Thimerosal, a widely used preservative, has been well known to induce intracellular calcium mobilization in various cell types. However, the mechanism of its calcium mobilization is not clearly understood yet. For studying the mechanism of thimerosal-mediated calcium release, we have used HL60 cells in calcium-free Lockes solution that has no extracellular calcium. Thimerosal significantly reduced the lag period of initial calcium release whereas it enhanced the rate and magnitude of the calcium release in a dose-dependent manner. At the same time, we found that thimerosal generated superoxide anion by activating NADPH oxidase in dose- and time-dependent manner. Interestingly, the kinetics and the dosedependency of superoxide anion generation were very similar to those of intracellular calcium mobilization. In inhibitors study, the thimerosal-induced superoxide anion generation was significantly suppressed by DMSO as well as superoxide dismutase but not by genistein or EGTA. Surprisingly, the pretreatment with N-Acetyl-$_{L}$-Cysteine blocked almost completely the thimerosal-induced calcium increase, indicating that ROS playa key role in the calcium mobilization. The present results suggest that thimerosal-induced calcium mobilization is possibly mediated by the activation of NADPH oxidase and subsequent ROS generation.n.
Most bacteria form biofilm as self-defence system, making efficient food sanitization, preservation, and instrument washing more difficult. Biofilm formation of Salmonella, E. coli, B. cereus, and S. aureus was observed during 24 hr food preservations by performing microtiter plate and glass wool assays. Most cells formed biofilm and attached onto glass wool. When biofilm formation and injury were analyzed on the microtiter plate, 10 and 20% acid-injured E. coli and S. aureus, respectively, 30-50% cold temperature $(4^{\circ}C)-injured$ B. cereus and E. coli, and 30-55% 6% sodium chloride solution-injured Salmonella showed significant biofilm formation. Results indicate biofilm formation level differed within species depending on type of stress.
This study was conducted to examine the effects of wet storage solution, transport period and temperature on vase life and quality of cut flowers in standard chrysanthemum 'Jinba'. Immediately after transport, the fresh weight and flower diameter of cut flowers did not show a difference according to wet storage solutions regardless of the transport period, but as the transport period increased, the fresh weight and flower diameter increased. The flower bud stage at harvest was maintained due to the small changes in flower diameter, and the freshness of leaves was better when transported at 5℃ than at 25℃. When transported at 25℃, the longer the transport period, the lower the quality of cut flowers as some petals opened up and showed early flowering after transport. In preservative solutions, quality of cut flowers transported at 25℃ was lower than that at 5℃ due to fresh weight and diameter according to the longer transport period. The vase life of cut flowers was 1.0 day, 0.8 day, and 7.3 days longer when transported for 3, 5, and 7 days respectively at 5℃ than at 25℃. The quality of cut flowers was better due to increase in fresh weight and flower diameter, as well as vase life in wet storage solutions of ClO2 and Chrysal OVB than in tap water, regardless of transport period and temperature. There was no difference in fresh weight and vase life between ClO2 and Chrysal OVB, but flower diameter was greater in ClO2 than in Chrysal OVB. Therefore, for long-term transport of cut standard chrysanthemum 'Jinba', wet storage transport in ClO2 at 5℃ was found effective in maintaining the quality and vase life of cut flowers.
This study was conducted to clarify the effects of chemicals and physical treatment on the split of flower stalk and the vase life of Lycoris squamigera and Lycoris radiata. There was no split of flower stalk for the Lycoris squamigera and the Lycoris radiata after the harvest of flowers and the immersion in 4 mM STA solution for 30 minutes. It was effective for the vase life of Lycoris squamigera that cut flower was pre-treated in 4 mM STS solution for 30 minutes and immersed in $5\~10\%$ sucrose + 150 ppm 8-HQS +4 ppm Rox preservative solution. The optimum periods of hot water treatments for the prevention of flower stalk split and the elongation of vase life for the cut flowers were 15 to 25 seconds for the Lycoris squamigera and 5 to 10 seconds for Lycoris radiata. Burning the cut parts of flower stalk for 10 to 30 seconds was effective for the prevention of flower stalk split and the elongation of vase life for the Lycoris squamigera, and 10 to 15 seconds for the Lycoris radiata. The vase lifes of Lycoris squamigera and Lycoris radiata were elongated when flower stalk was cut by an incline of 45 degrees compared with the horizontal cut. And Banding the flower stalks of cut flowers was effective for the prevention of flower stalk split and the elongation of vase life for the Lycoris squamigera and the Lycoris radiata.

This experiment was conducted to improve postharvest quality and extended vase life of Korean native Aster koraiensis Nakai for use as cut flowers. Aster koraiensis Nakai cut flowers were treated with various pretreatment and holding solution. Postharvest pretreatment with 3% sucrose + $200mg{\cdot}L^{-1}$ HQC + $50mg{\cdot}L^{-1}\;AgNO_3$ + $50mg{\cdot}L^{-1}$ BA and 3% sucrose + $200mg{\cdot}L^{-1}$ HQC + $50mg{\cdot}L^{-1}\;AgNO_3$ + $150mg{\cdot}L^{-1}$ citric acid for 16 hours extended vase life of cut Aster koraiensis Nakai flowers by 1.4 times as compared with the control (distilled water). Holding solution of 3% sucrose + $200mg{\cdot}L^{-1}$ HQC + $50mg{\cdot}L^{-1}\;AgNO_3$ + $50mg{\cdot}L^{-1}$ BA and 3% sucrose + $200mg{\cdot}L^{-1}$ HQC + $50mg{\cdot}L^{-1}\;AgNO_3$ + $150mg{\cdot}L^{-1}\;GA_3$ extended vase life of cut Aster koraiensis Nakai flowers by1.6 and 1.7 times as compared with control (distilled water). Aster koraiensis Nakai.flowers held in this preservative solution increased fresh weight and were maintained positive water balance for a long vase period. It was suggested that the vase life of cut Aster koraiensis Nakai flowers was closely related to fresh weight and water balance of the cut flower.
This study was conducted to investigate the antimicrobial effect of chlorine dioxide ($ClO_2$) on the vase life of cut rose 'Beast' (Rosa hybrida L.). Postharvest treatments to extend the vase life of cut roses were divided into two: holding solution treatment and pulsing solution treatment. In holding solution treatment, the cut roses were treated with preservative solutions containing tap water (TW, control), distilled water (DW), $ClO_2$ 2, 4, 6, and $8{\mu}L{\cdot}L^{-1}$, and compared with a commercialized antimicrobial compound of 8-HQS $200{\mu}L{\cdot}L^{-1}$. In pulsing solution treatment, cut roses were dipped into the $ClO_2$ solutions of 50, 100, 150, 200, and $250{\mu}L{\cdot}L^{-1}$ for 60 seconds and were placed in DW. The air temperature was $18.4^{\circ}C$, RH 51.5%, and light (photosynthetically active radiation, PAR) $3.6{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ with 12 hour day length. The longest vase life of cut roses was observed in the holding solution with $ClO_2$$4{\mu}L{\cdot}L^{-1}$ as 13.8 days and pulsing with $200-250{\mu}L{\cdot}L^{-1}$ as 13.5-13.7 days, where vase life were extended four days longer than TW. Whereas, the inclusion of 8-HQS $200{\mu}L{\cdot}L^{-1}$ in vase solution resulted in phytotoxicity. The relative fresh weight and water uptake have similar tendencies. Bacteria inhibition by $ClO_2$ and 8-HQS were very effective. But bacteria at TW and DW treatments on cut flower with stem were detected in $3.7{\times}10^5CFU{\cdot}L^{-1}$ and $6.3{\times}10^5CFU{\cdot}L^{-1}$, respectively (without stem in DW $1.4{\times}10^4CFU{\cdot}L^{-1}$). The $ClO_2$ contents in holding solution of all treatments were scavenged in two-four days after treatment. This study indicated that $ClO_2$$4{\mu}L{\cdot}L^{-1}$ holding solution treatment and $200-250{\mu}L{\cdot}L^{-1}$ pulsing solution treatment can be applied to extend the postharvest life of cut roses.

An annual plant, Eclipta prostrata (Linn) is a member of the Asteraceae plant family and inhabited in tropical or subtropical regions of the world. Through many previous researches, E. prostrata has been extensively studied for its hepatoprotective effect, antivenom potential against viper venom, antioxidant, hair-growth, wound-healing efficacy and so on. In this study, for better understanding of the potential of E. prostrata as skin protectant, we conducted the experiments evaluating the antioxidant and antiaging efficacy. To this end, 50% ethanolic extract of E. prostrata and its ethyl acetate fraction were prepared and investigated. For the evaluation of antioxidant capacity of the samples, $FSC_{50}$ and $OSC_{50}$ were estimated. As a result, $OSC_{50}$ of ethyl acetate fraction was 2.7 times superior to $OSC_{50}$ of L-ascorbic acid, a well known antioxidant agent. Futhermore E. prostrata showed notable reactive oxygen species (ROS) scavenging effect and protective effect against $H_2O_2$ in the celluar level as well. Especially, in the $^1O_2$ induced hemolysis test, $64{\mu}g/mL$ of ethyl acetate fraction showed greater than 6 times increased retardation effect compare to control which means E. prostrata has remarkable antioxidant capacity. To validate the antiaging effect of the samples, we conducted elastase inhibition assay using elastase solution extracted from human skin fibroblasts, Hs68. As a result, $16{\mu}g/mL$ of each sample showed 6.8% and 14.0% of elastase inhibition respectively. Finally, antimicrobial activity of E. prostrata was assessed to validate the possibility as alternative preservative. From the result, ethyl acetate fraction showed oustanding antimicrobial activity as of methyl paraben, a well known chemical preservative. In conclusion, these results suggest that E. prostrata can be used as natural skin protectant or preservative as natural ingredient in food or cosmetics industry.
This study was conducted to evaluate the pressure treatment characteristics of Japanese red pine and Japanese larch skin timber with ACQ-2, CUAZ-2 and CuHDO-1. The effect of moisture content (MC) on preservative treatability was investigated for Japanese red pine sapwood and Japanese larch heartwood, and fixation characteristics of CCA alternatives was also evaluated. Japanese red pine sapwood, which was dried below 30 percent MC, was fully penetrated with preservatives, and minimum requirement of preservative retention for the hazard class H3 was achieved. Through measuring preservative retention gradient in Japanese red pine sapwood, it was confirmed that the retention gradient of CuHDO-1 was steeper than that of both ACQ-2 and CUAZ-2. In particular, it was intensified at a higher MCs of wood samples (25∼30%). Japanese larch heartwood did not meet the minimum requirement of penetration and retention for the hazard class H3 over the range of pretreatment MCs tested. With presteaming under $121^{\circ}C$ for 12 hours, the treatability of Japanese larch heartwood was enhanced to meet the minimum requirement for the hazard class H3. The fixation rate of copper was much more faster under drying condition compared with nondrying condition; more than 95% of copper were fixed in 3~6 days and 1 day under drying conditions in Japanese red pine sapwood and Japanese larch heartwood, respectively. After 3-week fixation period at ambient temperature, the amount of mobile copper in treated wood sample that remains available for leaching from treated wood was the highest in the wood samples treated with ACQ-2, followed by CuHDO-1 and CUAZ-2. It was proportional to the amount of copper in treating solution.
Background: Graft vessel preservation solution in coronary artery bypass surgery is used to maintain the graft conduit in optimal condition during the perioperative period. Nafamostat mesilate (NM) has anticoagulation and anti-inflammatory properties. Therefore, we investigated NM as a conduit preservative agent and compared it to papaverine. Methods: Sprague-Dawley (SD) rat thoracic aortas were examined for their contraction-relaxation ability using phenylephrine (PE) and acetylcholine (ACh) following preincubation with papaverine and NM in standard classical organ baths. Human umbilical vein endothelial cells (HUVECs) were cultured to check for the endothelial cell viability. Histopathological examination and terminal deoxynucleotidyl transferase dUTP nick end labeling assay were performed on the thoracic aortas of SD rats. Results: The anti-contraction effects of papaverine were superior to those of NM at PE (p<0.05). The relaxation effect of NM on ACh-induced vasodilatation was not statistically different from that of papaverine. Viability assays using HUVECs showed endothelial cell survival rates of >90% in various concentrations of both NM and papaverine. A histopathological study showed a protective effect against necrosis and apoptosis (p<0.05) in the NM group. Conclusion: NM exhibited good vascular relaxation and a reasonable anti-vasocontraction effect with a better cell protecting effect than papaverine; therefore, we concluded that NM is a good potential conduit preserving agent.

This study was conducted to develop an analytical technique for determination of chlorite and chlorate concentrations in fresh-cut food and dried fish products by an ion chromatography/conductivity detection method using a hydroxide mobile phase. Deionized water was added to homogenized samples, which were then extracted by ultrasound extraction and centrifuged at high speed (8,500 rpm). Subsequently, a Sep-Pak tC18 cartridge was used to purify the supernatant. Chlorite and chlorate ions were separated using 20 mM KOH solution as the mobile phase and Dionex IonPac AS27 column as the stationary phase. Ethylenediamine was used as sample preservative and dibromoacetate was added to adjust for the disparity in extraction efficiencies between the food samples. The method detection limit) for chlorite and chlorate were estimated to be 0.2 mg/kg and 0.1 mg/kg, respectively, and the coefficient of determination ($r^2$) that denotes the linearity of their calibration curves were correspondingly measured to be 0.9973 and 0.9987. The recovery rate for each ion was 92.1 % and 96.3 %, with relative standard deviations of 7.47 % and 6.18 %, respectively. Although neither chlorite nor chlorate was detected in the food samples, the analytical technique developed in this study may potentially be used in the analysis of disinfected food products.
