  • The hydrolysis conditions (enzym $e_strate ratio, time and temperature) of the mixture of anchovy sauce residue (ASR) and soy sauce residue (SSR) after fermentation by Flavourzyme 500M $G^{TM}$ were optimized using response surface methodology (RSM) for pretreatment of processing functional enzyme-hydrolyzed sauce. A model equation obtained from RSM was hydrolysis ratio (%) = 28.157+1.929enzym $e_strate ratio+1.818time+2.038temperature-1.093temperatur $e^2$, whose stationary point showed saddle point. From the ridge analysis of the saddle point, the conditions producing the highest hydrolysis ratio was determined as follows: 0.49% enzym $e_strate ratio; 3.55hr hydrolysis time; 62.5$^{\circ}C$ hydrolysis temperature. Adding of SSR to the mixture of water and ASR improved sensory qualities of mixture, so it seemed that SSR has masking effects on off-flavor and taste of ASR.R.of ASR.R.

  • In order to lower a reaction temperature with high conversions for simultaneous catalytic reduction of NO and $N_2O$ over Pd-Rh supported mixed metal oxide honeycomb catalysts, $H_2$ or CO was utilized as a reductant. When using the reductants, the effects of reaction conditions were examined in NO and $N_2O$ conversions, where reaction temperatures, concentrations of the reductants and oxygen and the concentration ratio of $N_2O$ to NO were varied. In using $H_2$ reductant, larger than 50% of NO and $N_2O$ conversions was observed at the temperatures below $200^{\circ}C$ in absence of $O_2$. In using CO reductant, NO and $N_2O$ conversions increased from the temperatures higher than $200^{\circ}C$ and $300^{\circ}C$, respectively. However, in use of both reductants, NO and $N_2O$ conversions decreased with increasing oxygen concentration. As a result, $H_2$ reductant could reduce simultaneously NO and $N_2O$ at relatively lower reaction temperature than CO. Also, NO and $N_2O$ conversions were less influenced by using $H_2$ reductant than CO one. Concentration ratio between NO and $N_2O$ did not affect their conversions regardless the type of reductants. Pretreatment of the catalyst in $H_2$ was more effective in simultaneous reduction of NO and $N_2O$ at low reaction temperature than that in $O_2$.

Cause Analysis in Candle Fire Investigation (양초화재 원인 감정에 관한 연구)

  • Candle fires do not occur frequently, but can easily result in death. In this study, the thermal characteristics of candles and conditions and debris of candle fires were investigated to determine the causes of candle fires. The rates of decrease in weight of 10 candles were measured and found to be between 2.6 g/h and 6.7 g/h. Most candle fires are caused by the ignitiong of combustible materials close to them. The temperature near a candle ranges from about $200^{\circ}C$ to $400^{\circ}C$ at a distance of 1 cm and low ignition temperature materials such as papers can easily catch fire. The melting temperature of candles ranges between $50^{\circ}C$ and $70^{\circ}C$ and their major chemical components are fatty acids and normal hydrocarbons (over C20). Using pretreatment conditions involving the use of activated charcoal strips at $150^{\circ}C$ for 16 hours, the fire debris including candle residues were analyzed using a Gas-chromatograph/Mass-spectrometer (GC/MS).

Effects of Fouling on Permeate Flux during Ultrafiltration of Protein Solutions in a Hollow-Fiber Membrane Reactor (중공사 막반응기에서 단백질용액의 한외여과시 유출속도에 미치는 Fouling의 영향)

  • It is known that a key limiting factor to the use of ultrafiltration membranes is that of membrane fouling, which has been a major cause of permeate flux reduction. In this work, the effects of several factors (operating time, protein concentration, temperature and pH, etc.) influencing permeate flux during ultrafiltration of gelatin, casein and bovine serum albumin using a hollow fiber membrane(M. W. 10,000 cut off) reactor have been examined. The permeate flux of gelatin solution was maintained almost constant during the operation up to 6 hours, but those of casein and albumin solutions were decreased to 50% and 43% of initial value after an operation time of 60min. The permeate flux with increasing concentration and temperature of protein solutions increased, but the permeate flux showed a minimum value near the isoelectric point of proteins. The permeate fluxes of protein solution were enhanced by a temperature increase and pH control. Also, it is proposed that fouling can be decreased by the pretreatment of insoluble proteins with enzymes.

Preparation of Humidity Sensor Using Novel Photocurable Sulfonated Polyimide Polyelectrolyte and their Properties (광가교성 Sulfonated Polyimide 전해질 고분자를 이용한 습도센서의 제조 및 특성 분석)

  • Photocurable sulfonated polyimide (SPI) polyelectrolyte containing chalcone group was prepared and fabricated on an alumina electrode pretreated with chalcone-containing silane-coupling agent. SPI films with bis(tetramethyl)ammonium 2,2'-benzidinedisulfonate ($Me_4N$-BDS)/4,4'-diaminochalcone (DAC)/pyromellitic dianhydride (PA)= 90/10/100 possessed very linear response(Y = -0.04528X+7.69446, $R^2=0.99675$) and showed resistance changing from 4.48 to $2.1k{\Omega}$ between 20 and 95 %RH. The response time for absorption and desorption measurements between 33 and 94 %RH% was about 79 s, which affirmed the high efficiency of crosslinked SPI film for rapid detection of humidity. A negative temperature coefficient showing $-0.49%RH/^{\circ}C$ was found and proper temperature compensation should be considered in future applications. Moreover, pretreatment of the substrates with chalcone-containing silane-coupling agent was performed to improve the water durability and the stability of the humidity sensors at a high humidity and a high temperature and long-term stability for 480 h. The crosslinked SPI films anchored to electrode substrate could be a promising material for the fabrication of efficient humidity sensors with superior characteristics compared to the commercially available sensors.

Effects of Extrusion and Enzyme Treatment on Extraction of β-Glucan from Agaricus blazei Murill (압출성형과 효소처리가 신령버섯 β-Glucan의 추출에 미치는 영향)

  • This study analyzed changes in ${\beta}$-glucan content in Agaricus blazei Murill concentrates according to extrusion and extraction conditions. Screw speed and feed rate were fixed to 250 rpm, and 100 g/min, respectively. Moisture contents (20 and 30%) and barrel temperature (130 and $140^{\circ}C$) were adjusted. ${\beta}$-Glucan content of the extruded sample at a moisture content of 20% and barrel temperature of $130^{\circ}C$ was higher compared to other extrusion conditions. ${\beta}$-Glucan content of the extruded sample at a moisture content of 20% and barrel temperature of $130^{\circ}C$ treated with Rohament CL enzyme was higher compared to Viscozyme L, and Plantase TL enzyme treatments under the same extrusion conditions. In conclusion, extrusion and pretreatment with Rohament CL enzyme enhanced yield of ${\beta}$-glucan extract.

Preparation and Characterization of Pitch based Coke with Anisotropic Microstructure Derived from Pyrolysis Fuel Oil (열분해유 유래 피치로부터 이방성 미세구조 코크스 제조 및 특성 평가)

  • In this study, pitch was synthesized using pyrolysis fuel oil (PFO). Coke with mesophase microstructure was then prepared from the synthesized pitch and its properties were evaluated. Pitch was synthesized by poly-condensation reaction, which is an endothermic reaction at a temperature above 400 ℃ because the PFO was mainly composed of molecules with two to three aromatic rings. The Coke reactor was composed of the pretreatment reactor, preheater for applying heat energy, and coke drum for inducing microstructure of coke. Coke was prepared from synthesized pitch by controlling the temperature of the preheater to 400~490 ℃, and properties were evaluated by polarization microscope, XRD and Raman spectroscopy. The coke prepared at a preheater temperature of 460 ℃ identified flow anisotropic microstructure, and the electrical conductivity was 72.0 S/cm due to high crystallinity. And the flow anisotropic coke showed approximately 2.2 times higher electrical conductivity than that of Super-P, a conductive carbon material.

Effect of Temperature Conditions and Chemical Treatments on Seed Germination of Pseudolysimachion kiusianum var. diamantiacum (Nakai) T.Yamaz. (봉래꼬리풀의 종자 발아에 미치는 온도 조건과 화학적 처리의 영향)

  • The germination characteristics of Pseudolysimachion kiusianum var. diamantiacum (Nakai) T.Yamaz., a rare and endemic plant designated by the Korea Forest Service, were investigated according to light conditions, temperature, and pretreatment. As a result of the germination experiment according to light conditions, it was determined that P. kiusianum is a photoblastic seed that does not germinate at all in dark conditions. The optimum germination temperature of the seeds was found to be 20-25℃, considering the final germination rate and germination time. Three growth regulators (IAA, GA3, kinetin) and two inorganic salts (KNO3, KCl) were pretreated to improve the germination rate of P. kiusianum seeds. The growth regulators IAA and kinetin had no significant effect on improving the germination rate of P. kiusianum seeds. On the other hand, GA3 significantly increased the final germination percentage and germination rate regardless of the concentration, especially the treatment of more than 500 mg·L-1 at 20℃ was more than 4 times more effective than the untreated. The inorganic salts KNO3 and KCl had no significant effect on the seeds of P. kiusianum at low concentrations, but at higher concentrations (40 mM and 300 mM, respectively), they improved the germination rate and germination age by 2 times compared to the untreated. The results of this study will be useful for the mass propagation of P. kiusianum, which has the potential to be utilized as a native plant for restoration.

Seed Germination, Seedling Grwoth and Optimal Seedling Density of Phellodendron amurense Rupr. in Nursery (황벽나무의 종자발아(種子發芽)와 묘목생장(苗木生長) 및 적정생육밀도(適正生育密度)에 관(關)한 연구(硏究))

  • We examined the effect of pretreatments on seed germination, seedling growth, optimal seedling density of Phellodendron amurense. The seeds were carefully purified after collection in late Oct. from natural population at Mt. Chiri. For the evaluation of pretreatment effects, seeds were sowed in an experimental nursery after being stored in open ground($2{\sim}8^{\circ}C$) with soaking in 20% $H_2O_2$, 50-fold pon-pon solution, cold moist stratification in refrigerator($2{\sim}4^{\circ}C$) and dry storage in roam temperature($4{\sim}18^{\circ}C$), respectively, For the effect of growth density of seedlings, seedlings germinated in nursery were precisely controlled with 53, 104, 220, 380 seedlings per $m^2$. Among pretreatments for the promotion of germination rate, pon-pon treatment(68.8%) was highest, the others were higher than that in dry storage(2.3%), and these seed pretreatment methods were significantly different at 1% level. The height growth of seedlings showed the most vigor from Jun.12 to Jul. 8, and from Jul.20 to Aug. 2, and it reached to 72.8% of height growth. In case of 1-0 seedling, although seedling density per $m^2$ was increased, seedling growth was decreased. The optimal seedling density was 104 seedlings per $m^2$. The standard seedlings for plantation were above height 55cm, root collar diameter 8.1mm, and root length 21cm. These results demonstrated that seed of P. amurense requires stratification for the germination promotion.

Effects of Hydrothermal Pretreatment on the Nutritional Values and In Vitro Fermentation Characteristics of Tall Fescue (Festuca arundinacea) and Corn Silage (열수 전처리에 따른 톨페스큐와 옥수수 사일리지의 영양적 가치와 in vitro 발효특성에 미치는 영향)

  • This study examined the effects of a hydrothermal pretreatment (HP) on the nutritional values and in vitro fermentation characteristics of tall fescue and corn silage. This study was conducted through a factorial design of 2 (control or HP) × 2 (hay; tall fescue or silage; corn). For the HP, forage was placed into a glass bottle with 20% w/v of water, and the glass bottle was sealed and heated to reach a temperature of 121℃ (0.12 MPa). The solid residue and liquid were collected and oven-dried at 65℃ for three days. The dried materials were tested for in vitro fermentation at 39℃ for 24 and 48 h. The content of ADF increased significantly regardless of the forage type. After in vitro incubation for 24 h, the total VFA content was significantly lower after HP, regardless of the forage type (p ≤ 0.05), and the propionate concentration was increased in corn silage with HP (p ≤ 0.05). After 48 hours of in vitro incubation, the propionate content increased significantly (p ≤ 0.03) in corn silage with HP (p ≤ 0.05), but the butyrate content decreased significantly (p ≤ 0.05). There was no change in the in vitro dry matter and neutral detergent fiber digestibility by HP regardless of the forage type. Therefore, the use of hydrothermally pretreated corn silage could be advantageous for the supply of energy for ruminants.
