에 분포하였다. 제비동자꽃은 이 공간에서도 사초기둥의 상단부에 주로 분포하였다. 따라서 뚝사초가 형성하는 사초기둥의 생성, 성장 및 소멸은 이 공간에 분포하는 식물의 정착, 성장, 소멸과 밀접한 연관이 있는 것으로 판단되었다. 현재 용늪습지보호지역은 제비동자꽃이 분포하는 사초기둥이 발달한 공간이 넓게 분포하고 있고 소멸과 재정착의 과정이 잘 이루어지는 것으로 평가되었다. 그러므로 현재와 같은 생태적 과정이 잘 유지된다면 용늪습지보호지역의 제비동자꽃 개체군은 오랜 기간에 걸쳐 지속 가능할 것으로 예상되었다.

Effect of multi-enzymes supplementation on growth performance, meat quality, ileal digestibility, digestive enzyme activity and caecal microbiota in broilers fed low-metabolizable energy diet

  • Objective: This study was conducted to evaluate the effect of using low energy diet with multi-enzymes supplementation on different biological parameters in broilers. Methods: Three hundred Arbor Acres broiler chicks were randomly divided into three groups (Cont, standard metabolizable energy(ME); L-ME, ME reduced by 50 kcal/kg without enzyme; and L-ME-MES, L-ME diet was supplemented with multi-enzymes) with five replicates per group (20 chicks per replicate) at the start of second week. Grower and finisher diets were formulated according to breed specific guide and offered with free access in respective phase (two weeks for grower [8 to 21 d]; two weeks for finisher [22 to 35 d]). External marker method was used to measure the nutrient digestibility. After feeding trial, fifteen birds (one bird per replicate) were selected randomly and slaughtered for samples collection. Results: The results exhibited no effect (p>0.05) of dietary treatments on all parameters of growth performance, carcass traits, relative weight of internal organs except bursa and overall parameters of thigh meat quality. Relative weight of bursa was significantly (p<0.05) higher in L-ME than control. Multi-enzymes supplementation in low-ME diet significantly (p<0.05) improved the breast meat pH 24 h, digestibility of crude protein, duodenum weight and length, jejunal morphology, counts of Lactobacillus spp. and Bifidobacterium spp., lipase and protease activities than control. Jejunum length was increased in both L-ME and L-ME-MES treatments than that of the control (p<0.05). Breast meat cooking loss and color lightness was lower in L-ME (p<0.05) than control. Conclusion: It can therefore be concluded that broilers could be reared on low energy diet with supplementation of multi-enzymes without compromising the growth performance. In addition, it is beneficial for other biological parameters of broilers.

Half-castration is a newly effective method for increasing yield and tenderness of male cattle meat

  • Objective: For improving meat quality especially tenderness, male cattle are usually castrated to removes both the testicles. This study was conducted to evaluate the effect castration method (half- and complete-castration) on meat yield and quality characteristics of Hanwoo male cattle. Methods: Thirty-two similar age (5.9 months) Hanwoo male calves were divided into: half-castration (HC) and complete-castration (CC) groups (n = 16 per group). At 7 months of age, all the animals were castrated in which the HC calves had only one testicle surgically removed while, the CC calves had both testicles surgically removed. The castrated animals were reared under identical conditions until 25 months of age. After slaughter, the carcasses were evaluated for carcass traits and meat yield of primal cuts. For examination of the castration effect on meat quality, L. lumborum and semimembranosus muscles were used. The meat samples were analyzed for chemical composition, color, pH, shear force and water holding capacity, fatty acids, metabolites and volatile aroma compounds. Results: The HC group showed higher meat yields of all primal cuts (p<0.05). As a result, the total meat yield was higher by approximately 44 kg in the HC group (303.32 kg, corresponding to 67.88%) compared to the CC group (259.30 kg, corresponding to 62.11%) (p<0.05). In terms of meat quality, the HC resulted in two times greater fat content in both muscles examined compared to intact males. More importantly, the shear force values did not differ between HC and CC groups for L. lumborum muscles (p>0.05). The meat from HC animals exhibited higher amount of free amino acids associated with sweetness (p<0.05). Furthermore, the castration method only exhibited a negligible effect on metabolites and volatile aroma compounds in the cooked meat. Conclusion: Half-castration emerged as an alternative practice to be used for increasing the yield and tenderness of male cattle meat.

흰색 팽이 신품종 '설한' 육성 (Breeding of a new cultivar of White Flammulina velutipes, 'Seolhan')

  • 팽이(Flammulina velutipes)는 주름버섯목(Agaricales), 뽕나무버섯과(Physalacriaceae)에 속하는 담자균류로서, 국내 생산량은 연간 31,818톤('19)으로 전체 생산량의 약 18%이며 버섯 전체 수출액의 약 36%를 차지할 정도로 대표적인 식용버섯이다. 그러나 국내 재배되고 있는 팽이 국산품종의 비중이 31.6%(20')로 매우 낮으므로 국제 경쟁력이 우수한 국내육성 품종확보가 필요하며 수출 버섯으로서 보다 품질이 우수한 품종 육성이 필요하다. 이에 본 연구는 농진청에서 육성한 '백승'과 '우리1호'의 단핵균주를 확보하여 mon-mon교잡을 통해 '설한' 품종을 육성하였다. 균사배양 특성은 25℃에서 균사생장이 가장 좋았으며 20, 25℃에서 대조품종보다 균사 생장속도가 빨랐다. 병재배 시 배양 최적온도는 15~17℃로 배지온도가 20℃이상 되지 않게 관리해야하며 생육온도는 7~8℃로 대조품종에 비해 다소 높다. 또한 자실체 수량은 병당(850 mL) 226.5±8.0 g으로 대조품종에 비해 18.4% 증수하였으며 초발이일수가 2일 빨라 전체적인 생육기간이 단축되었다. 또한 설한의 외형은 대조품종보다 갓이 작고 대는 굵었으며 갓 색이 더 밝았다. 대치배양과 유전적 다형성 검정 결과, 설한이 다른 흰색 품종들과 유전적으로 구별되는 것을 확인하였다.

양식산업에 발전을 위한 유전체 분석 기술 적용 (The Application of Genome Research to Development of Aquaculture)

  • 수산업은 수산물의 남획, 국가 간의 제한, 기후변화로 인하여 수산물 개체 수 감소 등으로 전 세계 수산물 생산량이 정체되면서, 양식업은 항상 세계 식량 소비를 위해 오랫동안 지속되어온 패러다임을 뒤집고 인류가 바다, 호수, 강의 풍부함을 새로운 방식으로 확장할 수 있도록 하는 '푸른 혁명'의 가능성을 제시하고 있다. 양식산업은 이제 인간 소비를 위한 해산물의 원료로서 해양에서 얻는 어업생산량의 절반이상을 넘어섰다. 지속 가능한 양식산업의 발전을 위하여 다양한 최신 생물학적 연구 방법들이 적용되고 있다. 유전체학은 2011년 대서양 대구의 염기서열이 규명된 이래 최근 상당한 진전을 이루었으며 더 많은 종의 유전체가 해독되어 우수 형질의 육종 및 번식 기술, 질병 저항성 품질 개량, 양식 사료 및 사료방식의 최적화 등 양식산업을 위한 보다 견고하고 생산적인 지식 기반을 제공한다. 본 리뷰는 수산양식종의 유전체 연구를 위한 유전체 분석 기술과 수산양식종의 유전체 연구의 현황에 대해서 살펴보았다. 이와 같이 유전체 분석 기술과 유전체학의 발전은 수산양식산업의 과제를 해결하는 데 중요하며 지속 가능한 어업과 양식에 도움을 줄 것이다.

Probiotics Increase Intramuscular Fat and Improve the Composition of Fatty Acids in Sunit Sheep through the Adenosine 5'-Monophosphate-Activated Protein Kinase (AMPK) Signaling Pathway

  • This experiment aims to investigate the impact of probiotic feed on growth performance, carcass traits, plasma lipid biochemical parameters, intramuscular fat and triglyceride content, fatty acid composition, mRNA expression levels of genes related to lipid metabolism, and the activity of the enzyme in Sunit sheep. In this experiment, 12 of 96 randomly selected Sunit sheep were assigned to receive the basic diet or the basic diet supplemented with probiotics. The results showed that supplementation with probiotics significantly increased the loin eye area, and decreased plasma triglycerides and free fatty acids, increasing the content of intramuscular fat and triglycerides in the muscle and improving the composition of the fatty acids. The inclusion of probiotics in the diet reduced the expression of adenosine 5'-monophosphate-activated protein kinase alpha 2 (AMPKα2) mRNA and carnitine palmitoyltransferase 1B (CPT1B) mRNA, while increasing the expression of acetyl-CoA carboxylase alpha (ACCα) mRNA, sterol regulatory element-binding protein-1c (SREBP-1c) mRNA, fatty acid synthase mRNA, and stearoyl-CoA desaturase 1 mRNA. The results of this study indicate that supplementation with probiotics can regulate fat deposition and improves the composition of fatty acids in Sunit sheep through the signaling pathways AMPK-ACC-CPT1B and AMPK-SREBP-1c. This regulatory mechanism leads to an increase in intramuscular fat content, a restructuring of muscle composition of the fatty acids, and an enhancement of the nutritional value of meat. These findings contribute to a better understanding of the food science of animal resources and provide valuable references for the production of meat of higher nutritional value.

Effects of diet and castration on fatty acid composition and volatile compounds in the meat of Korean native black goats

  • Objective: This study determined the effects of dietary treatments and castration on meat quality, fatty acids (FAs) profiles, and volatile compounds in Korean native black goats (KNBG, Capra hircus coreanae), including the relationship between the population of rumen microbiomes and meat FA profiles. Methods: Twenty-four KNBG (48.6±1.4 kg) were randomly allocated to one of four treatments arranged into a 2×2 factorial structure. The factors were dietary forage to concentrate ratio (high forage [HF, 80:20] and low forage [LF, 20:80]), and a castration treatment (castration [CA] vs non-castration [NCA]). Results: Among meat quality traits, the CA group exhibited a higher percentage of crude fat and water holding capacity (p<0.05). The profiles of the saturated fatty acid (SFA) in meat sample derived from CA KNBG showed a significantly lower percentage compared to NCA individuals, due to the lower proportion of C14:0 and C18:0. Feeding a high-forage diet to KNBG increased the formation of C18:1n7, C18:3n3, C20:1n9, C22:4n6 in meat, and polyunsaturated fatty acid (PUFA) profiles (p<0.05). Consequently, the n6:n3 ratio declined (p<0.05). There was an interaction between dietary treatment and castration for formation of C20:5n3 (p<0.05), while C18:1n9, C22:6n3, monounsaturated fatty acid (MUFA) and the MUFA:SFA ratio were influenced by both diet and castration (p<0.05). Nine volatile compounds were identified and were strongly influenced by both dietary treatments, castration (p<0.05), and their interaction. In addition, principal component analysis (PCA) revealed distinctly different odor patterns in the NCA goats fed LF diets. Spearman correlation analysis showed a high correlation between rumen bacteria and meat PUFAs. Conclusion: These results suggest the essential effects of the rumen microbial population for the synthesis of meat FAs and volatile compounds in KNBG meat, where dietary intake and castration also contribute substantially.

Effects of β-glucan with vitamin E supplementation on the growth performance, blood profiles, immune response, pork quality, pork flavor, and economic benefit in growing and finishing pigs

  • Objective: This study was conducted to evaluate the effects of β-glucan with vitamin E supplementation on the growth performance, blood profiles, immune response, pork quality, pork flavor, and economic benefit in growing and finishing pigs. Methods: A total of 140 growing pigs ([Yorkshire×Landrace]×Duroc) were assigned to five treatments considering sex and initial body weight (BW) in 4 replications with 7 pigs per pen in a randomized complete block design. The experimental diets included a corn-soybean meal-based basal diet with or without 0.05% or 0.1% β-glucan and 0.02% vitamin E. The pigs were fed the diets for 12 weeks (phase I, 0 to 3; phase II, 3 to 6; phase III, 6 to 9; phase IV, 9 to 12). The BW and feed intake were measured at the end of each phase. Blood samples were collected at the end of each phase. Four pigs from each treatment were selected and slaughtered for meat quality. Economic benefit was calculated considering the total feed intake and feed price. Pork flavor was analyzed through inosine monophosphate analysis. Results: The average daily gain and feed efficiency were improved compared to the control when β-glucan or vitamin E was added. Supplementing 0.05% β-glucan significantly increased the lymphocyte concentration compared to the addition of 0.1% β-glucan and the content of vitamin E in the blood increased when 0.02% vitamin E was added. The treatment with 0.1% β-glucan and 0.02% vitamin E showed the most economic effect because it had the shortest days to market weight and the lowest total feed cost. The addition of β-glucan or vitamin E had a positive role in improving the flavor of pork when considering that the content of inosine monophosphate was increased. However, carcass traits and meat quality were not affected by β-glucan or vitamin E. Conclusion: The addition of 0.1% β-glucan with 0.02% vitamin E in growing and finishing pig diets showed great growth performance and economic effects by supplying vitamin E efficiently and by improving the health condition of pigs due to β-glucan.
