효소종류에 따른 불등풀가사리 유래 다당류의 이화학적 특성 및 생리활성 (Biological activities and physicochemical properties of polysaccharides from Gloiopeltis furcata prepared by using various enzymes)

  • 본 연구에서는 불등풀가사리 유래 다당류의 기능성식품소재로의 활용성을 향상시키고자 불등풀가사리에 5종의 상업용 효소를 처리한 다음 분리된 다당류의 이화학적 특성 및 생리활성을 조사하였다. 효소 분해 구간의 다당 수율은 52.8-66.4%로 무처리 구간 50.6%에 비해 유의적으로 증가하였다. 이화학적 특성으로 총당 및 단백질 함량은 각각 71.04% 및 7.22%, uronic acid 및 sulfate 함량은 23.18 g/100 g 및 28.27%로 효소 분해를 통해 증가하였다. DPPH radical 소거활성 및 FRAP에 의한 항산화 활성은 23.10% 및 $218.50{\mu}M$을 나타내어 무처리 구간에 비해 항산화 활성이 우수하였으며, L132 세포 사멸에 대한 보호효과는 viscozyme 효소처리 구간($1{\mu}g/mL$)에서 $H_2O_2$를 처리한 구간 대비 세포 보호효과는 85.64%로 세포 활성이 증가하여 높은 세포 보호효과를 나타내었다. NO 생산량은 viscozyme 효소 처리구간 $5{\mu}g/mL$ 농도에서 $32.13{\mu}M$ 함량을 나타내어 LPS 대비 90% 높은 생성량을 나타내었으며, 4종의 암세포(A549, SNU719, HeLa 및 MCF7) 생존율은 $25{\mu}g/mL$농도에서 각각 69.57%, 61.06%, 52.74% 및 68.64%의 유의적으로 낮은 암세포 생존율을 나타내었다. 따라서 불등풀가사리의 효소 분해를 통해 다당의 이화학적 특성 및 생리활성이 향상됨에 따라 기능성 식품소재 로 다양하게 활용 가능할 것으로 사료된다.

열처리 방법에 따른 가공 인삼 열수추출물의 이화학적 품질 특성 (Physicochemical quality characteristics of hot water extracts of processed ginseng based on different heat treatments)

  • 본 연구는 고온 고압의 autoclaving 조건에서 2시간 내외로 증삼한 인삼과 그 용출액으로 2단계 열풍건조법으로 제조한 홍삼을 분말로 하여 그 열수추출물에 대한 이화학적 특성을 조사하였다. 즉 시료로 사용한 홍삼의 제조는 고온 고압으로 증삼한 용출액을 건조시 증삼 표면에 바르고 건조하여 외형이 흑삼 모양으로 변하였다. 홍삼 분말로부터 열수추출물의 총 폴리페놀, 총 플라보노이드, 총당, 산성다당체, 조사포닌 등의 함량을 분석하고, 색차계와 전자코를 이용하여 향미 성분을 측정하였다. Autoclave를 이용한 홍삼 열수추출물(ARG)의 총 폴리페놀 함량은 9.06 mg GAE/g, 총 플라보노이드 3.38 mg NE/g, 총당 35.22 g/100 g, 산성다당체 10.90 g/100 g, 조사포닌 10.22 g/100 g으로 나타났다. 본 홍삼제조법에 의해 얻어진 홍삼을 이용한 열수추출물은 총 폴리페놀, 총 플라보노이드, 조사포닌 등의 최종 함량이 다른 홍삼 제조법에 비해 높게 나타났으며, 고온 고압으로 단시간에 얻은 증삼액인 용출액은 진세노사이드 조성이 다양하였다. Rb1을 포함한 인삼의 주요 진세노사이드 총 함량은 ARG에서 가장 낮은 10.69 mg/g으로 나타났으며 가공 조건이 홍삼 특유의 진세노사이드로의 전환에 영향을 주었다. Autoclave로 얻어진 홍삼 및 용출액은 홍삼성분의 저분자량으로의 변환을 통해 색의 강화 등 향미성분이 개선되어 식품 뿐만 아니라 비식품의 소재 및 제품을 위한 수요가 기대된다.

누에 실샘 가수분해물의 제조 및 특성 규명 (Manufacture and Characterization of Silkworm Gland Hydrolysate)

  • 본 연구에서는 인체에 유해한 용매를 사용하지 않고 누에 실샘으로부터 가수분해물을 제조하는 새로운 공정을 개발하였다. 제조된 누에 실샘 가수분해물의 특성 분석을 통해 누에고치 유래 세리신 가수분해물(CSH)과 구성 성분 및 물성이 상이함을 확인하였다. 또한 1 mg/ml 수용성 누에 실샘 가수분해물(SSGH)을 첨가한 경우 10% FBS를 첨가한 대조군과 동등한 세포 증식효과가 있음을 증명하였다. 결론적으로 누에 실샘 가수분해물은 혈청을 대체할 수 있는 우수한 소재임을 확인하였다. 본 연구에서 개발한 제조방법을 통해 생산된 누에 실샘 가수분해물은 향후 화장품 및 의료용 소재로 그 응용범위를 확대할 수 있을 것으로 사료된다.

Schisandrae Fructus ethanol extract attenuates particulate matter 2.5-induced inflammatory and oxidative responses by blocking the activation of the ROS-dependent NF-κB signaling pathway

  • BACKGROUND/OBJECTIVES: Schisandrae Fructus, the fruit of Schisandra chinensis Baill., has traditionally been used as a medicinal herb for the treatment of various diseases, and has proven its various pharmacological effects, including anti-inflammatory and antioxidant activities. In this study, we investigated the inhibitory effect of Schisandrae Fructus ethanol extract (SF) on inflammatory and oxidative stress in particulate matter 2.5 (PM2.5)-treated RAW 264.7 macrophages. MATERIALS/METHODS: To investigate the anti-inflammatory and antioxidant effects of SF in PM2.5-stimulated RAW 264.7 cells, the levels of pro-inflammatory mediator such as nitric oxide (NO) and prostaglandin E2 (PGE2), cytokines including interleukin (IL)-6 and IL-1β, and reactive oxygen species (ROS) were measured. To elucidate the mechanism underlying the effect of SF, the expression of genes involved in the generation of inflammatory factors was also investigated. We further evaluated the anti-inflammatory and antioxidant efficacy of SF against PM2.5 in the zebrafish model. RESULTS: The results indicated that SF treatment significantly inhibited the PM2.5-induced release of NO and PGE2, which was associated with decreased inducible NO synthase and cyclooxygenase-2 expression. SF also attenuated the PM2.5-induced expression of IL-6 and IL-1β, reducing their extracellular secretion. Moreover, SF suppressed the PM2.5-mediated translocation of nuclear factor-kappa B (NF-κB) from the cytosol into nuclei and the degradation of inhibitor IκB-α, indicating that SF exhibited anti-inflammatory effects by inhibiting the NF-κB signaling pathway. In addition, SF abolished PM2.5-induced generation of ROS, similar to the pretreatment of a ROS scavenger, but not by an inhibitor of NF-κB activity. Furthermore, SF showed strong protective effects against NO and ROS production in PM2.5-treated zebrafish larvae. CONCLUSIONS: Our findings suggest that SF exerts anti-inflammatory and antioxidant effects against PM2.5 through ROS-dependent down-regulating the NF-κB signaling pathway, and that SF can be a potential functional substance to prevent PM2.5-mediated inflammatory and oxidative damage.

캘러스 활용도를 향상시키기 위한 벼(Oryza sativa L.) 형질전환 시스템 구축 (Development of rice(Oryza sativa L.) transformation system to improve callus utilization)

  • 분자농업은 식물을 일종의 공장개념으로 확대 적용하여 산업적으로 가치가 높은 유용물질들을 대량생산하는 분야로 최근 많은 각광을 받고 있다. 그 중에서 벼 캘러스를 이용한 단백질 발현 시스템은 대량 배양이 가능하고 목적 단백질의 높은 발현율로 인한 산업화가 가능한 기술이다. 본 연구는 이러한 벼(Oryza sativa L.) 캘러스의 활용도를 높이기 위한 효율적인 형질전환 시스템을 구축하기 위해 수행되었다. 형질전환에 이용된 종자의 종피 제거 시 손을 이용함으로써 캘러스 유도율을 높여주었고, 6년 정도의 오래된 종자에서도 원활한 캘러스 유도가 가능하였다. 목적 유전자가 도입된 캘러스의 선발은 최소한 3주 이상의 배양기간을 필요로 하였고 가장 효율적인 것은 한번의 계대를 포함한 6주 배양 후 선발하는 것이었다. 이러한 선발은 유전자가 식물세포의 genomic DNA 안에 안정적으로 삽입되어 이루어졌음을 서던 블롯 분석 및 후대검정 등을 통하여 확인할 수 있었다. 이러한 장기적이고 대량의 배양이 가능한 벼 캘러스의 효율적인 선발 시스템은 벼를 이용한 유용 재조합 단백질의 산업화에 실속있는 기술로 기여할 수 있을 것이다.

<이춘풍 난봉기>와 <열하일기만보>를 통해 본 손진책의 '마당미학' (Sohn Jin-Chaek's 'Madang' Aesthetics in Playboy Lee Chunpoongjeun and Yulha Ilgyee Manbo)

  • Son Jinchaek got into his directing career since 1976 when he founded the theatre group Minye with Huhgyu and others. His experience in Minye was the turnaround of his life; Huhgyu was a teacher in his artist's life whereas Brecht was a teacher for his ideology to make 'Korean Theatre'. From these two teachers, he learned how and why Korean Theatre should be made. Since then, Korean theatre was his calling for 40 years of his directing career. As he established Michoo in 1986, it served a turning point in his art. His focus was on intrinsic attributes of Korean Theatre with Madang Jungshin. With Madang Jungshin, he tried to get over the former generation's fixation on external materials of Korean traditional theatre and folk culture to make Korean Theatre. Rather, he believed Korean Theatre could be realized when it grafted Korean social reality onto the stage, while the form was subsequent. He advocated Korean Theatre should mirror present social reality and circulate social energy. Also, he did not give up aesthetics. On the contrary, his aesthetic style was conspicuously evident in his productions. In spite of his life long career with noteworthy works, the critical discourses are strikingly scarce especially when compared with other senior and peer directors such as Hugh Gyu, Ahn Minsoo, Oh Taesok, and Lee Yoontaek. During his career he has crossed into various genres from Changeuk, Madangnori, and to theatres, which were too versatile to thread them into a discourse and caused a lack of theoretical greeting. Madangnori has anchored its artistic structure on its polished aesthetics which were acclaimed by the general audience for 30 years. For theatre, he concentrated on one production per year to grasp its own style. Theatre works also had revealed his own style of being opened and of being emptied which was certainly different from Madangnori, but had same aesthetic principle within it. This paper attempts to recompose his stylistic features with 'Madang aesthetics' which were based on open space, open acting style, and graphic ensemble. This paper tries to demonstrate how his 'Madang aesthetics' has refined his productions in scenography, acting style, and in more like metaphoric and metonymic symbolic expression of the graphic ensemble. To do this, two productions were explored: eLee Chunpoongjeun and Yulha Ilgyee Manbo. Madangnori was sorely explored by Son Jinchaek with his artistic colleagues Yoon Munshik, Kim Jongyeup, Kim Sungnyu, music designer Park Buhmhoon, and choreographer Guk Sooho. Though it has been ignored for its popular appeal by the doctrinaire theoretical opinions, it started to pull academic attention recently. His theatres are also getting sharp with his 'Madang aesthetics' as well as minimalistic expression in scenography, acting style, and graphic ensemble. Madang Jungshin is the soul and Madang aesthetics is the body in his artistic works. The Madang Jungshin animates the Madang aesthetics, so they become alive in his theatre.

한국 정치극의 전개 양상 - 1920년대부터 80년대까지의 정치극운동을 중심으로 - (The Development Aspects of Korean Political Theatre Movement)

  • This paper investigates the development and aesthetics of Korean political theatre from its quickening period 1920s to democratization era 1990s. Political theatre before 90s developed an antithesis resistant movement toward Korean modern history that had been scattered with suppressing political circumstances such as colonial era and dictatorial government, the movement has powerful activity and social influences. Just like the 20 century political theatre had been quickened under the influence of Marxism at Russia and Germany in 1920s, Korea's political theatre began in socialism theatre movement form around the same time. Proletarian theatre groups had been founded in Japan and Korea, and developed into practical movement with organized connection. However, the political theatre movement in Japanese colonial era was an empty vessel makes great sound but not much accomplishments. Most performance had been canceled or disapproved by suppression or censorship of the Japanese Empire. The political theatre in liberation era was the left drama inherited from Proletarian theatre of the colonial era. Korean Theatre alliance took lead the theatrical world unfold activities based on theatre popularization theory such as 'culture activists' taking a jump up the line and 'independent theatre' peeping into production spot as well as the important event, Independence Movement Day Memorial tournament theatre. Since 1947, US army military government in Korea strongly oppressed the left performances to stop and theatrical movement was ended due to many left theatrical people defection to North Korea. The political theatre in 1960s to 70s the Park regime, developed in dramatically different ways according to orthodox group and group out of power. The political theatre of institutional system handled judgment on sterile people and had indirect political theatre from that took history material and allegory technique because of censorship. In political theatre out of institution, it started outdoor theatre that has modernized traditional performance style and established deep relationship with labor spot and culture movement organizations. Madangguek(Outdoor theatre) is 'Attentive political theatre', satirizing and offending the political and social inconsistencies such as the dictatorial government's oppression and unbalanced distribution, alienation of general people, and foreign powers' pillage sharply as well as laughing at the Establishment with negative characters. The political theatre in 1980s is divided into two categories; political theatre of institutional system and Madangguek. Institutional Political theatre mainly performed in Korea Theatre Festival and the theatre group 'Yeonwoo-Moudae' led political theatre as private theatre company. Madangguek developed into an outdoor theatrical for indoor theatre capturing postcolonial historical view. Yeonwoo-Moudae theatre company produced representative political plays at 80s such as The chronicles of Han's, Birds fly away too, and so on by combining freewheeling play spirit of Madangguek and epic theatre. Political theatre was all the rage since the age of democratization started in 1987 and political materials has been freed from ban. However, political theatre was slowly declined as real socialism was crumbling and postmodernism is becoming the spirit of the times. After 90s, there are no more plays of ideology and propaganda that aim at politicization of theatre. As the age rapidly entered into the age of deideology, political theatre discourse also changed greatly. The concept 'the political' became influential as a new political possibility that stands up to neoliberalism system in the evasion of politics. Rather than reenact political issues, it experiments new political theatre that involves something political by deconstructing and reassigning audience's political sense with provocative forms, staging others and drawing discussion about it.

목이버섯으로부터 추출한 항혈전물질의 제품화와 동물실험을 통한 항혈전활성 검증 (Production of Antithrombotic Material Extracted from Auricularia auricular-judae and the Verification of Its Antithrombotic Activity via Animal Test)

  • 목이버섯으로부터 항혈전물질을 추출하여 제품화하기 위한 대량생산공정을 다음과 같이 확립하였다. 즉, 건조된 목이버섯을 분쇄하여 75% ethanol로 추출하여 원심분리한 침전물에 증류수에 첨가한 후 $76^{\circ}C$에서 2시간 추출한 후 압착여과하여 여과액을 얻는다. 이를 감압농축한 후 여기에 최종 농도가 80%가 되도록 ethanol을 첨가하여 추출한 침전물을 동결건조한다. 제품의 소비지 기호도 조사를 통해 부재료의 종류와 첨가량을 결정하여 배합비를 목이버섯 추출물 90.5%, 고과당 2.0%, $\beta$-cyclodextrin 1.5%, 올리고 당 2.0%, 배 농축액 4.0%로 결정하였다. 제품을 포장하여 25, 37, $45^{\circ}C$에서 저장하였을 경우 저장 8주에서도 수분활성도, 수분함량, pH, 산도에 큰 변화가 없었다. 미생물 생균수는 $25^{\circ}C$$37^{\circ}C$에서 저장하였을 경우 저장기간이 지남에 따라 약간 증가한 반면, $45^{\circ}C$에서 저장하였을 경우에는 저장 2개월 후에는 대부분 사멸하였다. 제품을 쥐에 정맥투여하였을 경우 1,000 mg/kg 투여 시 APTT 활성, TT 활성, PT 활성, FIB 활성이 대조구와 비교하여 유의적으로 높았으며 500 mg/kg을 경구투여하였을 경우 항혈소판활성이 아스피린과 동일한 수준을 나타내었다.

폐스팀을 이용한 가역 고체산화물 연료전지의 기술적 경제적 해석 (Techno-Economic Analysis of Reversible Solid Oxide Fuel Cell System Couple with Waste Steam)

  • Reversible solid oxide fuel cell (ReSOC) system was integrated with waste steam for electrical energy storage in distributed energy storage application. Waste steam was utilized as external heat in SOEC mode for higher hydrogen production efficiency. Three system configurations were analyzed to evaluate techno-economic performance. The first system is a simple configuration to minimize the cost of balance of plant. The second system is the more complicated configuration with heat recovery steam generator (HRSG). The third system is featured with HRSG and fuel recirculation by blower. Lumped models were used for system performance analyses. The ReSOC stack was characterized by applying area specific resistance value at fixed operating pressure and temperature. In economical assessment, the levelized costs of energy storage (LCOS) were calculated for three system configurations based on capital investment. The system lifetime was assumed 20 years with ReSOC stack replaced every 5 years, inflation rate of 2%, and capacity factor of 80%. The results showed that the exergy round-trip efficiency of system 1, 2, 3 were 47.9%, 48.8%, and 52.8% respectively. The high round-trip efficiency of third system compared to others is attributed to the remarkable reduction in steam requirement and hydrogen compression power owning to fuel recirculation. The result from economic calculation showed that the LCOS values of system 1, 2, 3 were 3.46 ¢/kWh, 3.43 ¢/kWh, and 3.14 ¢/kWh, respectively. Even though the systems 2 and 3 have expensive HRSG, they showed higher round-trip efficiencies and significant reduction in boiler and hydrogen compressor cost.

포그 스크린을 이용한 공간증강현실(SAR) 구현 (Implementation of Spatial Augmented Reality Using Fog Screen)

  • 이 연구는 디스플레이 장비와 사용자를 분리하여 현실 공간 전체, 또는 현실 공간 속 허공에 이미지를 디스플레이하는 '공간증강현실(Spatial Augmented Reality)' 구현에 적합한 포그 스크린의 활용 방안에 대하여 전체적으로 조망 하였다. 세 번의 전시와 한 번의 공연을 통하여 통과가 가능한 포그 스크린이 공간증강현실을 구현하는데 있어 적합한 소재라는 것과 포그 스크린을 이용한 공간증강현실 구현을 통하여 무대나 전시에서 홀로그램 연출이 이전보다 쉬워졌다는 것에 대해서도 증명 하였다. 전시와 함께 공연계 종사자들을 상대로 실시한 설문조사를 통하여 포그 스크린을 알고 있는 사람들이 절반에 불과하다는 것과 실제로 포그 스크린을 관람한 사람의 비율이 전체 응답자 중에서 10% 정도로 매우 낮다는 것을 확인 하였다. 또한, 포그 스크린이 주변 공간에 어떠한 영향을 미치는가를 알아보기 위하여 국립아시아문화전당 어린이문화원에서 처음 실시한 시간과 거리 변화에 따른 습도 변화 관찰 실험을 통하여 포그 스크린을 중심으로 반경 5m 이내의 습도가 2~3%($6,400m^3$ 기준) 정도 증가할 수 있다는 것을 확인 하였다. 이를 통하여 전시장에서 물감이나 종이, 나무와 같이 습기에 취약한 소재를 사용하는 전시물과 함께 전시할 경우, 포그 스크린과의 설치 유격에 참고할 수 있는 최소한의 근거를 마련하였다.
