Genetic Polymorphisms and Cancer Susceptibility of Breast Cancer in Korean Women

  • Breast cancer is the most prevalent cancer among women in Western countries, and its prevalence is also increasing in Asia. The major risk factor for breast cancer can be traced to reproductive events that influence the lifetime levels of hormones. However, a large percentage of breast cancer cases cannot, be explained by these risk factors. The identification of susceptibility factors that predispose individuals to breast cancer (for instance, if they are exposed to particular environmental agents) could possibly give further insight into the etiology of this malignancy and provide targets for the future development of therapeutics. The most interesting candidate genes include those that mediate a range of functions. These include carcinogen metabolism, DNA repair, steroid hormone metabolism, signal transduction, and cell cycle control. We conducted a hospital-based case-control study in South Korea to evaluate the potential modifying role of the genetic polymorphisms of selected low penetrance genes that are involved in carcinogen metabolisms (i.e., CYP1A1, CYP2E1, GSTM1/T1/P1, NAT1/2, etc.), estrogen synthesis and metabolism (i.e., CYP19, CYP17, CYP1B1, COMT, ER-$\alpha$, etc.), DNA repair (i.e., XRCC1/3, ERCC2/4, ATM, AGT, etc.), and signal transduction as well as others (i.e., TGF-$\beta$, IGF-1, TNF-$\beta$, IL-1B, IL-1RN, etc.). We also took into account the potential interaction between these and the known risk factors of breast cancer. The results of selected genes will be presented in this mini-review.

  • Since the introduction of pit and fissure sealants in the early 1970's, many studies have been published amply documenting the efficacy of these materials as caries-preventive agents. However, the acceptance of the technique by dentist has been questionable. This survey was investigated the attitudes toward and use of pit and fissure sealants by dentist in Chonbuk area. The results were as follows: 1. Utilization of pit and fissure sealants is very high. Only 15% of dentists never use pit and fissure sealants, whereas 85% generally use pit and fissure sealants. 2. Most dentists reported that their knowledge of pit and fissure sealants was gained through dental schools education. 3. Concern about difficult to sell' to patient, poor retention of pit and fissure sealants and unsubstantiated by research were given as the main reasons why pit and fissure sealants was not used. 4. Patient demands and various educate program were given as important factors that would lead to increase use of pit and fissure sealants.

Inhibition of Tyrosinase and Lipoxygenase Activities by Resveratrol and Its Derivatives from Seeds of Paeonia lactiflora

  • Previously, a methanol extract from seeds of Paeonia lactiflora was shown to have a potent inhibitory activities against tyrosinase and soybean lipoxygenase (SLO). Seven stilbenes, trans-resveratrol-4-Ο-$\beta$-D-glucoside, trans resveratrol, trans-$\varepsilon$-viniferin, cis-$\varepsilon$-viniferin, gnetin H, suffruticosol A and B were isolated from the seeds as active principles for inhibition of the enzymatic activity. Among them, the resveratrol trimer, gnetin H exhibited the most potent inhibitory activities against tyrosinase and SLO, respectively. Additionally, the resveratrol dimers, trans-$\varepsilon$-viniferin and cis-$\varepsilon$-viniferin exhibited significant inhibitory activity against the two oxidative enzymes. Meanwhile, three other stilbene derivatives, such as trans-resveratrol, suffruticosol A and suffruticosol B had also weak inhibition activity. The least inhibitory activity was observed in transresveratrol-4-Ο-$\beta$-D-glucoside. These results suggest that resveratrol dimers and trimer in the seeds of Paeonia lactiflora are potentially useful therapeutic agents against pathological disorders such as hyperpigmentation and inflammation.

Anti-microbial Activity of Saussurea lappa C.B. Clarke Roots

  • We investigated the total phenolic and flavonoid contents and the antimicrobial activity of ethanol extracts obtained from Saussurea lappa C.B. Clarke. The ethanol extracts of S. lappa C.B. Clarke were fractionated with various solvents (n-hexane, chloroform, and n-butanol). The antimicrobial activity of S. lappa C.B. Clarke was examined by disc-diffusion and micro-dilution susceptibility assays with six food-borne pathogens, and compared to that of the synthetic antibiotics. It is found that the S. lappa C.B. Clarke ethanol extract and n-hexane fraction have strong activity against B. cereus and V. parahaemolyticus strains compared to ampicillin. The inhibitory concentration ($IC_{50}$) values of hexane fraction against L. monocytogenes, B. cereus, and B. subtilis were 62.5, 250 and 500 ppm, respectively. Therefore, these data suggest that S. lappa C.B. Clarke may be useful as antimicrobial agents against food-borne pathogens.

Salicornia herbacea Prevents High Fat Diet-Induced Hyperglycemia and Hyperlipidemia in ICR Mice

  • Salicornia herbacea L. (Chenopodiaceae) has been used as a seasoned vegetable by living in coastal areas. S. herbacea (SH) has been demonstrated to stimulate cytokine production, nitric oxide release, and to show anti-oxidative effect. In a series of investigations to develop potential anti-diabetic and/or anti-hyperlipidemic agents from Korean indigenous plants, 50% ethanol extract of Salicornia herbacea was found to prevent the onset of the hyperglycemia and hyperlipidemia induced by high fat diet in ICR mice. At 6 week old, the ICR mice were randomly divided into five groups; two control and three treatment groups. The control mice were to receive either a regular diet (RD) or high-fat diet (HFD), and the treatment groups were fed a high fat diet with either 350 mg/kg, 700 mg/kg of SH (SH350 and SH700) or 250 mg/kg of met-formin (MT250) for a 10-week period. SH not only reduced body weight but also corrected associated hyperglycemia and hyperlipidemia in a dose dependent manner. SH exerted beneficial effects on the plasma glucose and lipid homeostasis possibly ascribed to its specific effects on lipogenesis related genes (SREBP1a, FAS, GAPT), and PEPCK, glucose 6-phosphatase gene expressions in liver. Ethanol extract of S. herbacea has potential as a preventive agent for type 2 diabetes (and possibly hyperlipidemia) and deserves future clinical trial.

Preventive Effects of Dairy Products on Dementia and Cognitive Decline (유제품의 치매 및 인지기능 저하 예방 효과)

  • The prevention of cognitive decline and dementia is an increasingly important global public health priority due to an increase in the percentage of the elderly population. Dementia, a severe cognitive disorder, not only negatively impacts the patients' quality of life but also creates a substantial burden for caregivers. This review introduced recent advances regarding the protective effects of dairy product intake against dementia and cognitive decline. Recent epidemiological studies have suggested that specific components of dairy products including bioactive peptides, colostrinin, proline-rich polypeptides, α-lactalbumin, vitamin B12, calcium, and probiotics might promote healthy brain function during aging. Additionally, oleamide and dehydroergosterol in Camembert cheese have been suggested as agents capable of reducing microglial inflammatory responses and neurotoxicity. The intake of neuroprotective and anti-inflammatory compounds in meals is safe and easy, hence nutritional approaches, including dairy product consumption, serve as a promising intervention for the prevention of neurodegenerative disorders.

Effect of Orostachys japonicus A. Berger on Apoptosis Induction of Human Leukemia HL60 Cells (와송의 HL60백혈병세포의 Apoptosis유도 효과)

  • Methanol extracts of Orostachys japonicus A. Berger (OAB) were found to exhibit apoptosis induction of HL60 human acute promyelocytic leukemia cells. Treatment of OAB exerted strong cytotoxicity against HL60 cells. OAB induced DNA fragmentation of HL60 cells in a dose dependent manner. Nitric oxide production were also increased in OAB-treated RAW264.7 macrophage cell lines. Treatment of OAB increased the expression of p53 and iNOS gene and the expression of p53, $NF-{\kappa}B$ and iNOS protein in cultured HL60 and RAW264.7 cells. These results suggest that OAB are effective on strong anti-cancer properties and can be useful as a chemo-preventive agents.

Measures against Security Safety and Hallucinogenic Drug Crimes (환각성 약물범죄와 경호안전상의 대응방안)

  • Troublesome problems are being caused by hallucinogenic drugs which come not only from illegal narcotics but also from unprohibited general drug-kinds such as cough-remedy agents of textromerupan, namely, Lamina, Rubi-king and the likes. Abuse and misuse of narcotics permeated into and threatens mental health of socially-known peoples, actor, actress, talents and businessmen as well. This study especially dealt with problems related with Mesampetamin, Rurajaepam, Texstrometropan and Karisofrodol which affects mental behaviour, and considered counter-measures through analysis and developed appropriate system of goods circulation for guidance to be complied by the medicine professionalists. The guidance sought here is recommended to the government ministries for further enforcement of crack-down of the drug crimes. Arquesa in method is recommend as a method of identifying behaviors and facial expressions of abusers and misusers of narcotics at the spot in place. This study is directed to curtailing hallucinogenic drugs use for absolute preventive security by detecting out black dealers of narcotics supplying to the abusers and misusers. There is a need of special education program to cut off the flow of narcotics or drugs crimes into official security force as well as private security personnel. This study introduced a priority and emphasis of real augmentation to theory and practice of the modern scientific security which can protect the mental health of the guarded persons as well especially in the light of security safety curriculum at the school.

Field Tests of Furamizole for Preventive Control of Avian Mycoplasmosis (닭의 호흡기성(呼吸器性) Mycoplasma 균병(菌柄)에 대한 Furamizole 의 예방시험(豫防試驗))

  • A number of chemotherapeutic agents, namely antibiotics, sulfonamides and nitrofuran derivatives have been used, as a fred additive, for the purposes of growth improvement of chicken, increase of feed efficiency, decrease of animal mortality, and disease prevention. In these experiments, furamizole which is one of nitrofuran derivatives and feed additive was test, in field, its antibacterial activity against Mycoplasma gallisepticum causing air sac disease, its effect on pullorum disease control, its effect on the lowering the mortality of chicken and finally the increase of feed efficiency. Throughout the studies, furamizole, in concentration of 0.025% in feed fed to baby chicken continuously resulted as following: 1. Tested chicken showed no avian mycoplasma infection compared to 3.7% outbreak in control chicken. 2. Tested chicken showed a low degree of outbreak of pullorum disease. However, its outbreak was much more surpresed compared to that of control chicken. 3. Total mortality rate of 5.5% and 30.8% were obtained in test and control chicken respectively. 4. Feed efficiency were 2.83 and 2.97 in test and control chicken respectively.

Induction of Quinone Reductase and Glutathione S-Transferase in Murine Hepatoma Cells by Flavonoid Glycosides

  • The potential of seven flavonoid glycosides to induce quinone reductase (QR), an anticarcinogenic marker enzyme, in murine hepatoma cells (hepalc1c7) and its mutant cells (BPRc1) was evaluated. Among test compounds, kaempferol-3-O-glucoside, luteolin-6-c-glucoside, and quercetin-3-O-glucoside (Q-3-G) induced QR in hepalc1c7 cells in a dose-dependent manner. However, in BPRc1 cells lacking arylhydrocarbon receptor nuclear translocator (ARNT), only Q-3-G caused a significant induction of quinone reductase at the concentration range of 0.5 to 8 ug/mL, suggesting that it is a monofunctional inducer. Q-3-G induced not only phase 2 enzymes, including QR and glutathione-S-transferase, but also nitroblue tetrazolium reduction activity in HL-60 cells, a biochemical marker for cell differentiation promoting agents. In conclusion, Q-3-G merits further study to evaluate its cancer chemopreventive potential.
