비장적출이 임신 Rat의 Progesterone농도에 미치는 영향 (Effect of Splenectomy on the Progesterone Concentration according to Gestation Periods in Rats)

  • The effect of splenectomy on the concentration of progesterone in pregnant wistar rats showing 4-day regular estrous cycles more than two cycles were investigated. The day after mating with the same male animal was designated day 0 of pregnancy. Splenectomy was conducted on day 0 (early-S), 6 (middle-S) and 13 (late-S) of pregnant rat, respectively. Blood sample were collected at day 1, 7, 14 and 21 of pregnancy. Concentraton of serum progesterone was determined by emzyme-linked immunosorbent assay (ELISA) using specific antibodies and emzyme conjugate(progesterone peroxidase labeled). In normal pregnant rats, serum progesterone concentrations were increased significantly (P<0.05) on day 7 and 14 of pregnancy compared with on day 1 of pregnancy and then dramatically decreased on day 21. However in early and middle splenectomized rats, progesterone concentrations were significantly (P<0.05) decreased on day 7 and 14 of pregnancy compared with same day of normal rats. These observations indicate that luteotropic action and steroidogenesis by corpus luteum of ovary may be regulated by splenocytes in rat.

Effects of IFN-γ on IL-18 Expression in Pregnant Rats and Pregnancy Outcomes

  • The present study focused on establishing the effects of interferon-gamma (IFN-${\gamma}$) on interleukin-18 (IL-18) expression patterns and pregnancy outcomes in pregnant rats. Pregnant rats at the post-implantation stage were randomized into control, low IFN-${\gamma}$ (L-IFN-${\gamma}$) and high IFN-${\gamma}$ groups (H-IFN-${\gamma}$) that received normal saline, 100 IU/g of IFN-${\gamma}$ and 500 IU/g of IFN-${\gamma}$ vaginal muscular injection, respectively. The effects of IFN-${\gamma}$ on IL-18 expression and pregnancy outcomes were assessed systematically using several methods, including immunohistochemistry streptavidin-perosidase (SP), image pattern analysis, enzyme-linked immune-sorbent assay (ELISA), whole blood count (WBC) count, microscopy and visual observation. IL-18 was detected in the uteri of all pregnant rats, and mainly distributed in the endometrium, decidual cells, vascular endothelium and myometrium. Immunohistochemistry and image pattern analyses revealed significantly lower IL-18 expression in the H-IFN-${\gamma}$ group compared to the L-IFN-${\gamma}$ and control groups (p<0.01), indicating that high doses of IFN-${\gamma}$ induce downregulation of IL-18 in the uterus of pregnant rats. ELISA results disclosed that IL-18 expression in peripheral blood of the H-IFN-${\gamma}$ group was lower than that of the L-IFN-${\gamma}$ group (p<0.05), and significantly reduced compared to the control group (p<0.01). Moreover, the number of peripheral leukocytes in the H-IFN-${\gamma}$ group was significantly higher than those in the control and L-IFN-${\gamma}$ groups (p<0.01). Morphology analysis showed no evident differences between the L-IFN-${\gamma}$ and control groups. However, for the H-IFN-${\gamma}$ group, uterine mucosa bleeding, necrosis and excoriation were observed using microscopy. Visual observation revealed marroon, swelling, crassitude and no embryo in the uterus, which are obvious indicators of abortion. These results indicate that IFN-${\gamma}$ plays a regulatory role in IL-18 expression in the uterus and peripheral blood of pregnant rats at the post-implantation stage. Moreover, high levels (500 IU/g) of IFN-${\gamma}$ influence normal pregnancy at the early stages in rats by downregulating IL-18 expression in the uterus and peripheral blood and increasing the number of peripheral leukocytes, consequently triggering termination of pregnancy.

운동이 F344계 임신쥐에서 골격근의 VAMP-2 및 GLUT-4 단백질 발현과 혈중 인슐린, 렙틴 농도에 미치는 영향 (Effects of maternal exercise on expression of GLUT-4, VAMP-2 in skeletal muscle and plasma insulin and leptin levels in pregnant rats)

  • 본 연구는 임신횐쥐에서 운동이 혈중 인슐린과 렙틴 농도, 그리고 골격근의 GLUT-4와 VAMP-2단백질 발현에 변화를 조사하기 위하여 수행되었다. 본 연구에서 비임신 횐쥐에 비해 임신 횐쥐에서 혈중 인슐린 농도가 유의하게 증가되었고, 렙틴 농도는 유의하게 감소하는 것으로 나타났다. 임신횐쥐에게 달리기 운동을 적용한 결과 임신으로 증가된 인슐린 농도를 유의하게 감소시키면서 렙틴 농도의 감소를 억제시키는 것으로 나타났다. 골격근에서 GLUT-4와 VAMP-2 단백질 발현량을 조사한 결과 비임신 횐쥐에 비해 임신 횐쥐에서 이러한 단백질 발현이 유의하게 감소되었지만, 임신 중 달리기운동을 경험한 횐쥐에서 GLUT-4와 VAMP-2 발현 모두 유의하게 증가되는 것으로 나타났다. 이와 잘이 임신으로 인한 혈중 인슐린 및 렙틴의 변화가 골격근에서 당을 근조직으로 흡수하는 신호 전달 경로와 수송체의 발현 손상과 연관되어있음을 보여주는 것이다. 하지만 임신 횐쥐에서 달리기 운동은 인슐린과 렙틴 농도의 변화를 완화시킴으로써 골격근의GLUT-4와 VAMP-2발현을 유의하게 증가시키는 것으로 나타났다.

임신 후반기 흰쥐의 인슐린 저항성과 그 기전 (Insulin Resistance in Late Pregnant Rats)

  • 임신시 발생하는 인슐린 저항성과 인슐린 분비 증가의 기전을 규명하기 위하여 Sprague-Dawley 종 암컷 흰쥐를 이용하여 정맥당부하 검사와 호르몬 및 지방 대사물질과 조직에서의 인슐린 수용체 결합, 당원질 합성효소를 분석한 결과를 요약하면 다음과 같다. 정맥당부하검사에서 임신군에서 전체적인 곡선이 대조군에 비하여 아래에 위치하였다. 그러나 당부하시 인슐린 분비는 현저히 증가하였으며 혈당에 대한 비(${\mu}U/mg$)로 비교하여 임신군에서 $56.9{\pm}8.9$였고 대조군에서 $23.6{\pm}2.8$로서 인슐린 저항성을 확인할 수 있었다. 인슐린 분비반응의 증가는 태반 호르몬인 progesterone의 증가와 강한 상관관계를 나타내었다. 당부하검사후 당원질 (mg/100mg tissue)은 골격근(soleus)에서는 임신군과 대조군간에 유의한 차이가 없었으나 간조직에서는 임신군에서 $4.7{\pm}0.9$으로 대조군의 $9.9{\pm}1.3$에 비하여 통계적으로 유의하게 감소하였다. 당원질로 합성된 $^{14}C$-glucose의 활동도도 마찬가지로 골격근에서는 임신군과 대조군간에 유의한 차이가 없었으나 간조직에서는 현저한 감소를 보였다. 당원질 합성 효소(glycogen synthase)는 골격근에서는 대조군이 높았고 간장조직에서는 임신군이 높았으나 유의한 차이는 없었다. 당부하검사후 간장조직의 crude membrane에서의 인슐린-인슐린 수용체 결합반응에서는 두 군 사이에 유의한 차이가 없었다. 이상의 결과로 보아 정상임신흰쥐에서 인슐린 저항성이 발생하였으나 인슐린 분비의 현저한 증가로 내당능의 감소는 나타나지 않았으며 인슐린의 분비증가는 progesterone의 증가와 상관관계가 있었다. 인슐린 저항성은 간에서 가장 현저하게 나타났으며 그 원인은 인슐린 수용체의 결합과정보다는 수용체 전 과정이거나 수용체 후 과정일 것으로 추정된다.

Sprague-Dawley 랫드에서 2-Bromopropane의 배자치사 및 최기형성 효과 (Embryo lethality and teratogenicity of 2-Bromopropane in the Sprague-Dawley rat)

  • The present study was undertaken to evaluate the potential adverse effects of 2-BP on pregnant dams and embryo-fetal development after maternal exposure during the gestational days (GD) 6 through 19 in Sprague-Dawley rats. The test chemical was administered subcutaneously to pregnant rats at dose levels of 0, 375, 750 and 1250 mg/kg/day. During the test period, clinical signs, mortality, body weights and food consumption were examined. All dams were subjected to caesarean section on GD 20 and their fetuses were examined for external, visceral and skeletal abnormalities. At above 750 mg/kg, toxic effects including signs of toxicity, suppressed body weight, decreased gravid uterine weight and reduced food intake were observed in pregnant dams. An increase in the fetal deaths, a decrease in the litter size, a reduction in the fetal body weight and an increase in the incidence of fetal morphological alterations were also found. There were no adverse effects on either pregnant dams or embryo-fetal development at a dose level of 375 mg/kg. These results suggest that a 14-day subcutaneous dose of 2-BP is embryolethal and teratogenic at above 750 mg/kg/day in pregnant rats. In the present experimental condition, the no-observed-adverse-effect level of 2-BP is considered to be 375 mg/kg/day for dams and embryo-fetuses, respectively.

임신 랫드에 투여한 Human Menopausal Gonadotropin이 임신에 미치는 영향 (Effects of the Administration of Human Menopausal Gonadotropin on the Pregnancy in Rats)

  • The effect of Human Menopausal Gonadotropin(hMG) on the implantation, pregnancy, and the concentration of plasma progesterone were studied in pregnant rats. HMG 75 or 150 IU were administered once on day 1, 2, 3, 4, 9, 12 or 14 of gestation, respectively. Rats were autopsied on day 7 or 18. A single dose of hMG prevented implantation and terminated pregnancy in all of the rats by injecting on either day 1 or day 2 and this abortifacient action was effective 82-98% of pregnant rats on day 3 or 4 and 14-20% on day 9 or 12. Administration of hMG had no effect on termination of pregnancy on day 14. Plasma progesterone concentration by injecting hMG on day 1, 2, 3 or 4 were very decreased.

십전대보탕과 육미지황탕이 실험동물의 모체에 미치는 생식독성학적 연구 (Sibjeondaebotang and Yugmijihwangtang's Toxicological Effects on Pregnant Rats)

  • Introduction : The experiments was undertaken to evaluate the effects of herbal medicine, Sibjeondaebotang, Yugmijihwangtang, in pregnant rats and fetuses. Female Sprague-Dawley rats were orally administered with the Sibjeondaebotang, Yugmijihwangtang at dose of 5mg/kg/day for 20 days. Pregnant rats were sacrificed at 20th day of gestation, and observed internal and reproductive organs. Approximately live fetuses in the 20th day of gestation were randomly selected and fixed in 95% ethanol. To observe skeletal malformations, fetuses were stained with alcian blue and alizarin red S. Effect on pregnant rats : Maternal body weight of Sibjeondaebotang, Yugmijihwangtang treated group has a tendency to increase compared to that of control group. There were no significant difference in internal and reproductive organs. There were no significant changes between two groups in blood chemistry and hematological values. There were no significant changes in number of corpus luteum, implantation, live fetuses and implantation rate, delivery rate, and sex ratio. But Yugmijihwangtang administered group showed higher early resorption rate than the control group(p<0.05). Also Sibjeondaebotang administered group showed higher late resorption rate than the control group(p<0.05). From the sex ratio, number of females, bigger than number of males in the control group, and more males than females in Yugmijihwangtang administered group(p<0.05).

임신 랫드에 투여한 GnRH와 HCG가 태아에 미치는 영향 (Effects of the Administration of GnRH and HCG on the Fetus in Pregnant Rats)

  • The effect of GnRH and/or hCG on the implantation, pregnancy, and the concentration of plasma estradiol and progesterone were studied in pregnant rats. GnRH 50, or 100ug and/or hCG 50 or 100 IU were administered once on day 2 or 9 of gestation, respectively. Rats were autopsied on days 8 or 16. Administration of GnRH on day 2 did not induce the prevention of implantation and termination of pregnancy but was able to induce termination of pregnancy administering on day 9. Administration of hCG induced delayed implantation on day 2 and termination of pregnancy on day 2 and 9. Administration of GnRH concomitant with hCG had no effect on prevention of implantation on day 2 but induced termination of pregnancy with a very increased fetal resorption on day 2 and with a moderate increased fetal resorption on day 9. Administration of GnRH concomitant with hCG on day 2 induced more increased termination of pregnancy compared to injection of GnRH or HCG and opposite result was observed on day 9. Plasma estradiol and progesterone concentrations by administering GnRH and/or hCG had no effect on the termination of pregnancy the pregnant rats.

Effects of Exposure Period on the Developmental Toxicity of 2-Bromopropane in Sprague-Dawley Rats

  • Recently we reported that 2-bromopropane (2-BP) has maternal toxicity, embryotoxicity, and teratogenicity in Sprague-Dawley rats. The aims of this study are to examine the potential effects of 2-BP administration on pregnant dams and embryo-fetal development, and to investigate the effects of metabolic activation induced by phenobarbital (PB) on developmental toxicities of 2-BP. Pregnant rats received 1000 mg/kg/day subcutaneous 2-BP injections on gestational days (GD) 6 through 10 (Group II and Group IIII) or 11 through 15 (Group IV). Pregnant rats in Group III received an intraperitoneal PB injection once daily at 80 mg/kg/day on GD 3 through 5 for induction of the liver metabolic enzyme system. Control rats received vehicle injections only on GD 6 through 15. All dams underwent caesarean sections on GD 20 and their fetuses were examined for external, visceral, and skeletal abnormalities. Significant adverse effects on pregnant dams and embryo-fetal development were observed in all the treatment groups, and the maternal and embryo-fetal effects of 2-BP observed in Group II were higher than those seen in Group IV. Conversely, maternal and embryo-fetal developmental toxicities observed in Group III were comparable to those seen in Group II. These results suggest that the potential effects of 2-BP on pregnant dams and embryo-fetal development are more likely in the first half of organogenesis (days $6{\sim}10$ of pregnancy) than in the second half and that the metabolic activation induced by PB pre-treatment did not modify the developmental toxic effects of 2-BP in rats.
