This paper describes a pattern recognition method of Magnoliae flos based on a gas chromatographic/mass spectrometric (GC/MS) analysis of the essential oil components. The botanical drug is mainly comprised of the four magnolia species (M. denudata, M. biondii, M. kobus, and M. liliflora) in Korea, although some other species are also being dealt with the drug. The GC/MS separation of the volatile components, which was extracted by the simultaneous distillation and extraction (SDE), was performed on a carbowax column (supelcowax 10; 30 m{\time}0.25 mm{\time}0.25{\mu}m$) using temperature programming. Variance in the retention times for all peaks of interests was within RSD 2% for repeated analyses (n = 9). Of the 74 essential oil components identified from the magnolia species, approximately 10 major components, which is $\alpha$-pinene, $\beta$-pinene, sabinene, myrcene, d-limonene, eucarlyptol (1,8-cineol), $\gamma$-terpinene, p-cymene, linalool, $\alpha$-terpineol, were commonly present in the four species. For statistical analysis, the original dataset was reduced to the 13 variables by Fisher criterion and factor analysis (FA). The essential oil patterns were processed by means of the multivariate statistical analysis including hierarchical cluster analysis (HCA), principal component analysis (PCA) and discriminant analysis (DA). All samples were divided into four groups with three principal components by PCA and according to the plant origins by HCA. Thirty-three samples (23 training sets and 10 test samples to be assessed) were correctly classified into the four groups predicted by PCA. This method would provide a practical strategy for assessing the authenticity or quality of the well-known herbal drug, Magnoliae flos.
In this study, the volatile compounds in 24 commercial Korean black raspberry wines were isolated by headspace solid-phase microextraction and analyzed by gas chromatography-mass spectrometry (GC-MS). A total of 43 volatile components, including 15 esters, 12 terpenes, 7 alcohols, 4 acids, 3 ketones, and 2 aldehydes, were identified. Ethyl esters and alcohols such as ethyl acetate, ethyl octanoate, isoamyl alcohol, and phenethyl alcohol were the most represented groups among the quantified volatiles. In particular, various terpenes such as DL-limonene, linalool, alpha-terpineol, and myrtenol were identified. The differences in volatile components among the 24 black raspberry wines and possible sample grouping were examined by applying principal component analyses to the GC-MS data sets. The first and second principal components explained 43.9% of the total variation across the samples. No apparent sample groupings were observed according to manufacturing locations. The samples KU, BH, SR, and MO showed higher overall levels in the concentrations of terpenes originating from black raspberry, while other samples such as BB and HB, showed higher in ethyl ester and alcohol contents produced by yeast fermentation, respectively.
We have performed analyses using ancient DNA extracted from 25 excavated human bones, estimating around the 1st century B.C. Ancient human bones were obtained from Nukdo Island, which is located off of the Korean peninsula of East Asia. We made concerted efforts to extract ancient DNA of high quality and to obtain reproducible PCR products, as this was a primary consideration for this extensive kind of undertaking. We performed PCR amplifications for several regions of the mitochondrial DNA, and could determine mitochondrial haplogroups for 21 ancient DNA samples. Genetic information from mitochondrial DNA belonged to super-haplogroup M, haplogroup D or its sub-haplogroups (D4 or D4b), which are distinctively found in East Asians, including Koreans or Japanese. The dendrogram and principal component analysis based on haplogroup frequencies revealed that the Nukdo population was close to those of the East Asians and clearly distinguished from populations shown in the other regions. Considering that Nukdo is geologically isolated in the southern part of the Korean peninsula and is a site of commercial importance with neighboring countries, these results may reflect genetic continuity for the habitation and migration of ethnic groups who had lived in a particular area in the past. Therefore, we suggest that phylogenetic analyses of ancient DNA have significant advantages for clarifying the origins and migrations of ethnic groups, or human races.
Objective: The aim of this study was to create a set of microsatellite markers with high polymorphism for the genetic monitoring and genetic structure analysis of local goose populations. Methods: Novel microsatellite markers were isolated from the genomic DNA of white Roman geese using short tandem repeated probes. The DNA segments, including short tandem repeats, were tested for their variability among four populations of geese from the Changhua Animal Propagation Station (CAPS). The selected microsatellite markers could then be used to monitor genetic variability and study the genetic structures of geese from local geese farms. Results: 14 novel microsatellite loci were isolated. In addition to seven known loci, two multiplex sets were constructed for the detection of genetic variations in geese populations. The average of allele number, the effective number of alleles, the observed heterozygosity, the expected heterozygosity, and the polymorphism information content were 11.09, 5.145, 0.499, 0.745, and 0.705, respectively. The results of analysis of molecular variance and principal component analysis indicated a contracting white Roman cluster and a spreading Chinese cluster. In white Roman populations, the CAPS populations were depleted to roughly two clusters when K was set equal to 6 in the Bayesian cluster analysis. The founders of private farm populations had a similar genetic structure. Among the Chinese geese populations, the CAPS populations and private populations represented different clads of the phylogenetic tree and individuals from the private populations had uneven genetic characteristics according to various analyses. Conclusion: Based on this study's analyses, we suggest that the CAPS should institute a proper breeding strategy for white Roman geese to avoid further clustering. In addition, for preservation and stable quality, the Chinese geese in the CAPS and the aforementioned proper breeding scheme should be introduced to geese breeders.
This study examines the role of mathematics anxiety as a mediator between self efficacy and mathematics skills using a series of regression analyses suggested by Baron RM & Kenny DA(1986). The participants include college students who enrolled in the Food Service Production and Operation course in a department of hotel and restaurant management at a college in the United States. Descriptive analysis, principal component analysis, reliability test, and a series of regression analyses were used for data analysis using SPSS 19.0. In order to collect data for the study, General Self Efficacy Scale(GSES) and Math Anxiety Rating Scale(MARS) were utilized, and they turned out to be reliable(${\alpha}$=.906 and ${\alpha}$=.890, respectively). A significant negative relationship was found between self efficacy and mathematics anxiety. In addition, it was found that self-efficacious students performed better mathematics skills than those who had lower level of self efficacy. However, the relationship was no longer significant when the concept of mathematics anxiety was added, which satisfies the condition of mediation.

Solid phase microextraction and gas chromatography-mass spectrometry (SPME/GC-MS), electronic nose, and electronic tongue were used to characterize the sensory profiles of cider vinegars from Korea (K1-2), China (C1-2), Japan (J1-2), and US (U1-2). SPME-GC/MS detected acetic acid as the common volatile compound in all vinegars, in addition to isovaleric acid, octanoic acid, and phenethyl acetate. Acids and acetic esters were the major components of Korean and US vinegar samples, respectively. Chinese vinegars had high ethyl acetate content, while Japanese samples were characterized by a low content of acetic acid. Principal component analysis (PCA) pattern provided a clear categorical discrimination of Chinese vinegars by E-nose and E-tongue analyses. The instrumental sensory scores and the taste attributes for flavor ($r^2=0.9431$), sourness ($r^2=0.9515$), and sweetness ($r^2=0.8325$) were highly correlated. Therefore, SPME/GC-MS, E-nose, and E-tongue analyses may be useful tools to discriminate the sensory profiles of cider vinegars of different origins.

The Jeju horse is an indigenous Korean horse breed that is currently registered with the Food and Agriculture Organization of the United Nations. However, there is severe lack of genomic studies on Jeju horse. This study was conducted to investigate genetic characteristics of horses including Jeju horse, Thoroughbred and Jeju crossbred (Jeju${\times}$Thoroughbred) populations. We compared the genomes of three horse populations using the Equine SNP70 Beadchip array. Short-range Linkage disequilibrium was the highest in Thoroughbred, whereas $r^2$ values were lowest in Jeju horse. Expected heterozygosity was the highest in Jeju crossbred (0.351), followed by the Thoroughbred (0.337) and Jeju horse (0.311). The level of inbreeding was slightly higher in Thoroughbred (-0.009) than in Jeju crossbred (-0.035) and Jeju horse (-0.038). $F_{ST}$ value was the highest between Jeju horse and Thoroughbred (0.113), whereas Jeju crossbred and Thoroughbred showed the lowest value (0.031). The genetic relationship was further assessed by principal component analysis, suggesting that Jeju crossbred is more genetically similar to Thoroughbred than Jeju horse population. Additionally, we detected potential selection signatures, for example, in loci located on LCORL/NCAPG and PROP1 genes that are known to influence body. Genome-wide analyses of the three horse populations showed that all the breeds had somewhat a low level of inbreeding within each population. In the population structure analysis, we found that Jeju crossbred was genetically closer to Thoroughbred than Jeju horse. Furthermore, we identified several signatures of selection which might be associated with traits of interest. To our current knowledge, this study is the first genomic research, analyzing genetic relationships of Jeju horse, Thoroughbred and Jeju crossbred.
Seed color is controlled by several genes and is a key trait in determining the metabolite content and biological activities of legume genotypes. In this study, 296 adzuki bean accessions, including 159 grey, 99 red, and 38 white adzuki beans, were grown in Korea. Variations in total phenolic content (TPC), total saponin content (TSC), DPPH• scavenging activity, ABTS•+ scavenging activity, and ferric reducing antioxidant power (FRAP) were assessed and were reported to be in the ranges of 1.52-8.24 mg GAE/g, 14.36-114.22 mg DE/g, 0.23-12.84 mg AAE/g, 1.05-17.66 mg TE/g, and 0.59-13.14 mg AAE/g, respectively, with a wide variation across adzuki beans. Except for DPPH• scavenging activity, the average values declined in the order gray > red > white adzuki beans, each demonstrating a significant variation (p < 0.05). White adzuki beans, which showed low metabolite content and antioxidant activity, were clearly separated from the gray and red genotypes using principal component and hierarchical cluster analyses. Moreover, TPC, TSC, and antioxidant activities were strongly correlated, regardless of seed color. Overall, the diversity of the TPC, TSC, and antioxidant activity in a broad population of adzuki bean genotypes was determined. Furthermore, this study found that seed color variation in adzuki beans had a significant effect on the metabolite content and antioxidant activity. Superior accessions with high levels of TPC, TSC, and antioxidant activity were also discovered and could be used for functional plant breeding and human consumption. The findings of this study may be useful for understanding the relationship between seed coat color and metabolite concentration in adzuki beans, paving the way for molecular-level analyses.
This study was carried out to provide proper management plans for small portable water supply system in the Nonsan area through water quality monitoring, hydrogeochemical investigation and multivariate statistical analyses. Nonsan area is a typical rural area heavily depending on small water supply system for portable usage. Geology of the area is composed of granite dominantly along with metasedimentary rocks, gneiss and volcanic rocks. The monitoring results of small portable water supply system showed that 13-21% of groundwaters have exceeded the groundwater standard for drinking water, which is 5 to 8 times higher than the results from the whole country survey (2.5% in average). The major components exceeding the standard limits are nitrate-nitrogen, turbidity, total coliform, bacteria, fluoride and arsenic. High nitrate contamination observed at southern and northern parts of the study area seems to be caused by cultivation practices such as greenhouses. Although Ca and $HCO_3$ are dominant species in groundwater, concentrations of Na, Cl and $NO_3$ have increased at the granitic area indicating anthropogenic contamination. The groundwaters are divided into 2 groups, granite and metasedimentary rock/gneiss areas, with the second principal component presenting anthropogenic pollution by cultivation and residence from the principal components analysis. The discriminant analysis, with an error of 5.56% between initial classification and prediction on geology, can explain more clearly the geochemical characteristics of groundwaters by geology than the principal components analysis. Based on the obtained results, it is considered that the multivariate statistical analysis can be used as an effective method to analyze the integrated hydrogeochemical characteristics and to clearly discriminate variations of the groundwater quality. The research results of small potable water supply system in the study area showed that the groundwater chemistry is determined by the mixed influence of land use, soil properties, and topography which are controlled by geology. To properly control and manage small water supply systems for central and local governments, it is recommended to construct a total database system for groundwater environment including geology, land use, and topography.

To better understand the morphological variation of the Perilla crop and their weedy types in East and Southeast Asia, we studied the morphological variation of 90 accessions by examining 10 morphological characteristics, such as flowering time, seed size, seed hardness, seed color, color of surface leaf, color of reverse side leaf etc. As a result, morphological variation determined that between cultivated var. frutescens and var. crispa, and between cultivated var. frutescens and its weedy type showed significant morphological differences in terms of seed size and seed hardness, whenever cultivated var. crispa and its weedy type could not showed significant differences in most morphological characters. In PCAs (principal component analysis), among 10 morphological characteristics, flower color (QL6), color of surface leaf (QL3), seed size (QN2), seed hardness (QL1), seed color (QL2), stem color (QL7), and color of reverse side leaf (QL4) contributed in negative direction on the first axis, while flowering time (QN1), leaf shape (QL5), and degree of pubescence (QL8) contributed in positive direction on the first axis. Among these morphological characters, particularly flower color (QL6), color of surface leaf (QL3), seed size (QN2), seed hardness (QL1), and degree of pubescence (QL8) were useful characters for discrimination between cultivated var. frutescens and weedy var. crispa, and between cultivated var. frutescens and its weedy type. However, most accession of cultivated and weedy types of var. crispa was not clearly discriminated by PCA analyses. Although the wild ancestral species of var. frutescens and of var. crispa are still unknown in East and Southeast Asia, the weedy types of Perilla crop may be the key taxon for our understanding of the origin of cultivated types of var. frutescens and var. crispa.
