The goal of interaction proteomics that studies the protein-protein interactions of all expressed proteins is to understand biological processes that are strictly regulated by these interactions. The availability of entire genome sequences of many organisms and high-throughput analysis tools has led scientists to study the entire proteome (Pandey and Mann, 2000). There are various high-throughput methods for detecting protein interactions such as yeast two-hybrid approach and mass spectrometry to produce vast amounts of data that can be utilized to decipher protein functions in complicated biological networks. In this review, we discuss recent developments in analytical methods for large-scale protein interactions and the future direction of interaction proteomics.
This tutorial explains the basic principles of mechanistic ligand-receptor interaction model, which is an operational model of agonism. A growing number of agonist drugs, especially immune oncology drugs, is currently being developed. In this tutorial, time-dependent ordinary differential equation for simple $E_{max}$ operational model of agonism was derived step by step. The differential equation could be applied in a pharmacodynamic modeling software, such as NONMEM, for use in non-steady state experiments, in which experimental data are generated while the interaction between ligand and receptor changes over time. Making the most of the non-steady state experimental data would simplify the experimental processes, and furthermore allow us to identify more detailed kinetics of a potential drug. The operational model of agonism could be useful to predict the optimal dose for agonistic drugs from in vitro and in vivo animal pharmacology experiments at the very early phase of drug development.
A multi-dimensioanl model is being increasingly used to predict the thermo-flow field in the gas turbine combustor. This article addresses an integrated survey of modeling of the liquid spray formation and fuel distribution in gas turbine with high-shear nozzle/swirler assembly. The processes of concern include breakup of a liquid jet injected through a hole type orifice into air stream, spray-wall interaction and spray-film interaction, breakup of liquid sheet into ligaments and droplet,5, and secondary droplet breakup. Atomization of liquid through hole nozzle is described using a liquid blobs model and hybrid model of Kelvin-Helmholtz wave and Rayleigh-Taylor wave. The high-speed viscous liquid sheet atomization on the pre-filmer is modeled by a linear stability analysis. Spray-wall interaction model and liquid film model over the wall surface are also considered.
The thermal stability of nitric acid solutions after contact with non-irradiated and irradiated tributyl phosphate (TBP) and its solution in Isopar-M has been studied. It has been established that exothermic processes occur during heating due to the interaction of soluble radiolysis products and the decomposition of the extractant with nitric acid. Such processes can occur at temperatures below 100 ℃, but unlike a thermal explosion that occurs in seconds, they are longer in time and are accompanied by weak heat evolution. Their intensity depends on the composition of the extractant, the concentration of HNO3, and the volume ratio of the organic and aqueous phases. The presence of extractant degradation products in raffinates does not pose a risk of a rapid evolution of gaseous products during evaporation, however, the presence of reducing agents can significantly increase the intensity of the exothermic decomposition of raffinates.
Within a framework of sociocultural approaches to writing process, this study examined six ESL graduate students' writing processes in depth based on individual interviews and their narratives of writing process. The narratives and interviews were analyzed to discover salient aspects of the students' writing processes and to understand the socially situated nature of the writing processes. First, it was observed that these six students displayed multiplicity in terms of their representations of writing process, episodes, textual practices, and concerns. Several factors including the writing task, students' familiarity with genre, literacy skills, attitude toward writing, and involvement in interaction contributed to individualized trajectories of writing process. It was also revealed that writing is unavoidably a socially situated practice. Students were situated in their cultural arenas as well as their disciplinary arenas, and these contexts helped the students serve as active agents producing and sharing knowledge. The confluence of personal, cognitive, and social factors observed in their writing processes suggests that writing process should be understood from multiple perspectives.
Multimodal interfaces are recognition-based technologies that interpret and encode hand gestures, eye-gaze, movement pattern, speech, physical location and other natural human behaviors. Modality is the type of communication channel used for interaction. It also covers the way an idea is expressed or perceived, or the manner in which an action is performed. Multimodal Interfaces are the technologies that constitute multimodal interaction processes which occur consciously or unconsciously while communicating between human and computer. So input/output forms of multimodal interfaces assume different aspects from existing ones. Moreover, different people show different cognitive styles and individual preferences play a role in the selection of one input mode over another. Therefore to develop an effective design of multimodal user interfaces, input/output structure need to be formulated through the research of human cognition. This paper analyzes the characteristics of each human modality and suggests combination types of modalities, dual-coding for formulating multimodal interaction. Then it designs multimodal language and input synchronization method according to the granularity of input synchronization. To effectively guide the development of next-generation multimodal interfaces, substantially cognitive modeling will be needed to understand the temporal and semantic relations between different modalities, their joint functionality, and their overall potential for supporting computation in different forms. This paper is expected that it can show multimodal interface designers how to organize and integrate human input modalities while interacting with multimodal interfaces.
생물 활동이 활발한 지하수-지표수 혼합구간에서 일어나는 생지화학 기작에 대한 관심은 지대하다. 지표수로부터 기인한 오염물질은 지하수-지표수 혼합구간을 통과할 때 이 구간의 특수한 환경 아래에서 생지화학 기작을 통해 오염물질이 제거되거나 자연저감 되기 때문이다. 본 연구의 목적은 혼합구간의 수직교환 흐름 유동률이 생지화학 과정에 미치는 영향의 상관성을 분석하는 것이다. 이를 위해 깊이별로 설치한 소형 관정을 통해 수직 수두구배를 측정하여 혼합구간의 수직 이동수의 방향을 조사하였으며, 연구지 토양시료에서 서식하는 미생물의 확인을 위해 중합효소연쇄반응 및 클로닝 방법이 수행되었다. 편상관 분석을 통해 수직 교환 흐름 유동률, 질산성 질소의 농도 그리고 미생물의 활성이 서로 영향을 주고 있음을 확인하고자 하였다. 그 결과 수직 흐름 교환 유동률이 질산성 질소의 농도 그리고 미생물의 활성 및 생지화학 기작에 영향을 미치는 것으로 조사되었다. 본 연구를 통해 수직 흐름 교환 유동률, 오염물의 농도 그리고 미생물의 활성을 통해 지하수-지표수 혼합구간의 생지화학 기작을 예상할 수 있음을 확인하였다.
Purpose: Level of meta-cognition of students has been regarded as one of the crucial factors on web-based learning. This study aimed to describe interaction type in small group discussion of the nursing graduate students and to investigate learning consequences and interaction types in group discussion on meta-cognition level. Method: Twenty six graduate nursing students attending the class on-line at the K university in Seoul were included in the study. We measured their meta-cognition level and learning attitude. We also scored their individual and group reports as well as analyzed interaction type by reviewing the dialogue of the group discussion. Results: The participants showed low frequency of exploratory interaction and high frequency of integrative interaction in the cognitive interaction category. They showed frequent modification interaction in the meta-cognitive interaction category. Interestingly, the students with lower level of meta-cognition achieved significantly greater scores in the individual assignments. High functioning group consisting of the students with high meta-cognitive level produced greater group report. Conclusion: A new strategy is needed to encourage in-depth interaction in a group discussion of nursing students. Meta-cognitive level of the students should be considered to form a small group for discussion in order to improve group activities.
A time-dependent folding process was used to determine whether or not protein disulfide isomerase (PDI) plays an important role in the maturation of nascent lactoferrin polypeptides. Interaction between lactoferrin and PDI was analyzed according to the co-immunoprecipitation of the two proteins. The results indicate that lactoferrin folding requires a significant interaction with PDI and its binding is relatively brief compared to other nascent polypeptides. The amount of lactoferrin interacting with PDI increases up to half a minute and sharply decreases beyond this time point. During the refolding process that follows reduction by DTT, lactoferrin polypeptides heavily interact with PDI and the interaction period was extended compared to the normal folding process. In terms of the temperature effect on PDI-lactoferrin interaction, PDI binds to lactoferrin polypeptides longer at a lower temperature (here, $25^{\circ}C$) than $37^{\circ}C$. The lactoferrin-PDI interaction was also studied in vitro. According to the in vitro experiment data, PDI was still functional in cell lysates assisting lactoferrin folding into the mature form. PDI interacts with lactoferrin polypeptides for an extended period during the folding in vitro. During the refolding process in vitro, intermolecular aggregates and refolding oligomers matured into a functional form after PDI binds to the lactoferrin. These results suggest that PDI provides a prolonged chaperoning activity in the refolding processes and that there appears to be a greater requirement for PDI chaperone activity in the refolding of lactoferrin polypeptides.
Information systems development entails planning, analysis, design and construction phases. The analysis phase identifying user requirements is the most important of these phases. Since unidentified defects in the early phase causes increased work and costs as development proceeds, the quality of analysis results affects the quality of the resultant system. Major tasks in the analysis phase are data modeling and process modeling. Research on building a knowledge-based system for data modeling have been conducted much, however, not sufficiently for process modeling. As a system environment with high user interaction increases, research on process modeling methods and knowledge- based systems considering such environment are required. In this research, a process modeling framework for information systems with high user interaction is suggested and a knowledge-based system for supporting the suggested framework is implemented. A proposed model consists of the following tasks: event analysis, process analysis, and event/process interaction analysis. Event analysis identifies business events and their responses. Process analysis break down the processes of an enterprise into progressively increasing details. Decomposition begins at the function level and ends when the elementary process level is reached. Event/process interaction analysis verifies the results of process analysis and event analysis. A knowledge-based system for supporting a proposed process modeling framework is implemented in a web-based environment.
